7. 'I feel good with my teachers'. The effects of positive teacher-student relationship on students’ self-esteem and perceptions about their future

Abstract

The school climate has long been acknowledged as a protective factor for children’s and adolescents’ well-being. In fact, research has demonstrated that learning environments based on positive relationships among various social actors (students, teachers, and parents) are essential for enhancing students’ global self-evaluations and fostering their positive adaptation. However, few studies have examined the role teachers’ play in students’ self-esteem and future expectations. To overcome this limitation present in the literature, particularly in the Italian context, this study investigated the effect of teachers-students relationship on students’ self-esteem and their expectations for the future. The study employed a sample of 287 high school students. The mediational models included the quality of the teachers-students relationship as the independent variable, students’ self-esteem as the mediating variable, and students’ future expectations as the dependent variable. The results indicated total mediation: positive relationships between teachers and students had a positive association with students’ self-esteem which, in turn, had a positive association with students’ future expectations. 11. OXYTOCIN RECEPTORS IN NEURODEVELOPMENTAL DISORDERS: INNOVATIVE APPROACHES FOR THE QUANTIFICATION OF SOPRAMOLECULAR RECEPTOR COMPLEXES

Abstract

In the brain, the oxytocinergic system is a key regulator of social behaviour. Oxytocin could therefore become a promising therapeutic molecule to treat behavioural alterations in neurodevelopmental disorders; however, in order to define its translational potential we need to understand more about the localization and signaling properties of its receptor (OTR) in the brain. The OTR is a G protein coupled receptor (GPCR) able to form homo or heterodimeric complexes with distinct pharmacological properties. Targeting GPCRs dimers in endogenous environments is often problematic: the techniques currently used to study them in native tissues can only prove molecular proximity between two protomers, not their actual dimerization. For these reasons, in my project I worked to develop new tools to study dimeric and monomeric OTRs in endogenous tissues. I characterized a series of bivalent ligands designed to pharmacologically target OTR homodimers, and started to develop an innovative ?Nanoruler? technique to label OTR homodimers in native tissues. In parallel I also run an extensive autoradiographic mapping of the OTR in mouse brains, looking for changes of OTR levels in a mouse model of neurodevelopmental disorder (Magel2-KO). Through my work I already found important sex and genotype-induced effects on OTR expression in Magel2-KO mice, and I showed that in some cases a neonatal OT treatment (that can rescue some autistic traits of this model) modifies OTR levels in several regions. Once the Nanoruler technique is fully optimized we will compare its results with this mapping, to gather new important information about the dynamics of OTR distribution in mouse brain.

Abstract

Aims : Brugada syndrome (BrS) is associated with an increased risk of sudden cardiac death due to ventricular tachycardia/fibrillation (VT/VF) in young, otherwise healthy individuals. Despite SCN5A being the most commonly known mutated gene to date, the genotype-phenotype relationship is poorly understood and remains uncertain. This study aimed to elucidate the genotype-phenotype correlation in BrS. Methods and results: Brugada syndrome probands deemed at high risk of future arrhythmic events underwent genetic testing and phenotype characterization by the means of epicardial arrhythmogenic substrate (AS) mapping, and were divided into two groups according to the presence or absence of SCN5A mutation. Two-hundred probands (160 males, 80%; mean age 42.6 ± 12.2 years) were included in this study. Patients harbouring SCN5A mutations exhibited a spontaneous type 1 pattern and experienced aborted cardiac arrest or spontaneous VT/VF more frequently than the other subjects. SCN5A-positive patients exhibited a larger epicardial AS area, more prolonged electrograms and more frequently observed non-invasive late potentials. The presence of an SCN5A mutation explained >26% of the variation in the epicardial AS area and was the strongest predictor of a large epicardial area. Conclusion : In BrS, the genetic background is the main determinant for the extent of the electrophysiological abnormalities. SCN5A mutation carriers exhibit more pronounced epicardial electrical abnormalities and a more aggressive clinical presentation. These results contribute to the understanding of the genetic determinants of the BrS phenotypic expression and provide possible explanations for the varying degrees of disease expression.

