Mycobacterium avium subsp. paratuberculosis (MAP) is responsible for Paratuberculosis mainly affecting domestic ruminants. The interaction between MAP and sperm and/or germ cells has not yet been established, however the adherence between MAP and the host cell surface is associated to the 85 complex proteins that bind to the host cell’s fibronectin. Therefore, this study aimed to evaluate the binding of MAP to bovine sperm and to verify changes in these cells by the presence of MAP before and after sperm cryopreservation. Polyclonal antibodies to MAP 85 complex proteins were produced and utilized in the analyzes. Two Nelore bulls were used for semen collection and MAP dilutions (103–108 CFU/mL) were inoculated in the samples; sperm motility and vigor were evaluated using light microscopy at different times before and after cryopreservation and in the presence and absence of the antibodies 85A and 85B. Interaction of MAP and sperm, interaction of MAP and sperm in the presence of Ab 85A and in the presence of Ab 85B were analyzed by scanning electron microscopy. The viability of MAP after sperm cryopreservation were evaluated by plating the samples after thawing. It was observed that sperm in the presence of MAP shows a decrease in motility and vigor, and that the higher the MAP concentration, the lower the sperm performance. It was possible to determine the viability of MAP after cryopreservation in samples of higher concentrations, which demonstrates the potential of transmission of this pathogen through artificial insemination. The interaction of MAP with bovine sperm occurs mainly in the midpiece and may be linked to the proteins 85A and 85B present in the MAP membrane.

Antibiotic resistance represents a widespread problem in milk production. The identification of compounds for a topically applied ointment used in mastitis therapy remains elusive. Compounds from the genus Hymenaea can be administered in cases of multi-drug-resistant Staphylococcus aureus infection for ruminant species, but the protective properties are not well known. Wi this research the aim is verify the protective effects of H. martiana against S. aureus infection in bovine mammary epithelial cell line (MAC-T) and to obtain an antioxidant profile evaluation in vitro. The MAC-T cells were challenged with S. aureus after being exposed to the extract of the H. martiana in the protective assay. For the verification of the viability of the MAC-T cells, the MTT assay was performed, and was used dilutions of the plant extract, starting at 2.5%. The extract of H. martiana was evaluated for antioxidant aspect in different diluition by FRAP, ORAC and DPPH. A variety of flavonoids (quercetin, luteolin, etc.) have been identified as the main components by using mass spectrometry, reinforcing our in vitro findings that flavonoids, especially quercetin, have a medicinal profile capable of killing mastitis-causing bacteria. An excellent antioxidant pattern was observed in the 2.5% solution; however, membrane integrity in MAC-T cells was compromised. Those findings suggest low dilutions of H. martiana extract has a desirable protective effect from S. aureus pathogenesis. Our in vitro studies can be gleaned upon for further in vivo studies.

Among the infectious diseases of the dairy herd, mastitis is the main cause of direct and indirect economic losses. Mastitis etiological agents are usually bacteria, but fungi and algae may be involved. Staphylococcus aureus is commonly isolated from mastitis and deserves special attention due to the numerous hospital infections related to methicillin resistant strains. Although the mammary gland presents different defense mechanisms, in many cases the etiologic agent can escape of the immune system and multiply, resulting in mastitis. Several nonspecific antimicrobial factors are found in the mammary gland, such as lactoperoxidase–thiocyanate¬–hydrogen peroxide, lysozyme, lactoferrin, and complement system. Adaptive or acquired immunity can lead to an immediate response in repetitive exposure to the same microorganism. Two types of adaptive responses are described: humoral and cellular immunities. Conventional diagnostic methods are widely considered for mastitis detection, like microbiological isolation and somatic cell count; however, alternative diagnosis methods are being increasingly used, like infrared thermography that measures the surface temperature of mammary gland skin and sphincters of the teats. Conventional therapy based on antibiotics is not always efficient, resulting in cases of persistent infections. Recent research using inhibitors of multidrug efflux systems and biofilms has been tested in vitro with reliable results. Studies have also demonstrated that bioactive substances extracted from plants with and without chemical modifications have shown synergistic associations with the traditional antimicrobials against bacteria causing mastitis. Other infectious diseases, such as brucellosis, tuberculosis, Johne's disease, leptospirosis, Salmonellosis, bovine viral diarrhea (BVD), and infectious bovine rhinotracheitis (IBR) can affect the dairy cattle herd, compromising the milk production chain in economic, social, and public health aspects.

