Weltweit stellt die künstliche Besamung eine der wichtigsten Reproduktionstechnologien dar und ist aus der modernen Rinderzucht nicht mehr wegzudenken. Bis heute hat sich die tiefe zervikale oder intrauterine Besamung als Standardbesamungsmethode durchgesetzt. Entgegen dem Trend der stetigen Reduktion des Besamungsvolumens und basierend auf der Frage, wie man aus einer Besamungsportion die meisten Kälber erzeugen kann, wurde in der jüngeren Vergangenheit eine high-volume-Besamungsmethode (HVB) entwickelt. Bei dieser wird das aufgetaute Sperma vor der Besamung mit bis zu 30 ml eines spermienfreundlichen Mediums verdünnt. Auf Grund des Aufbaus der Besamungsapparatur bietet die HVB daher auch die Möglichkeit, Spermaportionen unter hygienischen Bedingungen auf mehrere Tiere aufzuteilen. Ziel der Studie war es einerseits, den Einfluss vier verschiedener Puffermedien [BO-IVFTM (BIVF), BO-SemenPrepTM (BSPREP), EasyBuffer B (EBB), mo-difiziertes Fert-TALP (mFert-TALP)] auf Qualitätsparameter boviner Spermien zu untersuchen, um Rückschlüsse auf ihre potenzielle Eignung als Trägermedium für die HVB zu ziehen. Andererseits wurde im praktischen Teil die HVB bei routinemäßigen Besamungen angewendet und mit der Standardbesamung in Bezug auf das Besamungsergebnis verglichen. Während beim Anteil an Spermien mit intakter Plasmamembran kein signifikanter Unterschied zwischen den Puffermedien bestand (BIVF–BSPREP p=0,549, BIVF–EBB p=0,960, BIVF–mFert-TALP p=0,990, BSPREP–EBB p=0,267, BSPREP–mFert- TALP p=0,738, EBB–mFert-TALP p=0,854), wurde der Anteil an progressiv motilen Spermien (Zeitpunkt 0 h: BIVF–BSPREP p=0,207, BIVF–EBB p=0,559, BIVF–mFert-TALP p=0,968, BSPREP–EBB p=0,007, BSPREP–mFert-TALP p=0,076, EBB–mFert-TALP p=0,832; Zeitpunkt 1 h: BIVF–BSPREP p, Introduction: Due to its simplicity, economy and efficiency, artificial insemination (AI) has become one of the most important reproductive technologies worldwide and deep cervical or intrauterine insemination is now the standard insemination (SI) method. We tested a high-volume-insemination (HVI) method, where semen is diluted with up to 30 ml of medium before insemination, and compared the pregnancy rates with those attained with the SI method. Material and methods: In laboratory tests, we used computer-assisted sperm analysis and flow cytometry to examine four buffer media [BO-IVFTM (BIVF), BO-SemenPrepTM (BSRPEP), EasyBuffer B (EBB), modified Fert-TALP (mFert-TALP)] for sperm compatibility and potential suitability as a carrier medium for HVI using bovine frozen-thawed ejaculates (n=10). We then undertook routine inseminations (SI n=100, HVI n=100) during natural oestrus and compared the conception rates of the two insemination methods. Pregnancy status was diagnosed by transrectal palpation starting 35 days after AI. We used modified Fert-TALP as carrier medium for HVI. Results: The laboratory tests revealed several differences in semen quality depending on the buffer media. Deficiencies in the trace elements selenium and copper have been shown to negatively affect reproductive performance in cattle. Selenium has also been shown to play a role in inflammatory regulation. Selenium impacts both the production of prostaglandins and the anti-inflammatory response, particularly through the activity of glutathione peroxidase. There is some evidence that selenium deficiency reduces embryo survival during implantation, which takes place in the fourth week after estrus. Primary copper deficiency results from insufficient levels of copper in the diet, while secondary deficiencies are attributed to the reduction in copper absorption or utilization by the antagonistic effects of molybdenum, sulfur or iron. Secondary copper deficiency caused by molybdenum interference retards embryo development and leads to early embryonic death.
