Abstract

Enterprises have paid attention to consortium blockchains like Hyperledger Fabric, which is one of the most promising platforms, for efficient decentralized transactions without depending on any particular organization. A consortium blockchain-based system will be typically built across multiple organizations. In such blockchain-based systems, system operations across multiple organizations in a decentralized manner are essential to maintain the value of introducing consortium blockchains. Decentralized system operations have recently been becoming realistic with the evolution of consortium blockchains. For instance, the release of Hyperledger Fabric v2.x, in which individual operational tasks for a blockchain network, such as command execution of configuration change of channels (Fabric's sub-networks) and upgrade of chaincodes (Fabric's smart contracts), can be partially executed in a decentralized manner. However, the operations workflows also include the preceding procedure of pre-sharing, coordinating, and pre-agreeing the operational information (e.g., configuration parameters) among organizations, after which operation executions can be conducted, and this preceding procedure relies on costly manual tasks. To realize efficient decentralized operations workflows for consortium blockchain-based systems in general, we propose a decentralized inter-organizational operations method that we call Operations Smart Contract (OpsSC), which defines an operations workflow as a smart contract. Furthermore, we design and implement OpsSC for blockchain network operations with Hyperledger Fabric v2.x. This paper presents OpsSC for operating channels and chaincodes, which are essential for managing the blockchain networks, through clarifying detailed workflows of those operations. The implementation of OpsSC has been open-sourced and registered as one of Hyperledger Labs projects.

Abstract

The purpose of this study was to adapt a Japanese version of the Big Five Inventory-2 (BFI-2-J) to examine its factor structure, reliability, validity, and measurement invariance. The BFI-2-J assesses five domains and 15 facets of the Big Five personality traits. We analyzed two datasets: 487 Japanese undergraduates and 500 Japanese adults. The results of the principal component analysis and confirmatory factor analysis revealed that the domain-facet structure of the BFI-2-J was similar to that of other language versions. The reliability of the BFI-2-J is sufficient. The correlation coefficients between the BFI-2-J and the other Big Five and self-esteem measures supported convergent and discriminant validity. Moreover, we confirmed measurement invariance across age and sex groups in domain-level and facet-level models. The results suggest that the BFI-2-J is a good instrument for measuring the Big Five personality traits and their facets in Japan. The BFI-2-J is expected to be useful in Japanese personality research and international comparative research.

Abstract

Recently, enterprises have paid attention to permissioned blockchain (BC), where business transactions among inter-authorized organizations (forming a consortium) can automatically be executed on the basis of a distributed consensus protocol, and applications of BC have expanded as permissioned BC has adopted the features of the smart contract (SC), which is programmable user-defined business logic deployed in BC and executed with the consensus protocol. A single BC-based system will be built across multiple management domains (e.g., the data centers of each organization) having different operational policies (e.g., operational procedures, timing, etc.); although establishing system management and operations over BC-based systems (e.g., SC installation for updates) will be important for production uses, such multi-domain formation will trigger a problem in that executing system operations over BC-based systems will become time-consuming and costly due to the difficulty in unifying and/or adjusting operational policies. Toward solving the problem, we propose an operations execution method for BC-based systems; the primary idea is to define operations as a smart contract so that unified and synchronized cross-organizational operations can be executed effectively by using BC-native features. For the recent BC architecture in which participating nodes have different types of roles, we designed the proposed method as a hybrid architecture characterized with in-BC consensus establishment and execution status management and out-BC operations execution for all types of nodes operated by agents that listen to triggered events including operational instructions defined in SCs. We implemented a prototype with Hyperledger Fabric v1.2.0. Abstract

Cell culture methods are indispensable strategies for studies in biological sciences and for drug discovery and testing. Most cell cultures have been developed using two-dimensional (2D) culture methods, but three-dimensional (3D) culture techniques enable the establishment of in vitro models that replicate various pathogenic conditions and they provide valuable insights into the pathophysiology of various diseases as well as more precise results in tests for drug efficacy. However, one difficulty in the use of 3D cultures is selection of the appropriate 3D cell culture technique for the study purpose among the various techniques ranging from the simplest single cell type-derived spheroid culture to the more sophisticated organoid cultures. In the simplest single cell type-derived spheroid cultures, there are also various scaffold-assisted methods such as hydrogel-assisted cultures, biofilm-assisted cultures, particle-assisted cultures, and magnet particle-assisted cultures, as well as non-assisted methods, such as static suspension cultures, floating cultures, and hanging drop cultures. Since each method can be differently influenced by various factors such as gravity force, buoyant force, centrifugal force, and magnetic force, in addition to non-physiological scaffolds, each method has its own advantages and disadvantages, and the methods have different suitable applications. We have been focusing on the use of a hanging drop culture method for modeling various non-cancerous and cancerous diseases because this technique is affected only by gravity force and buoyant force and is thus the simplest method among the various single cell type-derived spheroid culture methods. We have found that the biological natures of spheroids generated even by the simplest method of hanging drop cultures are completely different from those of 2D cultured cells. 50. Case Report: Structurally Rare EML4-ALK Identified by Next Generation Sequencing in a Patient with NSCLC with Bilateral Ovarian Metastases.

Abstract

The EML4-ALK oncogene is a fusion of the EML4 and ALK genes and is found in approximately 5– 6% of the cases of non-small cell lung cancer (NSCLC). Herein, we present a unique case of lung adenocarcinoma with metastases to the bilateral ovaries harboring a rare EML4-ALK fusion gene variant in a 52-year-old patient. The patient had initially received a diagnosis of ovarian cancer, then had undergone neo-adjuvant chemotherapy followed by a surgical resection. Despite two cycles of adjuvant chemotherapy consisting of carboplatin and gemcitabine, CT revealed that the pleural effusion had increased from it before chemotherapy, and the shortness of breath worsened. Molecular profiling revealed an EML4-ALK rearrangement containing ALK -EML4 and ALK -NPR2 fusion genes. The diagnosis was changed to primary lung adenocarcinoma with metastases to the bilateral ovaries based on a pathological reevaluation. Treatment with alectinib, a second-generation ALK-tyrosine kinase inhibitor, led to a partial response of 18 months' duration, and the shortness of breath improved. No adverse events related to the alectinib therapy occurred. To assess the unique structure of the fusion genes, RNA sequencing was performed. An intronic sequence from both ALK and EML4 was found between ALK and EML4 exon, possibly because of an unusual insertion of a gene fragment derived from NRP2, indicated by the panel sequencing results. Variations in the drug response among EML4-ALK fusion variants highlight the importance of understanding their molecular structure. Further investigation is warranted to refine fusion gene detection methods and assess the therapeutic implications of rare fusion variants. [ABSTRACT FROM AUTHOR]