Excessive intake of nutrients can result in decreases in the reproductive efficiency of goat bucks and increase production costs in goat e nterprises. In this study, the aim was to compare the reproductive features of young bucks fed diets for maintenance of growth or were fed ad libitum. Ten bucks were used (n = 5/group), with a completely randomized design. Blood and semen were collected from each animal every 2 weeks for nine collections. Semen was frozen and later analyzed using flow cytometry to compare reactive oxygen species, plasma membrane integrity, acrosome integrity, and membrane lipid peroxidation. Body condition score, sexual behavior, testicular biometry and histomorphometry of the testis, as well as oxidative enzymes and carbonylated proteins, were also evaluated. Animals fed ad libitum had a greater body condition score (3.32 + 0.08 × 2.88 + 0.05), sperm volume (0.68 + 0.04 × 0.52 + 0.03 mL), and testis circumference (25.2 + 0.21 × 24.5 + 0.24 cm) and volume (121 + 5.83 × 107 + 4.38 cm³, left testicle; 116 + 5.26 × 100 + 3.75 cm³, right testicle). Ejaculates of the bucks fed diets to maintain growth rate had a larger number of minor and total sperm morphology defects. There were no differences in testosterone concentrations and sexual behavior. In conclusion, there can be feeding for growth rate maintenance instead of ad libitum feeding, for the reduction in production costs of young bucks without compromising semen quality and reproductive functions.

The aim of the study was to evaluate the effect of recombinant bovine somatotropin (rbST) and thyroxine (T4) on survival and growth of bovine preantral ovarian follicles (PAOF) cultured in vitro. Ovarian fragments were collected in local abattoirs and immediately fixed for classical histology and transmission electron microscopy (non-cultured control). The other fragments were then cultured in situ for seven days in minimum essential medium alone (MEM+ - cultured control) or in the presence of 1,000 ng/mL rbST and 20 ng/mL T4, isolated or associated. After seven days, there was a reduction (P0.01) of the medium used on the follicular diameter of the PAOF cultured for seven days. Ultrastructural analysis showed cell damage. In conclusion, the presence of rbST maintains the rate of morphologically normal follicles during the culture for seven days (observed by optical microscopy), but it does not exert beneficial effects on its ultrastructural integrity and oocyte and follicular growth. Resumo O objetivo do trabalho foi avaliar o efeito da somatotropina recombinante bovina (rbST) e da tiroxina (T4) sobre a sobrevivência e o crescimento de folículos ovarianos pré-antrais (FOPA) bovinos cultivados in vitro. Fragmentos ovarianos foram coletados em abatedouros locais e imediatamente fixados para histologia clássica e microscopia eletrônica de transmissão (controle não cultivado). Os demais fragmentos foram cultivados in situ por sete dias em meio essencial mínimo sozinho (MEM+ - controle cultivado), ou na presença de 1.000 ng/mL de rbST e 20 ng/mL de T4, isoladamente ou em associação. Após sete dias, houve redução (P0,01) do meio utilizado, no diâmetro folicular, dos FOPA cultivados por sete dias. A análise ultraestrutural demonstrou danos celulares. Conclui-se que, apesar da presença da rbST manter a taxa de folículos morfologicamente normais durante o cultivo por sete dias (observado por microscopia óptica), o uso de rbST e T4 em meio de cultivo in vitro de FOPA, nas concentrações utilizadas no presente experimento, não exerce efeitos benéficos quanto à integridade ultraestrutural e aos crescimentos oocitário e folicular.

