Technical development during the last two decades has brought new potential and new applications for ­mobile measurements. In this paper, we present six case studies where mobile measurement devices were used to acquire data for meteorological and climatological research. Three case studies deal with ground-based mobile measurements – on buses for urban climate measurements and on a vessel on a lake – and three with airborne platforms – on a cable car and on an unmanned aerial vehicle for vertical soundings and on a tethered balloon sonde for cloud physics. For each study, we describe the measurement set-up and address the potential and drawbacks of these applications. At the end, we discuss general aspects related to mobile observations especially concerning the time and space dimension of measurements.

In urban areas situated downstream of rural areas, cold air drainage flow contributes to reduced nighttime temperatures and reduced pollutant concentrations. In this study, the impact of historical, present and possible future land-use changes upon evening cooling by suburban cold air drainage flow is analysed by using the numerical cold air drainage model KLAM_21 of the German Weather Service (DWD). In order to do this, land-use patterns of the year 1810 and 1910 are reconstructed. Furthermore, potential land-use scenarios are developed, considering both a scenario with beneficial cold air drainage characteristics and a second adverse scenario. Past, present and future land-use data are used as model input. To validate model results in the present land-use situation, empirical data from field measurements are used to compare modelled and effective cold air drainage flow characteristics. Model data for 14 reference areas – including suburban and inner city sites – are compared with regard to the cooling rate 3 h after sunset. The results show distinctive effects of accumulated long-term urban land-use changes on the suburban cold air drainage flow. In the city centre, for example, the modelled mean cooling rate decreases from 3.6 K in 1810 to 2.8 K in 1910 and to 2.4 K in 2010. In contrast to this, appropriate measures to stop or even reverse the trend of decreasing cooling rates are shown. Intensified urban cooling by nocturnal cold air drainage can contribute to mitigating probable future global warming effects.

Understanding the role of structure and social aspects regarding heat stress of people in urban areas requires an interdisciplinary scientific approach that connects methods from both natural sciences and social sciences. In this study, we combine three approaches to provide an interdisciplinary analysis of the structure and social components of heat stress in the city of Aachen, Germany. First, we assess the overall spatial structure of the urban heat island using spatially distributed measurements from mobile air temperature recordings on public transport units combined with spatially distributed geo-statistical data. The results indicate that the time of day matters: During the afternoon, areas with a relative low building density, like the industrial area northeast of the inner city, are the warmest, while surfaces in high-building-density areas like the inner city heat up faster during the evening. Second, we combine these measurements with place-based survey data collected in 2010 from residents aged 50 to 92 regarding their individual housing conditions, medical history and social integration to examine the match among heat-based stress of older residents, social conditions and elevated temperatures in their residential quarter. We identify disadvantaged areas for specific already-disadvantaged demographic groups in the city, pointing to a cumulation of inequalities, including heat stress among the most vulnerable. Third, we compare data of biometeorological measurements on urban public squares during the afternoon with results of the micrometeorological model ENVI-met to examine the spatial variability of the inner-city heat load. We complement the modelling results with on-site interviews to evaluate people’s heat perception at the same public places. A simulation shows that additional vegetation would increase thermal comfort at these public places, whereby the heat load assessed using the predicted mean vote (PMV) value would decrease by approximately 60 %. Furthermore, we demonstrate the strengths and weaknesses of heat stress simulation. ENVI-met allows for an overall reasonable representation of heat load during stable atmospheric conditions. However, due to the setup and structure of ENVI-met, large-scale atmospheric changes that occur during the day cannot readily be integrated into ENVI-met simulations.

The neuromodulatory subcortical system (NSS) nuclei are critical hubs for survival, hedonic tone, and homeostasis. Tau-associated NSS degeneration occurs early in Alzheimer's disease (AD) pathogenesis, long before the emergence of pathognomonic memory dysfunction and cortical lesions. Accumulating evidence supports the role of NSS dysfunction and degeneration in the behavioral and neuropsychiatric manifestations featured early in AD. Experimental studies even suggest that AD-associated NSS degeneration drives brain neuroinflammatory status and contributes to disease progression, including the exacerbation of cortical lesions. Given the important pathophysiologic and etiologic roles that involve the NSS in early AD stages, there is an urgent need to expand our understanding of the mechanisms underlying NSS vulnerability and more precisely detail the clinical progression of NSS changes in AD. Here, the NSS Professional Interest Area of the International Society to Advance Alzheimer's Research and Treatment highlights knowledge gaps about NSS within AD and provides recommendations for priorities specific to clinical research, biomarker development, modeling, and intervention. HIGHLIGHTS: Neuromodulatory nuclei degenerate in early Alzheimer's disease pathological stages. Alzheimer's pathophysiology is exacerbated by neuromodulatory nuclei degeneration. Neuromodulatory nuclei degeneration drives neuropsychiatric symptoms in dementia. Biomarkers of neuromodulatory integrity would be value-creating for dementia care. Neuromodulatory nuclei present strategic prospects for disease-modifying therapies.

