In the analysis of water samples, the type of filtration membrane material can influence the recovery of Legionella species, although this issue has been poorly investigated. Filtration membranes (0.45 µm) from different materials and manufacturers (numbered as 1, 2, 3, 4, and 5) were compared: mixed cellulose esters (MCEs), nitrocellulose (NC), and polyethersulfone (PES). After membrane filtration of samples, filters were placed directly onto GVPC agar and incubated at 36 ± 2 °C. The highest mean counts of colony-forming units and colony sizes for Legionella pneumophila and Legionella anisa were obtained with PES filters (p < 0.001). All membranes placed on GVPC agar totally inhibited Escherichia coli and Enterococcus faecalis ATCC 19443 and ATCC 29212, whereas only the PES filter from manufacturer 3 (3-PES) totally inhibited Pseudomonas aeruginosa. PES membrane performance also differed according to the manufacturer, with 3-PES providing the best productivity and selectivity. In real water samples, 3-PES also produced a higher Legionella recovery and better inhibition of interfering microorganisms. These results support the use of PES membranes in methods where the filter is placed directly on the culture media and not only in procedures where membrane filtration is followed by a washing step (according to ISO 11731:2017).

Cognitive impairment is present in a broad spectrum of medical conditions and in aging. Here, we aimed to identify plasma proteins related to cognitive function in a sample of older adults with overweight/obesity and metabolic syndrome. A total of 129 subjects (mean age 64.7 years; 36% females) were grouped according to low (l-GCF, N=65) or high (h-GCF, N=64) global cognitive function and matched according to education, sex, age, and body mass index. Cognitive performance was assessed using neuropsychological tests. Plasma levels of 92 neurology-related proteins were assessed using a proximity extension assay. An elastic net regression analysis was used to identify proteins more associated with cognitive performance. Additionally, the protein expression levels were compared between the two groups by means of a t-test with false discovery rate correction. Pearson correlations were used to assess associations between the protein levels and scores from the neurocognitive tests. Six proteins (alpha-2-MRAP, HAGH, Siglec-9, MDGA1, IL12, and EDA2R) were identified as potential contributors to cognitive performance, remaining significantly increased in l-GCF compared to h-GCF participants after correction for multiple testing. Negative correlations (r= −0.23 to −0.18, i.e., lower protein levels, higher cognitive function) were found between global cognitive function and Siglec-9, NMNAT1, HAGH, LXN, gal-8, alpha-2-MRAP, IL12, PDGF-R-alpha, NAAA, EDA2R, CLEC1B, and LAT. Mini-mental state examination z scores showed the strongest correlations with protein levels, specifically negative correlations with CLEC1b, LXN, LAT, PLXNB3, NMNAT1, gal-8, HAGH, NAAA, CTSS, EZR, KYNU, MANF (r=−0.38 to −0.26) and a positive correlation with ADAM23 (r= 0.26). In summary, we identified several plasma proteins that were significantly associated with cognitive performance in older adults with obesity and metabolic syndrome, although further research is needed to replicate the results in larger samples and to include a predictive perspective.

Background: Marfan syndrome (MFS) is a rare disease that induces a connective tissue disorder. The variability of its manifestations makes difficult an unequivocal diagnosis.Results: In this paper, we propose a method for the analysis of genetic aggregation and segregation based on Mendel’s Laws to identify the inheritance pattern for a single-gene disease. The probabilistic method is combined with a classical Uniformly Most Powerful (U.M.P.) test for comparing composite hypothesis against a composite hypothesis, in order to establish whether a disease is caused by a single autosomal dominant gene or by a recessive one. A random decision rule could be used for small samples.Conclusions: This methodology was tested to evaluate the quality assurance of the clinical diagnosis in Marfan Syndrome that enables a more reliable decision combining a clinical criteria and a mathematical model.