O experimento foi realizado para estimar a exigência nutricional de sódio (Na) para codornas japonesas em postura. Foram utilizadas 245 codornas japonesas, com 187 dias de idade, peso de 189,4 + 6,1 g e taxa de produção de ovos de 91,05 + 5,5 %. As aves foram distribuídas em delineamento experimental inteiramente casualizado, constituído por cinco rações e sete repetições. Foram formuladas cinco rações experimentais à base de milho e farelo de soja, com cinco níveis de Na (0,10; 0,15; 0,20; 0,25 e 0,30%). Foram analisados: consumo de ração, produção de ovos por ave ao dia, por ave alojada, de ovos comercializáveis, de ovos viáveis por ave ao dia, peso do ovo, massa de ovos, conversão alimentar por massa de ovos e por dúzia de ovos, componentes dos ovos (gema (g e %), albúmen (g e %) e casca (g e %), variação de peso corporal, viabilidade das aves e gravidade específica. Não foi verificado efeito (P>0,05) para nenhum dos parâmetros estudados. Sintomas de deficiência de sódio como inapetência, redução da taxa de crescimento, produção e apetite depravado não foram observados, demonstrando que o menor nível de sódio utilizado, 0,10%, não foi prejudicial às aves. A suplementação de 0,10% de sódio na ração, correspondente ao consumo diário de 26,1 mg de Na por ave foi suficiente para proporcionar satisfatório desempenho produtivo e qualidade dos ovos de codornas japonesas. The experiment was conducted to estimate the nutritional requirement of sodium (Na) for laying Japanese quails. Were used 245 Japanese quails, with 187 days old, weight of 189.4 + 6.1 g and egg production rate of 91.05 + 5.5%. Birds were distributed in a completely randomized design, consisting of five diets and seven replicates. Five experimental diets were formulated based on corn and soybean meal, with five levels of Na (0.10, 0.15, 0.20, 0.25 and 0.30%). Were examined: feed intake, egg production per bird day, egg production per hen housed, commercial eggs, viable eggs per bird day, egg weight, egg mass, feed conversion by egg mass and per dozen eggs, components eggs (yolk, albumen and shell), body weight variation, viability of birds and specific gravity. There were no differences (P>0.05) for all parameters studied. Symptoms of deficiency as inappetence, reduction in the growth rate and depraved appetite were not observed in this study, demonstrating that the lower level of Na used, 0.10%, was not adverse to birds. Supplementation of 0.10% Na in diet of the birds, corresponding to a daily intake of 26.1 mg / bird was enough to provide good performance and egg quality of Japanese quails.

Com o objetivo de avaliar diferentes níveis de proteína bruta e de energia metabolizável para codornas de corte de um a 14 dias de idade, foi realizado um estudo utilizando 840 codornas de corte, mistas. As aves foram distribuídas em arranjo fatorial (2x5) constituído por 10 tratamentos (dois níveis de energia metabolizável - 2800 e 3000kcal/kg e cinco níveis de proteína bruta - 22,0; 24,0; 26,0; 28,0 e 30,0%), sete repetições com 12 aves cada. Foram avaliados os seguintes parâmetros: consumo de ração, ganho em peso aos sete e 14 dias de idade, conversão alimentar aos 14 dias e viabilidade. Verificou-se que codornas alimentadas com dietas com o nível energético mais baixo, 2800kcal/kg, apresentaram maior consumo. As aves que consumiram ração com menor nível de energia apresentaram maior peso aos sete dias de idade independentemente do nível de proteína bruta utilizado. O peso aos 14 dias de idade apresentou resposta quadrática com o acréscimo de proteína bruta na ração, sendo que dietas contendo 28,86% de PB proporcionaram melhor ganho independentemente do nível de energia utilizado. A conversão alimentar apresentou melhores resultados com o aumento do nível de proteína bruta da ração, independentemente do nível de energia metabolizável utilizado. Não foram observados efeitos significativos dos níveis de proteína e energia ou de sua interação sobre a viabilidade das aves. Rações contendo 30,0% de proteína bruta e 2800 ou 3000kcal/kg de energia metabolizável proporcionam satisfatórios resultados de desempenho para codornas de corte de um a 14 dias de idade.

