Abstract

Conservation translocations are becoming common conservation practice, so there is an increasing need to understand the drivers of plant translocation performance through reviews of cases at global and regional levels. The establishment of the Italian Database of Plant Translocation (IDPlanT) provides the opportunity to review the techniques used in 186 plant translocation cases performed in the last 50 years in the heart of the Mediterranean Biodiversity Hotspot. We described techniques and information available in IDPlanT and used these data to identify drivers of translocation outcomes. We tested the effect of 15 variables on survival of translocated propagules as of the last monitoring date with binomial logistic mixed-effect models. Eleven variables significantly affected survival of transplants: life form, site protection, material source, number of source populations, propagation methods, propagule life stage, planting methods, habitat suitability assessment, site preparation, aftercare, and costs. The integration of vegetation studies in the selection of suitable planting sites significantly increased the success of translocation efforts. Although posttranslocation watering had a generally positive effect on translocation outcome, other aftercare techniques did not always increase transplant survival. Finally, we found that how funds were spent appeared to be more important than the actual amount spent. Plant translocations in Italy and in the Mediterranean area should account for the complexity of speciation, gene flow, and plant migrations that has led to local adaptations and has important implications for the choice and constitution of source material.

Abstract

IDPlanT is the Italian Database of Plant Translocation, an initiative of the Nature Conservation Working Group of the Italian Botanical Society. IDPlanT currently includes 185 plant translocations. The establishment of a national database on plant translocation is a key step forward in data sharing and techniques improvement in this field of plant conservation. Abstract

Simplification of agricultural systems, farmland abandonment, uncontrolled urbanization, together with global scale drivers, determine fast and unpredictable phenomena of hydrogeological instability, biodiversity decline and identity loss. Men should hence promote socio-economic resilience and place identity, using biocultural landscapes as a resource. These landscapes are the outcome of the historical relationship between man and nature, resulting from complex interactions between biodiversity (at all levels, including species richness, ecosystem and biotope diversity) and cultural diversity, including material and immaterial aspects (architectural heritage, traditions, customs, local traditional agricultural practices, dialectal culture). Traditional water use is one of the main defining factors of traditional agricultural landscapes, characterized by significantly multiplied and enhanced functions and by the highest degree of biocultural diversity. The integrity of traditional agricultural landscapes is an indicator of environmental sustainability, socio-economic growth and habitat resilience. Land management and planning should promote the multi-functionality of biocultural landscapes through new interdisciplinary frameworks and innovative strategies, aimed at landscape conservation, enhancement and creative management.

Abstract

The work presents a biocultural diversity interpretation of traditional Mediterranean agricultural landscapes. These landscapes are the result of complex interactions between biodiversity (at all levels, including species richness, ecosystem and biotope diversity) and cultural diversity, including material and immaterial aspects (architectural heritage, traditions, customs, local traditional agricultural practices, dialectal culture, etc.). The main theme is the traditional use of water related to traditional hydraulic systems, which in some areas date back to medieval times. Traditional agricultural landscapes grant a variety of ecosystem services and play a key role in biodiversity conservation. They support the persistence of a rich heritage of cultivated plant varieties and, at the same time, determine an ecosystem infrastructure that preserves wild animal and plant biodiversity.

Abstract

AIM: Arrhythmogenic right ventrticular dysplasia/cardiomyopathy (ARVD/C) is an inherited cardiomyopathy characterized by fibrofatty replacement and a high risk of ventricular arrhythmias (VA) and sudden cardiac death (SCD). The aim of the present investigation is to examine the pathological profile and the clinical correlations in a group of ARVD/C patients. METHODS: We conducted a multicenter study evaluating 47 patients (31 men; mean age 37±14 years) with definite ARVD/C. Diagnosis was established according to the actual clinicomorphologic criteria at autopsy or clinically. We divided the study population in 2 different groups. First group included 28 alive patients and the second 19 patients dead suddenly. RESULTS: Age at presentation was different in the two groups (P=0.0015). We observed an important association regarding the risk of sudden death and the history of physical exercise (P=0.0017). Moreover patients with negative outcome (i.e., SCD, cardiac transplantation, congestive heart failure) had a significantly association with biventricular form of ARVD/C (P=0.0034) and age presentation (P=0.003). Left ventricular (LV) involvement was frequently observed in the two groups (17% and 32% respectively). Post-mortem examination revealed frequent inflammatory infiltrates (26%) indicating active myocarditis, which probably justify the fatal arrhythmic events occurred in these patients. CONCLUSION: Frequent LV involvement justifies the recent adoption of the broad term Arrhythmogenic Cardiomyopathy. Early age presentation, sport activity and the biventricular form of ARVD/C represent important predictors of adverse outcome that can be useful to early identify patients at high risk.