13. Simulation of equinus gait and effects of ankle stiffness compensation with a neuromusculoskeletal model

Abstract

Introduction: Pes equinus with increased ankle joint stiffness is a common impairment in stroke and Cerebral Palsy (CP) patients with structural and/or neural deficits of the ankle muscles. Equinus gait is characterized by toe-strike, abnormal ankle plantarflexion and decreased ankle range of motion (ROM). A new ankle-foot orthosis (AFO) with negative stiffness was previously developed to compensate for the increased ankle joint stiffness and improve equinus gait, reducing plantarflexion and increasing ankle ROM. The goal of this project was to predict the effects of ankle stiffness compensation on equinus gait simulations. Methods: Forward simulations of unimpaired and equinus gait were generated in the sagittal plane with a musculoskeletal model implemented in OpenSim and a neural controller implemented in SCONE. After validation of the unimpaired gait simulation, a sensitivity analysis on ankle kinematics was performed, introducing structural and/or neural alterations of the ankle muscles to achieve an equinus gait. Consecutively, equinus gait simulations were validated with previously collected data of CP patients. Finally, an AFO model was developed in OpenSim to simulate AFO-assisted gait. Results: The unimpaired gait simulation yielded realistic results, and was robust to all alterations, generating stable gaits. Shorter Gastrocnemius fiber length and/or increased plantarflexor muscles activations, with Tibialis Anterior weakness, had the largest effect on the ankle joint kinematics and resulted in realistic CP equinus gaits. Simulations of AFO-assisted gait resulted in reduced abnormal plantarflexion, and increased ankle ROM in the condition with shorter Gastrocnemius. Beta-arrestin and arrestin-3 not only promote the internalization of beta(sub-2)-adrenergic receptots (B2AR) but binds directly and stoichiometrically to clathrin as well. Beta-arrestin/arrestin chimeras which have defects in B2AR or clathrin binding exhibit reduced ability to promote B2AR endocytosis. Beta-arrestin serves as an adaptor in the endocytosis pathway.

Abstract

Aim: To describe clinical experience with the Straumann Orthosystem method for orthodontic anchorage. Material and Methods: The Straumann Orthosystem consists of a pure titanium implant, a healing cap with screw, and a set of burs and instruments for the insertion and removal of the implant. The elements of the recommended kit also include an octagonal plastic transfer, a laboratory analogue, and an octagonal steel cap for the laboratory phase. The step-by-step procedure is illustrated from diagnostic planning through the surgical phase. Conclusion: The Straumann Orthosystem represents a valid anchorage system for extraction and nonextraction cases, and it has proven to be an easy and noninvasive surgical technique that is comfortable for the patient. Abstract

We describe the world's first fully leadless cardiac resynchronization therapy pacing implant with transseptal approach, in a patient with pacemaker dependency, atrial fibrillation, ischemic hypokinetic cardiomyopathy, and a history of pocket infections. After lead extraction, we implanted a MicraTranscathether Pacing System® (Medtronic, Minneapolis, MN, USA) as the sole right ventricular pacemaker, and the WISECRT system (EBR Systems, Sunnyvale, CA, USA) to provide biventricular pacing. We performed the WISECRT implant procedure using the transseptal approach given the presence of a prosthetic aortic mechanical valve, achieving satisfactory periprocedural results. 49. Selective recruitment of arrestin-3 to clathrin coated pits upon stimulation of G proteincoupled receptors

Abstract

Nonvisual arrestins (arrestin-2 and arrestin-3) play critical roles in the desensitization and internalization of many G proteincoupled receptors. In vitro experiments have shown that both nonvisual arrestins bind with high and approximately comparable affinities to activated, phosphorylated forms of receptors. They also exhibit high affinity binding, again of comparable magnitude, to clathrin. Further, agonistpromoted internalization of many receptors has been found to be stimulated by exogenous overexpression of either arrestin2 or arrestin3. The existence of multiple arrestins raises the question whether stimulated receptors are selective for a specific endogenous arrestin under more physiological conditions. Here we address this question in RBL-2H3 cells, a cell line that expresses comparable levels of endogenous arrestin-2 and arrestin-3. When β2-adrenergic receptors are stably expressed in these cells the receptors internalize efficiently following agonist stimulation. However, by immunofluorescence microscopy we determine that only arrestin-3, but not arrestin-2, is rapidly recruited to clathrin coated pits upon receptor stimulation. Similarly, in RBL-2H3 cells that stably express physiological levels of m1AChR, the addition of carbachol selectively induces the localization of arrestin-3, but not arrestin-2, to coated pits. Thus, this work demonstrates coupling of G proteincoupled receptors to a specific nonvisual arrestin in an in vivo setting.