In Brazil, ethanol has been used as an alternative fuel in both road and air (agricultural airplanes) transport sectors. Due to that ethanol is volatile, therefore flammable in standard conditions, its explosive characteristics need to be investigated. In this work, it was made a detailed experimental assessment of Lower Flammability Limits (LFLs) from QAV – 1/anhydrous ethanol/air mixtures at different temperature and pressure conditions, where QAV – 1 is Jet fuel – A1. For the measures, it was used a flammability apparatus build according to American Standard ASTM E – 681. The experimental measures from mixtures both pure and binary were compared with the classical Le Chatelier's rule. Besides, two analytical expressions were proposed. One related to the stoichiometric concentration and another related to the specific states (geometrical approximation) of each fuel. The Absolute Relative Error (ARE) was used to identify the differences between the actual values and calculated values. However, to evaluate the goodness of fit of both distributions, two important statistic parameters were used, that is; the Average Absolute Relative Error (AARE) and Chi-Square test χ2 for the full observed data. The results have shown that, for different pressure conditions, the effect of temperature increase on the LFLs is slight in the range from 100 to 150 °C. Also, LFLs from binary blends depend strongly on both nature of fuels and the size of the combustion chamber used. Additionally, it was observed that Le Chatelier's rule applied to experimental data distribution is more adequate for the prediction of LFLs, improving in less presence of fuel molecules. Orientador: Fresia Soledad Ricardi Torres Branco Tese (doutorado) - Universidade Estadual de Campinas, Instituto de Geociências Resumo: Neste estudo se analisou a composição taxonômica e as características fisionômicas do material foliar de sete amostras de serapilheira da floresta ripária da Estação Ecológica de Mogi Guaçú, Nordeste do estado de São Paulo, assim como de cinco acumulações de macro-restos vegetais que se encontram ao longo de rios meandrantes do Nordeste, Leste e litoral Sul do estado de São Paulo, com a finalidade de se obter novas pautas para a análise e interpretação das associações fósseis de angiospermas. Também se analisou a relação entre a proporção de espécies sem dentes de uma flora e a temperatura média anual (TMA) de um conjunto de 121 localidades da América do Sul, com o objetivo de se obter uma nova equação baseada na Análise da Margem Foliar para a região. Ao analisar o material foliar das amostras de serapilheira observamos que a relação entre o número de espécies e de folhas/folíolos, assim como a composição florística, depende da localidade, do habitat e da dispersão produzida pelo vento. Deste material foliar 60-95% pertence a 2-3 espécies arbóreas. A maior parte das espécies identificadas pertence às famílias Bignoniaceae, Euphorbiaceae, Fabaceae, Myrtaceae e Sapindaceae. Das espécies identificadas 55-70% são arbóreas e o 20-40% restante são trepadeiras, as quais representam 8-35% da riqueza total de espécies arbóreas e 5-32% da riqueza total de espécies de trepadeiras da área. As principais características fisionômicas do material foliar nos permitiram estimar a TMA e a precipitação média anual (PMA). Os valores da TMA obtidos subestimaram o valor real em 0.8-1.8 °C e o superestimaram em 1.5-4.8 °C, e os valores da PMA obtidos subestimaram o valor real em 11-188 mm e o superestimaram em 44-83 mm. As acumulações de macro-restos vegetais estudadas podem ser consideradas um registro de alta resolução de El Niño. O material foliar que está presente nestas acumulações depende da localidade, do habitat, e do transporte produzido pelo vento e pela água. Deste material foliar 70-85% pertence a 2-3 espécies arbóreas. A maior parte das espécies identificadas pertence às famílias Bignoniaceae, Fabaceae e Sapindaceae. Das espécies identificadas 70% são arbóreas e o 30% restante são trepadeiras, as quais representam 25-35% da riqueza total de espécies arbóreas e 23-32% da riqueza total de espécies de trepadeiras da área. As principais características fisionômicas deste material foliar permitiram estimar a TMA e a PMA para o momento de sua deposição. Os valores da TMA obtidos subestimaram o valor real em 0.5-1.3 °C e o superestimaram em 2.2 °C, e os valores da PMA obtidos subestimaram o valor real em 332-1070 mm. A análise da relação entre a proporção de espécies sem dentes e a TMA de um conjunto de 121 localidades da América do Sul nos permitiu gerar uma nova equação de regressão linear, baseada na Análise da Margem Foliar, mais apta para a reconstrução da TMA de floras fósseis cenozoicas da região Abstract: In this study, we analyzed the taxonomic composition and physiognomic features of the leaf material of seven samples of leaf litter of the riparian forest of Mogi Guaçu Ecological Station, Northeastern of the Sao Paulo State, Brazil, as well as five accumulations of plant