22. Polyamine metabolism in prostate cancer : studies on localization, cell growth and apoptosis

Contains fulltext : 18924_polymeinp.pdf (Publisher’s version ) (Open Access) The involvement of polyamines in cellular growth and differentiation has prompted many studies of their possible role in cellular neoplasia, including prostatic carcinoma and benign hyperplasia, which represent increasingly important pathologies in men. The precise role of the polyamine system in cellular physiology remains to be clarified. Insight in the exact (sub)cellular localization of polyamine metabolic enzymes might greatly contribute to the elucidation of the specific functions of polyamines in (tumor) cell growth. As described in the first part, we developed cytochemical methods for the (sub)cellular localization of ornithine decarboxylase and polyamine oxidase, which are involved in the biosynthesis and catabolism of polyamines, respectively. The second objective of this thesis was to establish the use of polyamines as determinants of malignant cellular behavior and as potential targets for chemotherapy in prostate cancer. Biochemical analysis of various human prostatic tissues showed that concentrations of the polyamine spermine were substantially lower in prostatic carcinoma compared to normal or benign prostatic tissue. We explored if magnetic resonance spectroscopy (MRS) could be used for the non-invasive determination of spermine in the prostate. Studies on prostatic biopsy samples showed that spermine can be detected and identified with MRS. Recent studies indicate that analogs of polyamines have significant antitumor activity in several human solid tumor models. We have studied the antitumor effect of the polyamine analog N1,N11-di(ethyl)norspermine (DENSpm) on prostatic cancer cells. The in vitro studies showed that all tested human prostatic cancer cells lines were sensitive to DENSpm. An in vivo study in athymic nude mice showed that DENSpm was able to inhibit tumor growth of xenografts of a human prostatic cancer cell line. In conclusion, the polyamine system provides an attractive target to study, to predict and to manipulate the cell kinetic behavior (cell growth, apoptosis) of tumor cells In addition, this study provided new tools and/or techniques for the in situ monitoring of polyamine metabolism 205 p.

Abstract

The prostate and prostate secretions have played an important role in the initial identification of polyamines. One of the polyamines, spermine, was reported in human semen as early as the 17th century by Antoni Van Leeuwenhoek. After the elucidation of the chemical structure of polyamines, about 200 yr later, the biochemical mechanism of action of polyamines in the prostate gland and semen has been studied extensively. Apart from this historical perspective, the prostate has one of the highest polyamine concentrations of any tissue. In rats, the amount of polyamines, especially spermidine, is highest in the ventral prostate followed by the dorsal and lateral prostate, whereas the coagulating glands and seminal vesicles show low values (1). Abstract

To investigate whether polyamines may be valuable diagnostic and prognostic markers in prostate cancer, the presence of alyamines was studied in various human prostatic tissues using both proton magnetic resonance (MR) spectroscopy and righ-pressure liquid chromatography (HPLC). The HPLC results showed that normal and benign hyperplastic prostatic tissues re characterized by a high content of spermine. Spermine levels were reduced in tumor tissue, especially in prostatic carcinoma h metastases, and in xenografts of human prostatic carcinoma cells. These preliminary results indicate that spermine may be d as a biomarker for malignant behavior. The MR spectroscopy study showed that it is possible to detect spermine resonances prostatic biopsy material by one-dimensional and two-dimensional J-resolved MR spectroscopy at high field (600 MHz). ocalized one-dimensional in vitro MR spectra obtained at the clinical field strength of 1.5 T showed spermine signals in the region between 3.0 and 3.3 ppm. In in vivo MR spectra of the human prostate, however, these signals were obscured by esonances of choline (3.2 ppm) and creatine (3.0 ppm). [ABSTRACT FROM AUTHOR]

31. Salivary biomarkers associated with perceived satiety and body mass in humans.

Abstract

Regulation of food intake and energy homeostasis is controlled by a delicate balancing of numerous central and peripheral factors, including circulating peptide hormones. This study investigated the proteome of saliva using SELDI-TOF-MS in relation to satiety and body mass index (BMI) in humans. Within a longitudinal test session, 18 subjects were exposed to a lunch-induced hunger-satiety shift, while every 15 min collecting their whole saliva and rating their hunger and satiety. Saliva was analysed by SELDI-TOF-MS using IMAC arrays with a chromatographic copper surface (IMAC-Cu). From all subjects and time points measured, peptide and protein profiles showed 190 common peaks. Their interrelationships show that 37% of the variation was accounted for in one dimension. About 30 means had a strong association (0.70<|r|<0.95) with all subjective satiety ratings across time during the test session, and seven peaks were significantly correlated to BMI. Database MS searches indicated characterisation of some relevant metabolic peptide hormones. In conclusion, SELDI-TOF-MS on human saliva provides a valuable and noninvasive way of profiling that enables characterisation of novel and differently expressed peptides and proteins which can be used as biomarkers of satiety and overweight., (Copyright © 2007 WILEY-VCH Verlag GmbH & Co. Abstract

Tomoregulin-1, a type-I transmembrane protein with two follistatin modules, a unique epidermal growth factor (EGF) domain and a short, highly conserved cytoplasmic tail, was studied. A number of hematopoietic cell lines (L1210, CEM, Jurkat, U937, K562, JY, THP-1 and T2) express tomoregulin-1 endogenously. In these cells, apoptosis was induced by an antiserum (C29) and purified IgG against the follistatin modules, but not by antisera against the EGF-domain or the cytoplasmic tail. Furthermore, C29 induced apoptosis in tomoregulin-1-, but not in mock-transfected cells. Apoptosis was monitored through genomic DNA fragmentation, annexin-V staining and caspase-3 activation. Treatment of the cells with C29 in the presence of H89 (a SerlThr kinase inhibitor) or 8'-bromo-cyclicAMP revealed that apoptosis was mediated by a cAMP-dependent Ser/Thr kinase. Moreover, C29 increased [cAMP]i over 5-fold. Together, these data suggest that the C29 antiserum against tomoregulin-1 induces apoptosis of hematopoietic cells.
