The growth and development of children aged 7-10 years takes place under conditions of continuous learning, and the level of fatigue increases by the end of the school year. Environmental factors of the Kola Peninsula play a special role here. the process of adaptation of the organism takes place in all age and gender groups. The most obvious is the reactivity of the cardiovascular and sympathetic-adrenal systems to physical load, which enhances the correlation links between indicators of the two active systems. It happens in the following way: by the end of the academic year, in all age and gender groups before the polar day, a proven shift of the observed indicators is recorded. The results obtained provide a comprehensive picture of age-related adaptation changes and increase in the level of fatigue in the arctic climate conditions of the Kola Peninsula. Examinations of children aged 7-10 years showed that age dynamics of cardiovascular and sympathoadrenal systems indicators is non-linear and significantly depends on specific parts of the academic year. This may be due to the periodicity of the polar night, namely, as the functional capabilities of particular systems tend to increase with age, certain indicators have a wave-like character of changes.

BACKGROUND: In 2014, germline signal transducer and activator of transcription (STAT) 3 gain-of-function (GOF) mutations were first described to cause a novel multisystem disease of early-onset lymphoproliferation and autoimmunity. OBJECTIVE: This pivotal cohort study defines the scope, natural history, treatment, and overall survival of a large global cohort of patients with pathogenic STAT3 GOF variants. METHODS: We identified 191 patients from 33 countries with 72 unique mutations. Inclusion criteria included symptoms of immune dysregulation and a biochemically confirmed germline heterozygous GOF variant in STAT3. RESULTS: Overall survival was 88%, median age at onset of symptoms was 2.3 years, and median age at diagnosis was 12 years. Immune dysregulatory features were present in all patients: lymphoproliferation was the most common manifestation (73%); increased frequencies of double-negative (CD4-CD8-) T cells were found in 83% of patients tested. Autoimmune cytopenias were the second most common clinical manifestation (67%), followed by growth delay, enteropathy, skin disease, pulmonary disease, endocrinopathy, arthritis, autoimmune hepatitis, neurologic disease, vasculopathy, renal disease, and malignancy. Infections were reported in 72% of the cohort. A cellular and humoral immunodeficiency was observed in 37% and 51% of patients, respectively. Clinical symptoms dramatically improved in patients treated with JAK inhibitors, while a variety of other immunomodulatory treatment modalities were less efficacious. Thus far, 23 patients have undergone bone marrow transplantation, with a 62% survival rate. CONCLUSION: STAT3 GOF patients present with a wide array of immune-mediated disease including lymphoproliferation, autoimmune cytopenias, and multisystem autoimmunity. Patient care tends to be siloed, without a clear treatment strategy. Thus, early identification and prompt treatment implementation are lifesaving for STAT3 GOF syndrome.

Introduction. Cultural heritage: monuments, ensembles, buildings and structures, archaeological sites define the urban identity of a settlement, reflect the historical development path of the people and are one of the main priorities in urban development. Urban risks are considered as the possibility of something happening that has a negative impact on the sites, as well as any possible loss that affects the value of cultural heritage, which can be divided into several categories.Materials and methods. From the point of view of the Sets Theory, the description of changes in the totality of tangible and intangible cultural heritage, occurring as a result of natural and anthropogenic factors, which allows to determine the main risks of its loss, has been proposed. The suggested mathematical model serves as the basis for the development of information tools for identification, assessment and consideration of historical and cultural heritage in urban planning.Results. Based on a retrospective analysis of the historical development of Homs, a classification of cultural heritage sites has been proposed, and a diagram of the transformation of the collectivity of objects of tangible and intangible cultural heritage as a result of natural and human factors of the risk of their loss has been created.Conclusions. It was found that the highest risks of loss of immovable objects of cultural heritage include natural catastro­phic processes and phenomena (earthquakes), and military conflicts. As a result of which, in particular in the city of Homs, a new local architectural style “Homsi” was formed to replace the Byzantine culture.

We document that stocks that experience sudden increases in idiosyncratic volatility underperform otherwise similar stocks in the future, and we propose that this phenomenon can be explained by the Miller conjecture [Miller E (1977) Risk, uncertainty, and divergence of opinion. J. Finance 32(4):1151-1168]. We show that volatility shocks can be traced to unusual firm-level news flow, which temporarily increases the level of investor disagreement about the firm value. At the same time, volatility shocks pose a barrier to short selling, preventing pessimistic investors from expressing their views. In the presence of divergent opinions and short-selling constraints, prices initially reflect optimistic views but adjust downward in the future as investors' opinions converge.