macroremains that are found along meandering rivers of the Northeastern, Eastern and the South coastline of Sao Paulo State, with the purpose of obtaining new guidelines for the analysis and interpretation of the fossil associations angiosperms. We also analyzed the relationship in between the proportion of the untoothed species of a flora and the mean annual temperature (MAT) of a dataset of 121 localities the South America, in attempt to establishing a new equation based on the Leaf Margin Analysis for the whole region. When analyzing the foliar material from the leaf litter, we observed that the relationship in between the number of species and the number of leaves/leaflets, as well as the floristic composition, depends on the locality, habitat and wind-induced dispersion. The 60-95% of these leaf material belongs to 2-3 tree species. Most of the identified species belongs to the Bignoniaceae, Euphorbiaceae, Fabaceae, Myrtaceae and Sapindaceae families. The 55-70% of the identified species are from trees and the remaining 20-40% are climbing species. These species represent a 8-35% of all tree species richness and a 5-32% of all climbing species richness of the área. The main physiognomic features of the leaf material allowed us to estimate the MAT and the mean annual precipitation (MAP). The obtained MAT values underestimated the real value in 0.8-1.8 °C and overestimated in 1.5-4.8 °C, and the obtained MAP values underestimated the real value in 11-188 mm and overestimated in 44-83 mm. The accumulations of plant macroremains studied can be considered a high-resolution record of the El Niño. The leaf material that is present in these accumulations depends on the locality, habitat and transport produced by wind and water. The 70-85% of this leaf material belongs to 2-3 tree species. Most of them are identified as part of the Bignoniaceae, Fabaceae and Sapindaceae families. The 70% of the identified species are from tree and the remaining 30% are climbing species. Anabolic androgenic steroids (AAS) are synthetic compounds derived from testosterone, which are widely used in supraphysiological doses by people seeking an aesthetic effect. The objective of the present experiment was to evaluate the possible morphometric changes in the cardiac left ventricle caused by the administration of supraphysiological doses of the anabolic steroids testosterone cypionate and stanozolol in the hearts of young sedentary mice, to serve as a comparative parameter with young mice that were submitted to exercise. We have used 60 hearts of sedentary young Swiss mice, aged ∼ 90 days old (young-adult), with a body weight between 40 and 50 g. The animals were divided into three groups: the control group, the testosterone cypionate group, and the stanozolol group. For the analysis, 10 distinct sections of the apex, of the middle region, and of the base of the heart were selected, followed by an optical microscope measurement with a 2.5x magnification. The results obtained show an increase in both myocardial thickness and left ventricular cavity diameter in the two groups of male animals in relation to the control group; however, in females, an increase in the thickness of the left ventricular myocardium was observed only for the stanozolol group. These results suggest that the cardiac alterations observed in the present study may be directly related to some signs and symptoms already described in the literature, such as hypertension, arrhythmias, infarction, sudden death, and other cardiovascular diseases.

No. of bitstreams: 0 Previous issue date: 2022-05-15 Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Waste heat emissions derived from cooling systems of the thermal power plant often discharged into rivers, lakes and seas, have been the direct cause of environmental problems for marine life. To control these wastewater discharges into the receptor water body, government regulations have been created to act as limiting factors for additional power generation from these plants. The problem can be solved with an organic Rankine cycle (ORC), which is considered a promising technology in electricity generation and an alternative to avoid the thermal pollution of aquatic ecosystems. The present work analyzes the use of an ORC system aiming to increase the thermal and overall efficiency of conventional operating systems without causing an additional thermal impact on marine species. This evaluation was based on the first and second laws of thermodynamics, applied to seven organic fluids (toluene, methanol, benzene, R11, R12, R113, R134a) and equations that linked the overall efficiency of the power plant with parameters of life quality in the rivers. Results showed that among the chosen organic fluids benzene produced the highest thermal efficiency for the ORC. Besides, an application of the proposed modelling in a thermal power plant localized near to Tubarão river, SC – Brazil was presented. Results showed that by using R113 as working fluid, it is possible to generate