Current therapies cause adverse effects and are expensive, invasive, and ineffective for most patients. Annexin A1 (AnxA1) is a pivotal endogenous anti-inflammatory and tissue repair protein in IBD. Nanostructured compounds loading AnxA1 or its active N-terminal mimetic peptides improve IBD symptomatology.Methods: To further explore their potential as a therapeutic candidate, the AnxA1 N-terminal mimetic peptide Ac2-26 was incorporated into SBA-15 ordered mesoporous silica and covered with EL30D-55 to deliver it by oral treatment into the inflamed gut.Results: The systems SBA-Ac2-26 developed measurements revealed self-assembled rod-shaped particles, likely on the external surface of SBA-15, and 88% of peptide incorporation. SBA-15 carried the peptide Ac2-26 into cultured Raw 264.7 macrophages and Caco-2 epithelial cells. Moreover, oral administration of Eudragit-SBA-15-Ac2-26 (200 μg; once a day; for 4 days) reduced colitis clinical symptoms, inflammation, and improved epithelium recovery in mice under dextran-sodium sulfate-induced colitis.Discussion: The absorption of SBA-15 in gut epithelial cells is typically low; however, the permeable inflamed barrier can enable microparticles to cross, being phagocyted by macrophages. These findings suggest that Ac2-26 is successfully delivered and binds to its receptors in both epithelial and immune cells, aligning with the clinical results.Conclusion: Our findings demonstrate a simple and cost-effective approach to delivering Ac2-26 orally into the inflamed gut, highlighting its potential as non-invasive IBD therapy. Systemic neutrophilia or abundant neutrophils in the tumor contribute toward its worst prognosis, and the interplay of cancer and the immune system has been shown in tumor development and metastasis. We recently showed the in vivo efficacy of poly(ε-caprolactone) lipid-core nanocapsule (LNC) or LNC loaded with acetyleugenol (AcE-LNC) to treat B16F10-induced melanoma in mice. In this study, we investigated whether LNC or AcE-LNC toxicity could involve modifications on crosstalk of melanoma cells and neutrophils. Therefore, melanoma cells (B16F10) were pretreated with vehicle, LNC, AcE or AcE-LNC for 24 h, washed and, further, cocultured for 18 h with peritoneal neutrophils obtained from C57Bl/6 mice. Melanoma cells were able to internalize the LNC or AcE-LNC after 2 h of incubation. LNC or AcE-LNC pretreatments did not cause melanoma cells death, but led melanoma cells to be more susceptible to death in serum deprivation or hypoxia or in the presence of neutrophils. Interestingly, the production of reactive oxygen species (ROS), which causes cell death, was increased by neutrophils in the presence of LNC- and AcE-LNC-pretreated melanoma cells. LNC or AcE-LNC treatments reduced the concentration of transforming growth factor-β (TGF-β) in the supernatant of melanoma cells, a known factor secreted by cancer cells to induce pro-tumoral actions of neutrophils in the tumor microenvironment. In addition, we found reduced levels of pro-tumoral chemical mediators VEGF, arginase-1, interleukin-10 (IL-10) and matrix metalloproteinase-9 (MMP-9) in the supernatant of LNC or AcE-LNC-pretreated melanoma cells and cocultured with neutrophils. Overall, our data show that the uptake of LNC or AcE-LNC by melanoma cells affects intracellular mechanisms leading to more susceptibility to death and also signals higher neutrophil antitumoral activity. Abstract

