Mice made experimentally obese were observed to have an earlier appearance and a greater incidence of tumors than were the normal-weight mice. A fatenriched diet increased the observed tumor incidence in normal-weight mice to about that of obese mice given a regular diet, while a fat-enriched diet further enhanced the already augmented observed yield of mammary tumors in the obese mice given a regular diet. Statistical analysis supported the dependence of tumor incidence and appearance time on the type of diet. The calories consumed by normal mice on a fat-enriched diet was considerably below that of obese mice on a regular diet, yet produced about the same tumor incidence. SummaryMarked weight gains were produced in stock albino mice by single injections of goldthioglucose. Analysis of total body lipids, protein, water, and ash showed these weight gains to be primarily due to an increase in adipose tissue. This obesity occurred in approximately one-third of the survivors of sublethal doses of goldthioglucose. Except for centrolubular fatty infiltration of the liver in obese animals, no-anatomic lesions were found in the organs examined.

Six theophylline preparations have been tested for their absorption from the gastrointestinal tracts of animals as reflected by blood theophylline levels. These values were obtained first from the stomach, and then from the remainder of the gastrointestinal tract. The results showed satisfactory absorption with all medications, and the wide variation inherent in oral preparations. No real superiority was evident in the six preparations when the available theophylline was the same in each drug.

I N 1 954, Simeons reported that injection of 125 I.U. of chorionic gonadotropin daily enabled obese patients to adhere to a fat-free, 5(R) calorie diet for four to six weeks and to lose weight at a rate of 1 to 2 pounds daily during this period.’ . 2 Without the hormone, the patients experienced discomfort and would either eat secretly or discontinue the diet. Simeons suggested that chorionic gonadotropin acted by mobilizing fat from adipose tissue, making it more readily available for metabolic use by the body and thereby replacing the caloric deficit resulting from the rigid dietary restriction. Subsequently, Sohartm treated a group of obese patients for forty days with Simeons’ diet in combination with daily injections of either chorionic gonadotropin or saline solution. Both treated and control subjects showed a uniform average weight loss of 20 pounds. Sohar attributed the reduction in weight to diet rather than to the hormone. Theophylline and related xanthine derivatives are used extensively, at the present time, in the management of cardiovascular and respiratory diseases. However, there are conflicting opinions among investigators concerning the therapeutic value of these compounds in clinical conditions for which they are prescribed. Schack and Waxler1have shown that theophylline is restricted to the plasma, does not penetrate the red cell membrane and is only slightly bound to the proteins of the blood. Also, the drug appears to be only slightly bound to tissue proteins, since it was readily removed from the liver by simple perfusion with saline solution. It may therefore be suggested that any therapeutic effect of this drug will be closely related to its level in the blood.