up to 1365.02 kWelec of additional electrical energy and increase both thermal and overall efficiency of a thermal power plant by up to 22.34% and 11.01%, respectively, without causing a thermal impact on the aquatic ecosystem. The best energy use was achieved by the recuperative ORC configuration. The Dissolved Oxygen Concentration (DOC) was reduced to 6.14 %day−1, which is consistent with the regulation of the Brazilian government regarding effluent discharges in lagoons, rivers and seas. On the other hand, considering the electricity cost at 0.16 US$kWh−1 and 5% as an annual percentage rate (APR), results in a payback period of approximately 5.3 years. La situación energética y medioambiental global hace que los sistemas de generación de energía basados en energías renovables sean cada vez más importantes. Estos sistemas tienen una doble ventaja, ya que permiten la diversificación energética y reducen la dependencia del petróleo, al tiempo que se reducen las emisiones globales de CO2. La biomasa es una de las fuentes de energía más prometedoras en los países desarrollados y emergentes, pero aún no experimenta el desarrollo esperado. El sistema de gasificación de biomasa es un sistema con gran potencial, ya que permite la generación de energía tanto térmica como eléctrica, a través de tecnologías de bajo costo, como gasificadores de lecho fijo (potencia media-baja), o reactores de lecho. fluidizado con tecnología más exigente y más capacidad de generación. Transformar la biomasa en un combustible gaseoso, utilizable en motores de combustión interna convencionales o turbinas de gas como combustible, la biomasa requiere varios pasos por procesos termoquímicos compuestos de reacciones endotérmicas (secado, volatilización y reducción) y exotérmicas (oxidación y carbón volátil). En este trabajo se calcula la temperatura de la llama adiabática con el objetivo de obtener la materia prima que produce una mayor energía térmica, utilizando para nuestro caso cinco tipos de biomasa como son: Eucalyptus Globulus o Blue Eucalyptus, Cane Bagasse (subproducto de la industria azucarera), Cascara de Arroz, Sequoia Gigante (residual) y Álamo Negro, con mayor énfasis en este último por considerarse una buena alternativa por su aparentemente rápido crecimiento (aprox. 2 años). De esta forma, una comparación de la energía térmica producida por los cinco tipos de biomasa utilizados.

The Brazilian aviation sector aiming to reduce its greenhouse emissions up to 37% by 2030 and up to 43% by 2050 (compared to 2005) using alternative fuels. For this reason, the evaluation of potential feedstock was made for the biojet fuel production focused in the Brazilian context. Four biomass types were proposed (Sugarcane, Jatropha, Soybeans and Eucalyptus), considering three factors: feedstock abundance (without negative impact in population foods), advances in conversion technology, and blend limit already approved by the ASTM International for the use of biojet fuels into fossil jet fuel. Based on this study, it is concluded that Brazil has a great number of available lands for the culture of feedstock from which aviation alternative fuel can be produced, with possible substitution of up to 10% vol. Of fossil jet fuel consumed in the country. However, conversion technologies are still a challenge. Only, Synthetic Paraffinic Kerosene (SPK) obtained by both Fischer Tropsch (FT) process and Alcohol to Jet (ATJ) process offer competitive prices compared to petroleum-based jet fuel. Considering sugarcane as the main feedstock, a self-sustained integrated process was evaluated aiming to increase the production performance of biojet fuel. Four biojet fuels were obtained by this process (ATJ – SPK, FT – SPK, Farnesane and Hexanol), allowing the reduction of up to 19.16% in the number of cultivated lands for sugarcane and up to 56.12% for its forest residues. Finally, the payload versus range ratio was described using the Breguet range equation applied to a possible commercial flight, taking into account all biojet fuels produced from the proposed feedstock including their blends.

Studies in the accumulation of plants developed in tropical forests promote understanding the origin of elements that integrate the accumulation time of layers, and the physical and chemical processes that interfere in their formation. Thus, studies were conducted in meandering rivers located in the state of Sao Paulo. Accumulations of leaves found in point bars were collected in the Mogi-Guacu, Capivari and Itanhaem (Branco, and Preto rivers) hydrographic basins and 14C dated. The leaves were classified in morphotypes and taxonomically organized. In all assemblages, the leaves belong to arboreal vegetation near the deposit with well-preserved leaves, being in decimetric thickness layers deposited along the 1950s, and possibly trend associated with years of high precipitation. The preservation of the assemblages is related to changes in the level of the water table and the chemical composition of water.