Purpose Therapeutic strategies for prostate cancer (PCa) have been evolving dramatically worldwide. The current article reports on the evolution of surgical management strategies for PCa in Italy. Methods The data from two independent Italian multicenter projects, the MIRROR-SIU/LUNA (started in 2007, holding data of 890 patients) and the Pros-IT-CNR project (started in 2014, with data of 692 patients), were compared. Differences in patients' characteristics were evaluated. Multivariable logistic regression models were used to identify characteristics associated with robot-assisted (RA) procedure, nerve sparing (NS) approach, and lymph node dissection (LND). Results The two cohorts did not differ in terms of age and prostate-specific antigen (PSA) levels at biopsy. Patients enrolled in the Pros-IT-CNR project more frequently were submitted to RA (58.8% vs 27.6%,p < 0.001) and NS prostatectomy (58.4% vs. 52.9%,p = 0.04), but received LND less frequently (47.7% vs. 76.7%,p < 0.001), as compared to the MIRROR-SIU/LUNA patients. At multivariate logistic models, Lower Gleason Scores (GS) and PSA levels were significantly associated with RA prostatectomy in both cohorts. As for the MIRROR-SIU/LUNA data, clinical T-stage was a predictor for NS (OR = 0.07 for T3, T4) and LND (OR = 2.41 for T2) procedures. As for Pros-IT CNR data, GS >= (4 + 3) and positive cancer cores >= 50% were decisive factors both for NS (OR 0.29 and 0.30) and LND (OR 7.53 and 2.31) strategies. Conclusions PCa management has changed over the last decade in Italian centers: RA and NS procedures without LND have become the methods of choice to treat newly medium-high risk diagnosed PCa.

Abstract

Penelitian ini bertujuan untuk meningkatkan motivasi dan hasil belajar matematika siswa kelas 2 SD di Cita Hati Christian School melalui penerapan prosedur Polya, yaitu pemahaman masalah, menemukan rencana, melaksanakan rencana dan evaluasi. Prosedur Polya ini akan diterapkan dalam pembelajaran kooperatif tipe Teams Games Tournament (TGT). Penelitian ini menggunakan metode penelitian tindakan kelas (PTK) yang dilaksanakan dalam dua siklus. Setiap siklus terdiri dari perencanaan, pelaksanaan, observasi, dan refleksi. Subjek penelitian ini adalah siswa kelas 2 yang berjumlah 22 siswa. Instrumen yang digunakan dalam penelitian ini meliputi lembar observasi dan tes hasil belajar. Hasil penelitian menunjukkan bahwa penerapan prosedur Polya dalam pembelajaran kooperatif tipe TGT dapat meningkatkan motivasi dan hasil belajar siswa di pelajaran Matematika. Hal ini dibuktikan dengan adanya konsistensi skor rata-rata motivasi belajar siswa yaitu 100% pada siklus I dan siklus II. Selain itu, hasil belajar siswa juga mengalami peningkatan, dimana rata-rata nilai tes hasil belajar siswa meningkat dari 91% pada siklus I menjadi 95% pada siklus II. Dengan demikian, dapat disimpulkan bahwa penggunaan prosedur Polya dalam pembelajaran kooperatif tipe TGT efektif dalam meningkatkan motivasi dan hasil belajar matematika siswa kelas 2 di Cita Hati Christian School.

Abstract

A thermo-nuclear fusion experimental reactor produces a large amount of energy that is mainly transported from the plasma by neutrons and deposited in the machine components, generating nuclear activation. One of the elements used in the fusion reaction is tritium, the radioactive isotope of hydrogen, but in many instances, experimental facilities are based on deuterium only reactions and do not use tritium as fuel. High temperatures in the plasma-facing components and in other parts of these facilities require specific cooling systems that are subject to the neutron activation and that transport the radioactivity into their loops and components. Because of the above considerations, the radiation sources in an experimental fusion machine could be: • the primary neutronic field resulting from the fusion reactions occurring in the reaction chamber, • the gamma radiation generated from neutrons’ interaction with the machine components, • the X and gamma radiation due to the plasma currents, • the gamma radiation emitted by activated products in the machine components • loose contamination from activated dust generated in the machine components, • activated corrosion products generated in the cooling loops after the activation of the inner wall of cooling water pipes, • activation of the cooling water, • tritium used as fuel for the fusion reaction or produced in the D-D fusion reactions, • wastes, still containing tritium and gamma emitters, • activated air produced in the main hall atmosphere and released to the environment, • neutrons and secondary gamma radiation generated in Neutral Beam Injectors. The current analysis is a brief review of the studies on the subject, aimed to define the radiation protection approach to be applied to the next fusion experimental machines. Specific reference will be made to the DTT experimental fusion device which is in an advanced design phase in Italy. Activities developed at different experimental fusion machines, like TFTR in the USA, JET in England, JT60 in Japan, together with some minor experiments implemented in Italy, will be the basis for identifying the typical radiological source terms. Finally, the studies performed for designing the international project ITER and the Italian DTT will be considered for providing qualitative and quantitative information about the radiological source terms and the potential radioactive waste produced and released to the environment.

