1. Impedance-based real-time monitoring of neural stem cell differentiation

Abstract

We present here the first impedance-based characterization of the differentiation process of two human mesencephalic fetal neural stem lines. The two dopaminergic neural stem cell lines used in this study, Lund human mesencephalic (LUHMES) and human ventral mesencephalic (hVM1 Bcl-XL), have been developed for the study of Parkinsonian pathogenesis and its treatment using cell replacement therapy. We show that if only relying on impedance magnitude analysis, which is by far the most usual approach in, e.g., cytotoxicity evaluation and drug screening applications, one may not be able to distinguish whether the neural stem cells in a population are proliferating or differentiating. However, the presented results highlight that equivalent circuit analysis can provide detailed information on cellular behavior, e.g. simultaneous changes in cell morphology, cell-cell contacts, and cell adhesion during formation of neural projections, which are the fundamental behavioral differences between proliferating and differentiating neural stem cells. Moreover, our work also demonstrates the sensitivity of impedance-based monitoring with capability to provide information on changes in cellular behavior in relation to proliferation and differentiation. For both of the studied cell lines, in already two days (one day after induction of differentiation) equivalent circuit analysis was able to show distinction between proliferation and differentiation conditions, which is significantly earlier than by microscopic imaging. This study demonstrates the potential of impedance-based monitoring as a technique of choice in the study of stem cell behavior, laying the foundation for screening assays to characterize stem cell lines and testing the efficacy epigenetic control.

Abstract

We present the combined results on electron-pair production in 158 GeV/n {Pb-Au} ($\sqrt{s}$= 17.2 GeV) collisions taken at the CERN SPS in 1995 and 1996, and give a detailed account of the data analysis. The enhancement over the reference of neutral meson decays amounts to a factor of 2.31$\pm0.19 (stat.)\pm0.55 (syst.)\pm0.69 (decays)$ for semi-central collisions (28% $\sigma/\sigma_{geo}$) when yields are integrated over $m>$ 200 MeV/$c^2$ in invariant mass. The measured yield, its stronger-than-linear scaling with $N_{ch}$, and the dominance of low pair $p_t$ strongly suggest an interpretation as {\it thermal radiation} from pion annihilation in the hadronic fireball. The shape of the excess centring at $m\approx$ 500 MeV/$c^2$, however, cannot be described without strong medium modifications of the $\rho$ meson. The results are put into perspective by comparison to predictions from Brown-Rho scaling governed by chiral symmetry restoration, and from the spectral-function many-body treatment in which the approach to the phase boundary is less explicit., Comment: 39 pages, 40 figures, to appear in Eur.Phys.J.C. Abstract

We simulate the electrical properties of a device realized by a G protein coupled receptor (GPCR), embedded in its membrane and in contact with two metallic electrodes through which an external voltage is applied. To this purpose, recently, we have proposed a model based on a coarse graining description, which describes the protein as a network of elementary impedances. The network is built from the knowledge of the positions of the C-alpha atoms of the amino acids, which represent the nodes of the network. Since the elementary impedances are taken depending of the inter-nodes distance, the conformational change of the receptor induced by the capture of the ligand results in a variation of the network impedance. On the other hand, the fluctuations of the atomic positions due to thermal motion imply an impedance noise, whose level is crucial to the purpose of an electrical detection of the ligand capture by the GPCR. Here, in particular, we address this issue by presenting a computational study of the impedance noise due to thermal fluctuations of the atomic positions within a rhodopsin molecule. In our model, the C-alpha atoms are treated as independent, isotropic, harmonic oscillators, with amplitude depending on the temperature and on the position within the protein (alpha-helix or loop). Abstract

We report on a direct experimental evidence of shot noise in a linear macroscopic resistor. The origin of the shot noise comes from the fluctuation of the total number of charge carriers inside the resistor associated with their diffusive motion under the condition that the dielectric relaxation time becomes longer than the dynamic transit time. Present results show that neither potential barriers nor the absence of inelastic scattering are necessary to observe shot noise in electronic devices., Comment: 10 pages, 5 figures

