Long-term research in grassland biodiversity experiments has provided empirical evidence that ecological and evolutionary processes are intertwined in determining both biodiversity–ecosystem functioning (BEF) and biodiversity–stability relationships. Focusing on plant diversity, we hypothesize that multifunctional stability is highest in high-diversity plant communities and that biodiversity–stability relationships increase over time due to a variety of forms of ecological complementarity including the interaction with other biota above and below ground. We introduce the multiple-mechanisms hypothesis of biodiversity–stability relationships suggesting that it is not an individual mechanism that drives long-term biodiversity effects on ecosystem functioning and stability but that several intertwined processes produce increasingly positive ecosystem effects. The following six mechanisms are important. Low-diversity plant communities accumulate more plant antagonists over time (1), and use resources less efficiently and have more open, leaky nutrient cycles (2). Conversely, high-diversity plant communities support a greater diversity and activity of beneficial interaction partners across trophic levels (3); diversify in their traits over time and space, within and across species, to optimize temporal (intra- and interannual) and spatial complementarity (4), create a more stable microclimate (5), and foster higher top-down control of aboveground and belowground herbivores by predators (6). In line with the observation that different species play unique roles in ecosystems that are dynamic and multifaceted, the particular mechanism contributing most to the higher performance and stability of diverse plant communities might differ across ecosystem functions, years, locations, and environmental change scenarios. Diversity and community composition of earthworms, key drivers of ecosystem functions, are increasingly threatened by global change, including climate and land-use change. However, empirical evidence for interactions of these concurrent drivers in affecting earthworm communities is scarce. Here, we investigated the effects of an experimentally imposed climate change scenario, including warming and altered precipitation patterns, and land use with two croplands (both conventional farming and organic farming characterize a three-year crop rotation) and two grasslands (intensively-used meadow and extensively-used meadow) on earthworm communities across different seasons and years in a field experiment. Compared with grasslands, earthworms in croplands have lower species richness (-26 %), abundance (-80 %), and biomass (-73 %), particularly the abundance of juveniles (-83 %) and Aporrectodea rosea (-76 %) as well as the biomass of juveniles (-84 %), A. rosea (-72 %), Octolasion cyaneum (-47 %), and Lumbricus terrestris (-83 %). Due to extreme droughts in Central Europe from 2018 to 2020, earthworm abundance and biomass were low across land-use types, but in grassland they increased (abundance: +80 %; biomass: +85 %) in 2021 presumably due to increased moisture conditions. Main effects of experimental climate change and intensified management practices as well as interaction of experimental climate change and land use on abundance and biomass of earthworms were non-significant. Notably, experimental climate change and land use interactively altered earthworm community composition, with the most pronounced difference between ambient and future climate in croplands than in grasslands. This indicates that earthworm community composition more sensitively reflects changes in environmental conditions than earthworm abundance and biomass, but the latter two negatively responded to prolonged drought conditions. Soils, just like all other ecosystem compartments, change over time and, consequently, conditions for soil-inhabiting organisms are also changing, affecting their composition and diversity. Soil biodiversity is a critical component of ecosystems that supports many essential ecosystem functions and services, such as nutrient cycling, carbon sequestration, water regulation and biomass production for food, fodder, fibre and energy. However, and despite the importance of soil biodiversity for ecosystem health and human well-being, neither current state, drivers, potential consequences for ecosystem services nor options for sustainable governance of soil biodiversity are well understood. Here, we provide a framework for and argue that conducting a national assessment of soil biodiversity, albeit being a complex endeavour, is fundamental to building a baseline to understand the current state and trends of soil biodiversity, but also to identify the main drivers of change, the impacts of soil biodiversity loss and the potential pathways for conservation and sustainable governance of soil biodiversity.