Abstract

Heating neutral beam (HNB) injectors, necessary to achieve burning conditions and to control plasma instabilities in ITER, are characterized by such demanding parameters that a neutral beam test facility (NBTF) dedicated to their development and optimization is being realized in Padua (Italy) with direct contributions from the Italian government (through Consorzio RFX as the host entity) and the ITER international organization (with kind contributions from the ITER domestic agencies of Europe, Japan and India) and technical and scientific support from various European laboratories and universities. The NBTF hosts two experiments: SPIDER, devoted to ion source optimization for the required source performance, and MITICA, the full-size prototype of the ITER HNB, with an ion source identical to SPIDER. This paper gives an overview of the progress towards NBTF realization, with particular emphasis on issues discovered during this phase of activity and on solutions adopted to minimize the impact on the schedule and maintain the goals of the facilities. The realization of MITICA is well advanced; operation is expected to start in 2023 due to the long procurement time of the in-vessel mechanical components. The beam source power supplies, operating at 1 MV, are in an advanced phase of realization; all high-voltage components have been installed and the complex insulation test phase began in 2018. At the same time, construction and installation of SPIDER plant systems was successfully completed with their integration into the facility. The mechanical components of the SPIDER ion source were installed inside the vessel and connected to the plants. Integrated commissioning with the control, protection and safety systems ended positively and the first experimental phase has begun. The first results of the SPIDER experiment, with data from operational diagnostics, and the plans for the 1 MV insulation tests on the MITICA high-voltage components are presented.

Abstract

The “Divertor Tokamak Test facility, DTT” is a milestone along the international program aimed at demonstrating – in the second half of this century – the feasibility of obtaining to commercial electricity from controlled thermonuclear fusion.DTT is a Tokamak conceived and designed in Italy with a broad international vision. The construction will be carried out in the ENEA Frascati site, mainly supported by national funds, complemented by EUROfusion and European incentive schemes for innovative investments. The project team includes more than 180 high-standard researchers from ENEA, CREATE, CNR, INFN, RFX and various universities.The volume, entitled DTT Interim Design Report (“Green Book” from the colour of the cover), briefly describes the status of the project, the planning of the design future activities and its organizational structure. The publication of the Green Book also provides an occasion for thorough discussions in the fusion community and a broad international collaboration on the DTT challenge.

Abstract

This study aimed to analyze the community structure of reef fish in the small island waters of Central Bangka. The research was conducted in March 2022 in the waters of Bebuar Island, Ketugar Island, Ketawai Island, and Gusung Asam Central Bangka Regency, Bangka Belitung Islands Province. The coral reef survey method uses purposive sampling and the coral reef fish visual census with data analysis of abundance, diversity index, uniformity, and dominance. In-site water quality measurements included six parameters: temperature, salinity, current, brightness, depth, and pH. The results showed that coral reef cover from 44.60 - 87.00% with the medium to very good category. The abundance of reef fish from 1 to 3 individuals/m2 was dominated by 18 species of the Pomacentridae family. The reef fish diversity index ranged from 2.47 to 2.90, including the medium category. The uniformity index value ranged from 0.79 - 0.94, including the high category, indicating that the species found were evenly distributed across all stations. The dominance index value ranges from 0.07 to 0.15, including low, where no species dominates significantly and the fish are evenly distributed. Temperature, brightness, pH, and salinity are factors that support the growth of coral and reef fish. A water current speed of 0.05 – 0.20 m/s is quite helpful in cleaning coral reefs from mud and can supply oxygen. Water conditions were categorized as good and fulfilled the seawater quality standards.

