1. Locally Most Powerful Invariant Tests for Correlation and Sphericity of Gaussian Vectors

Abstract

In this paper we study the existence of locally most powerful invariant tests (LMPIT) for the problem of testing the covariance structure of a set of Gaussian random vectors. The LMPIT is the optimal test for the case of close hypotheses, among those satisfying the invariances of the problem, and in practical scenarios can provide better performance than the typically used generalized likelihood ratio test (GLRT). The derivation of the LMPIT usually requires one to find the maximal invariant statistic for the detection problem and then derive its distribution under both hypotheses, which in general is a rather involved procedure. As an alternative, Wijsman's theorem provides the ratio of the maximal invariant densities without even finding an explicit expression for the maximal invariant. We first consider the problem of testing whether a set of $N$-dimensional Gaussian random vectors are uncorrelated or not, and show that the LMPIT is given by the Frobenius norm of the sample coherence matrix. Second, we study the case in which the vectors under the null hypothesis are uncorrelated and identically distributed, that is, the sphericity test for Gaussian vectors, for which we show that the LMPIT is given by the Frobenius norm of a normalized version of the sample covariance matrix. Finally, some numerical examples illustrate the performance of the proposed tests, which provide better results than their GLRT counterparts.

Abstract

The problem of two-channel constrained least squares (CLS) filtering under various sets of constraints is considered, and a general set of solutions is derived. For each set of constraints, the solution is determined by a coupled (asymmetric) generalized eigenvalue problem. This eigenvalue problem establishes a connection between two-channel CLS filtering and transform methods for resolving channel measurements into canonical or half-canonical coordinates. Based on this connection, a unified framework for reduced-rank Wiener filtering is presented. Then, various representations of reduced-rank Wiener filters in canonical and half-canonical coordinates are introduced. An alternating power method is proposed to recursively compute the canonical coordinate and half canonical coordinate mappings. A deflation process is introduced to extract the mappings associated with the dominant coordinates. The correctness of the alternating power method is demonstrated on a synthesized data set, and conclusions are drawn.

Abstract

The Unified Protocol (UP) as a transdiagnostic intervention has primarily been applied in the treatment of anxiety disorders and in face-to-face-settings. The current study investigated the efficacy of a 10-week internet-based adaptation of the UP for anxiety, depressive, and somatic symptom disorders. N=129 participants were randomized to treatment or waitlist control. Linear mixed effect models revealed significant treatment effects for symptom distress, satisfaction with life, positive/negative affect and markers of anxiety, depression, and somatic symptom burden (within-group Hedges’ g = 0.32-1.38 and between-group g = 0.20-1.11). Treatment gains were maintained at 1- and 6-month-follow-up. Subgroup analyses showed comparable effects in participants with anxiety and depressive disorders. The results strengthen the application of the UP as an internet-based treatment for alleviating symptom distress across emotional disorders. More research on the applicability for single disorders and the mechanisms underlying the effects is needed.

Abstract

Background: Germline mutation screening of BRCA1 and BRCA2 genes is performed in suspected familial breast cancer cases, but a causative mutation is found in only 30% of patients. The development of additional methods to identify good candidates for BRCA1 and BRCA2 analysis would therefore increase the efficacy of diagnostic mutation screening. With this in mind, we developed a study to determine molecular signatures of BRCA1—or BRCA2—mutated breast cancers. Materials and Methods: Array-cgh (comparative genomic hybridization) and transcriptomic analysis were performed on a series of 103 familial breast cancers. The series included 7 breast cancers with a BRCA1 mutation and 5 breast cancers with a BRCA2 mutation. The remaining 91 cases were obtained from 73 families selected on the basis of at least 3 affected first-degree relatives or at least 2 affected first-degree relatives with breast cancer at an average age of 45 years. Array-cgh analyses were performed on a 4407 BAC-array (CIT-V8) manufactured by IntegraGen. Transcriptomic analyses were performed using an Affymetrix Human Genome U133 Plus 2.0 chip. Results: Using supervised clustering analyses we identified two transcriptomic signatures: one for BRCA1-mutated breast cancers consisting of 600 probe sets and another for BRCA2-mutated breast cancers also consisting of 600 probes sets. We also defined cgh-array signatures, based on the presence of specific genomic rearrangements, one for BRCA1-mutated breast cancers and one for BRCA2-mutated breast cancers. Conclusions: This study identified molecular signatures of breast cancers with BRCA1 or BRCA2 germline mutations. Genes present in these signatures could be exploited to find new markers for such breast cancers. We also identified specific genomic rearrangements in these breast cancers, which could be screened for in a diagnostic setting using fluorescence in situ hybridization, thus improving patient selection for BRCA1 and BRCA2 molecular genetic analysis.

Initial characterization of these antibodies often involves measurement of their neutralization activity against a panel of viruses, but such experiments do not generally lead to conclusive identification of an antibody’s epitope.

Abstract

Following infection by HIV-1, the host immune response is unable to clear the virus due to a variety of factors, including rapid viral mutation and the establishment of latent reservoirs. The only target of neutralizing antibodies is the trimeric envelope (Env) spike complex, but HIV-1 can usually evade anti-spike antibodies due to rapid mutation of its two spike glycoproteins, gp120 and gp41, and structural features that allow the spike to hide conserved epitopes. Efforts to design an effective antibody-based vaccine against HIV-1 would benefit from understanding how germ-line B-cell receptors recognize the gp120/ gp41 envelope spike. Recently isolated from several infected individuals, potent VRC01-like (PVL) HIV-1 antibodies derived from the VH1–2*02 germ-line allele target the CD4 binding site on gp120. These antibodies have unprecedented potency and breadth for HIV-1 neutralization and some protect against HIV-1 infection in animal models. Since the VH1–2*02 germ-line allele is present in up to 95% of the population, it may be possible to elicit by vaccination similar CD4 binding site antibodies in other individuals. VH1–2*02 germ-line B-cell receptor interactions with gp120 were uncharacterized, which was a bottleneck for designing immunogens to elicit PVL antibodies.

Abstract

We show that an arrangement A of n lines in general position in the plane has an inducing polygon of size O(n). Additionally, we present a simple algorithm for finding an inducing n-path for A in O(n log n) time and an algorithm that constructs an inducing n-gon for a special class of line arrangements within the same time bound.
