The increasing impact of severe weather over Europe on lives and weather-sensitive economies can be mitigated by accurate 0–6 h forecasts (nowcasts), supporting a vital‘last line of defence for civil protection and many other applications. Recognizing lack of skill in some complex situations, often at convective and local sub- kilometre scales and associated with rare events, we identify seven recommendations with the aim to improve nowcasting in Europe by the national meteorological and hydrological services (NMHSs) by2030. These recommendations are based on a review of user needs, the state of the observing system, techniques based on observations and high- resolution numerical weather models, as well as tools, data and infrastructure supporting the nowcasting community in Europe. Denser and more accurate observations are necessary particularly in the boundary layer to better characterize the ingredients of severe storms. A key driver for improvement is next-generation European satellite data becoming available as of 2023. Seamless ensemble pre-diction methods to produce enhanced weather forecasts with 0–24 h lead times and probabilistic products require further development. Such products need tobe understood and interpreted by skilled forecasters operating in an evolving forecasting context. We argue that stronger co-development and collaboration between providers and users of nowcasting-relevant data and information are key ingredients for progress. We recommend establishing pan-European now-casting consortia, better exchange of data, common development platforms and common verification approaches as key elements for progressing nowcasting in Europe in this decade

Hintergrund und Ziele: Die Dipeptidylpeptidase 4 (DPP4) ist als Enzym vor allem für ihre Rolle in der Regulation des Blutzuckerspiegels bekannt. Bei systemischer Inhibition der DPP4 konnten zusätzlich gewebsprotektive und antiinflammatorische Effekte nachgewiesen werden, unter anderem in Versuchen zur Transplantatabstoßung in Tiermodellen. Obwohl die protektiven Effekte besonders häufig in der Niere beschrieben wurden, gab es bisher noch keine Untersuchungen zur Auswirkung der DPP4 Inhibition im Rahmen der Nierentransplantation. Methoden: Um den Einfluss der extrarenalen DPP4 Inhibition im Rahmen der Nierentransplantation zu untersuchen, wurden Nieren mit vorhandener membrangebundener DPP4 in Wildtyp (wt) Ratten sowie in Ratten mit fehlender extrarenaler DPP4 Expression (ko) transplantiert und die Abstoßungsreaktion nach 3 Tagen histologisch ausgewertet. Als Kontrollgruppe (kt) dienten die nicht transplantierten Nieren von wt Ratten. Ergebnisse und Beobachtungen: In der histologischen Untersuchung der Nieren auf Kapillaruntergang, Zellproliferation, Apoptose, Fibrose und das Einwandern von Immunzellen ins Gewebe zeigten sich signifikant mehr ERG-1 positive Endothelzellen in den Ratten mit fehlender extrarenaler DPP4 sowie eine um mehr als 25% reduzierte Fibrosierung, die mittels TGF-β und Myofibroblasten nachgewiesen wurde. Außerdem ließen sich signifikant weniger CD161a positive NK-Zellen in den Transplantnieren der ko Guppe nachweisen. Die positiven Effekte der DPP4 Inhibition auf weitere Immunzellen oder Apoptose, die sowohl in Untersuchungen zur Nierenschädigung als auch in Transplantationsmodellen anderer Organe beobachtet wurden, konnten in unserem Modell der rein extrarenalen DPP4 Inhibition nicht nachgewiesen werden. Schlussfolgerungen: Die in anderen Modellen beschriebenen gewebsprotektiven und antiinflammatorische Effekte scheinen vor allem von der Unterdrückung der renalen DPP4 auszugehen, während die Inhibition der extrarenalen DPP4 in unserem Modell Auswirkungen auf die Fibrose und NK-Zellen zeigte. Aims: Systemic inhibition of dipeptidyl peptidase 4 (DPP4) showed a protective effect in several transplant models. Here we assessed the specific role of extrarenal DPP4 in renal transplant rejection. Methods: Kidneys from wildtype (wt) F344 rats were either transplanted in wt Dark Agouti or congenic rats not expressing DPP4. The remaining, not transplanted donor kidney served as healthy controls. To investigate early inflammatory events rats were sacrificed 3 days after transplantation and kidneys were evaluated for inflammatory cells, capillary rarefaction, proliferation, apoptosis and myofibroblasts by immunohistochemistry. Results: Capillary ERG-1-positive endothelial cells were significantly more abundant in renal cortex when transplanted into DPP4 deficient compared to wt recipients. In contrast, TGF-ß and myofibroblasts were reduced by more than 25% in kidneys transplanted into DPP4 deficient compared to wt recipients. Numbers of CD161a-positive NK-cells were significantly lower in allografts in DPP4 deficient compared to wt recipients. Numbers of all other investigated immune cells were not affected by the lack of extrarenal DPP4. Conclusion: In early transplant rejection extrarenal DPP4 is involved in the recruitment of NK-cells and early fibrosis. Beneficial effects were less pronounced than reported for systemic DPP4 inhibition, indicating that renal DPP4 is an important player in transplantationmediated injury.

