The efficiency of turmeric fractions, such as petroleum ether, chloroform, benzene, methanol and aqueous were evaluated for their inhibitory effect on clinical and standard strains of pathogenic bacteria and fungus. The methanolic fraction of Curcuma longa rhizome had high potential to inhibit some pathogenic bacteria i.e. S. aureus to a greater degree than other fractions of Curcuma longa. In our study the results show that the different fractions (petroleum ether, methanol etc.) of Curcuma longa rhizome were more effective antimicrobial agents than the crude extract of Curcuma longa. Methanolic extract of curcuma longa was evaluated against gram positive and gram-negative bacteria. The bacteria isolates include Bacillus subtilis and Escherichia coli. Antimicrobial activities were estimated by Well diffusion method and antifungal activity was evaluated against Candida Albicans and Aspergillus Niger. The results were pointed as the methanol extract was more effective on both bacteria and fungus then water also similar effective on both bacteria and fungus but against Aspergillus Nigerit has outstand results. Vegetables are one of the most preferred food commodities and can be consumed either raw or as processed due to their health-promoting nutrients. In the present work, analysis of pharmacognostical standards, antioxidant capacity, and separation of phytocompounds through thin layer chromatography (TLC) from cabbage, cauliflower, pea, carrot, and potato peels were carried out. Microscopic analysis revealed the presence of wood fibers, trichomes, crystals, and annular xylem vessels in the vegetable peels. Physicochemical analysis showed that all the vegetable peel samples which were analysed have low (7.08%-10%) moisture content. The total ash content of vegetable peels varied in cauliflower peels (1.95±0.58) to the peels of pea (19.86±1.9). The content of acid insoluble ash varied from 1.46±0.63 to 3.09±0.59 in cauliflower and pea. Potato peel has the lowest water-soluble ash content (1.16±1.90) as compared to other peels. The highest pH value was found in the peels of pea (7), while the lowest pH was found in the peels of cabbage (4). Among all extracts, the petroleum ether extract has shown the greatest yield (5.6±0.45). The fluorescence analysis showed various colours like green, brown, pale green, and yellow under different chemical treatments. Different types of pri-secondary metabolites were detected in small, moderate, and high amounts and notified to provide numerous health benefits to humans. In case of DPPH assay, aqueous extract of cauliflower has shown the low value of IC50 (24.82 µg/ml) in comparison to standard, suggested the higher antioxidant activity of the extract. Among all the extracts, aqueous and methanol extracts of cauliflower have shown the better reducing and total antioxidant activity in comparison to standard. TLC profiling of methanolic extract of cabbage and cauliflower peels revealed the presence of different compounds of varying Rf values. Above results indicate that the food waste consists of valuable components and may be utilized as noticeable and cheap source in pharmaceuticals for the treatment of several life-threatening diseases.

The present study was carried out for the evaluation of in-vitro antioxidant potential of alcoholic stem bark (ALSB) extract of Bauhinia variegata Linn. Dried stem bark extract of B. variegata Linn. was screened to evaluate its free radical scavenging effect. Various methods like DPPH assay, reducing power assay, percentage scavenging activity of hydrogen peroxide and superoxide radical scavenging activity were used for screening invitro antioxidant potential. Antioxidant potentials were concentration dependent which were compared with standard antioxidants such as butylated hydroxyanisole (BHA) and ascorbic acid. The maximum scavenging effect of B. variegata Linn. alcoholic stem bark extract on DPPH free radical, superoxide radical and hydrogen peroxide was 72.19 + 0.20, 81.60 + 0.22 and 76.06 + 0.16 respectively at a concentration of 2500 μg/mL. It was clearly indicated that the alcoholic extract of the stem bark has significant in vitro antioxidant activity. Currently available synthetic antioxidants like butylated hydroxyanisole (BHA), butylated hydroxyl toluene (BHT), Ascorbic acid and gallic acid appear to be associated with hepatotoxicity and many others negative health effects. The aim of this study was to explore the antibacterial and antioxidant efficacy of nickel oxide nanoparticles (NiO NPs). The Raphanus sativus ( R. sativus ) extract mediated NiO NPs were calcined at 100, 300, 600 and 900 ℃ in a muffle furnace for 3 h. The increased intensity of diffraction bands in the X-ray diffraction (XRD) spectrum suggest that the degree of crystallinity increases with increasing calcination temperature. The desired elements was depicted in the energy dispersive X-rays (EDX) spectrum confirm the purity of the NiO Sample. The variation in surface morphology and increase in the particles size from 12.78 to 51.42nm was determined from the transmission electron microscope (TEM) micrographs. The shift toward higher wavelength was observed in the diffuse reflectance spectroscopy (DRS) spectra with increasing calcination temperature, results into a clear decrease in band gap from 3.12 to 2.86 eV. The presence of hydroxyl group along with other organic moieties were confirm through Fourier transform infrared (FTIR) spectroscopy analysis. The biological potential of the calcined NiO NPs was examined during the antibacterial and antioxidant experiments. The antibacterial effect of NiO NPs was studied using the agar well diffusion process, and the ABTS free radical scavenging potential of NiO NPs was also assessed. The activity of NiO NPs calcined at 100 °C is greater than that of those calcined at higher temperatures.

Hydroxyl radical footprinting-mass spectrometry (HRF-MS) is a powerful technique for measuring protein structure by quantitating the solvent accessibility of amino acid side-chains; and when used in comparative analysis, HRF-MS data can provide detailed information on changes in protein structure. However, consistently controlling the amount of hydroxyl radical labeling of a protein requires the precise understanding of both the amount of radicals generated and half-life of the radicals in solution. The latter is particularly important for applications such as protein-protein and protein-ligand interactions, which may have different characteristics such as intrinsic reactivity and buffer components, and can cause differences in radical scavenging (herein termed "scavenging potential") between samples. To address this inherent challenge with HRF-MS analysis, we describe the comprehensive implementation of an internal standard (IS) dosimeter peptide leucine enkephalin (LeuEnk) for measuring the scavenging potential of pharmaceutically relevant proteins and formulation components. This further enabled evaluation of the critical method parameters affecting the scavenging potential of samples subjected to HRF-MS using fast photochemical oxidation of proteins. We demonstrate a direct correlation between the oxidation of the IS peptide and biotherapeutic target proteins, and show the oxidation of the IS can be used as a guide for ensuring equivalent scavenging potentials when comparing multiple samples. Establishing this strategy enables optimization of sample parameters, a system suitability approach, normalization of data, and comparison/harmonization of HRF-MS analysis across different laboratories.