Background: Research has highlighted that personal characteristics such as teachers' attitudes play an important role in the implementation of inclusive education. However, there are only a few studies that have shed empirical evidence on the relationship of attitudes and inclusive teaching approaches, such as differentiated instruction. In this vein, the present study tackles this research gap and aims to explore teachers' attitudes specifically towards the inclusive practice of DI in Germany. Aims: The present study aimed to investigate teacher profiles based on their attitudes towards differentiated instruction, as well as further to explore whether teachers differentiated instructional implementation varies between the teacher profiles. Sample: The sample consists of 450 teachers (M[subscript age] = 42.89, SD = 10.48, 65% female), from different school tracks in Germany. Methods A two-step cluster analysis was performed in order to identify teacher attitude profiles concerning their implementation of differentiated instruction. Moreover, an analysis of variance was conducted in order to identify variations in terms of the implementation of differentiated instruction across the three clusters. Results: Results from the cluster analyses indicate three distinct teacher attitude profiles: Cluster 1 'The valuing-teacher', Cluster 2 'The non-valuing-teacher' and Cluster 3 'The challenged-but-valuing-teacher'. Moreover, the findings reveal gender and school track differences between the three teacher attitude profiles. Lastly, an analysis of variance indicated that teachers' differentiated instruction practice varied significantly across the clusters. Conclusions: The findings from the present study indicate that teachers not only perceive the value of DI but also the insufficient resources. Thus, it can be assumed that teachers identify both the 'positive' and the 'negative' aspect of DI, and more importantly, they can recognize both attitude domains towards DI in a similar or different level. Consequently, the results show that attitudes in the context of inclusion cannot be characterised as continuum with two distinct poles.

Inflammation and dysfunction of the extrahepatic biliary tree are common causes of human pathology, including gallstones and cholangiocarcinoma. Despite this, we know little about the local regulation of biliary inflammation. Tuft cells, rare sensory epithelial cells, are particularly prevalent in the mucosa of the gallbladder and extrahepatic bile ducts. Here, we show that biliary tuft cells express a core genetic tuft cell program in addition to a tissue-specific gene signature and, in contrast to small intestinal tuft cells, decreased postnatally, coincident with maturation of bile acid production. Manipulation of enterohepatic bile acid recirculation revealed that tuft cell abundance is negatively regulated by bile acids, including in a model of obstructive cholestasis in which inflammatory infiltration of the biliary tree correlated with loss of tuft cells. Unexpectedly, tuft cell-deficient mice spontaneously displayed an increased gallbladder epithelial inflammatory gene signature accompanied by neutrophil infiltration that was modulated by the microbiome. We propose that biliary tuft cells function as bile acid-sensitive negative regulators of inflammation in biliary tissues and serve to limit inflammation under homeostatic conditions.

With the rapid spread of COVID-19, countries around the world implemented strict protocols ordering schools to close. As a result, educational institutions were forced to establish a new form of schooling by implementing emergency remote education. Learning from home during the COVID-19 pandemic brought numerous changes, challenges, and stressors to students' daily lives. In this context, major concerns have been raised based on the reports of students' negative experiences resulting from social distancing and isolation. Given the impact of COVID-19 on many aspects of students' lives, in particular their social and school experiences, research that provides insights into the consequences of this health crisis for students' well-being has become important. This study aims to explore students' experiences of social distancing and its relation to their negative emotional experiences during Germany's first COVID-19-related school closure. Findings indicate that both primary and secondary students missed their friends and classmates and that primary school students perceived higher levels of social distancing. However, a linear regression analysis indicated that the older the students were, the more negatively affected they were by social distancing. The implications of the study's results and further lines of research are discussed.