Foram utilizados três cães machos, adultos, sadios da raça Labrador do Retrievier, que foram submetidos a uma coleta de sêmen semanal durante o período de cinco semanas. Os três meios diluentes utilizados neste estudo foram: (D1)-tris-citrato, acrescido de 3% de dimetilformamida + 3% de glicerol, (D2)-3% de dimetilformamida e de trealose e (D3)-4% de glicerol. No descongelamento, metade das amostras de cada meio diluente foi descongelada pelo método rápido, em banho-maria a 75ºC por sete segundos, seguido de nova imersão a 37 ºC por 1 minuto. A outra metade foi descongelada pelo método lento, em banho-maria à 37 ºC por 1 minuto. O sêmen foi avaliado quanto à motilidade progressiva, vigor espermáticos e integridade de membrana. Pra isso, as amostras foram submetidas aos testes hipo-osmótico e de integridade da membrana plasmática e do acrossoma (fluorescência). Os resultados indicam que o uso do glicerol como crioprotetor em diluidor TRIS proporciona maior eficácia na criopreservação dos espermatozoides da espécie canina, quando comparados com a dimetilformamida associada ao glicerol ou trealose. The aim of this study was to evaluate the association between cryoprotectants little studied in Brazil such as dimethylformamide and trehalose amid thinner, using protocols of fast and slow defrosting. Three adult Labrador Retrievier male, healthy dogs, weekly submitted to one semen collection during five-weeks period, were used. The base diluent medium used in this study was tris-citrate added with 3% of dimethylformamide + 3% glycerol (D1), 3% dimethylformamide and trehalose (D2) and 4% glycerol (D3). At defrosting, half of the semen samples from each diluent medium was defrosted by rapid method in water-bath at 75 °C for seven minutes, followed by a new immersion at 37 °C for 1 minute. The other half of the samples was defrosted by slow method, in water-bath at 37 °C for 1 minute. The semen was evaluated for sperm progressive motility and vigor, besides membrane integrity. For this, the semen samples were submitted to either hyposmotic and membrane integrity tests of the plasmatic membrane and acrosome (fluorescence). The results indicated that the use of glycerol as cryoprotector in TRIS diluter provides greater efficacy in cryopreserving spermatozoa of the canine species, when compared to dimethylformamide associated with trehalose or glycerol. The aim of this study was to investigate the influence of the interaction between follicle stimulating hormone obtained from Human Pituitary (hFSH; 10 ng/mL), Insulin-like Growth Factor Type 1 (IGF-I; 100 ng/mL) and thyroxine (T4 ; 10, 20 ng/mL) on survival, activation and growth of preantral follicles (PAF) cultured in vitro. Fragments of ovarian cortex collected from 17 nonpregnant adult goats underwent different treatments that included α-MEM+ medium, IGF-I, hFSH, T4 and were processed for classic histology and transmission electron microscopy. The results showed a reduction in the percentage of normal follicles, after one and seven days of culture in media α-MEM+ (cultured control), T4 (10)/IGF-I, T4 (20)/IGF-I/hFSH and in media α-MEM+, IGF-I/hFSH, T4 (20)/IGF-I/hFSH, T4 (20)/IGF-I/hFSH, when compared to the non-cultured control. However, the medium containing T4 (20)/hFSH showed a follicular survival rate higher than the other treatments, when cultured for seven days. It was also observed that the follicular diameter did not change when cultured in this medium, when compared to the control (non-cultured). The ultrastructural analysis confirmed the integrity of follicles cultured in a medium containing T4 (20)/hFSH. In conclusion, the medium containing T4 (20)/hFSH showed the best results, when evaluating the survival, growth and ultrastructural aspects of caprine preantral follicles cultured in vitro.

