1. BET bromodomain inhibition potentiates radiosensitivity in models of H3K27-altered diffuse midline glioma

Abstract

Diffuse midline glioma (DMG) H3K27-altered is one of the most malignant childhood cancers. Radiation therapy remains the only effective treatment yet provides a 5-year survival rate of only 1%. Several clinical trials have attempted to enhance radiation antitumor activity using radiosensitizing agents, although none have been successful. Given this, there is a critical need for identifying effective therapeutics to enhance radiation sensitivity for the treatment of DMG. Using high-throughput radiosensitivity screening, we identified bromo- and extraterminal domain (BET) protein inhibitors as potent radiosensitizers in DMG cells. Genetic and pharmacologic inhibition of BET bromodomain activity reduced DMG cell proliferation and enhanced radiation-induced DNA damage by inhibiting DNA repair pathways. RNA-Seq and the CUT&RUN (cleavage under targets and release using nuclease) analysis showed that BET bromodomain inhibitors regulated the expression of DNA repair genes mediated by H3K27 acetylation at enhancers. BET bromodomain inhibitors enhanced DMG radiation response in patient-derived xenografts as well as genetically engineered mouse models. Abstract

In addition to conventional ground rovers, the Mars 2020 mission will send a helicopter to Mars. The copter's high-resolution data helps the rover to identify small hazards such as steps and pointy rocks, as well as providing rich textual information useful to predict perception performance. In this paper, we consider a three-agent system composed of a Mars rover, copter, and orbiter. The objective is to provide good localization to the rover by selecting an optimal path that minimizes the localization uncertainty accumulation during the rover's traverse. To achieve this goal, we quantify the localizability as a goodness measure associated with the map, and conduct a joint-space search over rover's path and copter's perceptual actions given prior information from the orbiter. We jointly address where to map by the copter and where to drive by the rover using the proposed iterative copter-rover path planner. We conducted numerical simulations using the map of Mars 2020 landing site to demonstrate the effectiveness of the proposed planner., Comment: 8 pages, 7 figures

Abstract

Background and Aim: Image enhancement endoscopy techniques, such as linked color imaging (LCI) and autofluorescence imaging (AFI), have shown promise in diagnosing mucosal inflammation in ulcerative colitis (UC). However, no studies have directly compared the diagnostic efficacy of LCI and AFI. This prospective observational study aimed to compare their diagnostic accuracy for histological healing in UC. Methods: This study included 81 UC patients, resulting in a total of 204 endoscopic images captured using LCI and AFI, respectively. Spearman's rank correlation coefficients assessed the correlation between LCI and AFI coloration and Geboes histopathology score (GHS). Six endoscopists, who were blinded to clinicopathological features, evaluated these images, and subsequently, the diagnostic accuracy was evaluated. Results: Spearman's rank correlation coefficients between LCI index, AFI index (reverse gamma value), and GHS were 0.324 and −0.428, respectively (P < 0.001), indicating a significant correlation between LCI and AFI coloration and histological healing. In LCI and AFI classifications, mean values for diagnostic accuracy, sensitivity, specificity, positive predictive value, and negative predictive value were 76.3 ± 2.2 versus 77.8 ± 2.7, 91.8 ± 4.0 versus 83.2 ± 7.6, 53.4 ± 10.0 versus 70.0 ± 5.3, 74.0 ± 3.5 versus 80.0 ± 1.6, and 82.9 ± 5.2 versus 75.5 ± 7.5, respectively. No significant difference in diagnostic accuracy existed between LCI and AFI classifications. However, LCI displayed higher sensitivity than AFI while AFI showed higher specificity compared with LCI (P < 0.05). Conclusions: LCI and AFI offer comparable diagnostic accuracy for histological healing. 22. Ansa Cervicalis External Branch Anastomosis to Restore High Tone Voice in Thyroid Surgery.

Abstract

Objectives: Injury to the external branch of the superior laryngeal nerve (EBSLN) causes low‐pitch voice and voice fatigue, particularly in female subjects, and available treatments are limited. Here, we assess a novel surgical procedure to restore a high‐tone voice: ansa cervicalis to EBSLN anastomosis (A‐E anastomosis). Methods: Between November 2012 and April 2022, 13 patients (12 female) underwent unilateral EBSLN resection and A‐E anastomosis, while 20 (16 female) underwent EBSLN resection during thyroid surgery. Patients (4494 women and 1025 men) with normal laryngoscopy scheduled for thyroid surgery served as normal controls. Phonatory function was examined using a Phonation Analyzer PA‐1000 preoperatively and intermittently postoperatively. Results: In patients who underwent A‐E anastomosis, high‐tone voice pitch decreased significantly postoperatively (673.9–471.5 Hz, p = 0.047), with restoration achieved within 5 months. The mean voice pitch in female patients who underwent A‐E anastomosis, EBSLN resection, and controls were 580.4, 522.8, and 682.0 Hz, respectively, indicating a significant decrease in EBSLN resection patients than controls (p = 0.002). The (mean – 1SD) of high‐tone voice pitch in female controls was 497 Hz; exceeding this may indicate recovery to a high‐tone voice. Overall, 73% (8/11) of A‐E anastomosis patients exceeded this value, which was marginally larger than the 43% (6/14) who underwent EBSLN resection. Data on male subjects are limited. There were no cases of adverse functional or cosmetic events. Conclusions: A‐E anastomosis, a novel simple procedure, restored high‐tone voice to some extent without any adverse events and thus warrants further investigation. Abstract

