Nanoparticles, especially from carbon, have great potential in biomedicine. However, prior to clinical use, biocompatibility and biodistribution of these nanoobjects have to be assessed. Currently, particle detection is mostly based on surface-bound labels, inevitably altering materials' properties by surface modification. Further obstacles include bleaching, dissociation of labels from the surface, weak emission of fluorophores due to insufficient tissue opacity or hampered light penetration or the need for specific excitation wavelengths. These characteristics greatly constrain employment of such nanoparticles to address complex analytical questions. To overcome these drawbacks, the use of intrinsic structural features of nanoobjects is highly desirable: the particle surface remains unchanged and the nanoobject exhibits its innate behavior. Thus, for sensitive detection and quantification, labels should be incorporated in the nanoparticle core, thereby avoiding cleavage and warranting unchanged surface characteristics. The incorporation of clinically approved radionuclide P-32 into the lattice of nanodiamond particles using highly defined ion implantation is described here. The properties, uptake, and biodistribution are studied in vivo, in the developing hen's egg model (hen's egg test on chorioallantoic membrane assay). It is found that P-32 labeling of diamond nanoparticles allows their reliable localization and sensitive quantification in a cost efficient, highly reliable, and safe way using available autoradiographic devices and analytical methods.

Nanodiamantpartikel (nDP) wurden in Hinsicht auf Biokompatibilität und Proteinträgereigenschaften untersucht und auf deren Anwendbarkeit in klinischen Therapien zur Knochenheilung getestet. Mesenchmyale Stammzellen spielen eine übergeordnete Rolle in der Erhaltung und Reparatur von Gewebsdefekten im alternden Organsismus, weswegen dieser Zelltyp als Modell zur Analyse der Partikeltoxizität verwendet wurde (in 2D und 3D Kultur getestet). Um die regenerative Kapazität der MSC zu erhöhen, wurden die Zellen mit dem Pentapeptid Met-Enkephalin (ME) behandelt und deren Proliferations- und Differenzierungsverhalten untersucht, weil preproenkephalin (PENK) indikativ mit erghöhter Stammzellfitness assoziert wurde. Da Gefäßneubildung einer der wichtigsten Aspekte der Knochenheilung darstellt, wurden die Wachstumsfaktoren Ang-1 und VEGF mit Iod markiert und auf deren Bindestabilität an die Diamantpartikeloberfläche (Fokis lag auf drei Partikel: ND, NS, LD)untersucht. Im Folgenden wurden Protein-Diamant Komplexe auf die Chorio-Allantoische Membran eines Hühnerembryos appliziert um die Aktivität der Wachstumsfaktoren zu testen. Die geringe Toxizität der Partikel konnte gezeigt werden, sowie die bindung der Proteine an Diamantoberfläschen, welche jedoch stark von der Protein-Diamanttyp Kombination abgängt. Die gefäßinduzierende Aktivität der diamantgebundenen Proteine war ebenfalls stark abhängig von der spezifischen Kombination. Die verteilung der Partikel in den Hühnerembryos konnte mittels P32-labeling nachgewiesen werden, diese wirketen sich jedoch nicht massiv auf den Embryo aus. Zusammenfassend konnte gezeigt werden, dass sich Nanodiamantparikel als Trägermaterial für Wachstumsfaktoren in der Knochenrekonstruktion mit Mesenchymalen Stammzellen eignen, jedoch zuvor individuell getestet werden müssen, Biofunctionalization of diamond nanoparticles (nDP) is being conceived to improve the outcome of established clinical therapies. We thus validated means and measures to characterize three nDPs for their potential in conjunction with ex vivo expanded mesenchymal stem cells (MSC), in particular to serve as carriers in bone healing and tissue engineering throughout aging. We also investigated potential adverse effects on chorionic allantois membrane of the developing hen’s embryo.We could demonstrate that nDPs can be functionalized with bioactive factors firstly revealing no adverse effects on cultured MSC at concentrations below 1 mg/ml. Growth factors bound through physisorption remained active on two of the three nDPs as shown by in ovo tests. First results of biodistribution of nDPs in the whole organism when inoculated during development of the hen embryo showed in ovo distribution of nDPs but no growth defects. Seeding of MSC into nDP coated scaffolds showed improved results compared to uncoated scaffolds, porosity and ridgidity of scaffold material is pivotal for cellular attachment and proliferation.Preproenkephalin (PENK) gene was additionally found being upregulated in fitter MSC. Triggering opioid signaling pathways in MSC using the PENK-derived peptide met-enkephalin increases MSC proliferation, which is in strong contrast to current literature [1, 2]. In conclusion, nDPs can be validated in a standardized fashion employing the here presented panel of tests; if passed successfully individual nDPs are thought to be best suited for subsequent clinical applications.
