Abstract

BackgroundTsetse flies (Glossina sp.) are the vectors of human and animal trypanosomiasis throughout sub-Saharan Africa. Tsetse flies are distinguished from other Diptera by unique adaptations, including lactation and the birthing of live young (obligate viviparity), a vertebrate blood-specific diet by both sexes, and obligate bacterial symbiosis. This work describes the comparative analysis of six Glossina genomes representing three sub-genera: Morsitans (G. morsitans morsitans, G. pallidipes, G. austeni), Palpalis (G. palpalis, G. fuscipes), and Fusca (G. brevipalpis) which represent different habitats, host preferences, and vectorial capacity.ResultsGenomic analyses validate established evolutionary relationships and sub-genera. Syntenic analysis of Glossina relative to Drosophila melanogaster shows reduced structural conservation across the sex-linked X chromosome. Sex-linked scaffolds show increased rates of female-specific gene expression and lower evolutionary rates relative to autosome associated genes. Tsetse-specific genes are enriched in protease, odorant-binding, and helicase activities. Lactation-associated genes are conserved across all Glossina species while male seminal proteins are rapidly evolving. Olfactory and gustatory genes are reduced across the genus relative to other insects. Vision-associated Rhodopsin genes show conservation of motion detection/tracking functions and variance in the Rhodopsin detecting colors in the blue wavelength ranges.ConclusionsExpanded genomic discoveries reveal the genetics underlying Glossina biology and provide a rich body of knowledge for basic science and disease control. They also provide insight into the evolutionary biology underlying novel adaptations and are relevant to applied aspects of vector control such as trap design and discovery of novel pest and disease control strategies.

Abstract

The present study examined the antimicrobial effects of betel leaf (Piper betle) extracts against Vibrio spp. along with the isolation of the most prevalent bioactive components using a bioassay-guided purification. Following a preliminary screening, the ethanolic extract proved the most effective and performed to silica gel chromatography, followed by reversed phase HPLC to isolate the active components. By means of subsequent UHPLC-orbitrap-ion trap mass spectrometry, two compounds were identified as (E)-3-phenylprop-2-enoic acid (cinnamic acid) and [4-(1-hydroxyprop-2-enyl)phenyl] acetate (1'−hydroxychavicol acetate). The compounds showed anti-microbial activity against Vibrio species including V. parahaemolyticus, V. harveyi, V. alginolyticus, V. vulnificus and V. anguillarum. Their antibacterial action was quantified by a microdilution assay showing that cinnamic acid displays more potent activity (IC50, 21–121 µg/mL) compared to 1'-hydroxychavicol acetate (IC50, 24–365 µg/mL). 24. Compound isolation through bioassay-guided fractionation of Tectona grandisleaf extract against Vibriopathogens in shrimp

Abstract

Tectona grandisLinn, commonly known as teak, is traditionally used to treat a range of diseases, including the common cold, headaches, bronchitis, scabies, diabetes, inflammation, and others. The present study was conducted with the purpose of isolating and identifying the active compounds in T. grandisleaf against a panel of Vibriospp., which may induce vibriosis in shrimp, using bioassay-guided purification. The antimicrobial activity was assessed using the microdilution method, followed by the brine shrimp lethality assay to determine toxicity. Following an initial screening with a number of different solvents, it was established that the acetone extract was the most effective. The acetone extract was then exposed to silica gel chromatography followed by reversed-phase HPLC and further UHPLC-orbitrap-ion trap mass spectrometry to identify the active compounds. Three compounds called 1-hydroxy-2,6,8-trimethoxy-9,10-anthraquinone, deoxyanserinone B, and khatmiamycin were identified with substantial anti-microbial action against V. parahaemolyticus, V. alginolyticus, V. harveyi, V. anguillarum, and V. vulnificus. The IC50values of the three compounds viz. 1-hydroxy-2,6,8-trimethoxy-9,10-anthraquinone, deoxyanserinone B, and khatmiamycin varied between 2 and 28, 7 and 38, and 7 and 56 μg/mL, respectively, which are as good as the standard antibiotics such as amoxicillin and others. The in vivo toxicity test revealed that the compounds were non-toxic to shrimp. The results of the study suggest that T. grandisleaf can be used as a source of bioactive compounds to treat Vibriospecies in shrimp farming.