Abstract

A study of the hydrophyte communities of some small ponds in the beech-wood belt of the higher Nebrodi Mts is presented. A new association of shallow stagnant water bodies, the Utriculario-Potametum natantis, is described. Its biological structure, biogeographical characteristics, syntaxonomic position, phytosociological relationships and dynamic links are discussed.

Abstract

The Mediterranean Basin is one of the most significantly altered World Biodiversity Hotspots with extensive habitat loss and fast genetic population erosion, for which urgent biodiversity reconnaissance and preservation actions are required. In particular, Sicily has about 600 taxa classified as threatened or near-threatened. The correct recognition and identification of such biodiversity is required for supporting further activities. The objective of this work is to assess the ability of the DNA barcoding approach to identify different taxonomic groups from a collection of the most threatened plant taxa, throughout natural Sicilian populations. The evaluation of the DNA barcoding core markers,rbcL andmatK, was carried out on 30 taxa belonging to 13 families. DNA barcode fragments were recovered from all taxa (100%). TherbcL gene was recovered from 97% of the taxa andmatK gene from 73%. In this test, 19 taxa overall (63%) were totally resolved at the specific or subspecific level, by at least one of the core markers. Fourteen of the 17 most threatened taxa (EN, CR) included in this work were totally discriminated. ThematK andrbcL locus, respectively, resolved 64% and 48% of the taxa successfully sequenced. ThematK gene expressed the highest genetic distance (K2P value), from 0.4% to 8.6%, against a range of 0.1–2% ofrbcL gene. However, therbcL gene appeared a good compromise between PCR, sequencing success and species-level resolution. Cryptic groups suggest the implementation of additional barcoding markers or different primer combinations, particularly formatK, in order to increase the performances. However, this preliminary result confirms the potential of the barcoding approach for quick identification of unknown and heterogeneous plant groups to generate a dedicated reference data-set of the threatened Sicilian flora for a wide range of applications. 29. Genetic analysis of <italic>Citrus aurantium</italic> L. (<italic>Rutaceae</italic>) cultivars by ISSR molecular markers.

Abstract

Eight cultivars of Citrus aurantium L., showing peculiar morphological traits of the fruits and leaves, were analyzed genetically. Inter simple sequence repeat (ISSR) was chosen as molecular markers because they represent a highly efficient system for investigating variability at intraspecific level. The particular morphological traits were discussed, the genetic identity and distance matrix based on Nei index was calculated, and the unweighted pair group method with arithmetic mean (UPGMA) dendrogram was generated. A total of 53 clearly distinguished DNA bands were considered for the ISSR analysis of which 24 were polymorphic. A basic C. aurantium fingerprinting pattern was obtained. The ISSR profiles allowed to discriminate only two cultivars, while the other six ones did not show polymorphisms, although their phenotypes were notably different, indicating that mutations must have occurred without significantly modifying the DNA length between the simple sequence repeats. The results showed a very low level of genetic variability among the cultivars; “Canaliculata” formed a separated cluster with C. sinensis Osbeck, suggesting a probable hybrid origin derived from crossing between sour and sweet orange. This study confirmed the reliability and reproducibly of ISSR markers for Citrus fingerprinting purposes, although some morphological differences could not be detected; therefore, both morphological and genetic characters must be considered for cultivar or variety identification. To investigate the biosystematic and genetic relationships among Sicilian species of Genista sect. Voglera, four populations of three species, Genista aristata C. Presl, Genista cupanii Guss., and Genista madoniensis Raimondo, have been studied. Genista madoniensis is tetraploid, with a chromosome number of 2n = 48, as are the other two species. An isozyme study showed that G. madoniensis is taxonomically closer to G. aristata than to G. cupanii, the latter possessing four unique alleles. Abstract

Eight cultivars of Citrus aurantium L., showing peculiar morphological traits of the fruits and leaves, were analyzed genetically. Inter simple sequence repeat (ISSR) was chosen as molecular markers because they represent a highly efficient system for investigating variability at intraspecific level. The particular morphological traits were discussed, the genetic identity and distance matrix based on Nei index was calculated, and the unweighted pair group method with arithmetic mean (UPGMA) dendrogram was generated. A total of 53 clearly distinguished DNA bands were considered for the ISSR analysis of which 24 were polymorphic. A basic C. aurantium fingerprinting pattern was obtained. The ISSR profiles allowed to discriminate only two cultivars, while the other six ones did not show polymorphisms, although their phenotypes were notably different, indicating that mutations must have occurred without significantly modifying the DNA length between the simple sequence repeats. The results showed a very low level of genetic variability among the cultivars; “Canaliculata” formed a separated cluster with C. sinensis Osbeck, suggesting a probable hybrid origin derived from crossing between sour and sweet orange. This study confirmed the reliability and reproducibly of ISSR markers for Citrus fingerprinting purposes, although some morphological differences could not be detected; therefore, both morphological and genetic characters must be considered for cultivar or variety identification. Abstract

