Human norovirus can accumulate in shellfish in contaminated waters through their filter-feeding mechanism, and they can retain the virus for extended periods. It is important to note that this bioaccumulation can pose a risk to human health if the shellfish are consumed raw or undercooked. Jeotgal is a salted fermented food made from various types of seafood and is consumed in Korea and certain Asian countries. However, jeotgal is not sterilized during preparation and is typically consumed raw after fermentation. Bivalve shellfish, such as oysters, mussels, and clams, are considered high-risk foods for HuNoV transmission due to the potential for contaminated water to lead to the accumulation of HuNoV in their digestive tissues. Invasive alien species pose grave threats to natural ecosystems and human society. The hemlock woolly adelgid (HWA; Adelges tsugae Annand) is an invasive alien species in several countries, causing severe damage to forest mainly composed of hemlock and spruce. This study employed a maximum entropy model to predict the global potential occurrence areas of HWA based on worldwide HWA occurrence data and four bioclimatic variables: mean diurnal range (BIO2), mean temperature of the wettest quarter (BIO8), mean temperature of the coldest quarter (BIO11), and annual precipitation (BIO12). Future climate change scenarios were used to assess the effects of climate change on HWA distribution. The developed model showed good predictive performance for HWA occurrence (AUC:0.992, F1 score: 0.876), with BIO11 being the most influential factor for HWA occurrence, followed by BIO12. The model identified high-risk areas in North America and eastern Asia including Korea, the Himalayan regions, central and western Europe, Oceania including southeastern Australia and New Zealand, and western part of South America. Additionally, most distribution areas of hemlock trees were in areas where HWA could potentially occur. According to climate change, the model predicted a northward shift in the average latitude of the predicted potential distribution areas showing that the range of HWA may potentially expand even into subarctic regions, where the occurrence probability is currently lower than that of temperate regions. The findings of this study provide valuable insights into the potential occurrence and invasion risk of HWA in current and future climates and emphasize the importance of proactive measures to monitor and manage its spread.

Transmembrane proteins known as hyperpolarization-activated cyclic nucleotide-gated (HCN) channels control the movement of Na+ and K+ ions across cellular membranes. HCN channels are known to be involved in crucial physiological functions in regulating neuronal excitability and rhythmicity, and pacemaker activity in the heart. Although HCN channels have been relatively well investigated in the brain, their distribution and function in the retina have received less attention, remaining their physiological roles to be comprehensively understood. Also, because recent studies reported HCN channels have been somewhat linked with the dysfunction of photoreceptors which are affected by retinal diseases, investigating HCN channels in the retina may offer valuable insights into disease mechanisms and potentially contribute to identifying novel therapeutic targets for retinal degenerative disorders. This paper endeavors to summarize the existing literature on the distribution and function of HCN channels reported in the vertebrate retinas of various species and discuss the potential implications for the treatment of retinal diseases. Then, we recapitulate current knowledge regarding the function and regulation of HCN channels, as well as their relevance to various neurological disorders.

IntroductionPain that arises spontaneously is considered more clinically relevant than pain evoked by external stimuli. However, measuring spontaneous pain in animal models in preclinical studies is challenging due to methodological limitations. To address this issue, recently we developed a deep learning (DL) model to assess spontaneous pain using cellular calcium signals of the primary somatosensory cortex (S1) in awake head-fixed mice. However, DL operate like a “black box”, where their decision-making process is not transparent and is difficult to understand, which is especially evident when our DL model classifies different states of pain based on cellular calcium signals. In this study, we introduce a novel machine learning (ML) model that utilizes features that were manually extracted from S1 calcium signals, including the dynamic changes in calcium levels and the cell-to-cell activity correlations.MethodWe focused on observing neural activity patterns in the primary somatosensory cortex (S1) of mice using two-photon calcium imaging after injecting a calcium indicator (GCaMP6s) into the S1 cortex neurons. We extracted features related to the ratio of up and down-regulated cells in calcium activity and the correlation level of activity between cells as input data for the ML model. The ML model was validated using a Leave-One-Subject-Out Cross-Validation approach to distinguish between non-pain, pain, and drug-induced analgesic states.Results and discussionThe ML model was designed to classify data into three distinct categories: non-pain, pain, and drug-induced analgesic states. Its versatility was demonstrated by successfully classifying different states across various pain models, including inflammatory and neuropathic pain, as well as confirming its utility in identifying the analgesic effects of drugs like ketoprofen, morphine, and the efficacy of magnolin, a candidate analgesic compound. In conclusion, our ML model surpasses the limitations of previous DL approaches by leveraging manually extracted features. This not only clarifies the decision-making process of the ML model but also yields insights into neuronal activity patterns associated with pain, facilitating preclinical studies of analgesics with higher potential for clinical translation.

