Objective To validate the World Health Organization (WHO) non-laboratory-based cardiovascular disease risk prediction model in regions of China. Methods We performed an external validation of the WHO model for East Asia using the data set of China Kadoorie Biobank, an ongoing cohort study with 512 725 participants recruited from 10 regions of China from 2004-2008. We also recalculated the recalibration parameters for the WHO model in each region and evaluated the predictive performance of the model before and after recalibration. We assessed discrimination performance by Harrell's C index. Findings We included 412 225 participants aged 40-79 years. During a median follow-up of11 years, 58 035 and 41 262 incident cardiovascular disease cases were recorded in women and men, respectively. Harrell's C of the WHO model was 0.682 in women and 0.700 in men but varied among regions. The WHO model underestimated the 10-year cardiovascular disease risk in most regions. After recalibration in each region, discrimination and calibration were both improved in the overall population. Harrell's C increased from 0.674 to 0.749 in women and from 0.698 to 0.753 in men. The ratios of predicted to observed cases before and after recalibration were 0.189 and 1.027 in women and 0.543 and 1.089 in men. Conclusion The WHO model for East Asia yielded moderate discrimination for cardiovascular disease in the Chinese population and had limited prediction for cardiovascular disease risk in different regions in China. Recalibration for diverse regions greatly improved discrimination and calibration in the overall population. Objectif Valider le modele de prevision hors laboratoire des risques de maladies cardiovasculaires de l'Organisation mondiale de la Sante (OMS) dans plusieurs regions de Chine. Methodes Nous avons effectue une validation externe du modele de l'OMS pour l'Asie orientale en nous fondant sur l'ensemble de donnees de la China Kadoorie Biobank, une etude de cohorte en cours qui compte 512 725 participants recrutes dans dix regions de Chine entre 2004 et 2008. Nous avons egalement recalcule les parametres de recalibrage du modele de l'OMS dans chaque region, et mesure les performances previsionnelles de ce dernier avant et apres recalibrage. Durante una mediana de seguimiento de 11 anos, se registraron 58 035 y 41 262 casos nuevos de enfermedades cardiovasculares en mujeres y hombres, respectivamente. El indice C de Harrell del modelo de la OMS fue de 0,682 en mujeres y 0,700 en hombres, pero vario entre regiones. El modelo de la OMS subestimo el riesgo de enfermedad cardiovascular a 10 anos en la mayoria de las regiones. Tras la recalibracion en cada region, tanto la discriminacion como la calibracion mejoraron en toda la poblacion. El indice C de Harrell aumento de 0,674 a 0,749 en las mujeres y de 0,698 a 0,753 en los hombres. The AlphaZero algorithm for the learning of strategy games via self-play, which has produced superhuman ability in the games of Go, chess, and shogi, uses a quantitative reward function for game outcomes, requiring the users of the algorithm to explicitly balance different components of the reward against each other, such as the game winner and margin of victory. We present a modification to the AlphaZero algorithm that requires only a total ordering over game outcomes, obviating the need to perform any quantitative balancing of reward components. We present a method for training a neural network to perform image denoising without access to clean training examples or access to paired noisy training examples. Our method requires only a single noisy realization of each training example and a statistical model of the noise distribution, and is applicable to a wide variety of noise models, including spatially structured noise. Our model produces results which are competitive with other learned methods which require richer training data, and outperforms traditional non-learned denoising methods. We present derivations of our method for arbitrary additive noise, an improvement specific to Gaussian additive noise, and an extension to multiplicative Bernoulli noise.