Fragmentos de tecido ovariano não cultivado e cultivados foram processados para histologia clássica e microscopia eletrônica de transmissão. Os resultados mostraram uma redução na taxa de sobrevivência em todos os meios testados (um a 28 dias) quando comparado ao controle fresco. Entretanto, o tratamento com T4/hFSH por sete dias de cultivo resultou em taxa semelhante ao controle. Os meios testados por 28 dias reduziram o percentual de folículos primordiais em todos os períodos de cultivo. Contudo, T4/hFSH no dia um de cultivo manteve-se semelhante ao controle fresco. Nenhum dos meios foi capaz de manter o percentual dos folículos em desenvolvimento. Observou-se que o diâmetro folicular cultivado no meio com T4/hFSH manteve-se semelhante ao controle fresco. As análises ultraestruturais confirmaram a integridade de folículos cultivados por sete dias em meio suplementado com T4/hFSH. Em conclusão, o meio com T4/hFSH é capaz de manter a sobrevivência, promover a ativação e a integridade ultraestrutural de folículos pré-antrais caprinos, por até sete dias. The aim of this study was to investigate the interaction of human FSH (10ng/ml) with T4 (20ng/mL) on survival, activation and growth of preantral follicles cultured in vitro for 28 days. Fragments of non-cultured and cultured ovarian tissue were processed for classic histology and transmission electron microscopy. The results showed a reduction in the survival rate in all the media tested (one to 28 days) when compared to the fresh control. However the treatment with T4/hFSH for seven days of culture maintained the rate similar to the control. The media tested by one and 28 days reduced the percentage of primordial follicles in all periods of culture. However, T4/hFSH on day one of culture remained similar to the fresh control. None of the media were able to keep the percentage of the developing follicles. It was observed that the follicular diameter in the medium with T4/hFSH remained similar to the fresh control. The ultrastructural analysis confirmed the integrity of follicles cultured for seven days in a medium supplemented with T4/hFSH. In conclusion, the medium with T4/hFSH is able to maintain the survival, promote the activation, and the ultrastructural integrity of caprine preantral follicles for until seven days. Conselho Nacional de Desenvolvimento Científico e Tecnológico The aim of this study was to evaluate the effect of T4, IGF-I and hFSH, alone or in combination, as well as the culture period (1, 7, 14, 21 and 28 days) on survival, activation and in vitro growth of caprine preantral ovarian follicles (PAOF). For Phase I, II and III, a fragment/follicle will be removed for classic histology and Transmission Electron Microscopy (TEM) constituting the fresh control. Then, the remaining fragments/follicles will be cultured in situ (included in ovarian tissue fragments) for 1 and 7 days (Phase I and II) and 1, 7, 14, 21 and 28 days (Phase III), (at 39°C in a kiln and 5% of CO2) in -MEM+ in the absence or presence of different concentrations of T4 (stage 1), IGF-1 (stage 2) and hFSH (stage 3). In Phase II, the fragments/follicles will be cultured on plates containing -MEM+ supplemented with T4, IGF-I and FSH, in combination at the best concentrations defined in Phase I. In Phase III, the fragments/follicles will be cultured in -MEM+ or in the best medium (media) obtained in Phase II for 1, 7, 14, 21 and 28 days. For the experiments conducted in Phase I, the results showed that after seven days of culture, 10 ng/ml of hFSH is able to promote the activation of primordial follicles and to maintain the ultrastructural integrity of follicles cultured for up to seven days. The addition of 100 ng/ml of IGF-I to the culture medium activate the developing follicles with one day of culture. Furthermore, the use of 20 ng/ml of T4 is effective in maintaining the survival, activation and growth of follicles during seven days of culture. In Phase II, the interaction of T4, hFSH and IGF-I performed during the culture showed that the combination of T4/hFSH (20/10ng/mL) is beneficial for the in vitro development of preantral follicles, during the survival, growth and ultrastructural aspects. The results presented in Phase III demonstrate that the long- duration culture at the concentration of 20/10 ng/ml of T4/hFSH promoted the survival, activation of primordial follicles and follicular growth during seven days of culture. In conclusion, the results of this study showed that the use of hFSH (10ng/ml), IGF-I (100ng/ml) and T4 (20ng/ml) promotes the maintenance of follicle survival and the development of caprine preantral follicles. Moreover, the association of T4 and hFSH xxiiensured the survival of