Curcumin has gained great attention in the last decades due to its fascinating properties for humans, such as anti-inflammatory or as cytotoxic against cancer. These effects are also claimed for pets such as cats and dogs, where curcumin administration is a daily practice routine. However, curcumin presents poor oral bioavailability, driving scientists to look for new delivery systems. In the last decades, several analytical methods for the quantification of curcumin in plasma have been published. To our knowledge there are no published reports on the effect of the level of hemolysis in the determination of this compound. In the present paper, a highly specific, sensitive and selective method is presented using Molecular Reaction Monitoring (SRM) using positive ionization (ESI+) mode. Curcumin and clopidogrel bisulfate – used as internal standard (IS) – were separated on an Acquity UPLC BECH Shield RP 18 column (1.7µm, 2.1 X 100mm) with 0.1% formic acid in acetonitrile and water in proportion of 60:40 (v/v). The analyte transitions were 369.3→177.06 m/z for curcumin and 322→212.05 m/z for IS. The method was fully validated and showed good linearity (R2 ≥ 0.999) over the range of 3-160 ng/mL. The Relative Standard Deviation (RSD) were less than 6% for intra-and inter-day analysis and recovery spanned 85-95%. We proved that the degree of hemolysis impaired curcumin quantitation. This method was applied to test curcumin bioavailability in both a mucoadhesive nanocapsule formulation and traditional capsules in dogs that attended routine veterinary consultation.Resumen. La curcumina ha ganado gran atención en las últimas décadas debido a sus propiedades terapéuticas para los humanos, como antiinflamatorio o citotóxico contra el cáncer. Estos efectos también se observan en pequeñas especies como gatos y perros, donde la administración de curcumina se ha vuelto una alternativa. Sin embargo, la curcumina presenta una baja biodisponibilidad oral, lo que impulsa a los científicos a buscar nuevos sistemas de administración. En las últimas décadas, se han publicado varios métodos analíticos para la cuantificación de curcumina en plasma. Actualmente, no hay informes publicados sobre el efecto del grado de hemólisis en la determinación de este compuesto. En este trabajo se desarrolló un método específico, sensible y selectivo utilizando el Monitoreo de reacción seleccionado (SRM) en modo de ionización positiva (ESI +). La curcumina y el bisulfato de clopidogrel, utilizado como patrón interno (IS), se separaron en una columna Acquity UPLC BECH Shield RP 18 (1,7 μm, 2,1 X 100 mm) con ácido fórmico al 0,1% en acetonitrilo y agua a una proporción de 60:40 (v/v). Las transiciones de los analitos fueron 369.3 → 177.06 m/z para curcumina y 322 → 212.05 m/z para IS. El método fue validado y demostró ser lineal (r2 ≥ 0.999) en el rango de 3-160 ng/mL. La desviación relativa estándar (RSD) fue inferior al 6% para el análisis intra e interdía y el porcentaje de recuperación fue 85-95%. Se descubrió que el grado de hemólisis afecta la cuantificación de curcumina. El método desarrollado se aplicó para evaluar la biodisponibilidad de curcumina tanto en una formulación de nanocápsulas mucoadhesivas como en cápsulas tradicionales en perros que asistieron a consultas veterinarias de rutina.

This paper presents an analysis of the basic organic Rankine cycle (ORC) using five types of working fluids: one wet fluid (R717), two isentropic fluids (R11 and R12) and two dry fluids (benzene and R113) with boiling points ranging from -33,35℃ to 79,85℃, to convert waste energy to power. These organic fluids are analyzed and compared, in the order to determine with which of them the best performance can be obtained. The evaluation was performed using a combined first and second law of thermodynamics and varying certain system operation parameters at saturation conditions. Results from these analyses have shown that the higher system thermal efficiency was obtained with the working fluids R11 and R113 compared with the others working fluids. In addition, the fluid that shows the best thermal efficiency is the one that the highest boiling point.