The occurrence of atrial fibrillation (AF), one of the most socially significant arrhythmias, is associated with the presence of areas of fibrosis. Fibrosis introduces conduction heterogeneity into the cardiac tissue and, thus, may be a substrate for spiral wave reentry, which provokes the onset of AF and is often associated with its persistence. Despite results from computer and animal models of cardiac tissues, data on the conditions under which microreentries occur in human tissues are limited. In this work, we conducted a study of the new approach to modeling the fibrous atrial tissue, which takes into account the cellular structure and conduction in fibrosis areas. Using the Potts model, we created a realistic texture of atrial tissues remodeled by fibroblasts and showed the presence of pathways in such a system with a low proportion of fibroblasts. Our study revealed the relationship between the shape of the cells' action potential, their location in the tissue, and the direction of the wave propagation. The wavefront obtained in the model creates a dynamic heterogeneity of the tissue, which affects the migration and pinning of spiral waves, and explains the formation of microreentries in the cardiac tissue. In the future, such a model can become a potential tool for predictive modeling of AF and the search for ablation target identification. [ABSTRACT FROM AUTHOR]

Induced pluripotent stem cells (iPSCs) constitute a potential source of patient-specific human cardiomyocytes for a cardiac cell replacement therapy via intramyocardial injections, providing a major benefit over other cell sources in terms of immune rejection. However, intramyocardial injection of the cardiomyocytes has substantial challenges related to cell survival and electrophysiological coupling with recipient tissue. Current methods of manipulating cell suspensions do not allow one to control the processes of adhesion of injected cells to the tissue and electrophysiological coupling with surrounding cells. In this article, we documented the possibility of influencing these processes using polymer kernels: biocompatible fiber fragments of subcellular size that can be adsorbed to a cell, thereby creating the minimum necessary adhesion foci to shape the cell and provide support for the organization of the cytoskeleton and the contractile apparatus prior to adhesion to the recipient tissue. Using optical excitation markers, the restoration of the excitability of cardiomyocytes in suspension upon adsorption of polymer kernels was shown. It increased the likelihood of the formation of a stable electrophysiological coupling in vitro. The obtained results may be considered as a proof of concept that the stochastic engraftment process of injected suspension cells can be controlled by smart biomaterials.

Based on international experience, the competitiveness of the container on barge system can be ensured by applying the different forms of cooperation between the business-players. In the case of horizontal cooperation, it’s raising the problem of distribution of structural elements of the container on barge system during the container-flows adoption. So, the paper proposed the mathematical decision to maximise the use of the linear ship's characteristics by the barge’s loading capacity and by the tug’s engine power.

На сьогоднішній день ринок абразивних армованих кругівпредставлений широкою номенклатурою та великою кількістю виробників. Круги масово застосовуються при виконанні будівельно-монтажних робіт для різання металевої продукції. Різання високоабразивних вогнетривів та штучних і природніх гірських порід міцністю до 60 МПа на будівельному майданчику виконується переважно машинами з дисковим алмазним робочим органом. Отже, дослідження механізму роботи абразивного армованого круга при різанні кам’яних матеріалів в залежності від умов його роботи з обезпиленням водою є актуальним питанням. В ході досліджень встановлено, що абразивний армований круг в процесі різання без охолодження має термомеханічне зношування. Це підтверджується вирішенням диференційного рівняння та визначенням глибини проникнення температури в круг. Встановлено необхідність уведення в полімерну матрицю круга модифікаторів, які дозволять підвищити теплостійкість круга і, відповідно, зменшити його термомеханічне зношування. Підвищення робочої температури руйнування зв’язки круга дозволяє абразивним зернам довше утримуватися в матриці круга. Обезпилення водою дозволяє не тільки прибрати із робочої зони пил, а й додатково охолодити круг, що також збільшує його ресурс. Розроблена технологія виготовлення абразивних армованих кругів, де як модифікатор використовується водний розчин полівінілового спирту у кількості 13,7%. Подальші експериментальні дослідження виготовлених кругів при різанні високоабрази вних вогнетривів та штучних і природних гірських порід міцністю до 60 МПа підтвердили правильність отриманих результатів досліджень. Ресурс круга зріс за рахунок покращення самозаточування. Зменшилася потужність привідного двигуна, що дозволило зменшити масу машини. Екологічні умови оператора поліпшилися за рахунок зменшення пилу.
