Abstract

It is debated whether primary progressive apraxia of speech (PPAOS) and progressive agrammatic aphasia (PAA) belong to the same clinical spectrum, traditionally termed non-fluent/agrammatic variant primary progressive aphasia (nfvPPA), or exist as two completely distinct syndromic entities with specific pathologic/prognostic correlates. We analysed speech, language and disease severity features in a comprehensive cohort of patients with progressive motor speech impairment and/or agrammatism to ascertain evidence of naturally occurring, clinically meaningful non-overlapping syndromic entities (e.g. PPAOS and PAA) in our data. We also assessed if data-driven latent clinical dimensions with aetiologic/prognostic value could be identified. We included 98 participants, 43 of whom had an autopsy-confirmed neuropathological diagnosis. Speech pathologists assessed motor speech features indicative of dysarthria and apraxia of speech (AOS). Quantitative expressive/receptive agrammatism measures were obtained and compared with healthy controls. Baseline and longitudinal disease severity was evaluated using the Clinical Dementia Rating Sum of Boxes (CDR-SB). We investigated the data's clustering tendency and cluster stability to form robust symptom clusters and employed principal component analysis to extract data-driven latent clinical dimensions (LCD). The longitudinal CDR-SB change was estimated using linear mixed-effects models. Of the participants included in this study, 93 conformed to previously reported clinical profiles (75 with AOS and agrammatism, 12 PPAOS and six PAA). The remaining five participants were characterized by non-fluent speech, executive dysfunction and dysarthria without apraxia of speech or frank agrammatism. No baseline clinical features differentiated between frontotemporal lobar degeneration neuropathological subgroups. The Hopkins statistic demonstrated a low cluster tendency in the entire sample (0.45 with values near 0.5 indicating random data). Cluster stability analyses showed that only two robust subgroups (differing in agrammatism, executive dysfunction and overall disease severity) could be identified. Three data-driven components accounted for 71% of the variance [(i) severity-agrammatism; (ii) prominent AOS; and (iii) prominent dysarthria]. None of these data-driven LCDs allowed an accurate prediction of neuropathology. The severity-agrammatism component was an independent predictor of a faster CDR-SB increase in all the participants. Higher dysarthria severity, reduced words per minute and expressive and receptive agrammatism severity at baseline independently predicted accelerated disease progression. Our findings indicate that PPAOS and PAA, rather than exist as completely distinct syndromic entities, constitute a clinical continuum. In our cohort, splitting the nfvPPA spectrum into separate clinical phenotypes did not improve clinical-pathological correlations, stressing the need for new biological markers and consensus regarding updated terminology and clinical classification.

Abstract

Neural Architecture Search (NAS) methods have been successfully applied to image tasks with excellent results. However, NAS methods are often complex and tend to converge to local minima as soon as generated architectures seem to yield good results. This paper proposes GEA, a novel approach for guided NAS. GEA guides the evolution by exploring the search space by generating and evaluating several architectures in each generation at initialisation stage using a zero-proxy estimator, where only the highest-scoring architecture is trained and kept for the next generation. Subsequently, GEA continuously extracts knowledge about the search space without increased complexity by generating several off-springs from an existing architecture at each generation. More, GEA forces exploitation of the most performant architectures by descendant generation while simultaneously driving exploration through parent mutation and favouring younger architectures to the detriment of older ones. Experimental results demonstrate the effectiveness of the proposed method, and extensive ablation studies evaluate the importance of different parameters. 17. A Systematic Review of Behavioral Interventions for Elementary School Children with Social, Emotional and Behavioral Difficulties: Contributions from Single-Case Research Studies

Abstract

Challenges arising from the classroom behavioral management of students with social, emotional and behavioral difficulties are a concern for educational professionals. The purpose of this study is to review common elements of behavior interventions for the disruptive behaviors of children with social, emotional and behavioral difficulties. A systematic review was conducted through an electronic search of studies (from 2000 to 2017) on ERIC, Web of Science, FRANCIS, and MEDLINE databases. The inclusion criteria involved: (i) an intervention improving behaviors at school of children with disruptive behaviors; (ii) elementary school children with the majority of the sample or average age between 6-11 years old; (iii) at least one measurable outcome focusing on social/emotional/behavioral outcomes; (iv) single-case designs. Of the 5339 articles that were identified in the initial screening, 27 met the criteria to be included in the review. Common characteristics of successful interventions are discussed to make recommendations for future implementation.