follicles and maintained follicular growth. Follicles cultured for a long duration period (28 days) is necessary for the understanding of the media to be used during the in vitro culture, demonstrating the need for further interactions between substances, for the discovery of a culture medium which allows the development of follicles during folliculogenesis in the preantral and antral stage. O objetivo deste estudo foi avaliar o efeito da T4, IGF-I e hFSH, isoladamente ou em associação, bem como o período de cultivo (1, 7, 14, 21 e 28 dias) sobre a sobrevivência, ativação e crescimento in vitro de folículos ovarianos pré-antrais (FOPA) caprinos. Para a Fase I, II e III, um fragmento/folículo foi retirado para histologia clássica e Microscopia Eletrônica de Transmissão (MET) constituindo o controle fresco. Em seguida, os demais fragmentos/folículos foram cultivados in situ (inclusos em fragmentos de tecido ovariano) por 1 e 7 dias (Fases I e II) e 1, 7, 14, 21 e 28 dias (Fase III), (a 39oC em estufa e 5% de CO2) em -MEM+ na ausência ou presença de diferentes concentrações de T4 (etapa 1), IGF-1 (etapa 2) e hFSH (etapa 3). Na Fase II, os fragmentos/folículos foram cultivados em placas contendo -MEM+ suplementado com T4, IGF-I e FSH, em associação nas melhores concentrações definidas na Fase I. Na Fase III, os fragmentos/folículos foram cultivados em -MEM+ ou no (s) melhor (es) meio (s) obtido (s) na Fase II por 1, 7, 14, 21 e 28 dias. Para os experimentos realizados na Fase I, os resultados mostraram que após sete dias de cultivo, 10 ng/mL de hFSH foi capaz de promover a ativação de folículos primordiais e manter a integridade ultraestrutural dos folículos cultivados por até sete dias. A adição de 100 ng/mL de IGF- I ao meio cultivo ativa os folículos em desenvolvimento com um dia de cultivo. Além disso, a utilização de 20 ng/mL de T4 é eficaz na manutenção da sobrevivência, ativação e crescimento de folículos, durante sete dias de cultivo. Na Fase II, a interação de T4, hFSH e IGF-I realizada durante o cultivo, demonstrou que a combinação de T4/hFSH (20/10ng/mL) é benéfica para o desenvolvimento in vitro de folículos pré-antrais, durante a sobrevivência, o crescimento e aspectos ultraestruturais. Os resultados apresentados na Fase III demonstram que o cultivo de longa duração na concentração de 20/10 ng/mL de T4/hFSH promoveu a sobrevivência, a ativação de folículos primordiais e o crescimento folicular durante sete dias de cultivo. Os resultados deste estudo mostraram que a utilização de hFSH (10ng/mL), IGF-I (100ng/mL) e T4 (20ng/mL) promovem a manutenção da sobrevivência folicular e o desenvolvimento dos folículos xxpré-antrais caprinos e que a associação de T4 e hFSH assegurou a sobrevivência dos folículos e manteve o crescimento folicular. Os folículos cultivados por um período de longa duração (28 dias) se faz necessário para o conhecimento dos meios a serem utilizados durante o cultivo in vitro, demonstrando a necessidade de mais interações entre as substâncias, para a descoberta de um meio de cultivo que permita o desenvolvimento dos folículos durante a foliculogênese na fase pré-antral e antral.

Conselho Nacional de Desenvolvimento Científico e Tecnológico This experiment has been developed in order to verify the thyroxine effect in the presence or absence of follicle stimulating hormone (FSH) on survival and growth of sheep preantral follicles. Fragments of ovarian tissue were grown for 1 or 7 days in a situation with a minimum essential (α-MEM +) alone or containing thyroxine (10, 20, 50 µg/mL) in the presence or absence of rFSH (50 ng/mL). The ovarian tissue non-grown (fresh control) and grown in different treatments were processed for analysis using the classical histology technique. When the days of culture were compared to the non-cultivated control, on the first day was found that the treatment with α-MEM+, FSH, T20 and T50 showed follicular survival percentages similar to control. When the treatments have been compared between each other within the growing season was identified that only on the 7th day, the treatment containing T50 presents a percentage of normal follicles significantly superior to the treatment with T10F. Related to the follicular diameter, the follicle in culture has not grown, since all treatments have had a similar behavior when controlling non-grown and α- MEM+, except the T10 treatment. When comparing the treatments within each growing season, it has been observed that on the first day, treatments T50, T10F and T50F have showed a follicular diameter greater when compared to T10 treatment. Similar to what happenedto follicular growth, oocitário growth has not grown either. For oocitário diameter, on the