A long time ago, you have heard about accidents by gas explosions and the destruction that they can cause. To be able to avoid it requires an understanding of what a gas explosion is and what can reduce the frequency and consequences of these events. The upper and the lower flammability limits (UFL and LFL, respectively) are the maximum and the minimum concentration of the fuel in the air, respectively, in which a flame propagate, they are considered key tools in the prediction of fire, evaluating the possibility of explosion and design of protection systems. There is an interest in finding the flammability limits of ethanol mixed with an inert gas at reduced pressure as a safety measure for the future use of this biofuel in aeronautical applications, taking into account the typical altitude of a commercial aeroplane (

La presente investigación tuvo como objetivo general, determinar la relación que existe entre satisfacción y el clima laboral en el puesto de control migratorio de la Superintendencia Nacional de Migraciones en el Aeropuerto Internacional Jorge Chávez- Callao ,2015; bajo el enfoque de la Teoría de las necesidades de McClelland y la Teoría de los factores de Herzberger. El método empleado en la investigación fue el hipotético-deductivo. Se usó el diseño no experimental, nivel correlacional, de corte transversal, que recogió datos en un período determinado, siendo la población limitada a aquellos que realizan solo control migratorio, a citar, 192 trabajadores; el muestreo elegido fue el muestreo probabilístico, el tamaño de la muestra fue de 96 trabajadores y se aplicó dos instrumentos para estudiar las variables satisfacción y clima laboral : Cuestionario de satisfacción laboral, el cual estuvo constituido por 30 preguntas en la escala tipo Likert y el Cuestionario clima laboral, el cual estuvo integrado por 33 preguntas, también en escala tipo Likert. La investigación finalmente brinda información acerca de ambas variables, transitando desde el plano teórico al plano de lo empíricamente contrastable y cuyos resultados se muestran gráfica y textualmente, se concluye por tanto que existe correlación entre las variables en estudio, habiendo obteniendo un valor Rho de Spearman 0,339, el cual evidencia la existencia de una relación baja entre las variables. Este trabalho apresenta um estudo das vantagens da correcao local da energia reativa nas unidades consumidoras residenciais, buscando otimizar os reativos circulantes na instalacao local e na rede de distribuicao secundaria, visto que a carga de uma instalacao residencial tem baixo fator de potencia. Tal situacao acarreta elevadas perdas na rede de distribuicao das concessionarias e nas instalacoes eletricas residenciais, que normalmente sao subdimensionadas e exigidas ao maximo devido ao grande numero de equipamentos eletroeletronicos utilizados atualmente. A compensacao dos reativos pode evitar a tarifacao destes em uma eventual cobranca por parte das concessionarias. Neste trabalho e feito um breve estudo sobre a natureza da energia reativa e das cargas residenciais, bem como um estudo de caso, no qual foram analisadas as cargas mais comuns em uma residencia. Em seguida, foram apresentados os procedimentos de calculo e dimensionamento do compensador (capacitor), visando demonstrar as vantagens da correcao de reativo residencial, avaliando tanto a sua viabilidade economica quanto os impactos na reducao das perdas na rede de distribuicao. Considerando que, no caso estudado, com a correcao do fator de potencia foi obtida uma reducao de 9,85% da corrente, utilizando um controlador automatico e capacitores de 20uF, 10uF e 5uF. Dentre varias vantagens, esta reducao de corrente circulante no sistema permitira uma menor utilizacao das usinas geradoras e uma reducao nas perdas no sistema de transmissao e distribuicao. Esses sao dois entre varios fatores que justificam a correcao do fator de potencia para as residencias, beneficiando de forma significativa o sistema de geracao e transmissao de energia. Abstract This paper presents a study of the advantages of local correction of reactive power in the residential consumer units, seeking to optimize the reactive circulating in the local installation and the secondary distribution network, since the load of a residential facility has a low power factor, this leads to high losses in the distribution network of dealers and residential electrical installations which are usually undersized and required the most due to the large number of electronic equipment used today. The compensation of reactive can avoid these charges in an eventual recovery by concessionaires. It made a brief study of the nature of the reactive power and residential loads, as well as a case study which analyzes the most common charges in a residence. Then presents the procedures for calculation and dimensioning the compensator (capacitor), the aim is to demonstrate the advantages of correction reactive residential, evaluating both its economic viability as the impacts in reducing losses in the distribution network. Whereas, in the case studied, correcting the power factor was obtained a reduction of 9.85% of the current, using an automatic controller and 20uF, 10uF and 5uF capacitors. Among various advantages, this reduction in system circulating current allows a smaller use of power plants and a reduction of system losses in transmission and distribution. These were two of many factors that justify the correction of power factor for residential, benefiting significantly the system of generation and transmission of energy.
