The interaction between electrons and positive ion beams and its application in accelerator physics are investigated. A space charge lens named Gabor lens was developed which confines electrons in a static column by external fields. The confined electrons are used for focusing and may support space charge compensation. In this structure the relative velocity between the ions and the electrons is maximal and corresponds to the beam velocity. An electron lens as at the Tevatron* is operated with a lower relative velocity in order to compensate the beam, to clean the beam abort gap or to excite the beam for beam dynamics measurements. In comparison electron cooling needs the same velocity of the ion and the electron beam. The following study contains the superposition of electric and magnetic self-fields and their impact on the density distribution of the ion beam and of the electron beam. Recombinations and ionisations are neglected. This is the beginning of an interface between these topics to find differences and similarities of the interaction between ions and electrons with different relative velocities. The 'Facility for Anti-Proton and Ion Research' (FAIR) presently under construction will deliver intense ion beams to its experimental users. The requested intensities require filling the existing synchrotron SIS18, which serves as injector to FAIR, up to the space charge (SC) limit. Operation under these conditions is challenging due to the large tune footprint of the beam, demanding delicate control of adverse effects caused by machine imperfections to avoid emittance growth and beam loss. To facilitate the high intensity operation, the installation of an electron lens for SC compensation into SIS18 is foreseen. This requires an electron beam of a current of several amperes with longitudinal and transverse distributions matched to those of the ion beam during the cycle. The electron beam needs to be RF modulated at a bandwidth of a few MHz with time varying amplitude ranging from DC to fully modulated, while the transverse size needs to be continuously adapted to the adiabatically shrinking ion beam. Space charge lenses use a confined electron cloud for the focusing of ion beams. The electron density gives the focusing strength whereas the density distribution influences the mapping quality of the space charge lens and is related to the confinement. The major role of the electron density with respect to the focusing quality has been pointed out many times in the past *,**. With an automated measurement system the radial light density profile, plasma stability and mean value of the electron density have been measured in respect to the confining fields and the pressure. The results are summarized in 3D-maps. The theoretical model approximations for space charge lenses predicts high electron densities then measured. With the automated system the realistic 3D-maps can be considered instead of an approximation of a theoretical density including knowledge of the most stable electron cloud achievable within the parameter range of the lens. The experimental results of the automated measurement system will be presented here and a concept of a control system for this type of space charge lenses will be explained., Proceedings of the 8th Int. Space-charge compensation provided by the accumulation of particles of opposing charge in the beam potential is an important effect occuring in magnetostatic low energy beam transport sections of high-intensity accelerators. An improved understanding of its effects might provide valuable input for the design of these beam lines. One approach to model the compensation process are Particle-in-Cell (PIC) simulations including residual gas ionisation. In simulations of a drifting proton beam, using the PIC code bender [1], some features of thermal equilibrium for the compensation electrons were found. This makes it possible to predict their spatial distribution using the Poisson-Boltzmann equation and thus the influence on beam transport. In my PhD thesis, I addressed two different topics. The first part dealt with the structural and biochemical characterization on the aryl hydrocarbon receptor (AhR). This protein belongs to the bHLH-PAS family of transcription factors that sense environmental signals. As all members