Abstract

Background: The COVID-19 pandemic has affected education at all levels in various ways. This paper provides a review of the literature on the challenges of online learning for children with special educational needs and disabilities (SEND). Method: In total, 17 studies from nine countries were analysed. The challenges of online learning for children with SEND reported by teachers and parents and the strategies applied to overcome the challenges were identified. Results: The challenges of online learning were the need of parental support, routine change, inequities of resources and access to technology, lack of accommodations, and social isolation. There was a considerable lack of interventions aimed at promoting the educational outcomes of children with SEND. Conclusions: The findings emphasise the importance of parent-teacher collaboration and communication. Suggestions for strategies to address challenges as well as for further research are also discussed.

Abstract

Introdução: A saúde mental da mulher é influenciada por particularidades e variações hormonais que podem predispor a diversos transtornos mentais. Com base na evidência das práticas de meditação de atenção plena (mindfulness) em populações gerais, estas intervenções têm o potencial de modular a psicopatologia feminina, promovendo melhoria na saúde mental. Objetivos: Este artigo tem por objetivo fazer uma revisão narrativa avaliando a relação entre a prática de atenção plena e a saúde mental da mulher. Métodos: Foram consultadas as bases de dados Pubmed, MEDLINE, Embase e Cochrane, selecionando os artigos em inglês publicados entre 2013 e 2023. Resultados e Discussão: Os resultados dos estudos incluídos nesta revisão apontam para a eficácia da atenção plena nos processos de manejo de estresse e na promoção da regulação emocional em mulheres. As práticas de meditação de atenção plena foram consistentemente associadas a uma atenuação nos sintomas de condições específicas à saúde mental da mulher, tais como: transtorno disfórico pré-menstrual; depressão, estresse e ansiedade na gestação e pós-parto; estresse na perimenopausa; disfunções sexuais e transtorno alimentar, especialmente episódios de compulsão alimentar e comer emocional. Evidenciam-se ainda algumas fragilidades metodológicas das pesquisas sobre meditação atenção plena em mulheres. Conclusão: A prática de mindfulness evidencia-se como uma intervenção benéfica e com amplas aplicações na saúde mental feminina. Destaca-se o impacto positivo no manejo de patologias associadas ao ciclo reprodutivo e nos transtornos alimentares, recomendando-se a expansão de pesquisas com melhor qualidade metodológica que tenham como enfoque os efeitos terapêuticos no espectro da saúde mental da mulher.

Abstract

In this paper, we propose G-EA, a novel approach for guided evolutionary NAS. The rationale behind G-EA, is to explore the search space by generating and evaluating several architectures in each generation at initialization stage using a zero-proxy estimator, where only the highest-scoring network is trained and kept for the next generation. This evaluation at initialization stage allows continuous extraction of knowledge from the search space without increasing computation, thus allowing the search to be efficiently guided. Moreover, G-EA forces exploitation of the most performant networks by descendant generation while at the same time forcing exploration by parent mutation and by favouring younger architectures to the detriment of older ones. Experimental results demonstrate the effectiveness of the proposed method, showing that G-EA achieves state-of-the-art results in NAS-Bench-201 search space in CIFAR-10, CIFAR-100 and ImageNet16-120, with mean accuracies of 93.98%, 72.12% and 45.94% respectively., Comment: Paper accepted at 35th Conference on Neural Information Processing Systems (NeurIPS) - New In ML. 9 pages, 2 figures, 1 table

29. Role of DNA replication timing in gene expression and chromatin organisation

Abstract

Eukaryotic DNA is duplicated according to an evolutionary conserved temporal pattern. This pattern of DNA replication is altered during development and differentiation and can be dysregulated in cancers. While temporal changes in genome duplication are associated with altered transcription and chromatin organisation, it is still unknown whether DNA replication timing (RT) is a cause or a consequence of cellular fate changes. During my thesis I used a conditional system to perturb DNA RT in a single cell cycle in budding yeast, in combination with whole-genome sequencing techniques such as replication profiles, RNA-Seq and MNase-Seq in order to understand the biological importance of a defined pattern of genome replication and the impact on the genome structure and function. Overall, dramatic changes in gene expression, chromatin structure and transcription-factor (TF) binding events were observed, and a significant number of genes affected are involved in differentiation processes such as sporulation. While some differentially expressed genes showed significant chromatin changes, there were also examples where this was not the case, as well as genes with changes in chromatin and no changes in expression, which illustrates the complex nature of the relationship between RT, gene expression and chromatin. Differential TF binding events explained some of the observed changes, supporting a role for RT to maintain the correct TF binding dynamics during S-phase. Additionally, the fact that budding yeast origins are defined by specific sequences allowed the local modulation of RT which showed a direct effect of RT on gene expression. Altogether, the work generated during this thesis provides insight into the complex relationship between replication timing, gene expression and the chromatin landscape.

