Abstract

Emerging technologies are considering the possible use of Terahertz radiation in different fields ranging from telecommunications to biology and biomedicine. The study of the potential effects of Terahertz radiation on biological systems is therefore an important issue in order to safely develop a variety of applications. This paper describes a pilot study devoted to determine if Terahertz radiation could induce genotoxic effects in human peripheral blood leukocytes. For this purpose, human whole blood samples from healthy donors were exposed for 20 min to Terahertz radiation. Since, to our knowledge, this is the first study devoted to the evaluation of possible genotoxic effects of such radiation, different electromagnetic conditions were considered. In particular, the frequencies of 120 and 130 GHz were chosen: the first one was tested at a specific absorption rate (SAR) of 0. 4 mW g-1, while the second one was tested at SAR levels of 0. 24,1. 4, and 2 mW g-1. Chromosomal damage was evaluated by means of the cytokinesis block micronucleus technique, which also gives information on cell cycle kinetics. Moreover, human whole blood samples exposed to 130 GHz at SAR levels of 1. 4 and 2 mW g-1 were also tested for primary DNA damage by applying the alkaline comet assay immediately after exposure. The results obtained indicate that THz exposure, in the explored electromagnetic conditions, is not able to induce either genotoxicity or alteration of cell cycle kinetics in human blood cells from healthy subjects.

Abstract

The aim of this study was to investigate the induction of genotoxic and cytotoxic effects in cultured human lymphocytes from healthy donors following combined exposure to a 50 Hz sinusoidal magnetic field (1 mT field intensity) and a chemical mutagen: mitomycin-C or hydroquinone. The induced effects were evaluated by applying the cytokinesis-block technique, because it offers the advantage of providing simultaneously information on both cell cycle progression (cytokinesis block proliferation index) and chromosomal damage (micronucleus frequency). The results obtained indicate that magnetic field exposure does not affect spontaneous micronucleus frequency, although a slight increase in cell proliferation was observed p 0.031). Both mutagens induce a statistically significant increase in micronucleus frequency p 0.0 and p 0.016 for mitomycin-C and hydroquinone, respectively). Moreover, the combined exposure to both magnetic field and chemicals does not increase the damage due to the chemical treatments alone, both in terms of genotoxicity and cell proliferation (p> 0.05 in all cases), suggesting that, in the experimental condition adopted, no cooperative effects are induced.

Abstract

In the present study, we investigated the effect of a 50-Hz sinusoidal magnetic field at 1 mT rms field intensity on cell proliferation and micro-nucleus frequency in peripheral blood lymphocytes exposed for 72 h. Lymphocyte cultures from 5 subjects affected by Turner's syndrome were tested; cultures from 5 healthy donors were also studied to make a comparison with a control group. No statistically significant differences in micronucleus frequency and cell proliferation were found between control and exposed cultures from healthy and TS donors. The results obtained indicate that the field tested does not exert any genotoxic effect nor interfere with cell cycle.

Abstract

The aim of this work was to ascertain whether microwave radiation (frequency 9 GHz; specific absorption rate 70 mW/gr, exposure duration 10 min) may produce genotoxic effects, assessed by cytokinesis-block micro-nucleus (MN) assay, in bovine (Bos taurus L.) peripheral blood lymphocyte cultures. As positive control for the MN induction, mitomycin-C (MMC) was used: it is known that this alkylating agent has a potent genotoxic (radiomimetic) action. To evaluate possible cooperative effects, some cultures from microwave-exposed samples were treated with MMC. The results obtained indicate that the optimal dose of MMC to induce MN frequency increase in this species is 0.044 µg/ml and that microwave radiation induces a statistically significant increase of MN frequency both with and without MMC.

Peripheral human blood from 23 healthy donors aged between 23 and 95 years was exposed to continuous wave (CW) or 50 Hz amplitude modulated (AM) microwave radiation and was cultured for 72 h. Other exposure parameters were: frequency 9 GHz, specific absorption rate (SAR) 90 mW/g, exposure duration 10 min. The possible genotoxic effect was evaluated by means of cytokinesis-block micronucleus method. A significant (p < 0.05) increase in micronuclei was found following AM microwave exposure.

Abstract

In the present study micronucleus induction and cell proliferation in human peripheral blood lymphocytes cultured in vitro and exposed to50 Hz sinusoidal magnetic fields for 72 h at different intensities (1.0, 0.75, 0.5, 0.25, and 0.05 mT rms) were investigated. The resultsobtained from 42 healthy donors aged between 26 and 54 y indicate that, for the field intensities tested, no genotoxic effects were found, as assessed by the cytokinesis-block micronucleus assay. Abstract

The present study investigated, using in vitro experiments on human lymphocytes, whether exposure to a microwave frequency used for mobile communication, either unmodulated or in presence of phase only modulation, can cause modification of cell proliferation kinetics and/or genotoxic effects, by evaluating the cytokinesis block proliferation index and the micronucleus frequency. In the GSM 1800 mobile communication systems the field is both phase (Gaussian minimum shift keying, GMSK) and amplitude (time domain multiple access, TDMA) modulated. The present study investigated only the effects of phase modulation, and no amplitude modulation was applied. Human peripheral blood cultures were exposed to 1.748 GHz, either continuous wave (CW) or phase only modulated wave (GMSK), for 15 min. The maximum specific absorption rate (5 W/kg) was higher than that occurring in the head of mobile phone users; however, no changes were found in cell proliferation kinetics after exposure to either CW or GMSK fields. As far as genotoxicity is concerned, the micronucleus frequency result was not affected by CW exposure; however, a statistically significant micronucleus effect was found following exposure to phase modulated field. These results would suggest a genotoxic power of the phase modulation per se.

Abstract

The occurrence of modulation-specific effects after co-exposures to radio-frequency (RF) and other agents has been discussed in the literature. In this paper, the influence of modulation and bandwidth in eliciting the DNA damage of RF alone and in combination with mitomycin-C (MMC) is analyzed in human lymphocytes. Blood cultures from healthy donors were exposed to 1950 MHz, and continuous wave (CW), wideband direct-sequence code division multiple access (WCDMA, 4.5 MHz bandwidth), and additive white Gaussian noise (AWGN, 9 MHz bandwidth) signals were considered. For each signal, specific absorption rate (SAR) values of 0.15, 0.3, 0.6, 1.25 W/kg were tested. RF exposure alone never induced DNA damage in the micronucleus assay. When RF exposure was followed by MMC treatment, the effect depended on modulation and bandwidth. CW exposure never altered the MMC-induced DNA damage, while such damage was reduced when either signals WCDMA at 0.3 W/kg SAR or AWGN at 0.15 and 0.3 W/kg were applied. These results indicate the influence of modulation for the occurrence of the protective effect, with a relation between the bandwidth and the power absorbed by samples. If confirmed in vivo , clinical applications using modulated RF signals could be devised to protect cells from side effects of therapeutic treatments.