of this family, AhR comprise a bHLH domain, necessary for DNA binding, a PAS A domain, necessary for dimerization, a PAS B domain, which in case of AhR is able to bind ligands and an unstructured TAD domain, involved in transactivation. Upon ligand binding, AhR heterodimerizes with the aryl hydrocarbon receptor nuclear translocator (ARNT), which also belongs to the bHLH-PAS family of transcription factors. Through this dimerization, the fully active heterodimeric transcription factor is formed. Depending on the ligand, transcriptional activation orchestrates the expression of genes required for detoxification, pro-tumor and anti-tumor pathway or immune response. In this study, AhR was functionally and structurally characterized to better understand the process of heterodimerization and DNA binding on a molecular level. This study showed for the first time successful co-expression and co- purification of the AhR/ARNT heterodimeric transcription factor complex comprising the bHLH and PAS A domains. The AhR/ARNT complex appeared properly folded and heterodimerized in a 1:1 stoichiometry. The affinity of the AhR/ARNT complex towards dsDNA containing the DRE was in the lower nano-molar range. This interaction was formed by one molecule AhR/ARNT complex and one molecule double stranded (ds)DNA. The AhR/ARNT complex bound to 12mer dsDNA was crystallized and crystals were extensively optimized. The optimized crystals diffracted anisotropically up to 3.5 Å and various phasing attempts were carried out in order to solve the structure of the AhR/ARNT complex bound to dsDNA. Although structure determination was not achieved within this study, the success of high yield purification of AhR/ARNT complex and biochemical characterization in vitro is a mile stone for further structural investigations. The second part of this study dealt with the structural characterization of the immunity-related GTPase Irga6. Immunity-related GTPases (IRGs) constitute a powerful cell-autonomous resistance systems against several intracellular pathogens in mice. In mice, 23 paralogs of IRG proteins are identified and Irga6 is one of the best studied members of the family. Irga6 is composed of a G domain flanked by a C-terminal and an N-terminal helical domain. Biochemical studies analyzed an interface, which spans over the nucleotide binding pocket of the G domains and is crucial for nucleotide-dependent oligomerization and cooperative GTP-hydrolysis. However, the already known crystal structures of Irga6 show a dimerization mode via the backside dimer but do not reveal this G domain interface. In this part of the study, the structure of a non-oligomerizing Irga6 mutant bound to GMPPNP was solved. This new structure reveals the G domain interface and thus a new mode of G domain dimerization for the IRG family. Beside the nucleotide, two additional interfaces participate in G domain dimerization. A hydrogen bonding network between the switch II and the trans stabilizing loop as well as hydrophobic interactions between the switch I loop and the trans activation loop and the G4 loop establish the G domain dimer interface. These findings are in excellent agreement with the previously published biochemical data. Based on the solved G dimer structure in combination with the published biochemical data and the structural information available for BDLP I proposed an oligomerization model of Irga6 with the G dimer as a nucleotide-dependent interaction interface., Innerhalb dieser Arbeit wurden zwei Themen behandelt. Zunächst befasst sich die Arbeit mit der strukturellen und biochemischen Charakterisierung des Aryl- hydrocarbon-rezeptors (AhR). Dieser gehört zu der bHLH-PAS-Domänen Familie der Transkriptionsfaktoren und reagiert auf unterschiedlichste Umwelteinflüsse. AhR besteht, wie alle Mitglieder dieser Proteinfamilie, aus einer bHLH Domäne, welche die DNA Bindung ermöglicht, aus einer PAS A Domäne, notwendig für die Heterodimerisierung, einer PAS B Domäne, welche im Falle von AhR Liganden binden kann, sowie einer unstrukturierten TAD Domäne. Durch Ligandenbindung wird AhR aktiviert und heterodimerisiert mit dem Aryl-hydrocarbon-rezeptor- nuclear-translocator (ARNT), ebenso Mitglied der oben genannten Proteinfamilie, zu einem aktiven, heterodimeren Transkriptionsfaktor. Dieser Komplex kann