Innate lymphoid cells (ILCs) are tissue-resident effectors poised to activate rapidly in response to local signals such as cytokines. To preserve homeostasis, ILCs must employ multiple pathways, including tonic suppressive mechanisms, to regulate their primed state and prevent inappropriate activation and immunopathology. Such mechanisms remain incompletely characterized. Here we show that cytokine-inducible SH2-containing protein (CISH), a suppressor of cytokine signaling (SOCS) family member, is highly and constitutively expressed in type 2 innate lymphoid cells (ILC2s). Mice that lack CISH either globally or conditionally in ILC2s show increased ILC2 expansion and activation, in association with reduced expression of genes inhibiting cell-cycle progression. Augmented proliferation and activation of CISH-deficient ILC2s increases basal and inflammation-induced numbers of intestinal tuft cells and accelerates clearance of the model helminth, Nippostrongylus brasiliensis, but compromises innate control of Salmonella typhimurium. Thus, CISH constrains ILC2 activity both tonically and after perturbation, and contributes to the regulation of immunity in mucosal tissue.

Programs defining tissue-resident macrophage identity depend on local environmental cues. For alveolar macrophages (AMs), these signals are provided by immune and nonimmune cells and include GM-CSF (CSF2). However, evidence to functionally link components of this intercellular cross talk remains scarce. We thus developed new transgenic mice to profile pulmonary GM-CSF expression, which we detected in both immune cells, including group 2 innate lymphoid cells and γδ T cells, as well as AT2s. AMs were unaffected by constitutive deletion of hematopoietic Csf2 and basophil depletion. Instead, AT2 lineage-specific constitutive and inducible Csf2 deletion revealed the nonredundant function of AT2-derived GM-CSF in instructing AM fate, establishing the postnatal AM compartment, and maintaining AMs in adult lungs. This AT2-AM relationship begins during embryogenesis, where nascent AT2s timely induce GM-CSF expression to support the proliferation and differentiation of fetal monocytes contemporaneously seeding the tissue, and persists into adulthood, when epithelial GM-CSF remains restricted to AT2s.

Recent findings position tuft cells as key mediators of intestinal immunity through their production of the cytokine interleukin (IL)-25 and activation of group 2 innate lymphoid cells (ILC2s). Though tuft cells are found in numerous epithelial tissues, their phenotype and function have been best characterized in the small intestine, where robust in vivo techniques have enabled the dissection of their cellular function, ontogeny, and key signaling pathways. We describe methods for the identification, quantification, and manipulation of tuft cells, focusing on analysis of ILC2s as a readout of tuft cell function. © 2021 Wiley Periodicals LLC. Group 2 innate lymphoid cells (ILC2s) are tissue-resident cells prominent at barrier sites. Although precursors are found in blood, mature ILC2s can enter the circulation after small intestinal perturbation by migratory helminths and move to distant tissues to influence the local reparative response. Using fate-mapping and methods to bypass the lung or intestinal phases of Nippostrongylus brasiliensis infection, we show that blood ILC2s comprise heterogeneous populations derived from distinct tissues that are dependent on alarmins matched to the receptor profile of the specific tissue ILC2s. Activation of local ILC2s by tissue-specific alarmins induced their proliferation, lymph node migration, and blood dissemination, thus systemically distributing type 2 cytokines. These studies uncover a possible mechanism by which local innate responses transition to systemic type 2 responses by extrusion of activated sentinel ILC2s from tissue into the circulation.

Tuft cells are rare, secretory epithelial cells that generated scant immunological interest until contemporaneous reports in 2016 linked tuft cells with type 2 immunity in the small intestine. Tuft cells have the capacity to produce an unusual spectrum of biological effector molecules, including IL-25, eicosanoids implicated in allergy (such as cysteinyl leukotrienes and prostaglandin D2) and the neurotransmitter acetylcholine. In most cases, the extracellular signals controlling tuft cell effector function are unknown, but signal transduction is thought to proceed via canonical, G protein-coupled receptor-dependent pathways involving components of the signalling pathway used by type II taste bud cells to sense sweet, bitter and umami compounds. Tuft cells are ideally positioned as chemosensory sentinels that can detect and relay information from diverse luminal substances via what appear to be stereotyped outputs to initiate both positive and aversive responses through populations of immune and neuronal cells. Despite recent insights, numerous questions remain regarding tuft cell lineage, diversity and effector mechanisms and how tuft cells interface with the immunological niche in the tissues where they reside.