Sumac (Rhus coriaria L.), a spice obtained by grinding whole sumac berries, is a complex natural product with a plethora of biological activities that can be favorably explored in nutraceutical and pharmaceutical fields. Sicilian sumac is herein chemically investigated by means of a combined NMR/HR MS-based profiling. A hydroalcoholic extract was prepared and its complexity unraveled following fractionation in an alcoholic and an aqueous fraction. The 1H-NMR spectrum of this latter fraction was dominated by the signals of gallic acid and its derivatives, whereas HR MS and HR MS/MS analyses highlighted the diversity in malic acid derivatives. Gallotannins and galloyl flavonol glycosides with quercetin and myricetin as the main aglycones were highly represented in the alcoholic fraction. To improve bioavailability of chemicals in sumac extract, the feasibility of a microencapsulation process by spray drying was investigated. In particular, the addition of maltodextrin, cyclodextrin and gum arabic as covering agents was evaluated. The tests were carried out at different temperatures and concentrations. The addition of maltodextrin was found to be suitable as a carrier for the spray drying of sumac extract, having the highest yields, always > 82%. The highest values of a* (red index) and Chroma were recorded with cyclodextrin and maltodextrin at the lowest concentrations and temperatures of spray drying (120°C). The correlations between the characteristics of the studied microcapsules (yields, moisture, total and surface polyphenols and colour) have been represented by Principal Component Analysis (PCA).

Abstract

Abies nebrodensis (Lojac.) Mattei is an endemic species of the north-west of Sicily located in an 84 ha area in the Madonie Regional park. The current population is limited to 30 relic adult trees and a fluctuating number of juveniles of natural regeneration. The species is defined as “Critically Endangered” in the Italian list of threatened plants and is classified as CR-D in the 2000 IUCN Red List of Threatened Species. This article reports the key action undertaken by the LIFE4FIR project aimed at preserving A. nebrodensis, and the results obtained so far in three years of activity. OpenArrays SNPs genotyping revealed a high rate of inbreeding in the natural population and that the adult trees are genetically related. Controlled cross-pollination was consequently performed to increase the genetic variability of the progeny. Outbred offspring are currently being grown in the nursery. Reforestation has been planned by using 4000 selected outbred seedlings in 10 areas within Madonie Park to create re-diffusion cores. Support and protection of the relic population have been implemented through regular phytosanitary surveys, as well as new fencing and video surveillance systems against grazing and wild herbivores. A seedbank and cryobank for the long-term germplasm conservation have been established.

Abstract

Sulla base dei dati di letteratura e di osservazioni in campo, gli autori analizzano la componente legnosa indigena della flora siciliana (fanerofite e nano-fanerofite) utile per interventi di recupero e restauro ambientale. Si tratta di 173 taxa, specifici e infraspecifici, afferenti a 72 generi di 34 famiglie, tra le quali sono particolarmente rappresentate Rosaceae (47), Fagaceae (18), Fabaceae (15) e Salicaceae(12). Per ogni taxon vengono evidenziati dati concernenti la natura del substrato e la fascia vegetazionale di pertinenza. Viene fornita, inoltre, una valutazione complessiva sulla potenzialità d’impiego in funzione della rispettiva plasticità ecologica.

Abstract

This paper presents an operational methodology to map and analyze the floristic richness of “target species” in Natura 2000 sites, making use of G.I.S. tools and procedures. A Floristic diversity map (scale 1:50,000), covering an area of 612 km2, was produced by a team of experts as part of the management plans of “Madonie Mountains” Sites of Community Importance (SCIs), located in Sicily (Italy). The primary grid map represents the richness of “target species”, which include species of Community interests, taxa on the National Red List, endemic and threatened, species protected under International Conventions, taxa of phytogeographic importance. Secondary data frames include a three-dimensional map representing the number of species present in each cell, a coarser species richness distribution (scale 1:400,000) and a reference map of endemism rate in the Mediterranean area. Such a cartographic document has proven to be an effective tool in biodiversity conservation planning. Furthermore, the knowledge of floristic richness and distribution is not only important for the management of protected areas, but it is also important for the sustainable management of cultural landscapes.

Abstract

Viscum album L., (Common Mistletoe), is a hemiparasitic shrub that lives on a wide range of woody plant species, known since Neolithic period for its ritual and for pharmacological properties. Up to now, no investigation has been reported on the chemical composition of the essential oils of V. album aerial parts in relation with the diverse host trees. Consequently, the essential oils compositions of three Sicilian accession of V. album, growing on Quercus suber L., Crataegus monogyna Jacq. and Olea europaea L. var. europaea cv. 'Santagatese', respectively, were evaluated by GC-MS. The three populations showed quite different profiles indicating that the nature of the host considerably influences the chemical composition of the guest plant, V. album.