first day, there was a reduction for T10 and T20F treatments when compared to non-grown. On the 7th day of grown, FSH, T10F and T50F treatments have reduced oocitário diameter, with a progression of culture from the first day to the 7th one and even when compared to control non-grown and α-MEM+. It was concluded that high T4 concentrations (50 g / ml) have provided promising about the survival and follicular development in vitro. The association of T4 with FSH, have not presented positive results in the survival and follicular development in vitro. Although it is known the thyroxine action on ovarian physiology, there is a need to verify its importance for the in vitro development of preantral sheep follicles. O presente experimento foi desenvolvido a fim de se verificar o efeito da tiroxina na presença ou ausência do hormônio folículo estimulante (FSH) sobre a sobrevivência e o crescimento de folículos pré-antrais de ovinos. Fragmentos de tecido ovariano foram cultivados por 1 e 7 dias em meio essencial mínimo (α-MEM+) suplementado ou não com diferentes concentrações de tiroxina (10, 20, 50 µg/mL) na presença ou ausência de rFSH (50 ng/mL). O tecido ovariano não-cultivado (controle fresco) e os fragmentos cultivados no dia 1 e 7 nos diferentes tratamentos foram processados e analisados utilizando a técnica de histologia clássica. Quando os dias de cultivo foram comparados ao controle não-cultivado, somente no dia 1 foi verificado que os tratamentos contendo α- MEM+, FSH, T20 e T50 mostraram percentuais de sobrevivência folicular semelhantes ao controle. Quando os tratamentos foram comparados entre si dentro de cada período de cultivo, foi observado que apenas no dia 7, o tratamento contendo T50 apresentou um percentual de folículos normais superior ao tratamento T10F. Com relação ao diâmetro folicular, não houve crescimento dos folículos em cultivo, uma vez que todos os tratamentos comportaram-se de maneira semelhante ao controle não-cultivado e ao α- MEM+, exceto o tratamento T10. Ao se comparar os tratamentos, dentro de cada período de cultivo, foi observado que no dia 1 os tratamentos T50, T10F e T50F apresentaram diâmetro folicular superior quando comparados ao tratamento T10. Semelhante ao crescimento folicular também não houve crescimento oocitário. Para o diâmetro oocitário, no dia 1, foi verificada uma redução para os tratamentos T10 e T20F quando comparados ao não cultivado. Já no dia 7 de cultivo os tratamentos FSH, T10F e T50F reduziram o diâmetro oocitário, com a progressão do cultivo do dia 1 para o dia 7 e ainda quando comparados ao controle não-cultivado e ao α-MEM+. Concluiu-se que altas concentrações de T4 (50 μg/ml) proporcionaram resultados promissores no que diz respeito à sobrevivência e desenvolvimento folicular in vitro. Quanto a associação da T4 com o FSH não demonstrou resultados positivos na sobrevivência e no desenvolvimento folicular in vitro. Embora seja conhecida a atuação da tiroxina na fisiologia ovariana são necessárias mais pesquisas que verifiquem sua importância para o desenvolvimento in vitro de folículos pré-antrais ovinos.

Fragmentos de córtex ovariano foram cultivados por um e sete dias na ausência ou presença de IGF-I (0, 50 e 100ng/mL). Os tecidos não cultivados e cultivados foram processados e analisados por histologia e microscopia eletrônica de transmissão. O cultivo por um dia em meio com 100ng/mL de IGF-I apresentou 86,7% de folículos morfologicamente normais. Estes resultados foram semelhantes (P>0,05) ao percentual de folículos normais encontrados no controle (96,7%). Verificou-se ainda que este meio aumentou o percentual de ativação folicular (folículos em desenvolvimento) com um dia de cultivo. Os diâmetros ovocitário e folicular mantiveram-se semelhantes ao controle ao cultivar por um dia em meio contendo 100ng/mL de IGF-I. As análises ultraestruturais não confirmaram a integridade folicular dos fragmentos em meio contendo IGF-I (100 ng/mL) após um e sete dias de cultivo. Em conclusão, esse estudo demonstrou que a adição de 100 ng/mL de IGF-I no meio de cultivo ativa o desenvolvimento de folículos pré-antrais de caprinos com um dia de cultivo. Entretanto, não é suficiente para manter a integridade folicular e a taxa de sobrevivência folicular após sete dias de cultivo. The aim of this study was to investigate the effects of the insulin-like growth factor -I (IGF-I) on survival, activation (transition from primordial to primary follicles) and growth of caprine preantral follicles cultured in vitro. Fragments of ovarian cortex were cultured for one and seven days in the absence or presence of IGF-I (0, 50 and 100ng/ml). The non-cultured and cultured tissues were processed and analyzed by