Abstract

Sago is a starch producer extracted from the sago tree (Metroxylon sp.), which is composed of two important fractions, namely amylose and amylopectin. Sago starch is a low-cost carbon source and is widely used in various applications. In this study, we aimed to chemically modify the different concentrations of sago starch through hydrolysis with hydrochloric acid. Acid hydrolysis was performed with different molarity of hydrochloric acid, and the hydrolysate was measured using dinitro salicylic acid (DNS) methods. Exopolysaccharide (EPS) production was conducted using modified MRS broth with hydrolysate sago starch as the only carbon source, compared to glucose and sucrose by fermentation Lactobacillus plantarum FU0811. Total carbohydrate was measured using the phenol-sulfuric method. In general, the highest sago starch hydrolysate (77 g/l) was obtained by sago starch with a concentration 10 mg/l and molarity of 0.5 M with an incubation time of 48 hours. EPS yield produced in sago starch hydrolysate was not significantly different compared to glucose and sucrose as a carbon source, ranging from 400-600 mg/l. Based on our results, the acid hydrolysis process will produce simple chain carbohydrates that can be used as a carbon source in the fermentation of exopolysaccharide-producing bacteria and are comparable to other commercial monosaccharides. Abstract

Naturally acquired immunity to the different types of malaria in humans occurs in areas of endemic transmission and results in asymptomatic infection of peripheral blood. The current study examined the possibility of naturally acquired immunity in Bornean orangutans, Pongo pygmaeus , exposed to endemic Plasmodium pitheci malaria. A total of 2140 peripheral blood samples were collected between January 2017 and December 2022 from a cohort of 135 orangutans housed at a natural forested Rescue and Rehabilitation Centre in West Kalimantan, Indonesia. Each individual was observed for an average of 4.3 years during the study period. Blood samples were examined by microscopy and polymerase chain reaction for the presence of plasmodial parasites. Infection rates and parasitaemia levels were measured among age groups and all 20 documented clinical malaria cases were reviewed to estimate the incidence of illness and risk ratios among age groups. A case group of all 17 individuals that had experienced clinical malaria and a control group of 34 individuals having an event of >2000 parasites μL−1 blood but with no outward or clinical sign of illness were studied. Immature orangutans had higher-grade and more frequent parasitaemia events, but mature individuals were more likely to suffer from clinical malaria than juveniles. The case orangutans having patent clinical malaria were 256 times more likely to have had no parasitaemia event in the prior year relative to asymptomatic control orangutans. The findings are consistent with rapidly acquired immunity to P. pitheci illness among orangutans that wanes without re-exposure to the pathogen. [ABSTRACT FROM AUTHOR]

Abstract

Lactic acid bacteria (LAB) have been widely known as group of exopolysaccharides (EPS) producers which is beneficial for gut health. This study aimed to obtain potential LAB as EPS producers from a laboratory collection of a total of 18 bacterial isolates from human feces. Only seven isolates that observed well grown in sago-starch based medium. The quantification of EPS yield was conducted in two different media and assessed using the phenol-sulfuric acid methods while amylase activity was measured by starch-iodine assay. The yield of EPS was varied from 155.8±7-574.4±4.5 mg/L and from 26.7±4.1-438.8±20 mg/L in glucose and sago starch-based medium, respectively. The amylase activity was ranged from 0.6-2.04 mU/mL. The 16S rRNA analysis showed that those isolates identified as Leuconostoc lactis MC01, Streptococcus equinus MH01 and Enterococcus faecium ME03, E. hirae MB01 and MB02 and E. faecalis ME02 and MK03. E. faecium ME02 and E. faecalis ME03 which showed the highest EPS yield in glucose and sago starch medium, respectively, may be potential for further development as EPS producers while Leuconostoc lactis MC01 which demonstrate the highest amylase activity may be potential as probiotic bacteria because they regarded as generally recognized as safe (GRAS) microorganism. Abstract

The severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) has caused the current pandemic of COVID-19 and is highly infectious with relatively high mortality risk. In this study, we reviewed and summarized miscellaneous COVID-19 cases from vulnerable groups, including children and those with comorbidity, reinfections, coinfections and vaccinated individuals, and rapid transmission cases to understand if there are any specific SARS-CoV-2 variants that infect those groups. We concluded that there were no specific variants associated with higher rates of infection within certain groups, rather, they may tend to infect any groups with any immunity levels, regardless of geographical boundaries. A high prevalence of SARS-CoV-2 infection is mainly driven by the ability of viruses with genetic markers that have been associated with higher risks of transmission and immune escape in vulnerable groups. Disease severity is not only influenced by the virus, but also host conditions such as gender, age, comorbidity, and obesity. The likelihood of new emerging SARS-CoV-2 variants resulting from infections in vulnerable groups is high, thus monitoring viral genetic markers and mutations is crucial for outbreak preparedness. Abstract