Abstract The purpose of this study was to automatically classify the three-dimensional (3D) positional relationship between an impacted mandibular third molar (M3) and the inferior alveolar canal (MC) using a distance-aware network in cone-beam CT (CBCT) images. We developed a network consisting of cascaded stages of segmentation and classification for the buccal-lingual relationship between the M3 and the MC. The M3 and the MC were simultaneously segmented using Dense121 U-Net in the segmentation stage, and their buccal-lingual relationship was automatically classified using a 3D distance-aware network with the multichannel inputs of the original CBCT image and the signed distance map (SDM) generated from the segmentation in the classification stage. The Dense121 U-Net achieved the highest average precision of 0.87, 0.96, and 0.94 in the segmentation of the M3, the MC, and both together, respectively. The 3D distance-aware classification network of the Dense121 U-Net with the input of both the CBCT image and the SDM showed the highest performance of accuracy, sensitivity, specificity, and area under the receiver operating characteristic curve, each of which had a value of 1.00. The SDM generated from the segmentation mask significantly contributed to increasing the accuracy of the classification network. The proposed distance-aware network demonstrated high accuracy in the automatic classification of the 3D positional relationship between the M3 and the MC by learning anatomical and geometrical information from the CBCT images.

Abstract The major neuropathologic feature of Parkinson’s disease is the presence of widespread intracellular inclusions of α-synuclein known as Lewy bodies. Evidence suggests that these misfolded protein inclusions spread through the brain with disease progression. Changes in synaptic function precede neurodegeneration, and this extracellular α-synuclein can affect synaptic transmission. However, whether and how the spreading of α-synuclein aggregates modulates synaptic function before neuronal loss remains unknown. In the present study, we investigated the effect of intrastriatal injection of α-synuclein preformed fibrils (PFFs) on synaptic activity in the somatosensory cortex using a combination of whole-cell patch-clamp electrophysiology, histology, and Golgi-Cox staining. Intrastriatal PFF injection was followed by formation of phosphorylated α-synuclein inclusions in layer 5 of the somatosensory cortex, leading to a decrease in synapse density, dendritic spines, and spontaneous excitatory post-synaptic currents, without apparent neuronal loss. Additionally, three-dimensional reconstruction of microglia using confocal imaging showed an increase in the engulfment of synapses. Collectively, our data indicate that propagation of α-synuclein through neural networks causes abnormalities in synaptic structure and dynamics prior to neuronal loss.

Abstract Intrinsic plasticity of the cerebellar Purkinje cell (PC) plays a critical role in motor memory consolidation. However, detailed changes in their intrinsic properties during memory consolidation are not well understood. Here, we report alterations in various properties involved in intrinsic excitability, such as the action potential (AP) threshold, AP width, afterhyperpolarization (AHP), and sag voltage, which are associated with the long-term depression of intrinsic excitability following the motor memory consolidation process. We analyzed data recorded from PCs before and 1, 4, and 24 h after cerebellum-dependent motor learning and found that these properties underwent dynamic changes during the consolidation process. We further analyzed data from PC-specific STIM1 knockout (STIM1PKO) mice, which show memory consolidation deficits, and derived intrinsic properties showing distinct change patterns compared with those of wild-type littermates. The levels of memory retention in the STIM1PKO mice were significantly different compared to wild-type mice between 1 and 4 h after training, and AP width, fast- and medium-AHP, and sag voltage showed different change patterns during this period. Our results provide information regarding alterations in intrinsic properties during a particular period that are critical for memory consolidation.

Multi-channel radio frequency (RF) signal generation, facilitated by photonic technology, offers significant potential for generating coherent signals with a high frequency and low phase noise, providing multifunctional capabilities across diverse platforms, including RF and photonic systems. Traditional methods for multi-channel photonic RF signal generation typically entail the integration of diverse optical components, such as filters and amplifiers. However, this integration often results in compromises related to power efficiency, cost-effectiveness, and implementation simplicity. To address these challenges, we propose a novel method for generating multi-channel photonic RF signals based on optical injection locking technology. This approach eliminates the necessity for traditional optical components, leading to a substantial enhancement in the performance of photonic RF signals. We present the design of an optical injection locking-based multi-channel photonic RF signal generation schematic and theoretically evaluate its Signal-to-Noise Ratio (SNR) and eye pattern performance for data modulation using the Lumerical INTERCONNECT simulator. Our results reveal a significant 1.3-dB and 3.6-dB enhancement in SNR for 30-GHz and 60-GHz signals, respectively. Furthermore, we observed an improved communication performance, as evidenced by enhanced eye patterns in 3-Gbps data transmission compared to passive photonic RF signal generation methods.