Nachhaltige Ressourcenverwendung und umweltgerechte Lebensstile., Cite as: Zürcher, Markus et al. (2012): Bulletin 4/12 «Nachhaltige Ressourcenverwendung und umweltgerechte Lebensstile».

An der Tagung «Nachhaltige Ressourcenverwendung und umweltgerechte Lebensstile» der Akademien der Wissenschaften Schweiz diskutierten Forschende den Stand der Ressourcenforschung sowie die Fragen, wo es noch weitere Forschung braucht und wie die Forschung für die Praxis verwendet werden kann. Im Hinblick auf die Tagung wurde im Auftrag der Schweizerischen Akademie der Geistes- und Sozialwissenschaften (SAGW) der Stand der geistes- und sozialwissenschaftlichen Forschung in der Schweiz zur Ressourcenverwendung erhoben. Dr. Michael Stauffacher und Dr. Franziska Schmid von der ETH Zürich stellten die Studie vor. Ergebnisse der Studie Gemäss Studie beschäftigen sich gegenwärtig 155 Forschende der Sozial- und Geisteswissenschaften in leitender Position mit natürlichen Ressourcen. Die Mehrheit der WissenschafterInnen forscht an den beiden ETH und deren Forschungsanstalten. An den kantonalen Universitäten befassen sich nur wenige Forschende im Rahmen der sozialwissenschaftlichen Kerndisziplinen (Soziologie, Ethnologie, Psychologie) mit diesem Forschungsfeld. Viele Forschende setzen sich mit ökonomischen und politischen Aspekten der Ressourcenforschung auseinander. Als Forschungslücken wurden Themen wie Rohstoffe, Bergbau, Landnahme, Abfall/Entsorgung und Biodiversität identifiziert. Bemängelt wurde an der Tagung, dass die Forschung an den Fachhochschulen und die Forschung der Juristen nicht hinreichend berücksichtigt wurde. Gemeinsam Lösungen suchen Es braucht integrierende Forschung an den Schnittstellen von Natur-, Technik-, Sozial- und Geisteswissenschaften, die gemeinsam mit Beteiligten und Betroffenen Probleme beschreibt, mögliche Lösungswege erarbeitet, bewertet und sich damit in die gesellschaftliche Diskussion einbringt. Darin waren sich Tagungsteilnehmer und Studienleiter einig. Gefordert wurde, dass sich die Geistes- und Sozialwissenschaften verstärkt in die Ressourcenforschung einbringen sollten. «Sozialwissenschaften können helfen, zu verstehen, wie Menschen als Individuum und im sozialen Umfeld funktionieren», erklärte Prof. Dr. Heinz Gutscher, Präsident der Akademien der Wissenschaften Schweiz. «Fragen des gesellschaftlichen Wandels, der Bedeutung von unterschiedlichen Kulturen, der Problematiken der Ungleichheit und Macht im Bereich der natürlichen Ressourcen, sowie des Umweltbewusstseins und –verhaltens sind zentral und können nur aus einer sozial- und geisteswissenschaftlichen Sicht sinnvoll bearbeitet werden», ist auch Studienleiter Stauffacher überzeugt. Einleitung: Jährlich fallen schweizweit ungefähr 40 Kinder aufgrund schwerer, erworbenen Hirnschädigungen in ein Wachkoma (Donis & Trojer, 2015). Sie können sich nicht zu ihrem Befinden äussern und so bleibt für das betreuende Fachpersonal unklar, ob die gewählten Massnahmen zur Zustandsverbesserung der Kinder beitragen. Zur Überprüfung dieser Massnahmen verwendet man deshalb im Praxisalltag Verlaufsparameter, die objektive Aussagen ermöglichen. Ziel: Das Ziel ist, reliable, valide und für die Physiotherapie relevante Schmerzassessments für Kinder im Wachkoma zu finden. Solche Assessments helfen, eine bessere und zielorientierte Empfehlung für eine physiotherapeutische Behandlung zu geben und den Therapieverlauf objektiv festzuhalten. Methodik: Mittels einer Literaturrecherche wurden Assessments gesucht, welche für Kinder mit schweren, erworbenen Hirnschädigungen anwendbar sind. Die inkludierten Validierungsstudien wurden bezüglich Methodik und psychometrischen Eigenschaften nach Lienert und Raatz (1998) beurteilt und kritisch diskutiert. Ergebnisse: Für die Schmerzerfassung zeigt die NCS-R die höchsten Werte für Reliabilität und Validität. Offen bleibt die Frage, inwiefern die Ergebnisse dieser Arbeit für Kinder anwendbar sind. Schlussfolgerungen: Da Kinder ab 2 Jahren bereits über entwickelte Schmerzbewältigungsstrategien (Deutscher Hebammenverband, 2010) verfügen, wird empfohlen, den Entwicklungsstand der Kinder immer miteinzubeziehen (Ashwal, 2004). Dadurch könnte die Möglichkeit bestehen, Kinder ab einer gewissen Reife, mit Assessments für Erwachsene zu bewerten.

