Der Einsatz textiler Carbonbewehrungen im Betonbau etabliert sich, aufgrund ihrer hervorragenden zug- und korrosionsmechanischen Eigenschaften, zunehmend als technische und wirtschaftliche Alternative zur Stahlbewehrung. Sie ermöglichen je nach Anwendung, eine signifikante Reduzierung des Betonmaterialeinsatzes um bis zu 70 % sowie der damit verbundenen CO2-Emissionen. Um das Leistungspo- tenzial von textilbewehrtem Beton voll auszuschöpfen, ist eine effiziente Kraftüber- tragung zwischen Textilbewehrung und Betonmatrix entscheidend. Je nach Bean- spruchung sind bisher materialineffiziente, unverhältnismäßig große Verankerungs- längen erforderlich. Gefordert werden daher Bewehrungsstrukturen mit ausgepräg- ter Oberflächenprofilierung analog zu geripptem Betonstahl. Konventionelle, sub- traktive und additive Profilierungsverfahren erzeugen jedoch einen unterbrochenen Faserverlauf, wodurch das hohe zugmechanische Leistungspotenzial der anisotro- pen Carbonfasern nicht effizient ausgenutzt werden kann. Ziel dieser Arbeit ist da- her die Entwicklung neuartiger profilierter Textilbetonbewehrungen mit material- und verbundgerechter Profilierung auf Garnebene und entsprechender Fertigungstech- nologien, auf Basis der Flecht- und Tränkumformtechnik. Dies umfasst die Verfah- rens- und Maschinenentwicklung sowie die Modellierung profilierter Bewehrungs- strukturen zur anforderungsgerechten Auslegung und deren Technologienachweis.

Adoptive T cell therapy (ACT) is a promising immunotherapeutic approach to fight cancer by utilising cytotoxic T lymphocytes (CTL), which specifically target and eradicate tumour cells. However, one major limitation of this therapy is the ability of tumours to interfere with the CTL through immune escape mechanisms. In a mouse model of B-cell lymphoma and ACT, we investigate the mechanisms underlying this failure. Our laboratory previously found that tumour antigen-specific CTL fail to eradicate lymphoma cells once the tumour burden reaches a certain threshold. In this case a major proportion of the CTL is lost while those remaining lose their effector function, a process we call “stunning”. Our tumour model enables us to investigate the underlying mechanisms of stunning by comparing characteristics of CTL and tumour cells in a small tumour setting, where CTL successfully eradicate tumour cells with a large tumour setting, where stunning occurs. In Chapter 3, we describe findings that suggest that the number of tumour cells presenting a CTL’s specific antigen rather than the total tumour burden determines ACT outcome in our murine lymphoma model. Once the number of antigen+ tumour target cells reaches a critical threshold, all CTL are rendered dysfunctional. We therefore hypothesize that a large heterogenous tumour may be more efficiently killed by using a number of CTL specific for several different antigens simultaneously, where each antigen is only expressed on a subpopulation of tumour cells to prevent CTL dysfunction due to high target cell density. We further showed that increasing numbers of antigen+ tumour cells at the time of adoptive transfer induce the rapid expansion of an Interleukin (IL)-18-receptor-alpha (IL18Ra) and Neuropilin-1 (NRP-1) double positive CTL population, with reduced cytotoxicity. The higher the number of antigen+ tumour cells encountered by the CTL upon transfer, the lower the cytotoxicity, cytokine production and T cell receptor (TCR)

With the aim of optimizing the interfacial adhesion of basalt fibres with thermoset matrices, in this research the plasma polymerization technique (PECVD) was used to synthesize polymeric coatings based on tetravinylsilane (TVS) or its mixtures with oxygen on the surface of basalt fibres. The successful deposition of the polymer sizing was confirmed by XPS analysis, which highlighted the increase in the intensity of the carbon and oxygen peaks. To evaluate the influence of polymer sizing on interfacial adhesion, basalt fibre/polyester resin composites were tested through the short beam shear (SBS) test. Compared to neat basalt fibres, the modified fibres showed a significant increase in the interlaminar shear strength (ILSS) higher than 180%. These results compared quite favourably with those on glass fibres used as baseline, with higher ILSS values as a function of oxygen content. The improvement in interfacial adhesion was correlated with the increase in basalt fibre surface energy by single fibre dynamic contact angle tests.

