A new process and mechanism are presented for controlling the shape memory response spatially within monolithic NiTi thin film structures. This technique is shown to effectively control the martensitic phase transformation temperature and exhibits control over aspects of the mechanical and shape memory responses as well. Specifically, the martensitic phase transformation temperature decreases with incident laser energy density. Concomitant modifications are observed in both the mechanical and shape memory responses in laser processed films. Analysis and characterization are performed via temperature controlled optical microscopy, x-ray diffraction, atomic force microscopy, and nanoindentation. [ABSTRACT FROM AUTHOR]

Introduction: Patients with spinal cord injury (SCI) and incomplete paralysis may experience muscle immobilization due to functional impairment of agonist and antagonist muscles. This can induce spasticity and pain, with the risk of contracture increasing as the period of immobilization increases. This could be aggravated by pain, which interferes with rehabilitation. There is no established treatment for pain and joint contractures caused by acute SCI. Botulinum therapy plays a role in relieving muscle tension. Here, we report a case of acute SCI in which botulinum therapy was administered., Case Presentation: The patient experienced a C5-cervical SCI due to a road traffic accident, with marked paralysis in the upper and lower limbs below the C5 level. The patient had persistent elbow flexion since the SCI and complained of excruciating pain, making adequate rehabilitation difficult. Botulinum toxin was administered to the biceps and brachialis muscles 15 days after the SCI. After administration, the patient experienced a reduction in pain with relaxation of the elbow flexor muscles, and rehabilitation treatment was resumed. This patient's contracture did not worsen, his pain decreased, and he was able to initiate self-feeding training., Discussion: In this case, occupational and physical therapy was administered with botulinum therapy. Together, they successfully reduced pain. Botulinum therapy provides analgesia and muscle relaxation and may lead to functional recovery, and we believe that treatment can be considered even in the acute phase., (© 2024. The efficiency of new anti-cancer drugs such as the S-1 system was demonstrated in a controlled study comparing treatment and non-treatment groups. We encountered a patient with gastric cancer demonstrating peritoneal dissemination, who was successfully treated by combination therapy using S-1 and docetaxel. A 62-year-old woman was admitted to the hospital due to appetite loss and nausea. Upper GI endoscopy demonstrated a type 3 gastric cancer extending from the upper to lower body of the stomach. In the pelvic cavity, an abdominal CT scan demonstrated massive ascites. An abnormally high CA72-4 (143.8 U/mL) level was detected in serum. Treatment with S-1 and docetaxel was started with the following regimen: daily oral administration of 80 mg/body S-1 for 14 days, followed by a 7-day rest and infusion of 40 mg/m2 docetaxel on day 1. After 4 courses, the sites of dissemination had disappeared, and the serum CA72-4 value returned to normal. The patient clinically achieved good QOL by this method, which was very effective for non-resected gastric cancer with peritoneal dissemination.

Using three-dimensional finite element analysis (3D-FEA), stress distributions in the remaining radicular tooth structure were investigated under the condition of varying diameters of fiber post for fiber post-reinforced composite resin cores (fiber post and core) in maxillary central incisors. Four 3D-FEA models were constructed: (1) fiber post (ø1.2, ø1.4, and ø1.6 mm) and composite resin core; and (2) gold-cast post and core. Maximum stresses in the tooth structure for fiber post and core were higher than that for gold-cast post and core. In the former models, stresses in the tooth structure as well as in the composite resin were slightly reduced with increase in fiber post diameter. These results thus suggested that to reduce stress in the remaining radicular tooth with a large coronal defect, it is recommended to accompany a composite resin core with a fiber post of a large diameter.

Amphioxus has neither elaborated brains nor definitive sensory organs, so that the two may have evolved in a mutually affecting manner and given rise to the forms seen in extant vertebrates. Clarifying the developmental and functional aspects of the amphioxus sensory system is thus pivotal for inferring the early evolution of vertebrates. Morphological studies have identified and classified amphioxus sensory cells; however, it is completely unknown whether the morphological classification makes sense in functional and evolutionary terms. Molecular markers, such as gene expression, are therefore indispensable for investigating the developmental and functional aspects of amphioxus sensory cells. This article reviews recent molecular studies on amphioxus sensory cells. Increasing evidence shows that the non-neural ectoderm of amphioxus can be subdivided into molecularly distinct subdomains by the combinatorial code of developmental cues involving the RA-dependent Hox code, suggesting that amphioxus epithelial sensory cells developed along positional information. This study focuses particularly on research involving the molecular phylogeny and expression of the seven-transmembrane, G protein-coupled receptor (GPCR) genes and discusses the usefulness of this information for characterizing the sensory cells of amphioxus.

