Natural fibres of plant and animal origin have been explored vastly over the last two decades and are gaining importance over synthetic counterparts owing to their biodegradability, renewability, lightweight and better biochemical and strength properties. Plant-based natural fibres such as banana, coir, sisal, jute, kenaf and many others have been studied for industrial applications. Among these, banana fibre is of major interest as banana is one of the most consumed fruit worldwide with annual production of 115.7 million tonnes in the year 2018 and is grown in 130 countries, which makes banana waste available locally. Various parts of banana plant such as fruit, fruit peel, flower bud, leaves and pseudo-stem (banana trunk) are utilized for different industrial purposes. Pseudo-stem is the major portion of banana waste biomass and yields good quality fibre that has the potential for industrial applications in the making of sanitary pads, textiles, pulp and paper, food and reinforced composite materials for automobiles, construction material, aerospace and other composite materials. Recently, Philippine Department of Science and Technology prepared masks from banana fibre which can cut the usage of single usage plastic. This review outlines various methods of extraction of fibre, biochemical and mechanical properties of banana fibre and its applications.

Coronavirus Disease-19 (COVID-19) is presently wreaking havoc on public health and socio-economic development. Besides the upper and lower respiratory tract involvement, gastrointestinal symptoms are also reported in COVID-19 patients through gut-lung axis. Finding its way through the feces of infected individuals and other sources, the genetic material of SARS-CoV-2 (ssRNA) is reported widely in wastewater and is being used as a fingerprint for its detection. With millions of cases arriving every day, there is a need to level up the testing speed efficiency. Due to the restricted sampling potential of testing laboratories, clinical testing is unable to track all the symptomatic and asymptomatic cases. Wastewater-based epidemiology (WBE) bestows an auxiliary monitoring tool that will contribute in community level screening. Sample collection, concentration, RNA extraction, quantification and data analysis are the main steps involved in implementation of WBE that can be relied upon as an alarm call for an upcoming wave, emergence of a new variant or any future pandemic. WBE can be a cheaper and more practical alternative to high end and sophisticated clinical testing for community transmission detection. Worldwide, there are more than 300 reports entailing the occurrence of SARS-CoV-2 in wastewater exhibiting unique temporal trends with five of them in India. This review aims to address the present knowledge on surveillance of SARS-CoV-2 in wastewater and its implications.

With the emergence of COVID-19 pandemic in 2019, the world saw a humungous loss of human life and economic resources globally but also the rapid appearance of SARS-CoV-2 variants which have exhibited a higher transmissibility and/or virulence and which also evade immune system to such an extent that it raises a big question mark on the efficacy of current diagnostics, vaccines and convalescent plasma and mAb therapies. This has been attributed to the emergence of huge spectrum of mutations, especially in the virus’s spike (S) protein, occurring in regions harboring high concentration of B cell epitopes thus allowing neutralizing antibody escape. The mutations resulting in ACE 2 receptor recognition failure (T19R), unfavorable electrostatic interactions (E484K), structural change (∆69-70), disruption of hydrogen bonds, salt bridges or hydrophobic interactions (K417N, N501Y, ∆Y145) and change in orientation (N501Y) cause strong immune evasion by these variants. Further, the recent emergence of Omicron with more than 30 mutations in the S protein VOC allows it to escape and fail diagnosis as well as immune system and the protection generated by different vaccination regimes. Yet Omicron may not be the end of the story. This review presents an insight of the immunity escape and its mechanisms followed by different SARS-CoV-2 variant of concerns.

COVID-19 pandemic continues to be a global threat, affecting more than 200 countries/territories at both human and economic level. This necessitates the rapid development of highly reliable diagnostic methods in order to effectively and accurately diagnose the pathology to prevent the spread of COVID-19. Currently, RT-PCR is the most widely used method worldwide for SARS-CoV-2 detection. Serological assays are being used for sero-surveys of SARS-CoV-2 antibody prevalence in the community. Radiology imaging has been useful in the clinical diagnosis of COVID-19. These methods have their own limitations and there are continued efforts to develop easier, economic, highly sensitive and specific, point-of-care methods. Reverse transcription-loop mediated isothermal amplification (RT-LAMP), nucleic acid sequence-based amplification (NASBA), CRISPR-Cas-based detection, and digital PCR are such techniques being employed in research laboratories, with many awaiting diagnostic approval from competent authorities. This review highlights the rapidly expanding array of existing and in-development diagnostic tests/strategies that may be used to diagnose SARS-CoV-2 infection in both clinical and research settings.