Abstract

A. nebrodensis (Nebrodi fir, Sicilian fir) is restricted to a small area of the Madonie Natural Park in Sicily. According to recent estimates, its only population consists of 30 adult individuals and a fluctuating number of juveniles derived from natural regeneration; besides, some hundreds of cultivated plants are preserved as ex situ collection. We used RAPD data from six 10-mer primers to examine the consequences of extensive historical clearance and human pressures on the extant population. Data from multiple life stages and different habitat conditions were considered, affording an opportunity to ascertain for the first time the structure of genetic variation in the extant uneven-aged population on both spatial and temporal scale. Consistent with previous works, we found that the history of disturbance and past land use did not affect drastically the biological diversity of the present-day population. Considerable levels of genetic variation were detected both in adult and juvenile sets. The non-significant correlation between genetic and physical distances (Mantel test; r = - 0.075) revealed a random distribution of genotypes in the natural stand. When the juvenile set was divided in 17 subsets, based on maternal provenance and growing conditions, Shannon's index (54%) and AMOVA analysis (84%) indicated that most of genetic variation resides within subsets. Neighbour-joining cluster analysis supported the hypothesis of high crossfertilization rates expected for a woody perennial species; nevertheless some grouping of related individuals suggested partial inbreeding and a weak pattern of genetic structure. Changes in structure can occur as the progeny ages and offspring thinning is responsible for moulding patterns of genetic diversity and population structure in time.

Abstract

Monumental olive trees, with their longevity and their remarkable size, represent an important information source for the comprehension of the territory where they grow and the human societies that have kept them through time. Across the centuries, olive trees are the only cultivated plants that tell the story of Mediterranean landscapes. The same as stone monuments, these green monuments represent a real Mediterranean natural and cultural heritage. The aim of this paper is to discuss the value of monumental trees as “biocultural heritage” elements and the role they play in the interpretation of the historical stratification of the landscape. We present the results of a survey of the most significant olive trees growing in Sicily. The selection was based on the “monumentality” aspects of trees, taking into account dendrometric parameters and environmental contexts. The collected dataset constitutes a heterogeneous sample of 367 specimens of considerable size that, in some cases, reach a circumference of about 19 m. Starting from the data presented here, the whole Sicilian territory shows a historical relationship between human and olive. The presence of these plant monuments is, therefore, evidence of long-term, often centennial, landscapes as a result of sustainable use of the territory.

Abstract

BACKGROUND: This study investigated the chemical profile and biological activity of Diplotaxis erucoides subsp. erucoides (L.) DC. (Brassicaceae) collected in Sicily (Italy). RESULTS: Liquid chromatography coupled with electrospray ionization and high-resolution mass spectrometry (LC-ESI/HRMS) analysis of the ethanol extract revealed the presence of 42 compounds – glucosinolates, hydroxycinnamic acids, flavonoids, and oxylipins. The extract was tested for its antioxidant activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic) acid (ABTS), ferric reducing ability power (FRAP), and β-carotene bleaching tests. Promising protection from lipid peroxidation was observed after 30 min of incubation in a β-carotene bleaching test (IC50 of 3.32 μg mL−1). The inhibition of carbohydrates-hydrolyzing enzymes resulted in IC50 values of 85.18 and 92.36 μg mL−1 for α-amylase and α-glucosidase, respectively. Significant inhibition against lipase enzyme was observed (IC50 of 61.27 μg mL−1). CONCLUSION: Diplotaxis erucoides can be considered a potential source of antioxidant, hypoglycemic, and hypolipidemic bioactives. © 2021 Society of Chemical Industry.

Abstract

The present work was designed to identify and characterize novel antimicrobial peptides (AMPs) from Charybdis pancration (Steinh.) Speta, previously named Urginea maritima, is a Mediterranean plant, well-known for its biological properties in traditional medicine. Polypeptide-enriched extracts from different parts of the plant (roots, leaves and bulb), never studied before, were tested against two relevant pathogens, Staphylococcus aureus and Pseudomonas aeruginosa. With the aim of identifying novel natural AMPs, peptide fraction displaying antimicrobial activity (the bulb) that showed minimum inhibitory concentration (MICs) equal to 30 &micro, g/mL against the above mentioned strains, was analysed by high-resolution mass spectrometry and database search. Seventeen peptides, related to seven proteins present in the investigated database, were described. Furthermore, we focused on three peptides, which due to their net positive charge, have a better chance to be AMPs and they were investigated by molecular modelling approaches, in order to shed light on the solution properties of their equilibrium structures. Some of new detected peptides could represent a good platform for the development of new antimicrobials in the fight against antibiotic resistance phenomenon.