This article reviews recent advances in comparative biological studies of vertebrate origins, with the aim of revisiting the long-standing controversy concerning these origins. Since early vertebrate evolution is paralleled by an evolutionary trend towards increasing activity, I focus on the evolution of respiratory and circulatory systems and discuss their potential roles in early vertebrate evolution. I give particular attention to the nasohypophyseal duct, an orifice characteristically found in agnathan vertebrates, and hypothesize that this duct originally functioned to convey oxygen dissolved in seawater to the respiratory gills. The chemosensory cell population that originated from the wall of the duct became the incipient olfactory organ and played a role in the organization of feeding behavior. An increase in chemosensory receptor genes via large-scale genomic evolution in the vertebrate lineage caused the repertoire of chemosensory cells to diversify and led to the appearance of the integrative center, including telencephalic structures typically lacking in protochordates.

Chemosensation is the primary sensory modality in almost all metazoans. The vertebrate olfactory receptor genes exist as tandem clusters in the genome, so that identifying their evolutionary origin would be useful for understanding the expansion of the sensory world in relation to a large-scale genomic duplication event in a lineage leading to the vertebrates. In this study, I characterized a novel GPCR (G-protein-coupled receptor) gene-coding locus from the amphioxus genome. The genomic DNA contains an intronless ORF whose deduced amino acid sequence encodes a seven-transmembrane protein with some amino acid residues characteristic of vertebrate olfactory receptors (ORs). Surveying counterparts in the Ciona intestinalis (Asidiacea, Urochordata) genome by querying BLAST programs against the Ciona genomic DNA sequence database resulted in the identification of a remotely related gene. In situ hybridization analysis labeled primary sensory neurons in the rostral epithelium of amphioxus adults. Based on these findings, together with comparison of the developmental gene expression between amphioxus and vertebrates, I postulate that chemoreceptive primary sensory neurons in the rostrum are an ancient cell population traceable at least as far back in phylogeny as the common ancestor of amphioxus and vertebrates., (Copyright (c) 2005 Wiley-Liss, Inc.)

Members of the T-box family of transcription factors share an evolutionarily conserved DNA-binding domain and play significant roles in various processes of embryonic development. Vertebrate T-box genes are categorized into the following five major subfamilies (eight groups), depending on sequence similarities: Brachyury, Tbx1 (Tbx1/10, Tbx15/18/22, Tbx20), Tbx2/3/4/5 (Tbx2/3 and Tbx4/5), Tbx6, and Tbr/Eomes/TBX21. Ascidians are primitive chordates, and their tadpole larva are considered to represent the simplified and basic body plan of vertebrates. In addition, it has been revealed that the ascidian genome contains the basic ancestral complement of genes involved in development. The present characterization of cDNAs and survey of the Ciona intestinalis draft genome demonstrated that the Ciona genome contains a single copy gene for each of the Brachyury, Tbx1/10, Tbx15/18/22, Tbx20, Tbx2/3, and Tbr/Eomes/TBX21 groups, and at least three copies of the Tbx6 subfamily. Each of the Ciona T-box genes shows a characteristic expression pattern, although that of Tbx20 was not determined in the present study. These results provide basic information that will be useful for future studies of the function of each gene, genetic cascades of different T-box genes, and genome-wide surveys of evolutionary changes in the T-box gene structure and organization in this primitive chordate., (Copyright 2004 Wiley-Liss, Inc.)

The notochord is one of the diagnostic features of the phylum Chordata. Despite the similarities in the early morphogenetic patterns of the notochords of various chordates, they are strikingly distinct from one another at the histological level. The amphioxus notochord is one example of an evolutionary novelty because it is made up of muscle cells. Our previous expressed sequence tag analysis, targeting messenger RNAs expressed in the adult amphioxus notochord, demonstrated that many muscle-related genes are expressed there. To characterize amphioxus notochord cells and to gain insights into the myogenic program in the notochord, we determined the spatial and temporal expre-ssion patterns of these muscle-related genes during amphioxus development. We found that BbNA1 (notochord actin), Amphi-Trop I (troponin I), Amphi-TPmyosin (tropo-myosin), Amphi-MHC2 (myosin heavy chain), Amphi-nMRLC (notochord-specific myosin regulatory light chain), Amphi-nTitin/MLCK (notochord-specific titin/myosin light chain kinase), Amphi-MLP/CRP3 (muscle LIM protein), and Amphi-nCalponin (notochord-specific calponin) are expres-sed with characteristic patterns in notochord cells, including the central cells, dorsally located cells, and ventrally located cells, suggesting that each notochord cell has a unique molecular architecture that may reflect its function. In addition, we characterized two MyoD genes (Amphi-MyoD1 and Amphi-MyoD2) to gain insight into the genetic circuitry governing the formation of the notochord muscle. One of the MyoD genes (Amphi-MyoD2) is expressed in the central notochord cells, and the coexistence of Amphi-MyoD2 transcripts along with the Amphi-MLP/CRP3 transcripts implies the participation of Amphi-MyoD2 in the myogenic program in the notochord muscle.

