Metal nanoclusters featuring tunable luminescence and high biocompatibility are receiving attention as fluorescent markers for cellular imaging. The recently discovered ability of gold clusters to scavenge cytotoxic reactive oxygen species (ROS) from the intracellular environment extends their applicability to biomedical theranostics and provides a novel platform for realizing multifunctional luminescent probes with engineered anti-cytotoxic activity for applications in bio-diagnostics and conceivably cellular therapy. This goal could be achieved by using clusters of strongly reactive metals such as silver, provided that strategies are found to enhance their luminescence while simultaneously enabling direct interaction between the metal atoms and the chemical surroundings. In this work, we demonstrate a synergic approach for realizing multifunctional metal clusters combining enhanced luminescence with strong and lasting ROS scavenging activity, based on the fabrication and in situ protection of Ag nanoclusters with a supramolecular mantle of thiolated-Au atoms (Ag/Au-t). Confocal imaging and viability measurements highlight the biocompatibility of Ag/Au-t and their suitability as fluorescent bio-markers. ROS concentration tests reveal the remarkable scavenging activity of Ag-based clusters. Proliferation tests of cells in artificially stressed culture conditions point out their prolonged anti-cytotoxic effect with respect to gold systems, ensuring positive cell proliferation rates even for long incubation time.

Intracellular pH is a key parameter in many biological mechanisms and cell metabolism and is used to detect and monitor cancer formation and brain or heart diseases. pH-sensing is typically performed by fluorescence microscopy using pH-responsive dyes. Accuracy is limited by the need for quantifying the absolute emission intensity in living biological samples. An alternative with a higher sensitivity and precision uses probes with a ratiometric response arising from the different pH-sensitivity of two emission channels of a single emitter. Current ratiometric probes are complex constructs suffering from instability and cross-readout due to their broad emission spectra. Here, we overcome such limitations using a single-particle ratiometric pH probe based on dot-in-bulk CdSe/CdS nanocrystals (NCs). These nanostructures feature two fully-separated narrow emissions with different pH sensitivity arising from radiative recombination of core- and shell-localized excitons. The core emission is nearly independent of the pH, whereas the shell luminescence increases in the 3-11 pH range, resulting in a cross-readout-free ratiometric response as strong as 600%. In vitro microscopy demonstrates that the ratiometric response in biologic media resembles the precalibralation curve obtained through far-field titration experiments. The NCs show good biocompatibility, enabling us to monitor in real-time the pH in living cells

Hybrid devices employing organic semiconductors interfaced with an aqueous solution represent a new frontier in bioelectronics and energy applications. Understanding of the energetics and photoinduced processes occurring at the organic/water interface is fundamental for further progress. Here, we investigate the interfacial electronic structure of poly-3-hexylthiophene (P3HT) sandwiched between an indium tin oxide (ITO) electrode and a liquid water electrolyte. The aqueous solution is found to polarize the polymer outermost layers, which together with the polymer p-(photo) doping by dissolved oxygen localizes photogenerated electrons at the P3HT/water interface, while holes can be transferred to the ITO electrode. Under illumination, the polymer/water interface is negatively charged, attracting positive ions from the electrolyte solution and perturbing the ion distribution in the aqueous solution. The observed mechanism is of general character and could underlie the behavior of a variety of devices characterized by an organic/water interface, such as prosthetic devices for artificial vision and organic-based systems for photoelectrochemical applications.

This paper reports a novel procedure using nanosomes, made of bola-hydroxyl and mercapto-palmitic acids, for the production of gold clusters with robust luminescent emissions and very large Stokes shifts. It shows that these results cannot be explained by the currently accepted mechanism based on ligand-to-metal charge transfer absorptions involving electron-rich ligands attached to the cluster core. Exhaustive characterization of the cluster samples using Mass Spectrometry, HR-TEM/STEM, XPS, EXAFS, and steady-state and time-resolved luminescence allows to deduce that a mixture of two cluster sizes, having non-closed shell electronic configurations, are firstly generated inside the nanosome compartments due to the difference in bonding strength of the two types of terminal groups in the fatty acids. This initial bimodal cluster size distribution slowly evolves into very stable, closed-shell Au cluster complexes (Au6-Au16 and Au5-Au14) responsible for the observed luminescent properties. The very small (≈1.2nm) synthesized cluster complexes are water soluble and suitable to be used for the conjugation of biomolecules (through the terminal COO- groups) making these systems very attractive as biomarkers and offering, at the same time, a novel general strategy of fabricating stable atom-level quantum dots with large Stokes shifts of great importance in many sensor applications.

Colloidal "dot-in-bulk" nanocrystals (DiB NCs) consist of a quantum confined core embedded into a bulklike shell of a larger energy gap. The first reported example of this class of nanostructures are CdSe/CdS DiB NCs that are capable of producing tunable two-color emission under both weak continuous-wave optical excitation and electrical charge injection. This property is a consequence of a Coulomb blockade mechanism, which slows down dramatically intraband relaxation of shell-localized holes when the core is already occupied by a hole. Here, we demonstrate electrochemical control of dual emission from DiB NCs. Spectro-electrochemical (SEC) experiments are used to tune and probe the photoluminescence (PL) intensity and branching between the core and the shell emission channels as a function of applied electrochemical potential (VEC). To interpret the SEC data we develop a model that describes the changes in the intensities of the shell and core PL bands by relating them to the occupancies of electron and hole traps. Specifically, application of negative electrochemical potentials under which the Fermi level is shifted upward in energy leads to passivation of electron traps at the surface of the CdS shell thereby increasing the total PL quantum yield by favoring the shell emission. Simultaneously, the emission color changes from red (VEC = 0) through yellow to green (VEC = -1). Time-resolved PL measurements indicate that as the Fermi level approaches the NC conduction band-edge electrons are injected into the NC quantized states, which leads to typical signatures of negative trions observed under optical excitation. Application of positive potentials leads to activation of electron traps, which quenches both core and shell PL and leads to the reduction of the overall PL quantum efficiency.