The perinatal period is a critical window for distribution of innate tissue-resident immune cells within developing organs. Despite epidemiologic evidence implicating the early-life environment in the risk for allergy, temporally controlled lineage tracing of group 2 innate lymphoid cells (ILC2s) during this period remains unstudied. Using complementary fate-mapping approaches and reporters for ILC2 activation, we show that ILC2s appeared in multiple organs during late gestation like tissue macrophages, but, unlike the latter, a majority of peripheral ILC2 pools were generated de novo during the postnatal window. This period was accompanied by systemic ILC2 priming and acquisition of tissue-specific transcriptomes. Although perinatal ILC2s were variably replaced across tissues with age, the dramatic increases in tissue ILC2s following helminth infection were mediated through local expansion independent of de novo generation by bone marrow hematopoiesis. We provide comprehensive temporally controlled fate mapping of an innate lymphocyte subset with notable nuances as compared to tissue macrophage ontogeny.

Sialic acids (Sias) cover vertebrate cell surface glycans. N-Acetylneuraminic acid (Neu5Ac) and its hydroxylated form N-glycolylneuraminic acid (Neu5Gc) are common Sia in mammals. Humans cannot synthesize Neu5Gc but accumulate it on cells through red-meat rich diets, generating numerous immunogenic Neu5Gc-neoantigens. Consequently, humans have diverse anti-Neu5Gc antibodies affecting xenotransplantation, cancer, atherosclerosis, and infertility. Anti-Neu5Gc antibodies circulate as IgG, IgM, and IgA isotypes; however, repertoires of the different isotypes in a large population have not been studied yet. Here, we used glycan microarrays to investigate anti-Neu5Gc IgGs and IgAs in intravenous immunoglobulin (IVIG) or pooled human IgA, respectively. Binding patterns on microarrays fabricated with Neu5Gc- and Neu5Ac-glycans, together with inhibition assays, revealed that different IVIG preparations have highly specific anti-Neu5Gc IgG reactivity with closely related repertoires, while IgAs show cross-reactivity against several Neu5Ac-glycans. Such different anti-Neu5Gc IgG/IgA repertoires in individuals could possibly mediate distinctive effects on human diseases.

Tuft cells-rare solitary chemosensory cells in mucosal epithelia-are undergoing intense scientific scrutiny fueled by recent discovery of unsuspected connections to type 2 immunity. These cells constitute a conduit by which ligands from the external space are sensed via taste-like signaling pathways to generate outputs unique among epithelial cells: the cytokine IL-25, eicosanoids associated with allergic immunity, and the neurotransmitter acetylcholine. The classic type II taste cell transcription factor POU2F3 is lineage defining, suggesting a conceptualization of these cells as widely distributed environmental sensors with effector functions interfacing type 2 immunity and neural circuits. Increasingly refined single-cell analytics have revealed diversity among tuft cells that extends from nasal epithelia and type II taste cells to ex-Aire-expressing medullary thymic cells and small-intestine cells that mediate tissue remodeling in response to colonizing helminths and protists.

In the ongoing era of digital transformation, it is imperative for teachers to equip learners with essential digital competences to navigate the intricacies of the digital landscape successfully. As future in-service teachers function as role models and educators for the proper use of digital technology, pre-service teachers must develop adequate digital proficiency. This holds particularly true in Germany, where the prevailing competence levels of pre-service teachers are reportedly suboptimal. To this end, an extracurricular training program for pre-service teachers, based on the DigCompEdu framework, was implemented from 2021 to 2024, coinciding with COVID-19 pandemic-related limitations. A total of 242 pre-service teachers registered for the program, and 40 completed it. Employing a pre–post design, we assessed (1) attitudes towards digital technology and digital learning, (2) competence beliefs, and (3) test-based competences. Pre–post comparisons show an improvement only in participants' confidence in deploying digital technologies for subject-specific purposes. Unexpectedly, no other statistically significant differences were observed. These findings point at shortcomings in the program, which are discussed to highlight potential areas for refinement and improvement in future programs and curricular implementation. The Corona virus (COVID-19) crisis forced many countries to follow strict protocols ordering schools to close. With schools under lockdown, homeschooling has become the only form of schooling available. Reports have indicated that parents and students have struggled with the challenges of homeschooling. Against this background, this study explored primary school students and parents' educational chances and challenges during homeschooling in two countries: Mexico and Germany. Comparing these two countries can shed light into potential differences of how inclusive approaches have been incorporated in homeschooling. Following a qualitative approach, thirteen semi-structured interviews were conducted with parents and school students. Results from a qualitative content analysis revealed that parents across both countries face challenges to organise homeschooling and motivate their children. However, they spent more time with their children. Primary school students in Germany and Mexico are challenged considerably by the loss of social contact.