Abstract

Nel presente lavoro si esamina il caso ipotetico di un incendio sviluppatosi in un laboratorio di ricerca in cui sono presenti quattro scatole a guanti, nelle quali sono avvenute lavorazioni di fabbricazione di combustibile a ossidi misti U-Pu Mixed OXide (MOX). L’analisi parte dalla valutazione del rischio incendio, che si basa sulla quantificazione del carico di incendio all’interno del laboratorio e tiene inoltre conto dell’efficacia dei presidi antincendio presenti (impianti di rilevazione, presidi mobili di spegnimento, impianti automatici di spegnimento). L’incendio, coinvolgendo le quattro scatole a guanti, rappresenta una potenziale fonte di contaminazione radioattiva con dispersione di particolato. Sulla base di questi presupposti si valuta la dose efficace impegnata ai gruppi di riferimento dei lavoratori (secondo i requisiti dell’art. 115-ter D.Lgs. 230/95) in relazione alla specifica granulometria del particolato contenuto nell’aerosol, stimando la velocità di rilascio, la concentrazione del contaminante in aria e la frazione respirabile del particolato. Specifica attenzione è rivolta al compartimento polmonare che, nello scenario incidentale considerato, rappresenta il distretto più critico in termini di potenziale conferimento di dose al soggetto esposto, a seguito di inalazione del particolato radioattivo

Abstract

Substantial progresses have been achieved in the realization of the ITER Neutral Beam Test Facility (NBTF) hosted in Padova, Italy; the buildings, completed by the end of 2015, are being progressively filled with new systems and components. The realization of SPIDER, the ITER full-size negative ion source, is well advanced and important progress is also recorded for MITICA, the full-scale prototype of the ITER HNB injector. The paper gives an overview of the achieved results, highlighting the main challenges faced.

Abstract

DTT (Divertor Tokamak Test) is an Italian scientific project in the field of nuclear fusion aimed at creating a European facility for the testing of power exhaust and helium removal strategies alternative to the one implemented in ITER, the international experiment in fusion that will prove the feasibility of fusion energy on a large scale from 2025. In the European Fusion Roadmap to the realisation of fusion energy within 2050, one of the main mission is indeed the study of the power released by the plasma core to the so-called divertor, a keycomponent of any present tokamak machine on which the thermal power is directed. This will be tested also in ITER on a "standard" divertor operating in a plasma fully detached condition, i.e. no contact between plasma and first wall of the vessel. Nonetheless, the baseline strategy implemented in ITER could not be extrapolated to DEMO and future power plants, then the problem of thermal loads on the divertor may remain particularly critical in the road to the realization of a reactor. For this reason, a facility like DTT, where a number of scaled experiments, fully integrated with the expected physical parameters and engineering solutions to be used in DEMO, could be tested, is of paramount importance. Indeed, DTT has been designed so far to retain the possibility of testing different divertor magnetic configurations, including liquid metal divertor targets, and other possible promising solutions to face the power exhaust problem. The construction, recently approved by the Italian government, is rapidly approaching on the base of a completely renovated design with respect to the original proposal in order to account also for double-null configurations (up-down symmetric) and to keep costs within the original budget. DTT will be a high field superconducting toroidal device (6 T) carrying plasma current up to 5.5 MA in pulses with length up to about 100s and with 45 MW of additional heating power. The nominal cross section is elongated with a major radius R=2.11 m and a minor radius a=0.64m. This paper describes the status of the design activities of DTT. In particular the integrated design approach adopted, the rationale for the design choices, the development of the integrated management system are illustrated. Also a look on the main components, namely magnet system, plasma scenarios, vacuum vessel, invessel components, thermal shield, neutron shield, and additional heating system will be taken.

Abstract

Purpose.In this article, the authors propose useful operational indications to approach in the best possible way the issues concerning the design of a facility for manufacturing radiopharmaceuticals, with focus on organizational and safety aspects. Methods.Several documents produced by authoritative bodies, national and international scientific institutions and associations were examined and referenced, to the purpose of reviewing all available information in the field. Results.Indications are gathered for the design stage, including the organization of accesses and routes and characteristics and requirements of premises and systems. Conclusions.Main goal is to guide the reader in evaluating and choosing the most suitable features and equipment to limit the risks due to ionizing radiation and to prevent contamination of the workers and the environment.