bestimmte DNA Motive binden und dadurch die Transkription aktivieren. Abhängig von der Art des Liganden wird die Transkription von unterschiedlichen Genen aktiviert, welche wiederum die Detoxification, das Tumorwachstum oder aber die Immunantwort beeinflussen können. Um die Heterodimerisierung und DNA-Bindung auf molekularer Ebene zu verstehen, wurde in dieser Studie AhR funktionell und strukturell charakterisiert. Erstmalig konnte hier die Coaufreinigung und Coexpression der bHLH und PAS A Domänen des AhR/ARNT Komplexes gezeigt werden. Die weitere Charakterisierung ergab, dass der AhR/ARNT Komplex korrekt gefaltet ist und aus jeweils einem Molekül AhR und ARNT geformt wird. Die Affinität des AhR/ARNT Komplexes zu doppelsträngiger DNA, welche ein DRE Motiv beinhaltet, liegt im niedrigen nanomolaren Bereich. Jeweils ein Molekül AhR/ARNT Komplex und ein Molekül doppelsträngige DNA sind an der Interaktion beteiligt. Der AhR/ARNT Komplex, gebunden an ein Molekül doppelsträngiger DNA, wurde kristallisiert und die Kristalle wurden umfangreich optimiert. Durch die Optimierung konnte eine Diffraktion der Kristalle bis zu einer Auflösung von 3.5 Å erreicht werden. Zahlreiche Phasierungsmethoden wurden angewendet, um die Kristallstruktur des AhR/ARNT Komplexes, gebunden an doppelsträngige DNA, zu lösen. Obwohl die Strukturbestimmung in dieser Studie nicht gelang, ist der Erfolg der Aufreinigung von AhR im Komplex mit seinem Interaktionspartner ARNT und dessen biochemischen Charakterisierung in vitro als hervorragende Basis für weiterführende strukturelle Studien zu sehen. Der zweite Teil dieser Arbeit behandelt die strukturelle Charakterisierung der immunity-related GTPase (IRG) Irga6. Diese stellen in Mäusen ein wirkungsvolles, zellautonomes Resistenzsystem gegen diverse interzelluläre Pathogene dar. In Mäusen sind insgesamt 23 Paraloge der IRG Proteine identifiziert, wobei Irga6 das wohl am besten studierte Mitglied der Proteinfamilie ist. Irga6 besteht aus einer N- und C-terminalen, helikalen Domäne sowie aus einer G Domäne, welche Nukleotid- abhängige Oligomerisation und kooperative GTP Hydrolyse bewirkt. Biochemische Studien zeigten, dass die für Oligomerisation und kooperative GTP Hydrolyse nötige Interaktionsfläche den Bereich der Nukleotidbindetasche der G Domäne von Irga6 mit einschließt. Die bisher bekannten Kristallstrukturen von Irga6 konnten jedoch diese G Domänen Dimerisierung nicht darstellen. In diesem Teil der Studie wurde eine oligomerisations-defizite Irga6 Mutante, gebunden an GMPPNP, strukturell charakterisiert, um die G Domänen Interaktion zu verstehen und die Oligomerisation von Irga6 zu erklären. In der Tat konnte die Kristallstruktur der Irga6 Mutante, gebunden an GMPPNP, gelöst und ein neuer Mechanismus der G Domänen Dimerisierung beschrieben werden, welcher das Nukleotid involviert. Neben dem Nukleotid konnten zwei weitere Interaktionselemente identifiziert werden, die die G Domänen Dimerisierung stabilisieren. Ein Wasserstoffbrückennetzwerk zwischen der switch-II-Schleife und der trans-Stabilisierungs-Schleife sowie hydrophobe Wechselwirkungen zwischen der Switch-I-Schleife und der G4-Schleife bzw. der trans- Stabilisierungs-Schleife tragen hier zur Stabilisierung bei. Insgesamt bestätigt die hier präsentierte Kristallstruktur die bekannten biochemischen Daten. Basierend auf der hier ermittelten Kristallstruktur von Irga6, den strukturellen Informationen zu BDLP und den bekannten biochemischen Daten schlage ich ein Oligomerisationsmodell für Irga6 vor, welches die neu entdeckte nukleotidabhängige G Domänen Dimerisierung zur Grundlage hat.

Die Interaktionen dieser relaxierenden und kontrahierenden Signalwege sollten zum einen mit Hilfe NO-GC1 KO-Mäusen, in denen das Enzym NO-sensitive Guanylyl-Cyclase genetisch ausgeschaltet ist, untersucht werden und zum anderen in WT und KO-Mäusen, die über einen Zeitraum von zwei Wochen mit Ang II oder Noradrenalin behandelt wurden, untersucht werden. Eine Reduktion der cGMP-Spiegel innerhalb der glatten Muskulatur führt zu einer Verschiebung des Gefäßtonus und stellt somit eine direkte Interaktion dar. Des Weiteren führt die Reduktion der cGMP-Spiegel zu einer Veränderung der autonomen Kreislaufregulation und stellt eine indirekte Interaktion dar, die über verschiedene Systeme stattfindet.