Given the increasing diversity of the student body, teachers are called to appropriately address students' various learning needs by means of differentiated instruction (DI). However, empirical research has yielded mixed evidence on teachers' reported use of DI. Using nationally representative data from the National Educational Panel Study in Germany, this article aimed to explore German (as native language) and Mathematics teachers' use of DI practices. In addition, this study took into consideration contextual factors, such as school track, and investigated the impact of teachers' constructivist beliefs on their DI implementation. Results from a mixed analysis of covariance indicated that teachers occasionally implement DI practices. Furthermore, between-subject effects reported differences across school tracks. It appears that advanced secondary school teachers implement less often DI practices. The covariate of teachers' constructivist beliefs was also positively linked to overall teachers' implementation of DI. Implications of the results, as well as further lines of research are discussed.

Group 2 innate lymphoid cells (ILC2s) are distributed systemically and produce type 2 cytokines in response to a variety of stimuli, including the epithelial cytokines interleukin (IL)-25, IL-33, and thymic stromal lymphopoietin (TSLP). Transcriptional profiling of ILC2s from different tissues, however, grouped ILC2s according to their tissue of origin, even in the setting of combined IL-25-, IL-33-receptor-, and TSLP-receptor-deficiency. Single-cell profiling confirmed a tissue-organizing transcriptome and identified ILC2 subsets expressing distinct activating receptors, including the major subset of skin ILC2s, which were activated preferentially by IL-18. Tissue ILC2 subsets were unaltered in number and expression in germ-free mice, suggesting that endogenous, tissue-derived signals drive the maturation of ILC2 subsets by controlling expression of distinct patterns of activating receptors, thus anticipating tissue-specific perturbations occurring later in life.

The small intestinal tuft cell-ILC2 circuit mediates epithelial responses to intestinal helminths and protists by tuft cell chemosensory-like sensing and IL-25-mediated activation of lamina propria ILC2s. Small intestine ILC2s constitutively express the IL-25 receptor, which is negatively regulated by A20 (Tnfaip3). A20 deficiency in ILC2s spontaneously triggers the circuit and, unexpectedly, promotes adaptive small-intestinal lengthening and remodeling. Circuit activation occurs upon weaning and is enabled by dietary polysaccharides that render mice permissive for Tritrichomonas colonization, resulting in luminal accumulation of acetate and succinate, metabolites of the protist hydrogenosome. Tuft cells express GPR91, the succinate receptor, and dietary succinate, but not acetate, activates ILC2s via a tuft-, TRPM5-, and IL-25-dependent pathway. Also induced by parasitic helminths, circuit activation and small intestinal remodeling impairs infestation by new helminths, consistent with the phenomenon of concomitant immunity. We describe a metabolic sensing circuit that may have evolved to facilitate mutualistic responses to luminal pathosymbionts.

In the small intestine, type 2 responses are regulated by a signaling circuit that involves tuft cells and group 2 innate lymphoid cells (ILC2s). Here, we identified the microbial metabolite succinate as an activating ligand for small intestinal (SI) tuft cells. Sequencing analyses of tuft cells isolated from the small intestine, gall bladder, colon, thymus, and trachea revealed that expression of tuft cell chemosensory receptors is tissue specific. SI tuft cells expressed the succinate receptor (SUCNR1), and providing succinate in drinking water was sufficient to induce a multifaceted type 2 immune response via the tuft-ILC2 circuit. The helminth Nippostrongylus brasiliensis and a tritrichomonad protist both secreted succinate as a metabolite. In vivo sensing of the tritrichomonad required SUCNR1, whereas N. brasiliensis was SUCNR1 independent. These findings define a paradigm wherein tuft cells monitor microbial metabolites to initiate type 2 immunity and suggest the existence of other sensing pathways triggering the response to helminths.