Ein unverzichtbarer Beitrag zur Erfüllung essenzieller Bodenfunktionen wird durch Bodenorganismen und deren Diversität geleistet. Der Kenntnisstand zur Bodenbiodiversität, die einer Vielzahl von Stressoren ausgesetzt ist und vielfach rückläufig oder gefährdet ist, muss derzeit als gering eingeschätzt werden. Das aktuelle Monitoring in Deutschland liefert keine belastbare Datengrundlage, um bundesweite Aussagen zum Zustand und zum Trend der Bodenbiodiversität treffen zu können. Die Weiterentwicklung des Bodenbiodiversitätsmonitorings ist daher eine Schwerpunktaufgabe des Nationalen Monitoringzentrums zur Biodiversität. Ein Fachgremium erarbeitet seit 2021 Leitlinien für ein bundesweit harmonisiertes Bodenbiodiversitätsmonitoring aufbauend auf bestehenden Monitoringaktivitäten und neueren Methodenentwicklungen. Dabei sollen gemeinsam mit den Monitoringakteuren Synergiepotenziale herausgearbeitet und genutzt werden. Die Identifizierung der wesentlichen Handlungsfelder und die Formulierung und Priorisierung der Ziele des Monitorings sind erste umgesetzte Schritte auf dem Weg zu einem bundesweit harmonisierten Bodenbiodiversitätsmonitoring.

Springtails (Collembola) inhabit soils from the Arctic to the Antarctic and comprise an estimated ~32% of all terrestrial arthropods on Earth. Here, we present a global, spatially-explicit database on springtail communities that includes 249,912 occurrences from 44,999 samples and 2,990 sites. These data are mainly raw sample-level records at the species level collected predominantly from private archives of the authors that were quality-controlled and taxonomically-standardised. Despite covering all continents, most of the sample-level data come from the European continent (82.5% of all samples) and represent four habitats: woodlands (57.4%), grasslands (14.0%), agrosystems (13.7%) and scrublands (9.0%). We included sampling by soil layers, and across seasons and years, representing temporal and spatial within-site variation in springtail communities. We also provided data use and sharing guidelines and R code to facilitate the use of the database by other researchers. This data paper describes a static version of the database at the publication date, but the database will be further expanded to include underrepresented regions and linked with trait data.

This article provides protocols with top ticks and pitfalls for preparation and successful generation of mouse and human DC from different cellular sources, such as murine BM and HoxB8 cells, as well as human CD34(+) cells from cord blood, BM, and peripheral blood or peripheral blood monocytes. We describe murine cDC1, cDC2, and pDC generation with Flt3L and the generation of BM-derived DC with GM-CSF. Protocols for human DC generation focus on CD34(+) cell culture on OP9 cell layers for cDC1, cDC2, cDC3, and pDC subset generation and DC generation from peripheral blood monocytes (MoDC). Additional protocols include enrichment of murine DC subsets, CRISPR/Cas9 editing, and clinical grade human DC generation. While all protocols were written by experienced scientists who routinely use them in their work, this article was also peer-reviewed by leading experts and approved by all co-authors, making it an essential resource for basic and clinical DC immunologists., 01 november 2023

Plant nutrient uptake and productivity are driven by a multitude of factors that have been modified by human activities, like climate change and the activity of decomposers. However, interactive effects of climate change and key decomposer groups like earthworms have rarely been studied. In a field microcosm experiment, we investigated the effects of a mean future climate scenario with warming (+ 0.50 °C to + 0.62 °C) and altered precipitation (+ 10% in spring and autumn, − 20% in summer) and earthworms (anecic—two Lumbricus terrestris, endogeic—four Allolobophora chlorotica and both together within 10 cm diameter tubes) on plant biomass and stoichiometry in two land-use types (intensively used meadow and conventional farming). We found little evidence for earthworm effects on aboveground biomass. However, future climate increased above- (+40.9%) and belowground biomass (+44.7%) of grass communities, which was mainly driven by production of the dominant Festulolium species during non-summer drought periods, but decreased the aboveground biomass (− 36.9%) of winter wheat. Projected climate change and earthworms interactively affected the N content and C:N ratio of grasses. Earthworms enhanced the N content (+1.2%) thereby decreasing the C:N ratio (− 4.1%) in grasses, but only under ambient climate conditions. The future climate treatment generally decreased the N content of grasses (aboveground: − 1.1%, belowground: − 0.15%) and winter wheat (− 0.14%), resulting in an increase in C:N ratio of grasses (aboveground: + 4.2%, belowground: +6.3%) and wheat (+5.9%). Our results suggest that climate change diminishes the positive effects of earthworms on plant nutrient uptakes due to soil water deficit, especially during summer drought.

