Micro and nanoplastics (MNPs) are ubiquitous in the environment and have been detected in most ecosystems, including remote regions. The class of contaminants under the MNP umbrella is quite broad and encompasses variable polymer types, shapes, and sizes. Fibers are the most frequently detected in the environment, followed by fragments, but still represent only a small fraction of laboratory studies. Many toxicity studies have been done using polystyrene microbeads which represent neither the polymer nor shape most present in the environment. Additionally, most of these studies are done using virgin particles when the majority of MNP pollution is from secondary microplastics which have weathered and broken down over time. To address these data gaps, we exposed the model fish Inland Silverside, Menidia beryllina, for 21-days to micro and nano cryo-milled tire particles, micro and nano polylactic acid, and polyester microfibers, both weathered and unweathered treatments were tested. We evaluated the impacts of these particles on growth, behavior, and gene expression to compare the relative toxicities of the different particles. We found that overall, the nanoparticles and weathered fibers had the greatest effect on behavior and gene expression. Gene ontology analysis revealed strong evidence suggesting MNP exposure affected pathways involved in muscle contraction and function. Unweathered microfibers decreased growth which may be a result of food dilution. Our results also suggest that under weathering conditions polyester microfibers breakdown into smaller sizes and induce toxicity similar to nanoparticles. This study highlights the variable effects of MNPs in fish and emphasizes the importance of considering particle shape and size in toxicity studies.

Drinking water is one of numerous sources of human exposure to microscale and nanoscale plastic particles. Here, using a mouse model, we tested enteric and hepatic cellular responses to nanoplastic ingestion. At 1.5 or 25.5 h after an oral dose of 70 mg polystyrene nanospheres (PSNS)/kg (nominal diameters of 20 and 200 nm) in aqueous suspension female mice exhibit no overt signs of toxicity. Routine histopathology on small intestine and liver reveals no acute toxicity. Immunohistochemistry detects an increase in the number of enterocytes with cleaved caspase-3 (active form) after PSNS exposure (p ≤ 0.05) indicating progression toward lytic cell death via a proinflammatory pathway. This is not evident in liver after PSNS exposure. Transmission electron microscopy detects lytic cell death in enterocytes at 25.5 h after 200 nm PSNS exposure. Putative endosomes in liver appear to sequester 20 and 200 nm particles 25.5 h after exposure. Both 20 and 200 nm PSNS appear in putative perinuclear autolysosomes 25.5 h after treatment. No significant changes in gene expression in the small intestine or liver 25.5 h were observed after dosing, but there was a trend toward altered expression of cyp1b1 in the liver. Analysis of the fecal microbiome shows loss of diversity after exposure to both 20 and 200 nm particles after 25.5 h. Taken together, these results suggest risk from ingestion of nanoscale plastic particles from drinking water, which deserves systematic investigation.

Changing salinity in estuaries due to sea level rise and altered rainfall patterns, as a result of climate change, has the potential to influence the interactions of aquatic pollutants as well as to alter their toxicity. From a chemical property point of view, ionic concentration can increase the octanol–water partition coefficient and thus decrease the water solubility of a compound. Biologically, organism physiology and enzyme metabolism are also altered at different salinities with implications for drug metabolism and toxic effects. This highlights the need to understand the influence of salinity on pesticide toxicity when assessing risk to estuarine and marine fishes, particularly considering that climate change is predicted to alter salinity regimes globally and many risk assessments and regulatory decisions are made using freshwater studies. Therefore, we exposed the Inland Silverside (Menidia beryllina) at an early life stage to seven commonly used pesticides at two salinities relevant to estuarine waters (5 PSU and 15 PSU). Triadimefon was the only compound to show a statistically significant increase in toxicity at the 15 PSU LC50. However, all compounds showed a decrease in LC50 values at the higher salinity, and all but one showed a decrease in the LC10 value. Many organisms rely on estuaries as nurseries and increased toxicity at higher salinities may mean that organisms in critical life stages of development are at risk of experiencing adverse, toxic effects. The differences in toxicity demonstrated here have important implications for organisms living within estuarine and marine ecosystems in the Anthropocene as climate change alters estuarine salinity regimes globally.