Abstract

PURPOSE: Evaluate the percentage of patients with prostate cancer treated with luteinizing hormone-releasing hormone analogues (LHRHa) that develop castration resistance after a follow-up period of 3 years. The secondary objective is to evaluate the variables potentially related to the progression to castration resistant prostate cancer (CRPC). METHODS: A post-authorization, nation-wide, multicenter, prospective, observational, and longitudinal study that included 416 patients treated with LHRHa between 2012 and 2017 is presented. Patients were followed for 3 years or until development of CRPC, thus completing a per-protocol population of 350 patients. A Cox regression analysis was carried out to evaluate factors involved in progression to CRPC. RESULTS: After 3 years of treatment with LHRHa 18.2% of patients developed CRPC. In contrast, in the subgroup analysis, 39.6% of the metastatic patients developed CRPC, compared with 8.8% of the non-metastatic patients. The patients with the highest risk of developing CRPC were those with a nadir prostate-specific antigen (PSA) > 2 ng/ml (HR 21.6; 95% CI 11.7-39.8; p < 0.001) and those receiving concomitant medication, most commonly bicalutamide (HR 1.8; 95% CI 1-3.1, p = 0.0431). CONCLUSIONS: The proportion of metastatic patients developing CRPC after 3 years of treatment with LHRHa is consistent with what has been previously described in the literature. In addition, this study provides new findings on CRPC in non-metastatic patients. Concomitant medication and nadir PSA are statistically significant predictive factors for the time to diagnosis of CRPC, the nadir PSA being the strongest predictor.

Abstract

The promise of good energy and spatial resolution coupled with high efficiency and room temperature operation has fuelled a large international effort to develop cadmium zinc telluride (CZT) for hard X-ray applications. We are involved on the development of a hard X-ray telescope based on multilayer optics and focal plane detector operative in the 10-80 keV energy range. This telescope requires a high efficiency focal plane providing both fine spatial resolution and spectroscopy with a compact and robust design. This paper reports preliminary results on the characterization both in spectroscopic and spatial response of two small pixellated CZT detectors (10 x 10 x 1 m[m.sup.3] and 10 x 10 x 2 m[m.sup.3] single crystals) with 0.45 mm pixel size. We present the results obtained using both standard commercial read-out electronics Readout Electronics for Nuclear Applications (RENA) and innovative low noise and low power dissipation ASICs developed within the collaboration. Abstract

Chemical vapor deposition (CVD) diamond is a radiation-hard sensor material that may be used for charged particle tracking near the interaction region in experiments at high-luminosity colliders. The goal of the work described in this paper is to investigate the use of several detector planes made of CVD diamond strip sensors for charged particle tracking. Toward this end, a tracking telescope composed entirely of CVD diamond planes has been constructed. The telescope was tested in muon beams, and its tracking capability has been investigated. Abstract

A MOSFET, connected as a diode with gate and drain short circuited and biased in saturation mode, is proposed as a substitute for the high-value feedback resistor to provide dc feedback and signal reset in integrated charge amplifiers with BJT input transistors. This paper describes the MOSFET design rules to set the desired value of the discharge time constant, to minimize its noise contribution, and to operate it in a low voltage circuit. The behavior of the MOSFET as an almost fixed value resistor is evidenced by the single time-constant discharge of the signal at the output of a prototype preamplifier for fast ( Index Terms - Integrated circuit design, MOSFET circuit, noise.

Abstract

The criteria underlying the design of low-noise front-end integrated electronics for radiation and particle detectors have been determined, taking into account the limits in the allowable power dissipation. The analysis specifically treats integrated amplifiers employing silicon bipolar transistors, whose performance has been studied to highlight the ultimate noise limit and the roles of the front-end device parameters such as the current gain, the base spreading resistance, the junction and diffusion capacitances, the transition frequency, and the device geometry. The relationships existing among the power dissipated in the front-end stage, the noise performance, and the characteristic of signal processing are derived. Abstract

A charge amplifier, especially designed for FET integrated on silicon radiation detectors, is presented. The circuit is based on the forward biased FET amplifier configuration, which does not require the feedback resistor nor additional devices for resetting. The severe constraints on the preamplifier design, imposed by the low transconductance of small size integrated FET's, are analyzed as far as loop-gain, bandwidth, and noise are concerned. A circuit topology which satisfies all the requirements even at relative fast shaping time (1 [[micro]second]) is proposed and experimentally tested.

An analysis of centroid of pulses enables the evaluation of drift detector pulse timing. Diffusion of electrons followed by drifting occurs in nonuniform fields. Time centroid of pulses is influenced by variations in the drift field for a constant time shift. Collecting field variations between the anodes necessitate the calibration of multianode detector channels.