Human-induced global environmental change is predicted to alter the stability and functioning of ecosystems worldwide. Most research in recent decades has focused on studying climate-change effects on aboveground systems, causing a poor understanding of belowground responses. However, gaining knowledge on environmental-change effects on soil biota is of crucial importance, as soil-ecosystem services are indispensable for human well-being and contribute fundamentally to the functioning of terrestrial ecosystems. Nematode communities play a central role in various soil ecosystem processes and are therefore commonly used as biological indicators to assess soil conditions and soil health. While causing overall shifts in community composition patterns, which are most often examined, climate change might also alter nematode population dynamics and the sex ratio (number of males per female). Previous studies on plant-parasitic nematode species suggest that changes to unfavorable environmental conditions trigger reduced development of females and favor sexual rather than parthenogenetic reproduction. Therefore, we are presenting the working hypothesis that predicted climate change causing reduced resource availability and enhanced environmental stress will lead to an increased proportion of males in soil nematode communities. Our systematic literature review revealed that climate- and environmental change effects on the sex ratio of free-living soil nematode populations are inconsistent, but heavily understudied. Data on sex ratios have been treated mostly as additional information, presented without any underlying theory and hypotheses, as well as limited discussion. In this perspective paper, we thus propose that future studies should include clear hypotheses and test if the sex ratio of free-living nematodes increases with climate change due to more stressful environmental conditions and low resource availability. The increasing production, use and emission of synthetic chemicals into the environment represents a major driver of global change. The large number of synthetic chemicals, limited knowledge on exposure patterns and effects in organisms and their interaction with other global change drivers hamper the prediction of effects in ecosystems. However, recent advances in biomolecular and computational methods are promising to improve our capacity for prediction. We delineate three idealised perspectives for the prediction of chemical effects: the suborganismal, organismal and ecological perspective, which are currently largely separated. Each of the outlined perspectives includes essential and complementary theories and tools for prediction but captures only part of the phenomenon of chemical effects. Links between the perspectives may foster predictive modelling of chemical effects in ecosystems and extrapolation between species. A major challenge for the linkage is the lack of data sets simultaneously covering different levels of biological organisation (here referred to as biological levels) as well as varying temporal and spatial scales. Synthesising the three perspectives, some central aspects and associated types of data seem particularly necessary to improve prediction. First, suborganism- and organism-level responses to chemicals need to be recorded and tested for relationships with chemical groups and organism traits. Second, metrics that are measurable at many biological levels, such as energy, need to be scrutinised for their potential to integrate across levels. Third, experimental data on the simultaneous response over multiple biological levels and spatiotemporal scales are required. These could be collected in nested and interconnected micro- and mesocosm experiments. Lastly, prioritisation of processes involved in the prediction framework needs to find a balance between simplification and capturing the essential complexity of a system. For example, in some

Affective disorders (AD) including major depressive disorder (MDD) and bipolar disorder (BD) are common mood disorders associated with increased disability and poor health outcomes. Altered immune responses characterized by increased serum levels of pro-inflammatory cytokines and neuroinflammation are common findings in patients with AD and in corresponding animal models. Dendritic cells (DCs) represent a heterogeneous population of myeloid cells that orchestrate innate and adaptive immune responses and self-tolerance. Upon sensing exogenous and endogenous danger signals, mature DCs secrete proinflammatory factors, acquire migratory and antigen presenting capacities and thus contribute to neuroinflammation in trauma, autoimmunity, and neurodegenerative diseases. However, little is known about the involvement of DCs in the pathogenesis of AD. In this review, we summarize the current knowledge on DCs in peripheral immune responses and neuroinflammation in MDD and BD. In addition, we consider the impact of DCs on neuroinflammation and behavior in animal models of AD. Finally, we will discuss therapeutic perspectives targeting DCs and their effector molecules in mood disorders.

Soil fauna plays a crucial role in plant litter decomposition. However, if and how increasing nitrogen (N) deposition and plant functional type jointly affect the contribution of soil mesofauna to litter decomposition remains poorly understood. A litterbag experiment with two meshed sizes (0.1 and 2 mm) was conducted to examine the effects of soil mesofauna on litter decomposition of three coexisting functional types of trees (evergreen broadleaf, deciduous broadleaf and coniferous) differing in litter identity (6 tree litter species for each plant functional type). Litterbags were exposed to three N addition levels (0, 20 and 40 kg N ha−1 yr−1) in a subtropical forest of southwest China. Overall, soil mesofauna significantly increased litter mass loss with the most profound effect in evergreen broadleaf litter. Further, the high N addition treatment stimulated the effects of soil mesofauna on litter decomposition, particularly in deciduous broadleaf litter. Soil mesofauna effects were positively correlated with initial leaf litter chemical characteristics, such as carbon and cellulose concentration and C/P ratio. The N effect was greater in low N than high N treatments in the 0.1 mm litterbags, but was similar in the two N addition levels in the 2 mm litterbags. Our results indicate that soil mesofauna effects on litter decomposition at low N addition are higher in evergreen broadleaf than in deciduous broadleaf and coniferous litter, and generally increase with N addition levels, particularly in deciduous litter species. The findings underscore the importance of soil mesofauna and plant functional types for litter decomposition in subtropical forests where N deposition is substantially increasing.