We introduce a new neural network model, together with a tractable and monotone online learning algorithm. Our model describes feed-forward networks for classification, with one output node for each class. The only nonlinear operation is rectification using a ReLU function with a bias. However, there is a rectifier on every edge rather than at the nodes of the network. There are also weights, but these are positive, static, and associated with the nodes. Our "rectified wire" networks are able to represent arbitrary Boolean functions. Only the bias parameters, on the edges of the network, are learned. Another departure in our approach, from standard neural networks, is that the loss function is replaced by a constraint. This constraint is simply that the value of the output node associated with the correct class should be zero. Our model has the property that the exact norm-minimizing parameter update, required to correctly classify a training item, is the solution to a quadratic program that can be computed with a few passes through the network. We demonstrate a training algorithm using this update, called sequential deactivation (SDA), on MNIST and some synthetic datasets. Upon adopting a natural choice for the nodal weights, SDA has no hyperparameters other than those describing the network structure. The neutrino mass can be extracted from a high statistics, high resolution calorimetric spectrum of electron capture in ${}^{163}$Ho. In order to better understand the shape of the calorimetric electron capture spectrum, a second isotope was measured with a close to ideal absorber-source configuration. ${}^{193}$Pt was created by irradiating a ${}^{192}$Pt-enriched platinum foil in a nuclear reactor. This Pt-in-Pt absorber was designed to have a nearly ideal absorber-source configuration. The measured ${}^{193}$Pt calorimetric electron-capture spectrum provides an independent check on the corresponding theoretical calculations, which have thus far been compared only for ${}^{163}$Ho. We present a method of creating high-density superconducting flexible wiring on flexible thin silicon substrates. The flexible wiring, called SOI flex, is created by depositing superconducting wiring on a silicon-on-insulator (SOI) wafer, selectively etching away the thicker silicon section handle layer, and bending the thinner silicon device layer. We show measurements of superconducting transition temperature and critical current for Mo, Nb, and Al on SOI flex. Background Higher body mass index (BMI) is associated with higher incidence of cardiovascular and some non-cardiovascular diseases (CVDs/non-CVDs). However, uncertainty remains about its associations with mortality, particularly at lower BMI levels. Methods The prospective China Kadoorie Biobank recruited >512 000 adults aged 30–79 years in 2004–08 and genotyped a random subset of 76 000 participants. In conventional and Mendelian randomization (MR) analyses, Cox regression yielded adjusted hazard ratios (HRs) associating measured and genetically predicted BMI levels with incident risks of major vascular events (MVEs; conventional/MR 68 431/23 621), ischaemic heart disease (IHD; 50 698/12 177), ischaemic stroke (IS; 42 427/11 897) and intracerebral haemorrhage (ICH; 7644/4712), and with mortality risks of CVD (15 427/6781), non-CVD (26 915/4355) and all causes (42 342/6784), recorded during ∼12 years of follow-up. Results Overall, the mean BMI was 23.8 (standard deviation: 3.2) kg/m
2 and 13% had BMIs of <20 kg/m2 . Measured and genetically predicted BMI showed positive log-linear associations with MVE, IHD and IS, but a shallower positive association with ICH in conventional analyses. Adjusted HRs per 5 kg/m2 higher genetically predicted BMI were 1.50 (95% CI 1.41–1.58), 1.49 (1.38–1.61), 1.42 (1.31–1.54) and 1.64 (1.58–1.69) for MVE, IHD, IS and ICH, respectively. These were stronger than associations in conventional analyses [1.21 (1.20–1.23), 1.28 (1.26–1.29), 1.31 (1.29–1.33) and 1.14 (1.10–1.18), respectively]. At BMIs of ≥20 kg/m2 , there were stronger positive log-linear associations of BMI with CVD, non-CVD and all-cause mortality in MR than in conventional analyses. Conclusions Among relatively lean Chinese adults, higher genetically predicted BMI was associated with higher risks of incident CVDs. 25. Examining the Teacher Pipeline: Will They Stay or Will They Go?

- Abstract
This study examines survey data from teacher education students in their final two years of preparation at a Midwestern university. We asked students to explain if they intend to stay in our state or move to other states to pursue teaching careers after graduation. We compiled all 137 responses using descriptive statistics and found that 59% of respondents intend to leave our state after graduating. Of the remaining respondents, 21% intend to stay in our state and 20% are unsure of their plans. We used qualitative methods to code students' responses, giving reasons for their intentions. While students mentioned financial reasons (low salary and debt) most often, students also valued a sense of place (home, family). Comparisons to populations studied in Carr and Kefalas (2009) and Petrin, Schafft, & Meece (2014) are made with implications for all concerned about the national teacher shortage and outmigration of graduates in rural states.

