Loss of function (LoF) of TAR-DNA binding protein 43 (TDP-43) and mis-localization, together with TDP-43-positive and hyperphosphorylated inclusions, are found in post-mortem tissue of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) patients, including those carrying LoF variants in the progranulin gene (GRN). Modeling TDP-43 pathology has been challenging in vivo and in vitro. We present a three-dimensional induced pluripotent stem cell (iPSC)-derived paradigm-mature brain organoids (mbOrg)-composed of cortical-like-astrocytes (iA) and neurons. When devoid of GRN, mbOrgs spontaneously recapitulate TDP-43 mis-localization, hyperphosphorylation, and LoF phenotypes. Mixing and matching genotypes in mbOrgs showed that GRN-/- iA are drivers for TDP-43 pathology. Finally, we rescued TDP-43 LoF by adding exogenous progranulin, demonstrating a link between TDP-43 LoF and progranulin expression. In conclusion, we present an iPSC-derived platform that shows striking features of human TDP-43 proteinopathy and provides a tool for the mechanistic modeling of TDP-43 pathology and patient-tailored therapeutic screening for FTD and ALS.

During recent years transformers architectures have been growing in popularity. Modulated Detection Transformer (MDETR) is an end-to-end multi-modal understanding model that performs tasks such as phase grounding, referring expression comprehension, referring expression segmentation, and visual question answering. One remarkable aspect of the model is the capacity to infer over classes that it was not previously trained for. In this work we explore the use of MDETR in a new task, action detection, without any previous training. We obtain quantitative results using the Atomic Visual Actions dataset. Although the model does not report the best performance in the task, we believe that it is an interesting finding. We show that it is possible to use a multi-modal model to tackle a task that it was not designed for. Finally, we believe that this line of research may lead into the generalization of MDETR in additional downstream tasks., Comment: 5 pages, 2 figures, 1 results chart

Objetivo: Describir las características clínicas, imagenológicas y la evolución de los pacientes trasplantados cardiacos por cardiotoxicidad inducida por antraciclinas. Métodos: Serie de casos descriptiva de pacientes consecutivos trasplantados cardiacos debido a cardiotoxicidad mediada por antraciclinas en el periodo de Noviembre de 2009 a Abril de 2021.Las características clínicas, los estudios complementarios pretrasplante y la información sobre la evolución posterior al trasplante fue recolectada de la historia clínica electrónica de forma retrospectiva. Resultados: Se incluyeron un total de 11 pacientes. La mediana de edad al diagnóstico de la patología oncológica fue de 15 años (RIC 10-37 años), mientras que la mediana de edad en la que recibieron el trasplante cardiaco fue de 56 años (RIC 39-62 años). Con respecto a la evolución posterior al trasplante, 3 pacientes murieron en el periodo del post operatorio inmediato. 1 paciente falleció a los 4 años del trasplante y los otros 7 pacientes tuvieron una evolución favorable. No se observó recaída oncológica en ningún paciente durante una mediana de seguimiento o de 2,5 años (RIC 1.86-3.85 años). Conclusión: La etapa final de la miocardiopatía inducida por antraciclinas puede ocurrir muchos años después del tratamiento con quimioterapia, por lo que es extremadamente importante un seguimiento cardiológico estricto. El trasplante cardiaco es una opción en este grupo de pacientes luego de una exhaustiva evaluación multidisciplinaria, con el fin de minimizar el riesgo de recaída oncológica.

The global challenge of feeding two billion more people by 2050, using more sustainable agricultural practices whilst dealing with uncertainties associated with environmental change, requires a transformation of food systems. We present a new perspective for how advances in network science can provide novel ways to better understand, harness, and restore multiple ecological processes in agricultural environments. We describe: (i) a network-focused framework for managing agro-ecosystems that accounts for the multiple interactions between biodiversity and associated ecosystem services; (ii) guidance for incorporating socio-economic factors into ecological networks; and (iii) the potential to upscale network methods to inform efforts to build resilience, including global food-supply chains. In doing so we aim to facilitate the application of network science as a systems-based way to tackle the challenges of securing an equitable distribution of food.