Escherichia coli sequence types 131 (ST131) and 1193 are multidrug-resistant extraintestinal pathogens that have recently spread epidemically among humans and are occasionally isolated from companion animals. This study characterized a nationwide collection of fluoroquinolone-resistant (FQR) E. coli isolates from extraintestinal infections in Australian cats and dogs. For this, 59 cat and dog FQR clinical E. coli isolates (representing 6.9% of an 855-isolate collection) underwent PCR-based phylotyping and whole-genome sequencing (WGS). Isolates from commensal-associated phylogenetic groups A (14/59, 24%) and B1 (18/59, 31%) were dominant, with ST224 (10/59, 17%), and ST744 (8/59, 14%) predominating. Less prevalent were phylogenetic groups D (12/59, 20%), with ST38 (8/59, 14%) predominating, and virulence-associated phylogenetic group B2 (7/59, 12%), with ST131 predominating (6/7, 86%) and no ST1193 isolates identified. In a WGS-based comparison of 20 cat and dog-source ST131 isolates with 188 reference human and animal ST131 isolates, the cat and dog-source isolates were phylogenetically diverse. Although cat and dog-source ST131 isolates exhibited some minor sub-clustering, most were closely related to human-source ST131 strains. Furthermore, the prevalence of ST131 as a cause of FQR infections in Australian companion animals was relatively constant between this study and the 5-year-earlier study of Platell et al. (2010) (9/125 isolates, 7.2%). Thus, although the high degree of clonal commonality among FQR clinical isolates from humans vs. companion animals suggests the possibility of bi-directional between-species transmission, the much higher reported prevalence of ST131 and ST1193 among FQR clinical isolates from humans as compared to companion animals suggests that companion animals are spillover hosts rather than being a primary reservoir for these lineages.

Abstract

Fluoroquinolones are often administered to pet rabbits given their perceived safety and limited effects on anaerobic gut microbiota. However, the pharmacokinetics and relative safety of pradofloxacin, a third‐generation veterinary fluoroquinolone with a much broader spectrum of activity, have not been reported in this species. Here, we determined the pharmacokinetic profile of a single dose of oral pradofloxacin in rabbits and evaluated effects on the faecal microbiome. Four mature female rabbits were administered pradofloxacin (25 mg/ml oral suspension), at a dose of 7.5 mg/kg. The pradofloxacin median (range) Tmax was 4.50 (2.00–5.00) h, Cmax 600.66 (395.85–886.72) ng/ml and t½ was 1.27 (0.12–1.39) h. These results indicated that oral absorption of pradofloxacin was slower, and elimination faster compared with other fluoroquinolones in healthy rabbits, as well as relative to cats and dogs. Following treatment with pradofloxacin, faecal microbiota profiling showed some compositional differences between treated and control animals. This was the result of a significant decrease in the abundance of Proteobacteria, in particular bacteria belonging to the Pseudomonas, Atopostipes and Parabacteroides genera. The pharmacokinetic profile of pradofloxacin in rabbits should be further studied by increasing the sample size and using multiple‐dose protocols (i.e. 7 days) to confirm safety. Further information on the effects of protein binding, higher dosages and disease on pradofloxacin pharmacokinetics in rabbits are needed before an accurate dosing regimen can be recommended. 23. Characterization of Staphylococcal Cassette Chromosome Elements from Methicillin-Resistant Infections in Australian Animals

Abstract

We examined the oxacillin resistance phenotype and genomic structure of staphylococcal cassette chromosome (SCC) elements from 77 veterinary methicillin-resistant (MRSP) isolates. Isolates were characterized by oxacillin broth microdilution, whole-genome sequencing, and bioformatics analysis. Five previously described SCC elements, and a sixth novel element, were identified: SCC III (also known as II-III), ΨSCC, and SCC (a SCC VII variant), all previously described in MRSP, and SCC IVg and SCC V, previously described in both methicillin-resistant (MRSA) and MRSP. The sixth element was novel and found among nine geographically clustered isolates. This novel pseudostaphylococcal cassette chromosome (ΨSCC) contained a class A gene complex but lacked genes. It also harbored heavy metal (cadmium) resistance determinants. The median oxacillin MIC values among ΨSCC, SCC III, and SCC V isolates were significantly higher than those determined for the SCC VII variant isolates and ΨSCC and SCC IVg isolates. ΨSCC was found exclusively in sequence type 497 (ST497), an MRSP clone that is locally successful in Victoria, Australia. Future studies are necessary to determine if this clone has disseminated further afield and if ΨSCC has moved into other MRSP lineages or staphylococcal species. is a significant veterinary pathogen and occasional cause of infections in humans. β-Lactams are an important group of antimicrobials used to treat staphylococcal infections in humans and animals. However, when staphylococci become methicillin resistant via the acquisition of a mobile genetic element called staphylococcal cassette chromosome (SCC), they become resistant to all β-lactams. This study detected a novel SCC element among a cluster of methicillin-resistant isolates from animals in Australia. It also detected SCC elements in that had high similarity to those identified in methicillin-resistant , demonstrating how human and animal pathogens can share the same resistance determinants.

