Abstract

In recent years, there has been increasing societal awareness of the crucial role that coastal dunes play in protecting against rising sea levels, mitigating climate change impacts, promoting biodiversity, and providing recreational opportunities. In some regions, dune management has been particularly focused on biodiversity and ecosystem restoration and the presence of alien species on dunes raises concerns about how these species become 'native,' 'invasive,' or 'hybrids' and whether they belong in their new ecosystems. These concerns illustrate how certain animals and plants assume different statuses according to normative categories associated with varying objectives. This article explores how perceptions of coastal dunes in the UK have transformed over time, from marginal resource frontiers to highly valued environments shaped by multispecies relations. In addition, this work explores how dunes around the UK emerge as 'patchy anthropocoasts,' that is, uneven landscapes designed by human purposes linked to economic activities, conservation, rabbit populations, unwanted vegetation, and the control of unpredictable sand movements. Bringing together diverse historical materials and scientific literature, this article links human and nonhuman histories with present debates on dune restoration from a transdisciplinary perspective rooted in anthropology, environmental history, and the natural sciences. Abstract

The LOng-Range Reconnaissance Imager (LORRI) is the high resolution imaging instrument for the New Horizons mission to Pluto, its giant satellite Charon, its small moons Nix and Hydra, and the Kuiper Belt, which is the vast region of icy bodies extending roughly from Neptune's orbit out to 50 astronomical units (AU). New Horizons launched on January 19, 2006 as the inaugural mission in NASA's New Frontiers program. LORRI is a narrow angle (field of view=0.29 deg), high resolution (4.95 microrad pixels), Ritchey-Chretien telescope with a 20.8 cm diameter primary mirror, a focal length of 263 cm, and a three lens field-flattening assembly. A 1024 x 1024 pixel (optically active region), thinned, backside-illuminated charge-coupled device (CCD) detector is used in the focal plane unit and is operated in frame transfer mode. LORRI provides panchromatic imaging over a bandpass that extends approximately from 350 nm to 850 nm. LORRI operates in an extreme thermal environment, situated inside the warm spacecraft with a large, open aperture viewing cold space. LORRI has a silicon carbide optical system, designed to maintain focus over the operating temperature range without a focus adjustment mechanism. Deficits are observed in multiple domains, including working memory, executive function, attention, and information processing. Disability caused by cognitive dysfunction is frequently as debilitating as the prominent emotional disturbances. Interactions between the hippocampus and the prefrontal cortex are increasingly appreciated as an important link between cognition and emotion. Recent developments in optogenetics, imaging, and connectomics can enable the investigation of this circuit in a manner that is relevant to disease pathophysiology. The goal of this review is to shed light on the contributions of this circuit to cognitive dysfunction in neuropsychiatric disorders, focusing on Alzheimer’s disease and depression. Abstract

Coastal sand dunes are multifunctional landscapes with rich biodiversity. In New Zealand, with the establishment of European settlement around 1840, dunes in the Manawatu-Whanganui region were affected due to the removal of their vegetation cover by human activities and animal grazing. As a result, sand drifted further inland affecting villages, infrastructure and agricultural areas. The main response was to introduce marram grass (Ammophila arenaria) used in Europe to stabilize dunes. This solution caused significant environmental impacts as marram grass turned invasive and native habitats of fauna and flora significantly decreased.This paper focused on the long-term analysis of aspects related to sand dune management in the region during two-time frames: 1) from the 19th to the late 20th century and 2) from then on to the early 21st century, using the innovative spiral DPSIR (Driver-Pressure-State-Impact-Response) framework. Data for this study comes from historical records, scientific literature and present management reports.The integrated spiral framework allows for establishing the connections between historical and future man-agement initiatives for mitigating and adapting to environmental impacts due to socio-economic drivers and their pressures. The study reinforces the paradigm shift from dune stabilization before the late 20th century to the restoration of stabilized dunes to make them active for enhancing native biodiversity should be again assessed in the context of sea-level rise during this century. Coastal managers should adopt an optimized solution between these two extreme solutions adopted from the 19th century to the present, by considering long-term and interdisciplinary analysis to better understand the systems' evolution and the full consequences of human actions. info:eu-repo/semantics/publishedVersion

Abstract

Purpose: Triple-negative breast cancer (TNBC) is a heterogeneous subgroup of breast cancer that is associated with a poor prognosis. We evaluated the activity of CDK4/6 inhibitors across the TNBC subtypes and investigated mechanisms of sensitivity. Experimental Design: A panel of cell lines representative of TNBC was tested for in vitro and in vivo sensitivity to CDK4/6 inhibition. A fluorescent CDK2 activity reporter was used for single-cell analysis in conjunction with time-lapse imaging. Results: The luminal androgen receptor (LAR) subtype of TNBC was highly sensitive to CDK4/6 inhibition both in vitro (P < 0.001 LAR vs. basal-like) and in vivo in MDA-MB-453 LAR cell line xenografts. Single-cell analysis of CDK2 activity demonstrated differences in cell-cycle dynamics between LAR and basal-like cells. Palbociclib-sensitive LAR cells exit mitosis with low levels of CDK2 activity, into a quiescent state that requires CDK4/6 activity for cell-cycle reentry. Palbociclib-resistant basal-like cells exit mitosis directly into a proliferative state, with high levels of CDK2 activity, bypassing the restriction point and the requirement for CDK4/6 activity. High CDK2 activity after mitosis is driven by temporal deregulation of cyclin E1 expression. CDK4/6 inhibitors were synergistic with PI3 kinase inhibitors in PIK3CA-mutant TNBC cell lines, extending CDK4/6 inhibitor sensitivity to additional TNBC subtypes. Conclusions: Cell-cycle dynamics determine the response to CDK4/6 inhibition in TNBC. CDK4/6 inhibitors, alone and in combination, are a novel therapeutic strategy for specific subgroups of TNBC. Abstract

International audience; Barley (Hordeum vulgare L.) is among the world's earliest domesticated and most important crop plants. It is diploid with a large haploid genome of 5.1 gigabases (Gb). Here we present an integrated and ordered physical, genetic and functional sequence resource that describes the barley gene-space in a structured whole-genome context. We developed a physical map of 4.98 Gb, with more than 3.90 Gb anchored to a high-resolution genetic map. Projecting a deep whole-genome shotgun assembly, complementary DNA and deep RNA sequence data onto this framework supports 79,379 transcript clusters, including 26,159 'high-confidence' genes with homology support from other plant genomes. Abundant alternative splicing, premature termination codons and novel transcriptionally active regions suggest that post-transcriptional processing forms an important regulatory layer. Survey sequences from diverse accessions reveal a landscape of extensive single-nucleotide variation. Our data provide a platform for both genome-assisted research and enabling contemporary crop improvement.