Although nitrogen (N) deposition is increasing globally, N availability still limits many organisms, such as microorganisms and mesofauna. However, little is known to which extent soil organisms rely on mineral‐derived N and whether plant community composition modifies its incorporation into soil food webs. More diverse plant communities more effectively compete with microorganisms for mineral N likely reducing the incorporation of mineral‐derived N into soil food webs. We set up a field experiment in experimental grasslands with different levels of plant species and functional group richness. We labeled soil with 15NH415NO3 and analyzed the incorporation of mineral‐derived 15N into soil microorganisms and mesofauna over 3 months. Mineral‐derived N incorporation decreased over time in all investigated organisms. Plant species richness and presence of legumes reduced the uptake of mineral‐derived N into microorganisms. In parallel, the incorporation of mineral‐derived 15N into mesofauna species declined with time and decreased with increasing plant species richness in the secondary decomposer springtail Ceratophysella sp. Effects of both plant species richness and functional group richness on other mesofauna species varied with time. The presence of grasses increased the 15N incorporation into Ceratophysella sp., but decreased it in the primary decomposer oribatid mite Tectocepheus velatus sarekensis. The results highlight that mineral N is quickly channeled into soil animal food webs via microorganisms irrespective of plant diversity. The amount of mineral‐derived N incorporated into soil animals, and the plant community properties affecting this incorporation, differed markedly between soil animal taxa, reflecting species‐specific use of food resources. Endogenous rhythmic growth (ERG) is displayed by many tropical and some major temperate tree species and characterized by alternating root and shoot flushes (RF and SF). These flushes occur parallel to changes in biomass partitioning and in allocation of recently assimilated carbon and nitrogen. To address how biotic interactions interplay with ERG, we cross-compared the RF/SF shifts in oak microcuttings in the presence of pathogens, consumers and a mycorrhiza helper bacterium, without and with an ectomycorrhizal fungus (EMF), and present a synthesis of the observations. The typical increase in carbon allocation to sink leaves during SF did not occur in the presence of root or leaf pathogens, and the increase in nitrogen allocation to lateral roots during RF did not occur with the pathogens. The RF/SF shifts in resource allocation were mostly restored upon additional interaction with the EMF. Its presence led to increased resource allocation to principal roots during RF, also when the oaks were inoculated additionally with other interactors. The interactors affected the alternating, rhythmic growth and resource allocation shifts between shoots and roots. The restoring role of the EMF on RF/SF changes in parallel to the corresponding enhanced carbon and nitrogen allocation to sink tissues suggests that the EMF is supporting plants in maintaining the ERG.

Contact hypersensitivity (CHS) is an established animal model for allergic contact dermatitis. Dendritic cells (DCs) play an important role in the sensitization phase of CHS by initiating T cell responses to topically applied haptens. The cannabinoid receptors 1 (CB1) and 2 (CB2) modulate DC functions and inflammatory skin responses, but their influence on the capacity of haptenized DCs to induce CHS is still unknown. We found lower CHS responses to 2,4-dinitro-1-fluorobenzene (DNFB) in wild type (WT) mice after adoptive transfer of haptenized Cnr2-/- and Cnr1-/-/Cnr2-/- bone marrow (BM) DCs as compared to transfer of WT DCs. In contrast, induction of CHS was not affected in WT recipients after transfer of Cnr1-/- DCs. In vitro stimulated Cnr2-/- DCs showed lower CCR7 and CXCR4 expression when compared to WT cells, while in vitro migration towards the chemokine ligands was not affected by CB2. Upregulation of MHC class II and co-stimulatory molecules was also reduced in Cnr2-/- DCs. This study demonstrates that CB2 modulates the maturation phenotype of DCs but not their chemotactic capacities in vitro. These findings and the fact that CHS responses mediated by Cnr2-/- DCs are reduced suggest that CB2 is a promising target for the treatment of inflammatory skin conditions.