Abstract

BackgroundCortical mean diffusivity is a novel imaging metric sensitive to early changes in neurodegenerative syndromes. Higher cortical mean diffusivity values reflect microstructural disorganization and have been proposed as a sensitive biomarker that might antedate macroscopic cortical changes. We aimed to test the hypothesis that cortical mean diffusivity is more sensitive than cortical thickness to detect cortical changes in primary progressive aphasia (PPA).MethodsIn this multicenter, case-control study, we recruited 120 patients with PPA (52 non-fluent, 31 semantic, and 32 logopenic variants; and 5 GRN-related PPA) as well as 89 controls from three centers. The 3-Tesla MRI protocol included structural and diffusion-weighted sequences. Disease severity was assessed with the Clinical Dementia Rating scale. Cortical thickness and cortical mean diffusivity were computed using a surface-based approach.ResultsThe comparison between each PPA variant and controls revealed cortical mean diffusivity increases and cortical thinning in overlapping regions, reflecting the canonical loci of neurodegeneration of each variant. Importantly, cortical mean diffusivity increases also expanded to other PPA-related areas and correlated with disease severity in all PPA groups. Cortical mean diffusivity was also increased in patients with very mild PPA when only minimal cortical thinning was observed and showed a good correlation with measures of disease severity.ConclusionsCortical mean diffusivity shows promise as a sensitive biomarker for the study of the neurodegeneration-related microstructural changes in PPA.

47. Brachypodium distachyon as a model to understand resistance to wheat root diseases

Abstract

The cultivation of wheat is an ever-increasing priority for the sustainability of food production worldwide, yet wheat is susceptible to various pathogens leading to considerable yield loss. Numerous studies have been performed to combine sources of resistance to these diseases without affecting yield. Most studies, however, are performed on visual infection phenotypes on aerial parts of plants, with considerable lack of disease studies on underground soil-borne diseases, like take-all and Fusarium root rot (FRR). Most wheat cultivars are considered susceptible to these diseases, with low differential in response. This factor combined with the high incidence of gene redundancy due to wheat's hexaploid genome, have resulted in a lack of knowledge on the genetic basis of resistance to root diseases of wheat. The purpose of this project was to further examine the model plant Brachypodium distachyon (Bd) to understand resistance to root diseases and identify potential resistance/susceptibility genes that play a significant role in response to FRR and take-all. The use of different Bd accessions lines, part of the recently re-sequenced Bd pangenome were used to examine responses to both diseases. A QTL analysis was performed in parent accessions of already developed Bd populations with promising resistance differential to FRR and Fusarium overall seedling death, demonstrating a novel source of resistance originating from chromosome 1 of Bd. A series of RNA-seq experiments were also conducted to characterize the response of susceptible line Bd21, to low concentrations of deoxynivalenol (DON) and reactive oxygen species (ROS). The results enabled definition of DON and DON-mediated ROS expression profiles within the plant in a non-toxic background. A characterisation of candidate genes involved in response to FRR and take-all was also performed, making use of Brachypodium T-DNA mutant lines, along with wheat Kronos TILLING mutant lines, establishing a model-to-crop system between Bd and wheat.

Abstract

Afirmar que se observan síntomas disruptivos en el actual modelo socioeconómico no significa que este esté acabado. Como señaló hace años Colin Crouch, la “extraña muerte del neoliberalismo”, sigue obstaculizando el surgimiento de un nuevo paradigma socioeconómico que refleja cambios sustanciales en el pensamiento humano. Desde esta perspectiva, se plantea avanzar en un nuevo paradigma social, económico y político que ilumine caminos alternativos para la organización productiva y el modelo de consumo y nos permita mantener un equilibrio estable con el mundo natural en el que vivimos. Esto pone de relieve la necesidad de ir más allá de supuestos tradicionales desarrollando formas innovadoras de entender complejos desafíos, para superar el antropocentrismo que ha dominado el pensamiento occidental en los últimos siglos. En este artículo se centran estas reflexiones, en una sociedad en transición, particularmente en el mundo del trabajo. Para, finalmente, introducir algunas prospecciones sobre lo que podría esperarse ante un cambio radical de paradigma socioeconómico, que contempla una paulatina desmercantilización del trabajo en respuesta a la emergencia planetaria que ya estamos viviendo.