A bacterial laccase having potential to work in industry without mediator is of special interest. In this work, gene (1.83kb) encoding a novel laccase from Rheinheimera sp., having potential to deink waste paper without mediator, was cloned, over-expressed and the induced 69kDa protein (RhLacc) was purified and characterized. rhlacc gene was mutated by error prone PCR and mutants were sequenced. One mutant showed protein truncation resulting in the absence of domain 3 that contains T1 copper center. It is known that redox potential of T1 is the key parameter for substrate oxidation. Overexpression of this mutant gene showed induced 41.1kDa protein (∆RhLacc) that exhibited laccase activity but in the presence of added copper, compared to RhLacc which showed activity without added copper ions. Optimum temperature for both was 55°C. However, optimum pH varied with substrates. Kinetic studies showed ∆RhLacc had lower affinity for substrates except for guaiacol and reduced kcat in comparison to RhLacc. Both were able to deink old newspaper and degrade indigo carmine without mediator. The study suggests that the novel property to deink waste paper without mediator may not depend on the redox potential of T1 but other mechanisms using domains 1 and 2 may be involved.

Increasing society awareness on sustainable development and environmental pollution has led to increase in utilization of industry-based secondary fibers such as old newsprint, mixed office waste, and old magazines for paper making. Paper recycling is necessary to reduce dependency on virgin wood pulp and to avoid disposal problem for waste paper. Deinking, the most important process in waste paper recycling, is traditionally handled by large quantities of chemicals, is expensive, and is damaging the environment. Microbial enzymes offer a better alternative for eco-friendly deinking of waste paper by reducing chemical consumption, which leads to cost-cutting, energy saving, and lower waste production. This chapter describes microbial enzymes reported for waste paper recycling and comparison of enzymatic deinking with other conventional methods.

An enzyme cocktail (xylanase-15U, pectinase-2U, α-amylase-2.5U, protease-2U and lipase- 1.8U) from Bacillus halodurans was applied for the biobleaching of unbleached and ODL (oxygen delignification) agro based pulp in the R&D set up of Shreyans Paper Mills Ludhiana, India, upscaling from 5 g (at lab scale) to 100 g odp (oven dried pulp). Enzyme pre-treatment was carried out at temperature and pH optima of 65 °C and 9–9.5 for 90 min followed by reduced chemical treatment. In case of unbleached agro pulp, as compared to the total chemical bleaching in mill, enzyme cocktail pre-treatment reduced the chemical usage by 50% without compromising brightness and rather with increased tensile strength (23.55%), burst factor (20.3%) and tear factor (3.17%) and reduced kappa number (19.5%). In case of ODL pulp, enzyme cocktail pre-treatment reduced the chemical usage by 40%, yet increasing brightness (2.24%), tensile strength (20.96%), burst factor (5.1%) and tear factor (8.2%) with reduction in kappa number (4.54%). The enzyme cocktail thus can make the paper manufacturing process greener, safer, economical and eco-friendly and may replace big budget ODL technologies.

Laccases have potential applications in industrial, biotechnological, and environmental set ups. Development of cost effective and efficient production technologies has gained significant attention in recent years. To enhance the laccase production from Rheinheimera sp. (Gram negative) using submerged fermentation (SmF) and from Lysinibacillus sp. (Gram positive) using solid-state fermentation (SSF), the inducing effect of various flavonoid-rich agro-industrial residues was investigated. Peels of citrus fruits, soybean meal, tofu dreg, lignin monomers, and lingo-cellulosic waste, used tea leaves and peels of onion and kiwi, paper, and dying industry effluents were tested as inducers. In SmF, 0.1% of soybean meal, tofu dreg, and powdered orange peel were best, enhancing the laccase production 2.57-, 2.11-, and 2.05-fold, respectively. In SSF, 10 mg (w/w) of used tata acti green tea leaves per 5 g of wheat bran, 1% pulp and paper industry effluent (agro based), and 1% wine made from Sygium cumini enhanced the laccase production 2.69-, 2.61-, and 2.09-fold, respectively. These results suggest the utilization of these flavonoid and phenolic-rich waste materials to be potential enhancers of industrially important laccase production.

A bacterium that utilizes 2,4,6-trichlorophenol (2,4,6-TCP) as a sole source of carbon and energy was isolated from an industrial dumpsite, the bacterium designated as strain S2. Degradation was routinely monitored by observing growth analysis, chloride release assay, and ring cleavage activity and was further confirmed by gas chromatography (GC) analysis. The bacterium was found to degrade up to 90% of 2,4,6-TCP at 1.5 mM concentration. The bacteria were characterized morphologically, biochemically, and by 16S rRNA gene sequencing, which showed 99% sequence similarity with Shigella sp. This is the first report that Shigella sp. was able to degrade 2,4,6-TCP. This strain was found to be novel and a potential 2,4,6-TCP degrader. Further, this strain may be used for bioremediation of 2,4,6-TCP–containing waste in the environment.