Abstract

This study investigated the ecology, epidemiology and plasmid characteristics of extended-spectrum cephalosporin (ESC)-resistant E. coliin healthy pigs over a period of 4 years (2013–2016) following the withdrawal of ESCs. High carriage rates of ESC-resistant E. coliwere demonstrated in 2013 (86.6%) and 2014 (83.3%), compared to 2015 (22%) and 2016 (8.5%). ESC resistance identified among E. coliisolates was attributed to the carriage of an IncI1 ST-3 plasmid (pCTXM1-MU2) encoding blaCTXM-1. Genomic characterisation of selected E. coliisolates (n= 61) identified plasmid movement into multiple commensal E. coli(n= 22 STs). Major STs included ST10, ST5440, ST453, ST2514 and ST23. A subset of the isolates belong to the atypical enteropathogenic E. coli(aEPEC) pathotype that harboured multiple LEE pathogenic islands. pCTXM1-MU2 was similar (99% nt identity) to IncI1-ST3 plasmids reported from Europe, encoded resistance to aminoglycosides, sulphonamides and trimethoprim, and carried colicin Ib. pCTXM1-MU2 appears to be highly stable and readily transferable. This study demonstrates that ESC resistance may persist for a protracted period following removal of direct selection pressure, resulting in the emergence of ESC-resistance in both commensal E. coliand aEPEC isolates of potential significance to human and animal health.

Objective: The number of COVID-19 cases in Indonesia reflects the disease severity and rapid dissemination. In response to the mounting threat, SARS-CoV-2 genomic surveillance and the investigation of naso-oropharyngeal bacterial communities in West Java were conducted, as dysbiosis of the upper respiratory tract microbiota might adversely affect the clinical condition of patients., Methods: We utilized the Oxford Nanopore sequencing platform to analyze genetic variation of 43 samples of SARS-CoV-2 and 11 selected samples for 16S rRNA gene sequencing, using samples collected from May to August 2021., Results: The prevalence of AY.23 (>82%) predominated among five virus lineages in the populations (AY.23, AY.24, AY.26, AY.42, B.1.1.7). The region in the SARS-CoV-2 genome found to have the highest number of mutations was the spike (S) protein (>20%). There was no association between SARS-CoV-2 lineages, mutation frequency, patient profile, and COVID-19 rapid spread-categorized cases. There was no association of bacterial relative abundance, alpha-beta diversity, and linear discriminant analysis effect size analysis with patient profile and rapid spread cases. MetagenomeSeq analysis showed eight differential abundance species in individual patient profiles, including Pseudomonas aeruginosa and Haemophilus parainfluenzae ., Conclusions: The data demonstrated relevant AY.23 dominance (the Delta variant) in West Java during that period supporting the importance of surveillance program in monitoring disease progression. Abstract

Six strains of anaerobic bacteria, C13EG70 T , C13EG118, C13EG186 T , C13GAMG5, C13GAMG28 and C13GAMG40, were isolated from the caecum of a healthy chicken bred in Bogor, Indonesia. Phylogenetic analysis showed the isolates were separated into two groups. Group I (C13EG70 T and C13EG118) showed nearly identical 16S rRNA gene sequences (99.9 % sequence similarity). Group II (C13EG186 T , C13GAMG5, C13GAMG28 and C13GAMG40) showed nearly identical 16S rRNA gene sequences (>99.4 % sequence similarity). The isolates showed low 16S rRNA gene sequence similarities to recognized species of the genus Bacteroides . High gene sequence similarities were found between type strains (C13EG70 T and C13EG186 T ) and Bacteroides salanitronis JCM 13657 T (87.9, 91.5 %, respectively). Physiological, biochemical and genotypic characteristics demonstrated that these strains could be separated from the type strain of B. salanitronis . It is concluded that Group I and Group II represent novel species. Two novel species of the genus Bacteroides are proposed as Bacteroides caecicola sp. nov. (type strain C13EG70 T  = LIPI12-4-Ck732 T  = JSAT12-4-Ck732 T  = InaCC B449 T  = NBRC 110958 T ) and Bacteroides gallinaceum sp. nov. (type strain C13EG186 T  = LIPI12-4-Ck844 T  = JSAT12-4-Ck884 T  = InaCC B451 T  = NBRC 110963 T ).

Abstract

Three strains of anaerobic Gram-stain-negative, short to longer rod-shaped bacteria isolated from the caecum of chicken in Indonesia were studied using a polyphasic taxonomic approach. These strains belonged to the genus Bacteroides, based on sequence analysis of 16S rRNA and hsp60 (groEL) genes, with similarities of 93.2-94.1 and 89.8-90.8 %, respectively, to the closest recognized species, Bacteroides coprocola JCM 17929T. Sugar fermentation and enzyme characteristics, cellular fatty acid profiles, menaquinone profiles and metabolic end products were also investigated. Furthermore, DNA-DNA hybridization studies confirmed that the three novel strains are different from the closest related species. The strains were also found to be distinct from each other on the basis of ribotype profiles. The DNA G+C contents of the three strains were 41.1-41.8 mol%. Based on phenotypic and phylogenetic characteristics, a novel species, Bacteroides caecigallinarum sp. nov., is proposed (type strain C13EG111T = LIPI12-4-Ck773T = JSAT12-4-Ck773T = InaCC B455T = NBRC 110959T).
