Abstract

Seascape ecology, the marine-centric counterpart to landscape ecology, is rapidly emerging as an interdisciplinary and spatially explicit ecological science with relevance to marine management, bio-diversity conservation, and restoration. While important progress in this field has been made in the past decade, there has been no coherent prioritisation of key research questions to help set the future research agenda for seascape ecology. We used a 2-stage modified Delphi method to solicit applied research questions from academic experts in seascape ecology and then asked respondents to identify priority questions across 9 interrelated research themes using 2 rounds of selection. We also invited senior management/conservation practitioners to prioritise the same research questions. Analyses highlighted congruence and discrepancies in perceived priorities for applied research. Themes related to both ecological concepts and management practice, and those identified as priorities include seascape change, seascape connectivity, spatial and temporal scale, ecosystem-based management, and emerging technologies and metrics. Highest-priority questions (upper tercile) received 50% agreement between respondent groups, and lowest priorities (lower tercile) received 58% agreement. Across all 3 priority tiers, 36 of the 55 questions were within a +/- 10% band of agreement. We present the most important applied research questions as determined by the proportion of votes received. For each theme, we provide a synthesis of the research challenges and the potential role of seascape ecology. These priority questions and themes serve as a roadmap for advancing applied seascape ecology during, and beyond, the UN Decade of Ocean Science for Sustainable Development (2021-2030).

Abstract

SERINC5 is the long-searched-for antiviral factor that is counteracted by the HIV-1 accessory gene product Nef. Here, we engineered, via CRISPR/Cas9 technology, T-cell lines that express endogenous SERINC5 alleles tagged with a knocked-in HA epitope. This genetic modification enabled us to study basic properties of endogenous SERINC5 and to verify proposed mechanisms of HIV-1 Nef-mediated counteraction of SERINC5. Using this unique resource, we identified the susceptibility of endogenous SERINC5 protein to posttranslational modulation by type I IFNs and suggest uncoupling of Nef-mediated functional antagonism from SERINC5 exclusion from virions., When expressed in virus-producing cells, the cellular multipass transmembrane protein SERINC5 reduces the infectivity of HIV-1 particles and is counteracted by HIV-1 Nef. Due to the unavailability of an antibody of sufficient specificity and sensitivity, investigation of SERINC5 protein expression and subcellular localization has been limited to heterologously expressed SERINC5. We generated, via CRISPR/Cas9-assisted gene editing, Jurkat T-cell clones expressing endogenous SERINC5 bearing an extracellularly exposed hemagglutinin (HA) epitope [Jurkat SERINC5(iHA knock-in) T cells]. This modification enabled quantification of endogenous SERINC5 protein levels and demonstrated a predominant localization in lipid rafts. Interferon alpha (IFN-α) treatment enhanced cell surface levels of SERINC5 in a ruxolitinib-sensitive manner in the absence of modulation of mRNA and protein quantities. Parental and SERINC5(iHA knock-in) T cells shared the ability to produce infectious wild-type HIV-1 but not an HIV-1 Δnef mutant. SERINC5-imposed reduction of infectivity involved a modest reduction of virus fusogenicity. An association of endogenous SERINC5 protein with HIV-1 Δnef virions was consistently detectable as a 35-kDa species, as opposed to heterologous SERINC5, which presented as a 51-kDa species. Nef-mediated functional counteraction did not correlate with virion exclusion of SERINC5, arguing for the existence of additional counteractive mechanisms of Nef that act on virus-associated SERINC5. In HIV-1-infected cells, Nef triggered the internalization of SERINC5 in the absence of detectable changes of steady-state protein levels. These findings establish new properties of endogenous SERINC5 expression and subcellular localization, challenge existing concepts of HIV-1 Nef-mediated antagonism of SERINC5, and uncover an unprecedented role of IFN-α in modulating SERINC5 through accumulation at the cell surface. Abstract

The range expansion of tropical fish into temperate waters is increasing markedly in response to climate change. Range-expanding fish encounter novel diets and environments, but we know little about how these conditions facilitate or hinder distribution shifts. Here, we quantified relative growth rate, morphometric condition and trophic niche of juvenile Acanthurus triostegus, a dominant range-expanding tropical surgeonfish, at four locations across 10° of latitude off eastern Australia. We related these metrics to differences in temperature and nutritional quality of dominant seaweeds and the epilithic algal matrix. Temperate food sources were richer in nitrogen than tropical diets. Stable isotope analysis (δ13 carbon and δ15 nitrogen) of fish muscle revealed a large trophic niche breadth at the highest latitude indicating a generalist foraging strategy, and more nitrogen-enriched isotopic signatures compared to tropical regions. Fish length was strongly correlated to δ13C in all regions, suggesting an ontogenetic shift in diet independent of latitude. Despite temperature differences of 4°C, fish growth and body condition were similar across tropical and temperate regions. These results suggest that more nutritious temperate diets may compensate for the effects of cooler water temperatures. Neither summer water temperatures nor dietary factors appear to limit the success of juvenile tropical vagrants as they continue to expand their range along eastern Australia.

Abstract

Understanding the processes driving species distributions is a key question in ecology, yet obtaining pertinent data for many species can be challenging. Tag-recapture data from voluntary recreational fisheries programmes is an undervalued citizen science data resource and can provide valuable observation records for data-poor species. Here, we highlight the utility of such data by compiling four tag-recapture datasets (n = 20 878 tags) to describe the movements and distribution of a piscivorous predator, tailor Pomatomus saltatrix, with respect to their environment. We compared species distribution models that explored a trade-off between temporal duration and spatial extent. Both models revealed sea surface temperature (SST) was an important factor in describing the distribution and density of tailor. However, the model with broader spatial extent indicated a non-linear response to SST, peaking at 21.5°C, while the model with temporal longevity showed a linear SST response. This disjunct showcases the importance of building models across an adequate geographic extent to ensure covariate responses are not truncated. This is the first study to comprehensively describe the distribution and migrations of adult tailor across their eastern Australian distribution. The results are discussed in light of ocean warming along eastern Australia, and the subsequent implications for using tag-recapture data in fisheries management.