Abstract This study was conducted to evaluate the biological activities of sweet potato tips and tubers. Antioxidant activity of 2,2‐azino‐bis 93‐ethlbenzothiazoline‐6‐sulphonic acid) diammonium salt (ABTS) and 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging activities had the highest value of 32.45 mg, AAE/g, and 15.10 mg AAE/g, respectively, in ‘Pungwonmi’ tips. Angiotensin converting enzyme I inhibitory activity ranged between 47.72% in ‘Sinjami’ tubers and 62.25% in ‘Pungwonmi’ tips. α‐Glucosidase inhibitory activity had the highest value of 78.81% and 62.93% in ‘Pungwonmi’ tips and ‘Juhwangmi’ tubers, respectively. In particular, ‘Pungwonmi’ tips had the most effective inhibiting effect on intracellular reactive oxygen species levels in HepG2 cells. Wound healing assay result revealed that ‘Sinjami’ showed 75% wound healing effect. For skin whitening, ‘Pungwonmi’ tips showed 63% activity at 10 mg/ml. These results suggest that sweet potato tips and tubers can be used to develop functional food and cosmetic materials.

It is essential to ensure the safety of spent nuclear fuel (SNF) transport cask in drop situation that is included in transport accident scenarios. The safety of the drop situation is affected by the impact absorption performance of impact limiters. Therefore, when designing an impact limiter, the uncertainty in the material properties that affect the impact absorption performance must be considered. In this study, the material properties of the wood inside the impact limiter were selected as the variables for a parametric study. The sensitivity analysis of the drop response of the SNF transport cask with impact limiter was performed. The minimum wood strength required to prevent a direct collision between the cask and floor was derived from the analysis results. In addition, the plastic strain response was analyzed and strain-based evaluation was performed. Based on this result, the critical values of wood properties that change the impact dynamic characteristics were investigated. Finally, the optimal material properties of wood were obtained to secure the structural safety of the SNF transport cask. The results of this study can contribute to the development of SNF transport cask, thereby ensuring safety in transport accident conditions.

Abstract The purpose of this study was to propose a continuity-aware contextual network (Canal-Net) for the automatic and robust 3D segmentation of the mandibular canal (MC) with high consistent accuracy throughout the entire MC volume in cone-beam CT (CBCT) images. The Canal-Net was designed based on a 3D U-Net with bidirectional convolutional long short-term memory (ConvLSTM) under a multi-task learning framework. Specifically, the Canal-Net learned the 3D anatomical context information of the MC by incorporating spatio-temporal features from ConvLSTM, and also the structural continuity of the overall MC volume under a multi-task learning framework using multi-planar projection losses complementally. Unsaponifiable matter (USM) from wheat bran, a by-product obtained from wheat milling, is abundant in health-promoting compounds such as phytosterols, tocopherols, policosanols, and alkylresorcinols. This study aimed to examine the effects of USM from the wheat bran of normal and waxy type wheat, Saekeumkang (SKK) and Shinmichal (SMC), on hepatic lipid accumulation in free fatty acid (FFA)-induced hepatocytes and to investigate the cellular mechanism. The total phytochemical contents were 46.562 g/100 g USM and 38.130 g/100 g USM from SKK and SMC, respectively. FFA treatment increased intracellular lipid accumulation by approximately 260% compared to the control group; however, treatment with USM from SKK and SMC significantly attenuated lipid accumulation in the hepatocytes in a dose-dependent manner. Moreover, USM downregulated the expression of lipogenic factors such as fatty acid synthase and sterol regulatory-element-binding protein 1c by approximately 40% compared to the FFA treatment group. Treatment with USM promoted lipolysis and positively regulated the expression of the proteins involved in β-oxidation, including peroxisome proliferator-activated receptor α and its downstream protein, carnitine palmitoyltransferase 1A. Moreover, the blockade of AMPK activation significantly abolished the inhibitory effects of USM on hepatic lipid accumulation. These results indicated that the USM from both SKK and SMC can alleviate lipid accumulation in hepatocytes in an AMPK-dependent manner. Therefore, USM from wheat bran may be useful as a therapeutic intervention for treating metabolic-dysfunction-associated fatty liver disease.