It is shown that under certain conditions it is possible to model a complex system in a way that leads to results that do not depend on system size. As an example of complex system an innovation diffusion model is considered. In that model a set of individuals (the agents), which are interconnected, must decide if adopt or not an innovation. The agents are connected in a member of the networks family known as small worlds networks (SWN). It is found that for a subfamily of the SWN the saturation time and the form of the adoption curve are invariants respect to the change in the size of the system., Comment: 10 pages

This paper progresses research on resistance in the context of financialisation by drawing on various social reproduction legacies. I explore how social reproduction theory offers conceptual and methodological tools that can deepen research on de‐financialisation and resistance against predatory finance. Expanding upon relational approaches to social reproduction, this paper frames "the financialisation of social reproduction" as the dialectics between the reproduction of financialised capitalism and the regeneration of life. This supports a perspective on resistance through the identification and analysis of the interplay between the objective and subjective limits to the financial extraction from life. I examine how methodologies like "counter‐topographies" can demystify financialisation while supporting the cultivation of alternative geographies of finance. Through "financial counter‐topographies", I explore how global reproductive entanglements contradictorily fragment working‐class interests across space while creating conditions to envision and cultivate de‐alienated financial landscapes. This paper investigates the esterase activity of minimalist amyloid fibers composed of short seven‐residue peptides, IHIHIHI (IH7) and IHIHIQI (IH7Q), with a particular focus on the role of the sixth residue position within the peptide sequence. Through computational simulations and analyses, we explore the molecular mechanisms underlying catalysis in these amyloid‐based enzymes. Contrary to initial hypotheses, our study reveals that the twist angle of the fiber, and thus the catalytic site's environment, is not notably affected by the sixth residue. Instead, the sixth residue interacts with the p‐nitrophenylacetate (pNPA) substrate, particularly through its −NO2 group, potentially enhancing catalysis. Quantum mechanics/molecular mechanics (QM/MM) simulations of the reaction mechanism suggest that the polarizing effect of glutamine enhances catalytic activity by forming a stabilizing network of hydrogen bonds with pNPA, leading to lower energy barriers and a more exergonic reaction. Our findings provide valuable insights into the intricate interplay between peptide sequence, structural arrangement, and catalytic function in amyloid‐based enzymes, offering potentially valuable information for the design and optimization of biomimetic catalysts. Emergency alert systems (EASs) have been deployed in some countries in order to broadcast emergency alerts and warning messages to the public. In this letter, we describe an innovative electronic embedded solution, named Firetec switch, intended to be placed in the FM radio stations. This switch will be supported by a server placed at the authorized officials, with the capability to generate the audio alerting messages. One interesting challenge is to deliver the messages only to the local public, in the catastrophe neighborhood, by using FM radio stations. Currently, research on microplastics (MPs) has increased due to their rapid distribution throughout the world and their harmful effects on the ecosystem. However, a detailed description of their dispersion and the methods for both detection and removal has not been given. The objective of this research is to carry out a bibliographic review that allows for a multidisciplinary analysis of microplastic contamination and current detection and removal methods. The method used is PRISMA in which articles from reliable databases such as Scopus, Web of science, and Google Scholar were collected and analyzed to finally provide details on the physical and chemical methods for detecting MPs, in addition to presenting the technologies for their removal. As a result of the analysis, critical information was obtained from the different studies on the impact of MPs on the ecosystem and the variation in detection and removal efficiency according to the type of pretreatment and methods applied to the sample. It is concluded that this research is essential to understand the consequences that MPs have on the ecosystem and provide tools to evaluate and improve current technologies, mainly detection and removal.

The “Lagunas de Montebello” National Park located in Chiapas, Mexico, is well known for its crystal blue water bodies, some of which, in 2003, started to change color from crystalline to cloudy brown, and occasionally emit a foul smell, contains white-yellowish supernatant debris and dead fish. To determine the causes of the changes in the water characteristics of the “Liquidambar” lagoon of the Montebello lagoon system, a physicochemical characterization was carried out over the first six meters of the water column, together with geochemical speciation analysis and the saturation index calculation for different minerals. Water was classified as calcium-sulfated and the main mechanism that controlled its chemistry was rocks dissolution. Sulfide was found at all sampled depths in the range of 0.11 to 1.13 mg.L-1. The concentration of sulfate in the water column ranged from 249.21 to 298.7 mg.L-1, carbonate ranged from 140.5 to 261.4 mg.L-1, calcium and magnesium ranged from 94.5 to 146.9 mg.L-1 and 34.2 to 38.3 mg.L-1, respectively. Likewise, oxygen was also found to be oversaturated on the surface with a value of 9.32 mg.L-1. The speciation results and SI indicated that the mineral phases calcite, aragonite, and dolomite were oversaturated, being greater on the surface. The results suggested the possibility that the turbidity, the coloration change, and the whitish supernatant were due to the precipitation of carbonate minerals, microbiologically influenced by the photosynthetic activity in the upper layer of the lagoon water.