Plant diversity is an important driver of belowground ecosystem functions, such as root growth, soil organic matter (SOM) storage, and microbial metabolism, mainly by influencing the interactions between plant roots and soil. Dissolved organic matter (DOM), as the most mobile form of SOM, plays a crucial role for a multitude of soil processes that are central for ecosystem functioning. Thus, DOM is likely to be an important mediator of plant diversity effects on soil processes. However, the relationships between plant diversity and DOM have not been studied so far.We investigated the mechanisms underlying plant diversity effects on concentrations of DOM using continuous soil water sampling across 6 years and 62 plant communities in a long‐term grassland biodiversity experiment in Jena, Germany. Furthermore, we investigated plant diversity effects on the molecular properties of DOM in a subset of the samples.Although DOM concentrations were highly variable over the course of the year with highest concentrations in summer and autumn, we found that DOM concentrations consistently increased with plant diversity across seasons. The positive plant diversity effect on DOM concentrations was mainly mediated by increased microbial activity and newly sequestered carbon in topsoil. However, the effect of soil microbial activity on DOM concentrations differed between seasons, indicating DOM consumption in winter and spring, and DOM production in summer and autumn. Furthermore, we found increased contents of small and easily decomposable DOM molecules reaching deeper soil layers with high plant diversity.Synthesis. Our findings suggest that plant diversity enhances the continuous downward transport of DOM in multiple ways. On the one hand, higher plant diversity results in higher DOM concentrations, on the other hand, this DOM is less degraded. Earth is home to over 350,000 vascular plant species that differ in their traits in innumerable ways. A key challenge is to predict how natural or anthropogenically driven changes in the identity, abundance and diversity of co-occurring plant species drive important ecosystem-level properties such as biomass production or carbon storage. Here, we analyse the extent to which 42 different ecosystem properties can be predicted by 41 plant traits in 78 experimentally manipulated grassland plots over 10 years. Despite the unprecedented number of traits analysed, the average percentage of variation in ecosystem properties jointly explained was only moderate (32.6%) within individual years, and even much lower (12.7%) across years. Most other studies linking ecosystem properties to plant traits analysed no more than six traits and, when including only six traits in our analysis, the average percentage of variation explained in across-year levels of ecosystem properties dropped to 4.8%. Furthermore, we found on average only 12.2% overlap in significant predictors among ecosystem properties, indicating that a small set of key traits able to explain multiple ecosystem properties does not exist. Our results therefore suggest that there are specific limits to the extent to which traits per se can predict the long-term functional consequences of biodiversity change, so that data on additional drivers, such as interacting abiotic factors, may be required to improve predictions of ecosystem property levels.

A large body of research shows that biodiversity loss can reduce ecosystem functioning. However, much of the evidence for this relationship is drawn from biodiversity–ecosystem functioning experiments in which biodiversity loss is simulated by randomly assembling communities of varying species diversity, and ecosystem functions are measured. This random assembly has led some ecologists to question the relevance of biodiversity experiments to real-world ecosystems, where community assembly or disassembly may be non-random and influenced by external drivers, such as climate, soil conditions or land use. Here, we compare data from real-world grassland plant communities with data from two of the largest and longest-running grassland biodiversity experiments (the Jena Experiment in Germany and BioDIV in the United States) in terms of their taxonomic, functional and phylogenetic diversity and functional-trait composition. We found that plant communities of biodiversity experiments cover almost all of the multivariate variation of the real-world communities, while also containing community types that are not currently observed in the real world. Moreover, they have greater variance in their compositional features than their real-world counterparts. We then re-analysed a subset of experimental data that included only ecologically realistic communities (that is, those comparable to real-world communities). For 10 out of 12 biodiversity–ecosystem functioning relationships, biodiversity effects did not differ significantly between the full dataset of biodiversity experiments and the ecologically realistic subset of experimental communities. Although we do not provide direct evidence for strong or consistent biodiversity–ecosystem functioning relationships in real-world communities, our results demonstrate that the results of biodiversity experiments are largely insensitive to the exclusion of unrealistic communities and that the conclusions drawn from biodiversity experiments
