6. The biological evaluation of inhibitors of the Keap1-Nrf2 protein-protein interaction

Abstract

The Keap1-Nrf2 pathway has been identified as a key regulator of the cytoprotective response of cells when exposed to oxidative stress. The induction of detoxification gene products by increasing the activity of the transcription factor nuclear factor erythroid 2-related factor 2 (Nrf2) can be used as a chemopreventive approach to cancer treatment. Natural and synthetic agents can disrupt the interaction between the substrate adaptor protein Kelch-like ECH-associated protein 1 (Keap1) and Nrf2, which allows nuclear Nrf2 translocation and accumulation. The aim of this project was to characterize the behaviour of direct and indirect Nrf2 inducer molecules by applying several biological methods that were specifically optimised or developed for the Keap1-Nrf2 pathway. Potential inhibitors of the Keap1-Nrf2 complex were screened using a cellular NQO1 induction assay and an in vitro fluorescence polarisation (FP) assay for direct inhibitors. A novel in vitro Förster resonance energy transfer (FRET) assay was developed as an additional screening tool to identify Keap1 binding partners. Further biological evaluation was conducted with the most promising molecules using western blotting analysis and a new intracellular Nrf2 staining method for flow cytometry. The natural product and indirect Nrf2 inducer sulforaphane was used as a reference compound (NQO1 CD = 0.4 μM). From a library of Michael acceptor-containing cyclohexadienone analogues, a 3-chlorophenyl compound (6.49) was identified as a potent inducer (NQO1 CD = 1 μM). Stearoyl-capped Nrf2-derived peptides were potent inhibitors of the direct Keap1-Nrf2 protein-protein interaction and showed promising cell penetrating properties. Direct inhibitor small molecules included two bissulphonamide derivatives (7A4, NQO1 CD = 8 μM and 7B1, NQO1 CD = 4 μM) and a 1,2,3-triazole derivative (7C55, NQO1 CD = 0.6 μM) with FP IC50 values in the nano- to micromolar range. The selectivity of the compounds was assessed by revealing potential aryl hydrocarbon receptor (AhR) transcription factor cross-talk.

Abstract

Coronary computed tomography angiography (CCTA) does not allow the quantification of reduced blood flow due to coronary artery disease (CAD). In response, numerical methods based on the CCTA image have been developed to compute coronary blood flow and assess the impact of disease. However to compute blood flow in the coronary arteries, numerical methods require specification of boundary conditions that are difficult to estimate accurately in a patient-specific manner. We describe herein a new noninvasive flow estimation method, called Advection Diffusion Flow Estimation (ADFE), to compute coronary artery flow from CCTA to use as boundary conditions for numerical models of coronary blood flow. ADFE uses image contrast variation along the tree-like structure to estimate flow in each vessel. For validating this method we used patient specific software phantoms on which the transport of contrast was simulated. This controlled validation setting enables a direct comparison between estimated flow and actual flow and a detailed investigation of factors affecting accuracy. A total of 10 CCTA image data sets were processed to extract all necessary information for simulating contrast transport. A spectral element method solver was used for computing the ground truth simulations with high accuracy. On this data set, the ADFE method showed a high correlation coefficient of 0.998 between estimated flow and the ground truth flow together with an average relative error of only 1 % . Comparing the ADFE method with the best method currently available (TAFE) for image-based blood flow estimation, which showed a correlation coefficient of 0.752 and average error of 20 % , it can be concluded that the ADFE method has the potential to significantly improve the quantification of coronary artery blood flow derived from contrast gradients in CCTA images.

18. Pile installation fatigue damage from impact driving records – A case study of offshore monopiles

Abstract

Currently, impact pile driving is the preferred method of installation for offshore wind monopile foundations, thanks to the good stability of the foundation achieved after driving and fairly reliable driveability predictions possible for this installation method. Despite these benefits, a possible drawback of impact pile driving can be the high forces exerted on the foundation during installation. Since roughly 90% of steel structure failures during service life is due to fatigue or fracture, in order to promote circularity in the renewable energy sector and to allow a safe re-use of the wind turbine foundations, a correct estimation of the fatigue damage cumulated during the installation process is needed. The purpose of this paper is to address the topic of cumulative fatigue damage of offshore monopiles installed by impact driving based on driveability prediction results and actual driving records. The paper shows that the monopile fatigue damage predicted in the design stage, often based on numerical models considering constant maximum impact energy, can be overly conservative when compared to the fatigue damage calculation performed based on the actual driving records. Abstract

De Europese Habitatrichtlijn schrijft voor dat de Europese lidstaten de verslechtering van de biodiversiteit aan belangrijke plant- en diersoorten in Natura 2000-gebieden zoveel mogelijk moeten voorkomen. Sinds de uitspraak van de Raad van State in 2019 mogen economische activiteiten alleen worden ontplooid als deze de Natura 2000-gebieden niet (verder) aantasten. De schade door stikstofdepositie is in deze gebieden echter nu al zo groot dat er op korte termijn actie nodig is om zowel de Natura 2000- gebieden te behouden als nieuwe economische activiteiten in Nederland te kunnen uitvoeren. Het land zit hiermee op slot en de overheid is op zoek naar oplossingen om uit deze impasse te komen.

Abstract

Atmospheric levels of ammonia (NH3) have substantially increased during the last century, posing a hazard to both human health and environmental quality. The atmospheric budget of NH3, however, is still highly uncertain due to an overall lack of observations. Satellite observations of atmospheric NH3 may help us in the current observational and knowledge gaps. Recent observations of the Cross-track Infrared Sounder (CrIS) provide us with daily, global distributions of NH3. In this study, the CrIS NH3 product is assimilated into the LOTOS-EUROS chemistry transport model using two different methods aimed at improving the modeled spatiotemporal NH3 distributions. In the first method NH3 surface concentrations from CrIS are used to fit spatially varying NH3 emission time factors to redistribute model input NH3 emissions over the year. The second method uses the CrIS NH3 profile to adjust the NH3 emissions using a local ensemble transform Kalman filter (LETKF) in a top-down approach. The two methods are tested separately and combined, focusing on a region in western Europe (Germany, Belgium and the Netherlands). In this region, the mean CrIS NH3 total columns were up to a factor 2 higher than the simulated NH3 columns between 2014 and 2018, which, after assimilating the CrIS NH3 columns using the LETKF algorithm, led to an increase in the total NH3 emissions of up to approximately 30 %. Our results illustrate that CrIS NH3 observations can be used successfully to estimate spatially variable NH3 time factors and improve NH3 emission distributions temporally, especially in spring (March to May). Moreover, the use of the CrIS-based NH3 time factors resulted in an improved comparison with the onset and duration of the NH3 spring peak observed at observation sites at hourly resolution in the Netherlands. Assimilation of the CrIS NH3 columns with the LETKF algorithm is mainly advantageous for improving the spatial concentration distribution of the modeled NH3 fields. Compared to in situ observations, a combination of both methods led to the most significant improvements in modeled monthly NH3 surface concentration and NH4+ wet deposition fields, illustrating the usefulness of the CrIS NH3 products to improve the temporal representativity of the model and better constrain the budget in agricultural areas.