We introduce a new type of graphical model that we call a "memory factor network" (MFN). We show how to use MFNs to model the structure inherent in many types of data sets. We also introduce an associated message-passing style algorithm called "proactive message passing"' (PMP) that performs inference on MFNs. PMP comes with convergence guarantees and is efficient in comparison to competing algorithms such as variants of belief propagation. We specialize MFNs and PMP to a number of distinct types of data (discrete, continuous, labelled) and inference problems (interpolation, hypothesis testing), provide examples, and discuss approaches for efficient implementation., Comment: 35 pages, 13 figures

OBJECTIVE: Integrated analyses of plasma proteomics and genetic data in prospective studies can help assess the causal relevance of proteins, improve risk prediction, and discover novel protein drug targets for type 2 diabetes (T2D). RESEARCH DESIGN AND METHODS: We measured plasma levels of 2,923 proteins using Olink Explore among ∼2,000 randomly selected participants from China Kadoorie Biobank (CKB) without prior diabetes at baseline. Cox regression assessed associations of individual protein with incident T2D (n = 92 cases). Proteomic-based risk models were developed with discrimination, calibration, reclassification assessed using area under the curve (AUC), calibration plots, and net reclassification index (NRI), respectively. Two-sample Mendelian randomization (MR) analyses using cis -protein quantitative trait loci identified in a genome-wide association study of CKB and UK Biobank for specific proteins were conducted to assess their causal relevance for T2D, along with colocalization analyses to examine shared causal variants between proteins and T2D. RESULTS: Overall, 33 proteins were significantly associated (false discovery rate <0.05) with risk of incident T2D, including IGFBP1, GHR, and amylase. The addition of these 33 proteins to a conventional risk prediction model improved AUC from 0.77 (0.73–0.82) to 0.88 (0.85–0.91) and NRI by 38%, with predicted risks well calibrated with observed risks. MR analyses provided support for the causal relevance for T2D of ENTR1, LPL, and PON3, with replication of ENTR1 and LPL in Europeans using different genetic instruments. Moreover, colocalization analyses showed strong evidence (pH4 > 0.6) of shared genetic variants of LPL and PON3 with T2D. CONCLUSIONS: Proteomic analyses in Chinese adults identified novel associations of multiple proteins with T2D with strong genetic evidence supporting their causal relevance and potential as novel drug targets for prevention and treatment of T2D. Summary: Background: There is limited evidence on the associations of dietary factors and patterns with risk of later‐onset ulcerative colitis (UC) in Chinese adults. Aims: To investigate the associations of dietary factors and patterns with risk of later‐onset UC in Chinese. Methods: The prospective China Kadoorie Biobank cohort study recruited 512,726 participants aged 30–79. Dietary habits were assessed using food frequency questionnaires. Dietary patterns were derived by factor analysis with a principal component method. Cox regression analysis was used to estimate hazard ratios (HRs) and 95% confidence intervals (CIs). Results: During a median follow‐up of 12.1 years, 312 cases of newly diagnosed UC were documented (median age of diagnosis 60.1 years). Egg consumption was associated with higher risk of UC (HR for daily vs. never or rarely: 2.29 [95% CI: 1.26–4.16]), while spicy food consumption was inversely associated with risk of UC (HR: 0.63 [0.45–0.88]). The traditional northern dietary pattern, characterised by high intake of wheat and low intake of rice, was associated with higher risk of UC (HR for highest vs. lowest quartile of score: 2.79 [1.93–4.05]). The modern dietary pattern, characterised by high intake of animal‐origin foods and fruits, was associated with higher risk of UC (HR: 2.48 [1.63–3.78]). Population attributable fraction was 13.04% (7.71%–19.11%) for daily/almost daily consumption of eggs and 9.87% (1.94%–18.22%) for never/rarely consumption of spicy food. Objective: There is little evidence on the genetic associations between life‐course adiposity (including birth weight, childhood BMI, and adulthood BMI) and severe liver disease (SLD; including cirrhosis and liver cancer). The current study aimed to examine and contrast these associations. Methods: Genetic variants were obtained from genome‐wide association studies. Two‐sample Mendelian randomization (MR) analyses were performed to assess the genetic associations of life‐course adiposity with SLD and liver biomarkers. Cox regression was used to estimate adjusted hazard ratios for SLD associated with genetic risk scores of life‐course adiposity and adulthood weight change in the China Kadoorie Biobank. Results: In observational analyses, genetic predispositions to childhood adiposity and adulthood adiposity were each associated with SLD. There was a U‐shaped association between adulthood weight change and risk of SLD. In meta‐analyses of MR results, genetically predicted 1‐standard deviation increase in birth weight was inversely associated with SLD at a marginal significance (odds ratio: 0.81 [95% CI: 0.65–1.00]), whereas genetically predicted 1‐standard deviation higher childhood BMI and adulthood BMI were positively associated with SLD (odds ratio: 1.27 [95% CI: 1.05–1.55] and 1.79 [95% CI: 1.59–2.01], respectively). The results of liver biomarkers mirrored those of SLD. Conclusions: The current study provided genetic evidence on the associations between life‐course adiposity and SLD. [ABSTRACT FROM AUTHOR]