Short fibres of different materials are used for crack bridging in strain-hardening cement-based composites (SHCC). Their mechanical properties and the fibre-matrix interphase on the micro level have a significant influence on the macroscopic component properties. Investigations on the specific modification and adaptation of fibre properties in relation with the failure mechanisms at different strain rates hardly exist so far, since mainly commercially available fibres are used. In the frame of this work, two different fibre types – polypropylene (PP) fibres and alkali-resistant (AR) glass fibres – were produced on lab spinning devices and the properties were adapted in such a way that fundamental correlations between the influence of fibre geometry, mechanical properties, chemical functionalities and surface structure on the behaviour during fibre pull-out from the concrete matrix can be derived. The PP fibres were produced with different degrees of stretching, cross-sectional geometries (circular, trilobal) and fibre diameters, as well as without and with sizing. The resulting changes in the crystallinity of the PP structure, surface roughness and wetting behaviour could be demonstrated by differential scanning calorimetry (DSC), roughness measurements by atomic force microscopy (AFM), and contact angle measurements. AR glass fibres were used in the unsized state and various chemical surface treatments were applied. Aqueous polymer dispersions of different materials were characterized in detail regarding particle size, pH value, solid content, and surface tension. In addition, their film-forming properties were evaluated using prepared polymer films. Furthermore, the influence of cross-linking agents on the thermal and mechanical stability of polyurethane sizings was investigated using thermal analysis methods. After application of the sizings to the AR glass surface, changes in surface structure and roughness could be observed by scanning electron microscopy (SEM, Für die Rissüberbrückung in hochduktilen Betonen (Strain-Hardening Cement-based Composites – SHCC) werden Kurzfasern verschiedener Materialien eingesetzt. Ihre mechanischen Eigenschaften und die Faser-Matrix-Grenzschicht auf der Mikroebene beeinflussen die makroskopischen Bauteileigenschaften deutlich. Untersuchungen zur gezielten Veränderung und Anpassung von Fasereigenschaften im Zusammenhang mit den Versagensmechanismen bei unterschiedlichen Dehnraten existieren bisher kaum, da überwiegend kommerziell verfügbare Fasern eingesetzt werden. Im Rahmen dieser Arbeit wurden daher zwei verschiedene Fasertypen – Polypropylen (PP)-Fasern und alkaliresistente (AR)-Glasfasern – an Laborspinnanlagen selbst hergestellt und die Eigenschaften so angepasst, dass sich grundlegende Zusammenhänge zwischen dem Einfluss von Fasergeometrie, mechanischen Eigenschaften, chemischen Funktionalitäten und Oberflächenstruktur auf das Verhalten bei Faserauszug aus der Betonmatrix ableiten lassen. Die PP-Fasern wurden mit verschiedenen Verstreckungsgraden, Querschnittsgeometrien (rund, trilobal), Faserdurchmessern sowie ohne und mit Schlichte hergestellt. Die dadurch hervorgerufenen Eigenschaftsveränderungen hinsichtlich Kristallinität der PP-Struktur, der Oberflächenrauheit und des Benetzungsverhaltens konnten durch dynamische Differenzkalorimetrie (DSC), Rauheitsmessungen mittels Rasterkraftmikroskopie (AFM) und Kontaktwinkelmessungen nachgewiesen werden. AR-Glasfasern wurden im ungeschlichteten Zustand verwendet und verschiedene chemische Oberflächenbehandlungen durchgeführt. Es wurden wässrige Polymerdispersionen verschiedener Materialien detailliert hinsichtlich ihrer Partikelgröße, pH-Wert, Feststoffgehalt und Oberflächenspannung charakterisiert sowie ihre Filmbildungseigenschaften anhand hergestellter Polymerfilme bewertet. Weiterhin wurde der Einfluss von Vernetzern auf die thermische und mechanische Stabilität von Polyurethanschlichten mit Methoden der thermischen Analyse untersucht.