Bone homeostasis is maintained by osteoblasts (bone formation) and osteoclasts (bone resorption). While there have been numerous studies investigating mesenchymal stem cells and their potential to differentiate into osteoblasts as well as their interaction with different bone substitute materials, there is only limited knowledge concerning in vitro generated osteoclasts. Due to the increasing development of degradable bone-grafting materials and the need of sophisticated in vitro test methods, it is essential to gain deeper insight into the process of osteoclastogenesis and the resorption functionality of human osteoclasts. Therefore, we focused on the comparison of osteoclastogenesis and resorption activity on tissue culture polystyrene (TCPS) and bovine extracellular bone matrices (BMs). Cortical bone slices were used as two-dimensional (2D) substrates, whereas a thermally treated cancellous bone matrix was used for three-dimensional (3D) experiments. We isolated primary human monocytes and induced osteoclastogenesis by medium supplementation. Subsequently, the expression of the vitronectin receptor (V3) and cathepsin K as well as the characteristic actin formation on TCPS and the two BMs were examined. The cell area of human osteoclasts was analyzed on TCPS and on BMs, whereas significantly larger osteoclasts could be detected on BMs. Additionally, we compared the diameter of the sealing zones with the measured diameter of the resorption pits on the BMs and revealed similar diameters of the sealing zones and the resorption pits. We conclude that using TCPS as culture substrate does not affect the expression of osteoclast-specific markers. The analysis of resorption activity can successfully be conducted on cortical as well as on cancellous bone matrices. For new in vitro test systems concerning bone resorption, we suggest the establishment of a 2D assay for high throughput screening of new degradable bone substitute materials with osteoclasts.

Despite advances in the development of in-vitro-tissue-models, the number of endpoints in toxicity-testing which can be addressed with these models is limited. This is due to a lack of key cellular components and a restricted live time of the models (Groeber et al., 2011). In addition, the manual production process of current tissue-models results in significant inter- and intra-lot deviations. One reason for these limitation is that suitable technologies for the culture and production of tissue-models are missing. To overcome these pitfalls, the Fraunhofer IGB is achieving advanced culture systems and biomaterials which allow long term culture of complex tissue-equivalents. Using these technologies, we have developed the first full thickness skin-model with a perfused vascular network (Groeber et al., 2012). Furthermore, within an interdisciplinary consortium of biologist and engineers, we have established a fully automated production facility to generate epidermal models based on an open source reconstructed epidermis (OS-REp) (Lemper et al., 2013; Poumay et al., 2004)). During the product manufacture, all media changes and manipulations are automatically preformed which allows a maximum output of 8000 OS-REp per month (http://www.tissue-factory.com). In our work we could create new technologies for the generation and production of tissue-models which is a vital requirement to increase the success of in-vitro-test-methods.

Introduction: In the field of orthopaedics and traumatology, the improvement of implant materials plays a crucial role for a better treatment of bone defects. Newly developed biodegradable materials have the potential to provide enhanced mechanical properties and reduce the need for implant removal surgery. The initial evaluation of newly designed materials in vitro to predict the outcome in vivo is strongly desired. Therefore, to approach the in vivo situation we established co-culture conditions for primary human mesenchymal stem cells (hMSCs) and monocytes (MCs) to address the bone remodelling process. Our aim of this study was the evaluation of the osteoclastogenesis of monocytes in consequence to the interaction and cross talk with hMSCs without any differentiation factors by media supplementation. Materials and Methods: Human monocytes were isolated of buffy coats and were characterized by FACS analysis, histochemical staining and differentiation studies. For co-culture experiments, hMSCs were seeded either on tissue culture plates (TCPS) or on bovine cortical bone slices. Two set ups were considered: proliferative media and the induction of the osteogenic differentiation for 14 days. Subsequently, monocytes were seeded on top of the pre-cultured hMSCs and were evaluated after another 14 days for an osteoclastic phenotype by immunofluorescence staining and REM analysis. Results and Discussion / References: MCs were successfully differentiated to osteoclasts by media supplementation including RANKL and M-CSF. Co-culture of hMSCs and MCs led to the expression of an osteoclastic phenotype after 14 days. Cathepsin K, VNR and Actin/Vinculin were expressed and analyzed by immunofluorescence staining revealing multinucleated cells within the co-culture set up. The co-culture of hMSCs and hMCs lead to an osteoclastic phenotype after 14 days. The interaction of both cell types is therefore sufficient to generate osteoclast like cells. The bone remodelling process can be simulated in vitro by using monocytes and mesenchymal stem cells. However, the activity of the osteoclasts must be further investigated e.g. by the detection of resorption pits in order to test efficiently newly designed materials. The coculture could further be transferred into a three-dimensional setting to consider the influence of a multilayer culture.