Centenarians have a decreased susceptibility to ageing-associated illnesses, chronic inflammation and infectious diseases.sup.1-3. Here we show that centenarians have a distinct gut microbiome that is enriched in microorganisms that are capable of generating unique secondary bile acids, including various isoforms of lithocholic acid (LCA): iso-, 3-oxo-, allo-, 3-oxoallo- and isoallolithocholic acid. Among these bile acids, the biosynthetic pathway for isoalloLCA had not been described previously. By screening 68 bacterial isolates from the faecal microbiota of a centenarian, we identified Odoribacteraceae strains as effective producers of isoalloLCA both in vitro and in vivo. Furthermore, we found that the enzymes 5[alpha]-reductase (5AR) and 3[beta]-hydroxysteroid dehydrogenase (3[beta]-HSDH) were responsible for the production of isoalloLCA. IsoalloLCA exerted potent antimicrobial effects against Gram-positive (but not Gram-negative) multidrug-resistant pathogens, including Clostridioides difficile and Enterococcus faecium. These findings suggest that the metabolism of specific bile acids may be involved in reducing the risk of infection with pathobionts, thereby potentially contributing to the maintenance of intestinal homeostasis. Abstract

Temperature dependence of pion and sigma-meson screening masses is evaluated by the Polyakov-loop extended Nambu--Jona-Lasinio model with the entanglement vertex (EPNJL model). We propose a practical way of calculating meson screening masses in the NJL-type effective models. The method based on the Pauli-Villars regularization solves the well-known difficulty that the evaluation of screening masses is not easy in the NJL-type effective models. The method is applied to analyze temperature dependence of pion screening masses calculated with state-of-the-art lattice simulations with success in reproducing the lattice QCD results. Abstract

We investigated the phase diagram of $SU(3)$ gauge theory in four dimension with one compact dimension by using the perturbative one-loop effective potential. Effects of the adjoint and fundamental fermions are investigated and then the rich phase structure in the quark-mass and compact-size scale is realized. Our results are qualitatively consistent with the recent lattice calculation and clearly show that the lattice calculation can be understood from the Hosotani mechanism. Abstract

We investigate chemical-potential ($\mu$) dependence of the static-quark free energies in both the real and imaginary $\mu$ regions, using the clover-improved two-flavor Wilson fermion action and the renormalization-group improved Iwasaki gauge action. Static-quark potentials are evaluated from Polyakov-loop correlators in the deconfinement phase and the imaginary $\mu=i\mu_{\rm I}$ region and extrapolated to the real $\mu$ region with analytic continuation. As the analytic continuation, the potential calculated at imaginary $\mu=i\mu_{\rm I}$ is expanded into a Taylor-expansion series of $i\mu_{\rm I}/T$ up to 4th order and the pure imaginary variable $i\mu_{\rm I}/T$ is replaced by the real one $\mu_{\rm R}/T$. At real $\mu$, the 4th-order term weakens $\mu$ dependence of the potential sizably. Also, the color-Debye screening mass is extracted from the color-singlet potential at imaginary $\mu$, and the mass is extrapolated to real $\mu$ by analytic continuation. Abstract

As the analytic continuation, the potential calculated at imaginary \mu=i\mu_{\rm I} is expanded into a Taylor-expansion series of i\mu_{\rm I}/T up to 4th order and the pure imaginary variable i\mu_{\rm I}/T is replaced by the real one \mu_{\rm R}/T. At real \mu, the 4th-order term weakens \mu dependence of the potential sizably. At long distance, all of the color singlet and non-singlet potentials tend to twice the single-quark free energy, indicating that the interactions between heavy quarks are fully color-screened for finite \mu. For both real and imaginary \mu, the color-singlet q{\bar q} and the color-antitriplet qq interaction are attractive, whereas the color-octet q{\bar q} and the color-sextet qq interaction are repulsive. The attractive interactions have stronger \mu/T dependence than the repulsive interactions. The color-Debye screening mass is extracted from the color-singlet potential at imaginary \mu, and the mass is extrapolated to real \mu by analytic continuation. Abstract

We determine the quark-hadron transition line in the whole region of temperature (T) and baryon-number chemical potential (\mu_B) from lattice QCD results and neutron-star mass measurements, making the quark-hadron hybrid model that is consistent with the two solid constraints. The quark part of the hybrid model is the Polyakov-loop extended Nambu-Jona-Lasinio (PNJL) model with entanglement vertex that reproduces lattice QCD results at mu_B/T=0, while the hadron part is the hadron resonance gas model with volume-exclusion effect that reproduces neutron-star mass measurements and the neutron-matter equation of state calculated from two- and three-nucleon forces based on the chiral effective field theory. The lower bound of the critical mu_B of the quark-hadron transition at zero T is mu_B = 1.6 GeV.