Hochduktile Betone (Engl.: Strain-Hardening Cement-based Composites – SHCC) und Textilbetone (engl.: Textile Reinforced Concrete – TRC) sind zwei neuartige Faserbetone, die ein duktiles und dehnungsverfestigendes Zugverhalten aufweisen. SHCC bestehen aus feinkörnigen Zementmatrizen und kurzen Hochleistungspolymerfasern, während TRC eine Kombination aus feinkörnigen Zementmatrizen und kontinuierlichen zwei- oder dreidimensionalen Textilschichten darstellt. Letztere bestehen aus Multifilamentgarnen aus Kohlenstoff, alkalibeständigem Glas oder Polymerfasern. Die hohe elastische Verformbarkeit beider Verbundwerkstoffe bis zum Erreichen der Zugfestigkeit entsteht aus der sukzessiven Bildung multipler feiner Risse. Neben der hervorragenden Risskontrolle und Duktilität weisen diese Verbundwerkstoffe ein hohes Energieabsorptionsvermögen auf, was in Bezug auf kurzzeitdynamische Belastungen eine durchaus erstrebenswerte Eigenschaft darstellt. Obwohl SHCC eine höhere Dehnungskapazität als herkömmliche TRC zeigen, weisen die Textilbetone eine erheblich höhere Zugfestigkeit auf. Darüber hinaus besitzen die textilbewehrten Betone deutlich niedrigere Einflüsse von Anwendungstechnologie und Maßstab auf das Zugverhalten, d. h. eine bessere Robustheit. Daher stellt die Kombination dieser beiden Bewehrungskonzepte einen vielversprechenden Ansatz dar. Während die Kurzfasern für eine bessere Risskontrolle und Erstrissfestigkeit sorgen, sichern die Textilgelege eine hohe Zugfestigkeit sowie Steifigkeit im gerissenen Zustand und eine gleichmäßige Verteilung der Kräfte in der Verstärkungsschicht bzw. im Bauteil. Dieser synergetische Effekt kann jedoch nur durch eine zielgerichtete Materialentwicklung erreicht werden, die eine grundlegende Materialcharakterisierung unter verschiedenen Belastungsszenarien erfordert. Im Rahmen des DFG-finanzierten Graduiertenkollegs GRK 2250 „Impaktsicherheit von Baukonstruktionen durch mineralisch gebundene Komposite“ werden duktile und Impakt resistente Kom, Strain-hardening cement-based composites (SHCC) and textile-reinforced concrete (TRC) are two novel types of fiber-reinforced cementitious composites that exhibit ductile, strain-hardening tensile behavior. SHCC comprises fine-grained cementitious matrices and short, high-performance polymer fiber, while TRC is a combination of a fine-grained, cementitious matrix and continuous two- or three- dimensional textile layers of multi-filament yarns, usually made of carbon or alkali-resistant glass. Both composites yield high inelastic deformations in a strain-hardening phase due to the successive formation of multiple fine cracks. Such cracking behavior stands for high energy absorption of the composites when exposed to extreme loading, without abrupt loss of load-bearing capacity. In comparative terms, SHCC shows superior strain capacity, while TRC yields considerably higher tensile strength. The addition of short fibers strengthens the matrix by efficiently restraining the micro-cracks’ growth and reducing spallation, while the textile reinforcement ensures a secure confinement of the reinforced concrete element (substrate to be strengthened), as well as a favorable stress distribution. The combination of SHCC and textile reinforcement is expected to deliver high tensile strength and stiffness in the strain-hardening stage along with pronounced multiple cracking. In order to achieve a favorable synergetic effect, a purposeful material design is required based on a clear understanding of the mechanical interactions in the composites. In the framework of the DFG Research Training Group GRK 2250, which aims at enhancing structural impact safety through thin strengthening layers made of high-performance mineral-based composites, this work focuses on developing hybrid fiber-reinforced cementitious materials to be applied on the impact rear side. Concrete is a predominant construction material due to several advantages; however, the pure cementitious composites have shown quasi-brittle behavior with undesirable typical large cracks under tensile loading conditions. Thus, the addition of a small volume of short fibers is a well-known strategy to increase the ductility and toughness of cementitious matrices besides optimization of the crack opening. Strain-hardening cement-based composites (SHCCs) is a particular class of fiber-reinforced concretes (FRCC) that can develop controlled multiple cracks while subjected to incremental tensile loading conditions. However, a proper composition design, especially concerning fiber and bond properties, still follows a trial and error approach. This work presents a newly developed model to simulate SHCC at the meso-scale level. This model is based on Finite-Element-Method and allows for nonlinear behavior for cement matrix, fiber material, and bond laws. Concerning three complexities of target FRCC, i.e., crack formation in the cement matrix, a large number of explicit fibers with arbitrary random distribution, and fibers’ interaction with the cement matrix via the bond, extra features are added to standard FE consist of: • Further development of the Strong Discontinuity Approach (SDA) to model discrete cracking of continuum elements on the element level • Discretization of single fibers by truss elements with truss nodes independently placed of continuum nodes • Connecting SDA elements to explicit truss elements by particular bond elements. In this research study, first, theoretical basics and special implementation issues were described. Later, this newly developed model was calibrated with several simple configurations. The bond law utilized in the simulation was derived from single fiber pullout test and calibrated with several analyses. In the next step, 2D SHCC dumbbell specimens under tensile loading condition were simulated, and a series of numerical case studies