A cDNA for a novel T-box containing gene was isolated from the amphioxus Branchiostoma belcheri. A molecular phylogenetic tree constructed from the deduced amino acid sequence of the isolated cDNA indicates that this gene belongs to the T-Brain subfamily. In situ hybridization reveals that the expression is first detected in the invaginating archenteron at the early gastrula stage and this expression is down-regulated at the neurula stage. In early larvae, the expression appears again and transcripts are detected exclusively in the pre-oral pit (wheel organ-Hatschek's pit of the adult). In contrast to the vertebrate counterparts, no transcripts are detected in the brain vesicle or nerve cord throughout the development. These results are interpreted to mean that a role of T-Brain products in vertebrate forebrain development was acquired after the amphioxus was split from the lineage leading to the vertebrates. On the other hand, comparison of the tissue-specific expression domain of T-Brain genes and other genes between amphioxus and vertebrates revealed that the pre-oral pit of amphioxus has several molecular features which are comparable to those of the vertebrate olfactory and hypophyseal placode., (Copyright 2002 Wiley-Liss, Inc.)

The development of the nervous system of amphioxus was investigated at the cellular level based on the expression of the pan-neuronal marker gene Hu/elav. In situ hybridization analysis showed that an amphioxus Hu/elav homolog (AmphiHu/Hel) was expressed in individual cells within the neural plate, but the cells exhibited no obvious arrangements in early embryos without distinct somites. However, in neurulae with somites, AmphiHu/Hel-positive cells were clustered along the D-V axis in close register with the boundaries of somites, resulting in reiterated cell arrangements that became evident along the neuraxis. Furthermore, AmphiHu/Hel-positive cell clusters appeared one by one along with the development of underlying somites. Double-staining in situ hybridization analysis with the islet gene revealed that the cell clusters contain presumptive motoneurons. In addition, AmphiHu/Hel expression was also observed outside the CNS, probably in the epithelial ectoderm, suggesting that amphioxus has a large number of putative sensory cell precursors as early as the early neurula stage. We investigated whether expressions of MHC class I and class II antigens relevant to tumor antigen presentation were changed on human tumor cells cultured with or without recombinant human IL-12(rhIL-12). We showed that the expression of MHC class I antigen on UTC-8, 28-1Cl and SBC-3 cells was augmented when these cancer cells were cultured with rhIL-12. The expression of class II antigen was slightly raised on UTC-8 and 28-1Cl cells by rhIL-12, but not enhanced on SBC-3 cells. These results suggest that rhIL-12 may provide possible enhancement of immunologic tumor recognition, and cytotoxic activity of lymphocytes against tumors through the enhanced expression of MHC class I antigen.

Purpose: We evaluated the usefulness of helical CT in the preoperative assessment of ruptured cerebral aneurysms during the acute stage of subarachnoid hemorrhage (SAH)., Methods: Nine patients with 13 aneurysms were included in this study. Helical CT scanning was performed before preoperative angiography. The section thickness and the table-movement speed were 1-2 mm/sec. Helical CT scanning was started 20 sec after the start of injection of 100 ml of contrast agent at a rate of 3 ml/sec., Results: The average CT value of SAH was 51.9 Hounsfield units (HU) and that of the aneurysm was 191.7 HU. Nine of 13 aneurysms greater than 3.0 mm in diameter were identified by three-dimensional CT angiography (3DCTA). In seven cases, the information provided by 3DCTA images, maximum intensity projection images, and multiplanar images, was very useful in surgical planning, providing information concerning the configuration of the neck and relationships between the aneurysm and brain parenchyma. Four aneurysms in two cases were not depicted by 3DCTA because they were located outside the imaging volume. This problem was overcome by changing the imaging volume according to the extent of origin of SAH., Conclusion: We conclude that helical CT in patients with SAH is useful for surgical planning, providing valuable information that cannot be obtained by conventional angiography.

The findings of three-dimensional computed tomography (3DCT) and two-dimensional computed tomography (2DCT) with helical CT scanning were compared for 21 patients with maxillofacial bone fractures. The results of this study suggest that the 3DCT evaluation can be divided into 3 groups. The first group, in which 3DCT is superior to 2DCT, includes severe complicated midface fractures, for example, tripod fractures and complicated maxillary bone fractures. The second group, in which 3DCT is equal to 2DCT, includes simple fractures, for example, nasal bone fractures and isolated zygomatic fractures. In this group, patients and their families could easily understand the nature of the fracture and clinical course shown by 3DCT as compared with conventional X-ray and 2DCT. The third group, in which 3DCT is inferior to 2DCT, includes blowout fractures. Although 3DCT does not provide additional information is blowout fractures, helical scanning permits clear observation of multiplanar images without artifacts arising from metal prostheses by excluding lower slices during image reconstruction. We conclude that 3DCT provides useful information, especially in regard to the extent of complex fracture lines, as in tripod fractures.