- Abstract
The objective of this study was to compare the effectiveness of a second tumor necrosis factor inhibitor (TNFi) versus a non‐TNFi biologic following discontinuation of a TNFi for patients with polyarticular‐course juvenile idiopathic arthritis (pJIA). Using the Childhood Arthritis and Rheumatology Research Alliance Registry, patients with pJIA who started receiving a second biologic following a first TNFi were identified. Patients were required to have no active uveitis on the index date and a visit six months after the index date. Outcome measures included Clinical Juvenile Arthritis Disease Activity Score with a maximum of 10 active joints (cJADAS10), cJADAS10 inactive disease (ID; ≤2.5) and cJADAS10 minimal disease activity (MiDA; ≤5). Multiple imputation was used to account for missing data. Adjusted odds ratios (aORs) were calculated using propensity score quintiles to compare outcomes at six months following second biologic initiation. There were 216 patients included, 84% initially received etanercept, and most patients stopped receiving it because of its ineffectiveness (74%). A total of 183 (85%) started receiving a second TNFi, and 33 (15%) started receiving a non‐TNFi. Adalimumab was the most common second biologic received (71% overall, 84% of second TNFi), and tocilizumab was the most common non‐TNFi second biologic received (9% overall, 58% of non‐TNFi). There was no difference between receiving TNFi versus non‐TNFi in cJADAS10 ID (29% vs 25%; aOR 1.23, 95% confidence interval [CI] 0.47–3.20) or at least MiDA (43% vs 39%; aOR 1.11, 95% CI 0.47–2.62) at six months. Most patients with pJIA started receiving TNFi rather than non‐TNFi as their second biologic, and there were no differences in disease activity at six months.

ABSTRACT Bacurizeiro (Platonia insignis) is a fruit of the Amazon that, due to anthropic actions, has been suffering serious damage to its genetic diversity. We analyzed the genetic diversity and structure of seven bacurizeiro populations distributed among four Brazilian states and two biomes, the Amazon and Cerrado, based on eight nuclear (ncSSR) and three chloroplast (cpSSR) microsatellite markers. Higher heterozygosity values were found for the Northern populations of Japurá and Marapanim, and the Northeast population of Chapadinha when using ncSSR. Higher diversity indices were also observed for these populations with cpSSR, although higher haplotypic diversity parameters were detected for the two Northern populations. Genetic structure analysis showed the formation of two well defined groups: I - populations from Maranhão and Piauí (Cerrado) and II - populations from Amazonas and Pará (Amazon). Positive and significant correlations were identified between genetic and geographical distances for both ncSSR (r = 0.09; p = 0.0010) and cpSSR (r = 0.11; p = 0.0008), in agreement with the genetic structure analysis. The high genetic structure among populations probably reflects the divergent natural and human selection pressures to which bacurizeiros are subjected in both the Amazon and Cerrado biomes, with higher diversity maintained in the Amazon populations.

A micro-level agent-based model of innovation diffusion was developed that explicitly combines (a) an individual's perception of the advantages or relative utility derived from adoption, and (b) social influence from members of the individual's social network. The micro-model was used to simulate macro-level diffusion patterns emerging from different configurations of micro-model parameters. Micro-level simulation results matched very closely the adoption patterns predicted by the widely-used Bass macro-level model (Bass, 1969). For a portion of the domain, results from micro-simulations were consistent with aggregate-level adoption patterns reported in the literature. Induced Bass macro-level parameters and responded to changes in micro-parameters: (1) increased with the number of innovators and with the rate at which innovators are introduced; (2) increased with the probability of rewiring in small-world networks, as the characteristic path length decreases; and (3) an increase in the overall perceived utility of an innovation caused a corresponding increase in induced and values. Understanding micro to macro linkages can inform the design and assessment of marketing interventions on micro-variables - or processes related to them - to enhance adoption of future products or technologies., Comment: 20 pages, 4 figures and a table of supplementary data. The well-known Ising model used in statistical physics was adapted to a social dynamics context to simulate the adoption of a technological innovation. The model explicitly combines (a) an individual's perception of the advantages of an innovation and (b) social influence from members of the decision-maker's social network. The micro-level adoption dynamics are embedded into an agent-based model that allows exploration of macro-level patterns of technology diffusion throughout systems with different configurations (number and distributions of early adopters, social network topologies). In the present work we carry out many numerical simulations. We find that when the gap between the individual's perception of the options is high, the adoption speed increases if the dispersion of early adopters grows. Another test was based on changing the network topology by means of stochastic connections to a common opinion reference (hub), which resulted in an increment in the adoption speed. 50. Weakly Supervised Video Anomaly Detection Combining Deep Features with Shallow Neural Networks

Deep features have outgrown hand-sketched features in many applications. The availability of pre-trained deep feature extractors helps to overcome one of the deep learning main drawbacks, which is the need for large volumes of data for training. Multiple Instance Learning (MIL) has become an attractive solution for video surveillance literature once it allows working with weakly labeled bases. This work evaluates a video anomaly detection approach based on the MIL paradigm combining deep features with shallow Neural Networks. For computational efficiency, we apply Principal Component Analysis (PCA) for dimensionality reduction before classification. We performed the experiments from a set of I3D (Inflated 3D) features, which corresponds to the ShanghaiTech benchmark dataset, and the MLP and SVM shallow classifiers achieved competitive results.
