Het Louis Bolk Instituut is door het Wereld Natuur Fonds Nederland (WWF-NL) gevraagd om de potentie van peulvruchten voor natuurpositieve productie en consumptie te verkennen. Voor het WWF-NL is het behoud en herstel van biodiversiteit een belangrijk thema. De landbouw heeft wereldwijd een groot effect op biodiversiteitsverlies. Ook in Nederland speelt dit en het WWF-NL werkt hier op verschillende fronten aan om de impact van de landbouw om te buigen van een negatieve impact naar een positieve. Iedere consument speelt daarin een rol en heeft middels het aankoopgedrag invloed op de biodiversiteit. De eiwittransitie (minder vlees, meer plantaardig) is hierin een belangrijk onderdeel om de impact van de voedselconsumptie op de natuur te verminderen. Dit rapport dient als kennisbron waarmee WWF-NL middels haar lobby de juiste dingen kan doen.

Dit overzicht brengt de resultaten uit het onderzoeksprogramma Slim Landgebruik samen. Het doel van Slim Landgebruik is het leveren van kennis die nodig is om te komen tot een jaarlijkse vastlegging van 0,5 Mton CO2-equivalenten vanaf 2030 voor minerale landbouwbodems in Nederland, waarbij tevens een bijdrage wordt geleverd aan het doel om vanaf 2030 alle landbouwbodems in Nederland duurzaam te beheren. In Slim Landgebruik wordt onderzoek gedaan naar het effect van maatregelen op koolstofvastlegging, lachgasemissie, bodemfuncties en kosten en baten. In deze samenvatting lees je wat het effect is van de maatregelen op deze indicatoren (Tabel 1 & 2). De maatregelen worden kort toegelicht en de resultaten geven een gemiddeld beeld van het verwachtte effect in Nederland. De resultaten komen voort uit modelstudies, lange termijn experimenten (LTE’s), economische analyses en expertkennis. Dit overzicht kan worden gebruikt door beleidsmakers, maar is ook geschikt voor mensen die graag meer inzicht willen verkrijgen in het effect van klimaatmaatregelen.

De ecologische waterkwaliteit wordt voor de Europese Kaderrichtlijn Water (KRW) gemonitord in oppervlaktewateren die in het beheer zijn van de waterschappen en het Rijk maar niet in agrarische bedrijfssloten. Hoe verhouden de totale concentraties stikstof (N) en fosfor (P) in die sloten zich tot die in de regionale oppervlaktewateren? Verschilt dit per grondsoortregio? En verschilt de verhouding van de componenten die samen totaal N en P vormen? Nieuwe (zomer)metingen in de sloten maken vergelijking mogelijk.

Duurzaam beheer van landbouwbodems kan bijdragen aan het behalen van beleidsdoelen voor bodem-, water- en luchtkwaliteit. Toch richten huidige beleidskaders in relatie tot de bodem zich hoofdzakelijk op emissiebeperking naar lucht en water. Wij onderzochten voor welke indicatoren van bodemfuncties en emissies beleidskaders elkaar versterken en voor welke niet.

Het energielandschap is fundamenteel aan het veranderen. Alternatieve, klimaat neutrale, energiebronnen zullen belangrijker worden en het gebruik van aardgas zal afnemen. Naast restwarmte, geothermie, PV en windenergie, is hernieuwbaar gas één van de energiebronnen die een plek zullen krijgen in het toekomstige energiesysteem. Hoe het hernieuwbaar gas zal worden ingezet, is met name afhankelijk van de situaties waarin er weinig alternatieven voor gasverbruik zijn. Gas dat wordt opgewerkt tot groen gas, gelijk aan aardgaskwaliteit, is een aantrekkelijke route omdat het kan worden ingezet via bestaande infrastructuren en installaties van gebruikers. Een rol voor groen gas lijkt weggelegd voor hybride warmtevoorzieningen, het verduurzamen van industriële processen, verwarming van oudere gebouwen, transport en mobiliteit.

PTP-BL is a cytosolic multidomain protein tyrosine phosphatase that shares homologies with several submembranous and tumor suppressor proteins. Here we show, by transient expression of modular protein domains of PTP-BL in epithelial MDCK cells, that the presence of a FERM domain in the protein is both necessary and sufficient for its targeting to the apical side of epithelial cells. Furthermore, immuno-electron microscopy on stable expressing MDCK pools, that were obtained using an EGFP-based cell sorting protocol, revealed that FERM domain containing fusion proteins are enriched in microvilli and have a typical submembranous location at about 10-15 nm from the plasma membrane. Immunofluorescence microscopy suggested colocalization of the FERM domain moiety with the membrane-cytoskeleton linker ezrin. However, at the electron microscopy level this colocalization cannot be confirmed nor can we detect a direct interaction by immunoprecipitation assays. Fluorescence recovery after photobleaching (FRAP) experiments show that PTP-BL confinement is based on a dynamic steady state and that complete redistribution of the protein may occur within 20 minutes. Our observations suggest that relocation is mediated via a cytosolic pool, rather than by lateral movement. Finally, we show that PTP-BL phosphatase domains are involved in homotypic interactions, as demonstrated by yeast two-hybrid assays. We report on the physical ordering of genes in a relatively small area of chromosome 19, segment q13, containing the locus for myotonic dystrophy (DM), the most frequent heritable muscular dystrophy of adulthood in man. DNAs from somatic cell hybrids with der 19q products that carry a breakpoint across the muscle-specific creatine kinase (CKMM) gene were analyzed by Southern blotting using probes for CKMM, APOC2, and the repair genes ERCC1 and ERCC2. Results were combined with data from CHEF and field inversion-gel-electrophoresis separation of large-sized DNA restriction fragments to establish a map localizing both DNA-repair genes and the CKMM gene within the same 250 kb of DNA, the order being cen-CKMM-ERCC2-ERCC1-ter, with APOC2 being at more than 260 kb proximal to CKMM. Transcriptional start sites of the CKMM and DNA-repair genes are all on the telomeric side of the genes. Our results provide a framework for the construction of a larger physical map of the area, which will facilitate the search for the DM gene.