Centella asiatica possess various health-promoting activities owing to its bioactive compounds such as triterpenes, flavonoids, and vitamins. Ultrasound treatment during the post-harvest process is a good strategy for eliciting secondary metabolite in plants. The present study investigated the effect of ultrasound treatment for different time durations on the bioactive compounds and biological activities of C. asiatica leaves. The leaves were treated with ultrasound for 5, 10, and 20 min. Ultrasound elicitation (especially for 10 min) markedly elevated the accumulation of stress markers, leading to enhanced phenolic-triggering enzyme activities. The accumulation of secondary metabolites and antioxidant activities were also significantly improved compared with that in untreated leaves. In addition, ultrasound-treated C. asiatica leaves protected myoblasts against H2O2-induced oxidative stress by regulating reactive oxygen species production, glutathione depletion, and lipid peroxidation. These findings indicate that elicitation using ultrasound can be a simple method for increasing functional compound production and enhancing biological activities in C. asiatica leaves.

The fermentation and quality characteristics of yogurt were investigated according to the inoculation concentration of Bifidobacterium longum. The total sugar content of yogurt decreased as the fermentation time increased, and with an increased concentration of B. longum treatment the fermentation time decreased rapidly. As fermentation progressed, the lactose content decreased rapidly at the beginning and gradually decreased as the pH decreased. Depending on the B. longum treatment concentration, the lactose content varied from 0.29 ± 0.01 to 0.47 ± 0.01% and was 0.5% or less in all experimental groups. The experimental group inoculated with 0.0015% of B. longum displayed the best results in all categories, including pH, total acidity, lactic acid content, solid non-fat content, and total lactic acid bacteria count, which are factors that determine the quality of yogurt. In summary, the experimental group inoculated with 0.0015% of B. longum was determined to be the highest quality yogurt.

Purpose To assess patient radiation doses during diagnostic and therapeutic neurointerventional procedures from multiple centers and propose dose reference level (RL). Materials and Methods Consecutive neurointerventional procedures, performed in 22 hospitals from December 2020 to June 2021, were retrospectively studied. We collected data from a sample of 429 diagnostic and 731 therapeutic procedures. Parameters including dose-area product (DAP), cumulative air kerma (CAK), fluoroscopic time (FT), and total number of image frames (NI) were obtained. RL were calculated as the 3rd quartiles of the distribution. Results Analysis of 1160 procedures from 22 hospitals confirmed the large variability in patient dose for similar procedures. RLs in terms of DAP, CAK, FT, and NI were 101.6 Gy·cm2, 711.3 mGy, 13.3 minutes, and 637 frames for cerebral angiography, 199.9 Gy·cm2, 3,458.7 mGy, 57.3 minutes, and 1,000 frames for aneurysm coiling, 225.1 Gy·cm2, 1,590 mGy, 44.7 minutes, and 800 frames for stroke thrombolysis, 412.3 Gy·cm2, 4,447.8 mGy, 99.3 minutes, and 1,621.3 frames for arteriovenous malformation (AVM) embolization, respectively. For all procedures, the results were comparable to most of those already published. Statistical analysis showed male and presence of procedural complications were significant factors in aneurysmal coiling. Male, number of passages, and procedural combined technique were significant factors in stroke thrombolysis. In AVM embolization, a significantly higher radiation dose was found in the definitive endovascular cure group. Conclusion Various RLs introduced in this study promote the optimization of patient doses in diagnostic and therapeutic interventional neuroradiology procedures. Proposed 3rd quartile DAP (Gy·cm2) values were 101.6 for diagnostic cerebral angiography, 199.9 for aneurysm coiling, 225.1 for stroke thrombolysis, and 412.3 for AVM embolization. Continual evolution of practices and technologies requires regular updates of RLs.

Sensorimotor information provided by mossy fibers (MF) is mapped to high-dimensional space by a huge number of granule cells (GrC) in the cerebellar cortex’s input layer. Significant studies have demonstrated the computational advantages and primary contributor of this expansion recoding. Here, we propose a novel perspective on the expansion recoding where each GrC serve as a kernel basis function, thereby the cerebellum can operate like a kernel machine that implicitly use high dimensional (even infinite) feature spaces. We highlight that the generation of kernel basis function is indeed biologically plausible scenario, considering that the key idea of kernel machine is to memorize important input patterns. We present potential regimes for developing kernels under constrained resources and discuss the advantages and disadvantages of each regime using various simulation settings.