histology and transmission electron microscopy. The culture for one day in a medium with 100ng/ml of IGF-I showed 86.7% of morphologically normal follicles. These results were similar (P>0.05) to the percentage of normal follicles found in the control (96.7%). It was also found that this medium increased the percentage of follicular activation (developing follicles) with one day of culture. The oocyte and follicular diameters remained similar to the control by culturing for one day in a medium containing 100ng/ml of IGF-I. The ultrastructural analysis did not confirm the integrity of the follicular fragments in a medium containing IGF-I (100ng/ml) after one and seven days of culture. In conclusion, this study demonstrated that the addition of 100 ng/ml of IGF-I in the culture medium enables the development of preantral follicles of goats with one day of culture. However, it is not sufficient to maintain the follicular integrity and the follicular survival rate after seven days of culture. A bactéria Mycobacterium avium subsp. paratuberculosis (MAP) é o agente responsável pela doença de Johne ou paratuberculose caracterizada por uma enterite crônica, levando à diminuição da produção, descarte de animais, desordens reprodutivas e susceptibilidade a outras doenças, principalmente em ruminantes. Possui o complexo de antígenos 85, constituída por três proteínas 85A, 85B e 85C, presentes no envelope externo, responsável por ligar-se a fibronectina da célula hospedeira. MAP consegue sobreviver às baixas temperaturas da criopreservação, no entanto pouco se sabe da sua viabilidade e sua interação com os espermatozoides, de como esta bacteria pode alterar a viabilidade do material genético antes e pós- congelamento. Assim, este estudo propôs determinar características do espermatozoide bovino na presença de MAP e sua viabilidade pós congelamento bem como estudar esta interação. Foi utilizado um bovino apto para a reprodução para coleta de sêmen e foi inoculado concentrações de baixa e alta carga bacteriana de MAP, avaliando-se quanto a motilidade e vigor, potencial da atividade mitocondrial antes e pós congelamento, e interação de MAP com as células espermáticas através de microscopia de varredura. Além disso foi avaliado a viabilidade de diferentes concentrações de MAP pós congelamento. Foram produzidos anticorpos a partir de proteínas do complexo 85 de MAP (85A e 85B) e utilizados nestas análises. Foi possível determinar a viabilidade de MAP após a criopreservação em amostras de concentrações maiores, o que demonstra o potencial de transmissão deste patógeno através da inseminação artificial (IA). Observou-se que o sêmen bovino na presença de MAP apresenta diminuição na motilidade e vigor proporcional a concentração de MAP e que proteínas de membrana de MAP, 85A e 85B, podem estar relacionados a interação/adesão com as células espermáticas, que ocorre na cauda do espermatozoide principalmente na peça intermediaria. The bacteria Mycobacterium avium subsp. paratuberculosis (MAP) is Johne's disease or paratuberculosis agent characterized by chronic enteritis, leading to decreased production, animal discard, reproductive disorders and susceptibility to other diseases, especially in ruminants. It has the complex 85 antigen, consisting of three proteins 85A, 85B and 85C, present in the external envelope, responsible for binding to fibronectin of the host cell. MAP can survive the low temperatures of cryopreservation, however little is known about its viability and its interaction with sperm, how this bacterium can alter the viability of the genetic material before and after freezing. Thus, this study proposed to determine characteristics of bovine spermatozoa in the presence of MAP and its viability after freezing as well as to study this interaction. A bovine was used for semen collection and low and high bacterial MAP concentrations were inoculated, evaluating the motility and vigor, potential mitochondrial activity before and after freezing, and MAP interaction with the cells spermatozoa by scanning microscopy. In addition, the viability of different concentrations of MAP after freezing was evaluated. Antibodies were produced from MAP complex 85 (85A and 85B) proteins and used in this analyzes. It was possible to determine the viability of MAP after cryopreservation in samples of higher concentrations, which demonstrates the potential of transmission of this pathogen through AI. It was observed that bovine semen in the presence of MAP shows decrease in motility and vigor proportional to MAP concentration and that MAP membrane proteins, 85A and 85B, may be related to the interaction / adhesion with sperm cells, which occurs in spermatozoa intermediate piece.
