Biomechanical cues such as shear stress, stretching, compression, and matrix elasticity are vital in the establishment of next generation physiological in vitro tissue models. Matrix elasticity, for instance, is known to guide stem cell differentiation, influence healing processes and modulate extracellular matrix (ECM) deposition needed for tissue development and maintenance. To better understand the biomechanical effect of matrix elasticity on the formation of articular cartilage analogs in vitro, this study aims at assessing the redifferentiation capacity of primary human chondrocytes in three different hydrogel matrices of predefined matrix elasticities. The hydrogel elasticities were chosen to represent a broad spectrum of tissue stiffness ranging from very soft tissues with a Young’s modulus of 1 kPa up to elasticities of 30 kPa, representative of the perichondralspace. In addition, the interplay of matrix elasticity and transforming growth factor beta-3 (TGF-β3) on the redifferentiation of primary human articular chondrocytes was studied by analyzing both qualitative (viability, morphology, histology) and quantitative (RT-qPCR, sGAG, DNA) parameters, crucial to the chondrotypic phenotype. Results show that fibrin hydrogels of 30 kPa Young’s modulus best guide chondrocyte redifferentiation resulting in a native-like morphology as well as induces the synthesis of physiologic ECM constituents such as glycosaminoglycans (sGAG) and collagen type II. This comprehensive study sheds light onto the mechanobiological impact of matrix elasticity on formation and maintenance of articular cartilage and thus represents a major step toward meeting the need for advanced in vitro tissue models to study both re- and degeneration of articular cartilage. Im Jahr 1907 beschrieb Silvestri zum ersten Mal eine neue Klasse der Hexapoda, die Protura. Diese Tiere leben in der Laubstreu und ihre Morphologie ist, aufgrund der geringen Körpergröße (0,5 - 2mm) und des langgestreckten, runden Körperbaus, sehr gut an ihre Lebensweise angepasst. Als Besonderheit weisen sie weder Augen noch Antennen auf, sie benutzten ihr erstes Beinpaar zur Orientierung. Von der Erstbeschreibung bis heute wurden nur wenige morphologische Arbeiten über diese Tiere verfasst und im Besonderen ist die abdominale Muskulatur nur unzureichend beschrieben. Beim Schlüpfen haben Proturen acht abdominale Segmente plus Telson, als Adulte elf plus Telson. Diese spezielle Ontogenie wird als Anamerie (Anamorphose) bezeichnet und ist einzigartig in der Klasse der Hexapoda. Während der Entwicklung der letzten Segmente wird auch die Genitalapertur (Squama genitalis) gebildet, ihre Funktionsweise ist unbekannt. In dieser Diplomarbeit wird die Muskulatur der abdominalen Segmente von Acerentomon maius beschrieben, sowie jene der Genitalanhänge. Die Muskulatur in den ersten sechs Segmenten ist Großteils gleich angeordnet. Man kann sie in dorsale, dorsoventrale, sowie ventrale Muskulatur einteilen, welche sowohl mono- als auch multisegmental verlaufen kann. Im 8. Segment kommen zwei weitere Muskeln für die Öffnung der Abdominaldrüse hinzu. Die Genitalapertur liegt in einer Einstülpung, der Genitalkammer, die ventral, am 11. Segment, ihre Öffnung hat. Die Genitalanhänge sind bei Männchen und Weibchen von unterschiedlicher Größe, außerdem ist die Muskulatur bei Männchen komplexer ausgebildet als beim Weibchen. Für ein besseres Verständnis der untersuchten Art und um systematisch relevante Erkenntnisse zu gewinnen ist ein Vergleich mit der Muskulatur anderer basaler Hexapoda unerlässlich., In 1907 Silvestri was the first who described a new class of Hexapoda called Protura. These animals live in soil and have a typical body morphology that is very well adjusted to their environment. Only a few anatomical studies on this order are available until know, that is why the musculature of the abdominal region of these animals is still poorly investigated. After hatching Protura have only eight abdominal segments plus Telson, whereas adult specimen have eleven segments plus Telson. Interestingly no stage with ten segments plus Telson is known. This special ontogeny is known as anamerie (anamorphosis) and unique within the class Hexapoda. During the formation of the last stage the exterior genital appendages (Squama genitalis) are developed. Their function remains unknown due to the poor investigation status. In this diploma thesis the musculature of the abdominal segments of Acerentomon maius as well its genital appendage`s musculature will be described. The muscles in the first six segments are arranged almost identical. They can be subdivided into dorsal, dorso- ventral, and ventral muscles which are either mono- or multisegmental. In addition two muscles in the 8. segment are known which are responsible for opening the abdominal glands. The Squama genitalis lies inside the genital chamber, which has its opening on the ventral side of the 11. segment. The genital appendages are different in both sexes whereas the male appendages and the appending musculature is much more complex compared to the female one. A comparison with the musculature of other basal Hexapoda is an important aspect for a better understanding of the investigated specie`s anatomy and gives new systematic aspects.
