This article examines the differences in aortic elasticity and histological patterns between male and female patients with aortopathy. The study included 96 patients who underwent thoracic aortic surgery, and measurements were taken using transthoracic echocardiography. The results showed that there were no significant differences in arterial distensibility, arterial strain, or most histological parameters between male and female patients. Therefore, the study suggests that changes in aortic elasticity and aortic wall patterns do not explain the higher risk of aortic dissection in female patients. [Extracted from the article]

The cause for the striking association between 3p13 loss and ERG fusion is unknown. Methods: Here, we made use of a preexisting heterogeneity prostate cancer tissue microarray including ten tissue spots from ten different tumor areas of 317 cancers to examine the spatial distribution of 3p13 deletions (determined by fluorescence in situ hybridization) in prostate cancer areas with and without ERG overexpression (determined by immunohistochemistry). Results: 3p13 deletions were found in 61 of 299 (20.4%) and ERG positivity in 174 of 317 (54.9%) interpretable cancers. The likelihood of 3p13 loss was twice as high in ERG-positive cancers (39/152, 25.7%) than in ERG-negative cancers (17/124, 13.7%, P=0.010). At least three tissue spots were interpretable for 3p13 deletion status in 279 cancers: only these were used for heterogeneity assessment. Among these tumors, 58 (20.8%) had a 3p13 deletion and 221 (79.2%) were undeleted. The majority of 3p13-deleted cancers showed marked intratumoral heterogeneity. Areas with and without 3p13 loss were found in 50 (18%) of 279 cancers with three or more interpretable tissue spots, while only eight (3%) tumors had a homogeneous 3p13 loss. Comparison with ERG data revealed that ERG fusion usually precede 3p13 deletions. In total, 26 (66.7%) of 39 cancers with ERG and 3p13 alteration had only focal 3p13 deletions in an otherwise ERG-positive background. In contrast, none of the cancers showed a pattern that would be consistent with 3p13 deletion preceding ERG fusion. Conclusion: Our study identifies 3p13 deletion as a highly heterogeneous alteration in prostate cancer preferentially developing at rather late stages of progression in TMPRSS2:ERG fusion-positive tumors. Zusammenfassung: Die Wiedereinführung des Oberflächenersatzes hat zu einer überraschend schnellen Reetablierung dieses Verfahrens nach den unbefriedigenden Ergebnissen in der Vergangenheit geführt. Die relativ hohe Rate an Frühversagern gibt jedoch noch Anlass zu Bedenken. Im Rahmen der vorliegenden Studie wurden 256 Revisionspräparate tribologisch, radiologisch, morphologisch und histologisch analysiert, um Ursachen für das Versagen bzw. die Revision zu identifizieren. Mehr als zwei Drittel der Versager wurden aufgrund einer Fraktur revidiert – Halsfrakturen traten hierbei deutlich früher auf als Fakturen innerhalb des Kopfes und Pfannenlockerungen. Die Versagensrate war innerhalb der ersten 16 Wochen nach Implantation und bei den ersten 10 Operationen eines Operateurs am höchsten. Revisionen ohne Vorliegen einer Fraktur oder Pfannenlockerung erfolgten im Durchschnitt nach 2 Jahren. Revisionen, bedingt durch femurseitige Probleme wie Notching oder überhöhte Implantationskräfte, traten vor pfannenseitigen Problemen wie Pfannenlockerung oder erhöhtem Verschleiß durch Pfannenfehlpositionierung auf. Die Anteversion der Pfanne spielt hierbei eine wichtige Rolle, die im Rahmen der Studie jedoch nicht quantifiziert werden konnte.

Both oncogenic and tumor suppressor functions have been described for junction plakoglobin (JUP), also known as γ‐catenin. To clarify the role of JUP in prostate cancer, JUP protein expression was immunohistochemically detected in a tissue microarray containing 11 267 individual prostatectomy specimens. Considering all patients, high JUP expression was associated with adverse tumor stage (P = 0.0002), high Gleason grade (P, Junction plakoglobin (JUP) is a component of desmosomes and adherens junctions and reported to play divergent roles in tumorigenesis. JUP expression was analyzed in prostate cancer in a large‐enough patient cohort to reflect the molecular heterogeneity of the disease. Overall, high JUP expression was associated with adverse tumor features, while low JUP expression indicated poor prognosis in CHD1‐deleted patients.

Publisher's version (útgefin grein), Sex differences have been observed in multiple facets of cancer epidemiology, treatment and biology, and in most cancers outside the sex organs. Efforts to link these clinical differences to specific molecular features have focused on somatic mutations within the coding regions of the genome. Here we report a pan-cancer analysis of sex differences in whole genomes of 1983 tumours of 28 subtypes as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium. We both confirm the results of exome studies, and also uncover previously undescribed sex differences. These include sex-biases in coding and non-coding cancer drivers, mutation prevalence and strikingly, in mutational signatures related to underlying mutational processes. These results underline the pervasiveness of molecular sex differences and strengthen the call for increased consideration of sex in molecular cancer research., We thank all the members of the Boutros lab for insightful discussions.
