Abstract

Stool is a mixture of excrement, microbiota, enzymes, undigested material, and small molecules. Fecal metabolomics has gained interest recently, owing to advances in metabolomics and growing research into both the host's physiology and the gut microbiome. One challenge with fecal metabolomics is preserving the sample integrity from collection until analysis, as the microbiota and enzymes continue to alter the metabolome following defecation. Currently, flash-freezing or lyophilization are utilized to minimize post-collection metabolome changes; however, this requires complex equipment and immediate processing, precluding the possibility for at-home sampling. Commercial devices containing stabilizing solvents have been developed to facilitate at-home collection, ambient transport, and sample storage. Here, we explore the efficacy of a commercially available stool collection device with a stabilization reagent tailored to fecal metabolomics. Stool samples from six donors were either processed shortly post-collection or stored at room temperature for seven days in the tube, with and without the stabilization reagent. Comprehensive two-dimensional gas chromatography time-of-flight mass spectrometry (GC×GC-TOFMS)-based untargeted metabolomics was utilized for analyzing extracted metabolites. Chemometric analysis was used to evaluate the performance of the device. We found that the device with the stabilization reagent minimized changes in the metabolite profile relative to unstabilized stool left at room temperature for one week. [ABSTRACT FROM AUTHOR]

Abstract

Volatile organic compounds (VOCs) are ubiquitous chemicals of great interest in the study of aromas and flavours of foods. Many recent studies present optimized headspace (HS) and dynamic headspace (DHS) methods for specific sample types; however, the literature does not present (to the best of our knowledge) a generalized procedure for the thorough optimization of a DHS extraction.This article presents an approach using design of experiments (DoE) for the optimization of DHS extraction parameters.The approach is demonstrated for two different food sample types with diverse populations of VOCs: active sourdough colony as an example with a high moisture content, and sourdough bread as an example with a lower moisture content. Optimized methods are assessed for VOC extraction reproducibility and exhaustiveness; guidelines for DHS optimization are presented. 6. More than Men in White Sheets: Seven Concepts Critical to the Teaching of Racism as Systemic Inequality

Abstract

Most college students tend to view racism as individual bias and prejudice and lack an understanding of the broader systemic nature of the problem. The purpose of this article is to examine how to help undergraduate students move from viewing racism as individual bigotry to recognizing its complex nature as a systemic phenomenon that pervades every aspect of United States society. Seven concepts are presented as necessary ingredients for any college course that seeks to address systemic racism. The seven critical concepts are: race as a social construction; dominant and subordinated groups; levels of racism; white privilege; internalized racism; multiple social group memberships; and historical inequality. The discussion of each concept includes a description, brief historical context, and an example of how the concept can be taught in the college classroom.