The concept of the Gabor lens goes back to an idea by Dennis Gabor, who proposed a magnetron-type trap as an effective diverging lens for electron beams (collecting lens for positive ion beams). Electrons confined inside the lens volume by orthogonal magnetic and electric fields, create an electric space charge field that causes a radial symmetric focusing force on an ion beam passing through the lens volume. Since the beginning of the 1990s, a new design of this lens type as well as numerical models to describe the confined plasma cloud have been developed at the Institute for Applied Physics (IAP, Johann Wolfgang Goethe-University Frankfurt). Thanks to an improved understanding of the plasma confinement as a function of the external fields, two lenses have successfully been tested for low beam currents and remain in operation. In the scope of this work, the performance of a prototype Gabor lens for the transport of intense, i.e. space charge dominated ion beams, was investigated at the High Current Test Injector (HOSTI) of GSI Helmholtzzentrum für Schwerionenforschung GmbH for the first time. To ensure an optimal focusing performance of the Gabor lens a homogeneous and stable electron confinement is required. Therefore, new non-interceptive diagnostic methods were developed to investigate the parameters and state of the confined nonneutral plasma column as a function of the external fields. An essential part of the studies was the time-resolved diagnostic of an occurring plasma instability and the determination of the electron temperature via optical spectroscopy. The latter necessitated the detailed investigation of atomic excitation as well as the measurement of optical-emission cross sections. A comparison of the results from both experiments i.e. the beam transport measurements at GSI and the diagnostic experiments performed at IAP concerning the plasma state, gave first indications of possible interaction processes between the nonneutral plasma and the ion beam. Das Konzept der Gabor-Linse wurde 1947 erstmals von Dennis Gabor vorgeschlagen, der in seiner Veröffentlichung „A Space-Charge Lens for the Focusing of Ion Beams“ die starke Fokussierwirkung einer eingeschlossenen Elektronenwolke auf Ionenstrahlen beschrieb. Die Gabor-Linse dient dabei als Falle für Elektronen, die durch eine spezielle Konfiguration aus elektrischen und magnetischen Feldern eingeschlossen werden. Sind die Elektronen homogen im Linsenvolumen verteilt, wird ein passierender Ionenstrahl durch das von ihnen erzeugte, lineare, elektrische Feld fokussiert. Am Institut für Angewandte Physik (IAP, Johann Wolfgang Goethe-Universität Frankfurt) wurde das Design der Gabor-Linse seit den 1990er Jahren stetig weiterentwickelt und darüber hinaus numerische Modelle zur Beschreibung des nichtneutralen Plasmas erarbeitet. Durch das bessere Verständnis des Plasmaeinschlusses und des Systemzustands in Abhängigkeit von den externen Parametern wurden zwei solcher Linsen auch sehr erfolgreich als Niederenergietransportsektion, allerdings bei vergleichsweise geringen Strahlströmen, getestet und befinden sich dort auch weiterhin im Einsatz. Im Rahmen der vorliegenden Arbeit wurde die Anwendung einer Gabor-Linse erstmals im Bereich raumladungsdominierter Ionenstrahlen am Hochstromtestinjektor (HOSTI) der GSI Helmholtzzentrum für Schwerionenforschung GmbH, im Zusammenhang mit dem geplanten FAIR Projekt, untersucht. Ein homogener und zeitlich wie auch räumlich stabiler Plasmaeinschluss ist von großer Bedeutung, um eine gute Abbildungsqualität der Gabor-Linse zu gewährleisten. Daher wurden Diagnosemethoden entwickelt, um die Parameter der eingeschlossenen Plasmawolke zu bestimmen und die Abhängigkeit ihres Zustands von den externen Parametern zu untersuchen. Ein wesentlicher Teil dieser Untersuchungen bestand in der zeitaufgelösten Diagnose von Plasmainstabilitäten und der Bestimmung der Elektronentemperatur durch optische Diagnose. Letztere erforderte eine dedizierte Untersuchung der atomaren Anregungsprozesse sowie die Vermessung optischer Wirkungsquerschnitte. Ein Vergleich der Ergebnisse beider Experimente, d.h. der Strahltransportmessungen bei GSI und der durchgeführten Diagnoseexperimente am IAP, in Bezug auf den Plasmazustand, ergab erste Hinweise auf die möglichen Wechselwirkungsprozesse zwischen nichtneutralem Plasma und Ionenstrahl.

