A plethora of in vitro bioassays is developed in the context of chemical risk assessment and clinical diagnostics to test effects on different biological processes. Such assays can also be implemented in effect-based monitoring (EBM) of (drinking) water quality in parallel to chemical analyses. EBM can provide insight in the risks for environmental and human health associated with exposure to (unknown) complex low-level mixtures of micropollutants, which fits in the risk-based approach that was recently introduced in the European Drinking Water Directive. Some challenges remain, in particular related to selection and interpretation of bioassays. For water quality assessment, carcinogenesis, adverse effects on reproduction and development, effects on xenobiotic metabolism, modulation of hormone systems, DNA reactivity and adaptive stress responses are considered as the most relevant toxicological endpoints. An evaluation procedure of the applicability and performance of in vitro bioassays for water quality monitoring based on existing information has been developed, which can be expanded with guidelines for experimental evaluations. Besides, a methodology for the interpretation of in vitro monitoring data is required, since the sensitivity of specific in vitro bioassays in combination with sample concentration may lead to responses of chemicals (far) below exposure concentrations that are relevant for human health effects. Different approaches are proposed to derive effect-based trigger values (EBTs), including EBTs based on 1) relative ecotoxicity potency, 2) health-based threshold values for chronic exposure in humans and kinetics of reference chemicals, and 3) read-across from (drinking) water guideline values. EBTs need to be chosen carefully in order to be sufficiently but not over-conservative to indicate potential health effects. Consensus on the crucial steps in the selection and interpretation of in vitro bioassay data will facilitate implementation, as well as legal embedding in the context of water quality monitoring of such assays in EBM strategies. Watermonsters van innamepunten (zg. bronnen) én drinkwater zijn onderzocht met een combinatie van bioassays en chemische analyses. Met name in de bronnen lieten de chemische analyses een breed spectrum organische microverontreinigingen zien en de bioassays een spectrum aan biologische activiteit.Figuur 13 uit het rapport toontverschillen tussen de geïntegreerde respons van de toegepaste CALUX bioassays (weergegeven door gesommeerde bioassay punten) en de gesommeerde analytische concentraties. Deze verschillen worden verklaard door het feit dat de zoetstoffen, complexvormers en metformine -die in relatief grote hoeveelheden worden aangetoond met de chemische analyses- geen biologische activiteit vertonen en dus geen responsgeven in de bioassays. Hieruit blijkteentoegevoegde waarde van bioassays: chemische stoffen die biologisch niet relevant zijn, worden ook niet opgepikt.

SOLUTIONS is a European Union Seventh Framework Programme Project (EU-FP7) that aims to deliver a solution-oriented conceptual framework for the evidence-based development of environmental policies with regard to water quality and its protection against contamination. This will integrate innovative chemical and effect-based monitoring tools with a full set of exposure, effect and risk assessment models and strategies to assess abatement options. Uniquely, SOLUTIONS takes advantage of (i) expertise of leading European scientists of major FP6/FP7 projects on chemicals in the water cycle, (ii) access to the infrastructure necessary to investigate the large basins of the Danube and Rhine as well as relevant Mediterranean basins as case studies, and (iii) innovative approaches for stakeholder dialogue and support. In particular, the EU Water Framework Directive (WFD) Common Implementation Strategy (CIS) working groups, International River Commissions, and water works associations will be directly supported with consistent guidance for the early detection, identification, prioritization, and abatement options for chemicals in the water cycle. A set of predictive models and tools will support stakeholders’ management decisions by benefiting from the wealth of data generated from monitoring and chemical registration. SOLUTIONS will provide a specific emphasis on concepts and tools for the impact and risk assessment of complex mixtures of emerging pollutants, their metabolites and transformation products. Analytical and effect-based screening tools will be applied together with ecological assessment tools for the identification of toxicants and their impacts. Beyond state-of-the-art monitoring and management, tools will be elaborated allowing risk identification for aquatic ecosystems and human health. The SOLUTIONS approach will provide transparent and evidence-based suggestions of River Basin Specific Pollutants for the case study basins and support future review of priority pollutants under the WFD as well as potential abatement options., JRC.H.1-Water Resources