In order to compare the effect of partial demineralization with root planing and partial demineralization of cementum with that of dentin on healing, the ultrastructural morphology of the interface between the layer of human periodontal ligament-derived, fibroblast-like cells (HPF) and the treated root surface was studied in an in vitro culture system. Sixty (60) pairs made from transversally-cut root slices, 500 microns thick, were obtained from extracted human periodontally diseased teeth. Thirty (30) pairs of the root slices were preliminarily root planed (RP). The remaining half were root planed and then partially demineralized in a solution of citric acid (RP+CA). The opposite surface of paired slices was made uniform by using either cementum or dentin. Consequently, all root slices were classified into four experimental groups: RP-cementum and RP-cementum pairs (group 1), RP-dentin and RP-dentin pairs (group 2), RP+CA-cementum and RP+CA-cementum pairs (group 3), and RP+CA-dentin and RP+CA-dentin pairs (group 4). Each pair of root slices was placed on the floor of a 35-mm culture dish. HPF were seeded at a concentration of 4 x 10(5) cells/dish. Co-cultures of HPF and the root slices were examined using phase contrast and electron microscope after 4, 6, and 10 weeks. Electron-dense material covered non-demineralized root surfaces and the lining cells in accumulating cell layers were oriented parallel to the root surface and attached to the material in groups 1 and 2.(ABSTRACT TRUNCATED AT 250 WORDS)

A case of cerebral cysticercosis is reported in a 49-year-old female who presented with headache, nausea, and vomiting. Lumbar CSF showed the findings of mild aseptic meningitis. CT scan revealed an enlargement of the left ambient cistern and a small cystic lesion in the left frontal cortex. MRI demonstrated multilobular cysts in the left ambient cistern with enhancement of their capsules, which compressed and distorted the midbrain. The cysts were surgically excised, and histological examination of the specimen disclosed characteristic features of cysticercus with viable larva. An excellent value of MRI in the diagnosis of intracisternal cysticercosis is emphasized.

The influence of vasospasm on the permeability of the major cerebral arteries was studied using horseradish peroxidase (HRP). Experimental vasospasm was produced in canine basilar arteries by successive injections, 2 days apart, of autologous blood into the cisterna magna. HRP was injected intravenously or intracisternally 48 hours after the second injection of autologous blood, and all animals were killed by perfusion fixation 60 minutes after the injection of HRP. The distribution of HRP was observed by transmission electron microscopy. In 10 dogs injected intracisternally with HRP, 5 control dogs demonstrated a moderate amount of HRP in the intermuscular space through the adventitia. Five dogs with subarachnoid hemorrhages (SAHs) showed a moderate amount of HRP in the intermuscular space. In 11 dogs injected intravenously with HRP, 5 control dogs showed no leakage of HRP into vessel walls. Six dogs with SAHs showed HRP in the interendothelial space. These findings suggest that, despite SAHs, HRP seems to be able to circulate in the cerebrospinal fluid and makes contact with the cerebral vessels. This study suggests the possibility that spasmogenic substances may penetrate the vessel wall from the extraluminal side more easily than from the intraluminal side.

We performed arterial infusion chemotherapy and embolization on 22 patients with unresectable hepatocellular carcinoma via an intraarterial catheter, which was inserted in the hepatic artery under laparotomy. The administration frequency of intraarterial chemotherapy on a patient was 1 to 155 with a median of 17, while that of embolization was 1 to 6 with a median of 2.5. The maximal and average durations of a catheter in place were 15 and 4 months, respectively. The survival rates were 50% in one year, 18% in two years, and 12% in three years. Insertion technique and management of the catheter are relatively safe and easy, so catheterization of the hepatic artery would enable us to develop new treatment modalities.

Mice (C57BL/6) with established intradermal tumors (melanoma B-16 or glioma 203G1) and inbred Swiss mice (SWM/Ms) with established intradermal tumors (3-methylcholanthrene-induced sarcoma) were treated by either surgical excision or laser irradiation. Laser irradiation is effective in preventing the growth of dermal melanoma, and laser therapy may be more efficacious than surgical therapy for induction of a reaction against melanoma in mice. The results also suggest that determination of a suitable laser wavelength may be required to obtain a good therapeutic effect on each tumor.