A prototype Gabor lens has successfully been tested at the GSI High Current Test Injector (HOSTI)*. The experiments comprised the investigation of an emittance dominated and a space-charge dominated beam transport. In particular, the high-current measurements represent a necessary step towards evaluating the focusing performance of the lens and to gain experience in a real accelerator environment. Besides the evaluation of the technical feasibility, the behavior of the electron cloud was characterized by the parameter analysis of the confined non-neutral plasma during beam transport measurements as well as subsequently performed diagnostic experiments. This contribution will present experimental results as well as numerical studies on an improved Gabor lens design for the possible application at the GSI High Current Injector (HSI) in the context of an upgrade program for FAIR**., Proceedings of the 5th Int. Space charge lenses using a stable electron cloud for focusing low energy heavy ion beams are an alternative concept to conventional ion optics. Due to external fields electrons are confined inside the lens’ volume. In case of a homogeneously distributed electron cloud the linear electric space charge field enables beam focusing free of aberration. Since the mapping quality of the lens is related to the confinement, non-destructive diagnostics has been developed to determine the plasma parameters and to characterize the collective behavior of the confined nonneutral plasma. Moreover, a scaled up space charge lens was constructed for a detailed investigation of the nonneutral plasma properties as well as beam interactions with a stable confined electron cloud. Experimental results will be presented in comparison with numerical simulations.

Gabor lenses are plasma traps that can be used for focusing an ion beam linearly without aberrations* by the electric field of a confined electron cloud. They combine strong electrostatic focusing with the possibility of space charge compensation and provide an attractive alternative to conventional ion beam optics in a LEBT section. The focusing performance strongly depends on the density and distribution of the enclosed electron plasma*. As the Gabor lens is usually operated close to the ion source, residual gas ionization is supposed to be the central electron generation mechanism. An electron source is introduced in order to investigate the possibility of boosting the electron density in plasma traps using the example of a Gabor lens. This way, a Gabor lens could be operated under XUHV conditions, where residual gas ionization is suppressed. The particle in cell code bender** was used to simulate the injection into the confining fields of the space charge lens in different geometrical configurations and a prototype experiment was constructed consisting of a Gabor lens and an electron source system. The design of many Low-Energy Beam Transport sections relies on the presence of space-charge compensation by particles of opposing charge. To improve understanding of the processes involved in the built-up and steady-state, simulations using the Particle-in-Cell code bender were made. We will present the influence of various system parameters on the results. Furthermore, the electron velocity distribution was found to be approximately thermal. The spatial distribution can then be found by solving the Poisson-Boltzmann equation. Such a model for the electron distribution was implemented in a 2D PIC code and applied to typical beam transport situations. Background The immunity-related GTPases (IRGs) constitute a powerful cell-autonomous resistance system against several intracellular pathogens. Irga6 is a dynamin-like protein that oligomerizes at the parasitophorous vacuolar membrane (PVM) of Toxoplasma gondii leading to its vesiculation. Based on a previous biochemical analysis, it has been proposed that the GTPase domains of Irga6 dimerize in an antiparallel fashion during oligomerization. Results We determined the crystal structure of an oligomerization-impaired Irga6 mutant bound to a non-hydrolyzable GTP analog. Contrary to the previous model, the structure shows that the GTPase domains dimerize in a parallel fashion. The nucleotides in the center of the interface participate in dimerization by forming symmetric contacts with each other and with the switch I region of the opposing Irga6 molecule. The latter contact appears to activate GTP hydrolysis by stabilizing the position of the catalytic glutamate 106 in switch I close to the active site. Further dimerization contacts involve switch II, the G4 helix and the trans stabilizing loop. Conclusions The Irga6 structure features a parallel GTPase domain dimer, which appears to be a unifying feature of all dynamin and septin superfamily members. This study contributes important insights into the assembly and catalytic mechanisms of IRG proteins as prerequisite to understand their anti-microbial action.
