11. Fate of As(V) and Cr(VI) adsorbed on goethite in a mangrove‐microcosm experiment.

Abstract

Mangroves provide fundamental ecosystem services; however, the growing impact of human activities has resulted in increased pollution pressure, such as chemical contaminants. The redox processes are major biogeochemical players in the fate of contaminants. We investigate the effects of the redox environment on As(V) and Cr(VI) adsorbed on goethite (i.e., Fe(III)‐oxide) over 40 days of incubation in columns containing mangrove soil subjected to seawater saturation cycles. Our spectroscopic data highlighted a less Fe(III)‐ordered arrangement on goethite over time; As(V) is strongly bound to goethite, which delayed until 20 days its remobilization and reduction to As(III). After 40 days, the goethite held ∼50% of the initial As, but it was 15% as As(III). On the other hand, Cr(VI) was desorbed almost completely in less than 10 days, and the residual Cr ions bound to goethite were almost totally converted to Cr(III). Our study stresses the importance of individual time‐dependence in evaluating chemical speciation changes among potential toxic elements in wetland systems, such as mangroves and artificial wetlands designed to water treatment or soil remediation. Abstract

We introduce and study the block voter model with noise on two-dimensional square lattices using Monte Carlo simulations and finite-size scaling techniques. The model is defined by an outflow dynamics where a central set of $N_{PCS}$ spins, here denoted by persuasive cluster spins (PCS), tries to influence the opinion of their neighbouring counterparts. We consider the collective behaviour of the entire system with varying PCS size. When $N_{PCS}>2$, the system exhibits an order-disorder phase transition at a critical noise parameter $q_{c}$ which is a monotonically increasing function of the size of the persuasive cluster. We conclude that how large the PCS is more power of persuasion it has. It also seems that the resulting critical behaviour is Ising-like independent of the range of the interactions., Comment: 10 pages, 7 figures

Since 2011, FDA (Food and Drug Administration) approved several novel therapeutic strategies, such as MAPK pathway targeted therapies, to treat cutaneous melanoma patients. However, their improvements in overall survival were limited, due to the development of resistance.Methods: In this work, several combinations of therapies, including the metabolic modulator DCA, were tested in melanoma cell lines, considering that MAPK and PI3K/AKT/mTOR pathways are deregulated and interconnected in melanoma and that the presence of the Warburg effect in melanoma cells may influence the response to therapy. The effect of the treatments was assessed in the proliferation and survival of melanoma cell lines with different genetic profiles. Also, the possibility to overcome resistance to the treatment with vemurafenib was tested.Results: In general, higher decrease in cell viability and cell proliferation and increase in apoptosis were obtained after the combination treatments, comparing with single treatments, in all the studied cell lines. The combination of cobimetinib and everolimus appear to be the best treatment option. The BRAFV600E -vemurafenib resistant melanoma cell line showed to retain sensitivity to both everolimus and DCA.Discussion and Conclusion: Our results suggest that the combination of MAPK pathway inhibitors with mTOR pathway inhibitors and DCA should be considered as therapeutic options to treat melanoma patients, as the combinations potentiated the effects of each drug alone. In a cell line resistant to vemurafenib, we verified that combined MAPK inhibitors with inhibition of mTOR pathway and/or DCA metabolism modulation might constitute possible strategies in order to overcome resistance to MAPK inhibition.Keywords: melanoma, vemurafenib, cobimetinib, everolimus, DCA, metabolism

31. Confidence and Accuracy in the Recall of Deceptive and Nondeceptive Sentences

Abstract

Two experiments were carried out to study the metamemory theory of confidence for the domain of sentence recall. Experiment 1 used nondeceptive sentences and deceptive synonym substitution sentences. Experiment 2 used nondeceptive sentences and deceptive schema inference sentences. In both experiments there was a strong positive relationship between memory confidence and memory accuracy for nondeceptive items and a much reduced relationship for deceptive items. The results were interpreted as showing that participants generated their confidence judgments using the metamemory belief that complete recalls are accurate recalls. This metamemory belief was valid for the nondeceptive sentences, which led to the strong positive relationship between confidence and accuracy. The reduced relationship for the deceptive items was interpreted as being due to the participants not being aware of making errors on the deceptive items, and thus using their metamemory belief about completeness to assign inappropriately high confidence to recall errors. Overall the results were taken to support the metamemory theory of confidence.

Background: Huntington's disease (HD) is a rare neurodegenerative disorder with protean clinical manifestations. Its management is challenging, consisting mainly of off-label treatments. Objectives: The International Parkinson and Movement Disorder Society commissioned a task force to review and evaluate the evidence of available therapies for HD gene expansion carriers. Methods: We followed the Grading of Recommendations Assessment, Development and Evaluation (GRADE) approach. Eligible randomized controlled trials were identified via an electronic search of the CENTRAL, MEDLINE, and EMBASE databases. All eligible trials that evaluated one or more of 33 predetermined clinical questions were included. Risk of bias was evaluated using the Cochrane Risk of Bias tool. A framework was adapted to allow for efficacy and safety conclusions to be drawn from the balance between the GRADE level of evidence and the importance of the benefit/harm of the intervention. Results: Twenty-two eligible studies involving 17 interventions were included, providing data to address 8 clinical questions. These data supported a likely effect of deutetrabenazine on motor impairment, chorea, and dystonia and of tetrabenazine on chorea. The data did not support a disease-modifying effect for premanifest and manifest HD. There was no eligible evidence to support the use of specific treatments for depression, psychosis, irritability, apathy, or suicidality. Similarly, no evidence was eligible to support the use of physiotherapy, occupational therapy, exercise, dietary, or surgical treatments. Conclusions: Data for therapeutic interventions in HD are limited and support only the use of VMAT2 inhibitors for specific motor symptoms. © 2021 International Parkinson and Movement Disorder Society.
