Abstract

The discovery of the quantum Hall effect (QHE) marked a turning point in condensed-matter physics. The measurement of the Hall resistance showed that electronic resistance could be defined precisely in terms of fundamental constants, even in a disordered and irregular sample. Over the past 40 years, the QHE has inspired new theories and led to experimental discoveries in a range of fields going beyond solid-state electronics to photonics and quantum entanglement. In this Viewpoint, physicists reflect on how the QHE has influenced their research.

Abstract

Summary Background Patients with chronic obstructive pulmonary disease (COPD) have few options for treatment. The efficacy and safety of the phosphodiesterase-4 inhibitor roflumilast have been investigated in studies of patients with moderate-to-severe COPD, but not in those concomitantly treated with longacting inhaled bronchodilators. The effect of roflumilast on lung function in patients with COPD that is moderate to severe who are already being treated with salmeterol or tiotropium was investigated. Methods In two double-blind, multicentre studies done in an outpatient setting, after a 4-week run-in, patients older than 40 years with moderate-to-severe COPD were randomly assigned to oral roflumilast 500 μg or placebo once a day for 24 weeks, in addition to salmeterol (M2-127 study) or tiotropium (M2-128 study). The primary endpoint was change in prebronchodilator forced expiratory volume in 1 s (FEV 1 ). Analysis was by intention to treat. The studies are registered with ClinicalTrials.gov, number NCT00313209 for M2-127, and NCT00424268 for M2-128. Findings In the salmeterol plus roflumilast trial, 466 patients were assigned to and treated with roflumilast and 467 with placebo; in the tiotropium plus roflumilast trial, 371 patients were assigned to and treated with roflumilast and 372 with placebo. Compared with placebo, roflumilast consistently improved mean prebronchodilator FEV 1 by 49 mL (p 1 was noted in both groups. Furthermore, roflumilast had beneficial effects on other lung function measurements and on selected patient-reported outcomes in both groups. Nausea, diarrhoea, weight loss, and, to a lesser extent, headache were more frequent in patients in the roflumilast groups. These adverse events were associated with increased patient withdrawal. Interpretation Roflumilast improves lung function in patients with COPD treated with salmeterol or tiotropium, and could become an important treatment for these patients. Funding Nycomed.

15. Simultaneous Determination of Human Plasma Levels of Citalopram, Paroxetine, Sertraline, and Their Metabolites by Gas Chromatography—Mass Spectrometry

Abstract

A gas chromatography—mass spectrometry method is presented which allows the simultaneous determination of the plasma concentrations of the selective serotonin reuptake inhibitors citalopram, paroxetine, sertraline, and their pharmacologically active N-demethylated metabolites (desmethylcitalopram, didesmethylcitalopram, and desmethylsertraline) after derivatization with the reagent N-methyl-bis(trifluoroacetamide). No interferences from endogenous compounds are observed following the extraction of plasma samples from six different human subjects. The standard curves are linear over a working range of 10-500 ng/mL for citalopram, 10-300 ng/mL for desmethylcitalopram, 5-60 ng/mL for didesmethylcitalopram, 20-400 ng/mL for sertraline and desmethylsertraline, and 10-200 ng/mL for paroxetine. Recoveries measured at three concentrations range from 81 to 118% for the tertiary amines (citalopram and the internal standard methylmaprotiline), 73 to 95% for the secondary amines (desmethylcitalopram, paroxetine and sertraline), and 39 to 66% for the primary amines (didesmethylcitalopram and desmethylsertraline). Intra- and interday coefficients of variation determined at three concentrations range from 3 to 11 % for citalopram and its metabolites, 4 to 15% for paroxetine, and 5 to 13% for sertraline and desmethylsertraline. The limits of quantitation of the method are 2 ng/mL for citalopram and paroxetine, 1 ng/mL for sertraline, and 0.5 ng/mL for desmethylcitalopram, didesmethylcitalopram, and desmethylsertraline. No interferences are noted from 20 other psychotropic drugs. This sensitive and specific method can be used for single-dose pharmacokinetics. It is also useful for therapeutic drug monitoring of these three drugs and could possibly be adapted for the quantitation of the two other selective serotonin reuptake inhibitors on the market, namely fluoxetine and fluvoxamine

50. Surface Organometallic Chemistry on Metals Applied to the Environment: Hydrogenolysis of AsPh3with Nickel Supported on Alumina

Abstract

The reaction of AsPh3with Ni/Al2O3at 100 to 200°C under 6 to 30 bars of hydrogen has been followed by kinetic analysis of both the consumption of AsPh3and the evolution of benzene and cyclohexane as well as magnetic and XRD measurements of the metallic and intermetallic phase(s). The reaction proceeds via complete hydrogenolysis of the As–Ph bonds, with final formation of a well-characterized NiAs alloy (XRD). The mechanism of alloy formation is complex: at low coverage of the nickel surface, stepwise hydrogenolysis of the As–Ph bonds leads to the formation of the surface organometallic fragments Nis[AsPhx]y(kinetic studies and analytical data). Further hydrogenolysis of these fragments progressively leads to the formation of arsenic “adatoms” which quickly migrate inside the nickel particles to form NiAs0.45amorphous intermetallic phases (XRD and magnetic measurements). The reaction then proceeds slowly up to the As/Ni ratio of unity with formation of the crystalline NiAs “nickeline” phase (XRD).
