Die konventionelle Behandlung von Krebs mit Bestrahlung oder Chemotherapeutika hat mit der Entwicklung resistenter Tumorzellen zu kämpfen und schädigt darüber hinaus gesundes Gewebe. In den letzten Jahren wurden selektive Therapieansätze entwickelt, die biologische Mechanismen nutzen, um gezielt Tumorzellen anzugreifen und deren Resistenz zu umgehen. Für die Konstruktion der hier untersuchten zytotoxischen Immunkonjugate wurden die codierenden Sequenzen der Antigen-bindenden Domänen von Antikörpern mit den entsprechenden Sequenzen von Todesliganden aus der TNF-Superfamilie genetisch fusioniert. Im Fall von FasL/CD95L basiert das Funktionsprinzip auf der Beobachtung, daß dieser Todesligand nur in membranständiger Form eine ausgeprägte apoptotische Wirkung besitzt, in löslichem Zustand jedoch keine nennenswerte zytotoxische Aktivität aufweist. Solche rekombinanten Fusionsproteine verfügen über ein latentes zytotoxisches Potenzial, das erst mit spezifischer Bindung an ein membranständiges Antigen aktiviert wird. Eine zielgerichtete und lokal begrenzte zytotoxische Aktivität von FasL/CD95L kann somit durch geeignete Wahl des Antikörpers erreicht werden. Dieses Konzept wurde mit dem Fusionsprotein scFv40-Flag-FasL umgesetzt, dessen Antigen-bindende Domäne scFv40 Spezifität für das Tumorstromaantigen "fibroblast activation protein" (FAP) besitzt. In dieser Arbeit wurde eine funktionelle Charakterisierung von scFv40-Flag-FasL in vitro vorgenommen und die antitumorale Wirkung in vivo untersucht. Mit HEK293-Zellen, die stabil mit dem Expressionsplasmid für scFv40-Flag-FasL transfiziert worden waren, wurde das rekombinante Protein im Labormaßstab produziert. Die Bioaktivität des Fusionsproteins wurde in vitro an HT1080- und HT1080-FAP-Zellen untersucht. Mittels FACS-Analyse konnte gezeigt werden, daß scFv40-Flag-FasL spezifisch an HT1080-FAP-Zellen bindet, nicht aber an HT1080-Zellen. In Bioassays wirkte scFv40-Flag-FasL auf HT1080-FAP-Zellen um den Faktor 1000 zytotoxischer als auf parentale HT1080-Zellen, obwohl beide Zell-Linien grundsätzlich eine vergleichbare Sensitivität für FasL-induzierten Zelltod besitzen. Die Präinkubation mit dem Caspaseinhibitor z-VAD-fmk schützt jedoch HT1080-FAP-Zellen vor der zytotoxischen Wirkung von scFv40-Flag-FasL. Eine genauere Charakterisierung zeigte, daß die Stimulation mit scFv40-Flag-FasL selektiv in HT1080-FAP-Zellen die Prozessierung von Procaspase-8 und die Aktivierung von Caspase-3 induziert. Die Behandlung mit scFv40-Flag-FasL stimulierte auch die geninduktiven Fähigkeiten von Fas/CD95, insbesondere dann, wenn die Aktivität der Caspasen mit z-VAD-fmk blockiert wurde. Unter diesen Bedingungen konnte exklusiv in FAP-positiven HT1080-Zellen die Induktion von IL8 im ELISA demonstriert werden. IL8 ist ein bekanntes Zielgen des Transkriptionsfaktors NFkB, der über CD95/Fas aktiviert werden kann. Mit der Bindung und Immobilisierung an membranständigem FAP wird scFv40-Flag-FasL in einen vollwertigen bioaktiven Zustand versetzt und kann, ähnlich rekombinantem quervernetztem FasL, sowohl die proapoptotischen als auch die geninduktiven Funktionen des Rezeptors CD95/Fas kompetent induzieren. Die Anwendung Fas-spezifischer Antikörper oder bestimmter rekombinanter FasL-Varianten in der Tumortherapie scheiterte bisher an den letalen Schädigungen der Leber, die durch deren systemische Applikation in vivo ausgelöst werden. Wie die Untersuchung der systemischen Toxizität von scFv40-Flag-FasL in vivo demonstrierte, ist hier die Konstruktion eines Fas-Agonisten gelungen, der nach einmaliger intravenöser Administration selbst in höchsten Dosen keine lebensbedrohlichen Komplikationen in Mäusen auslöst. Die antitumorale Wirkung von scFv40-Flag-FasL wurde durch Behandlung von HT1080- und HT1080-FAP-Zellen untersucht, die subkutan in nu/nu NMRI-Mäuse transplantiert worden waren. Durch wiederholte Applikation des Immunkonjugates nahe der Implantationsstelle wurde selektiv das Wachstum der HT1080-FAP-Zellen inhibiert, nicht jedoch das parentaler HT1080-Zellen. In vivo beruht die antitumorale Wirkung von scFv40-Flag-FasL möglicherweise nicht allein auf einer unmittelbaren proapoptotischen Funktion. Im Zusammenhang mit der "tumor counter-attack"-Hypothese wurde beobachtet, daß FasL-exprimierende Tumorzellen auch in syngenen Mäusen durch neutrophile Granulozyten abgestoßen werden. In anderen Studien wurde die Eignung der ektopischen FasL-Expression in pankreatischen Inselzellen zum Schutz vor Abstoßung in allogenen Rezipienten untersucht, es wurde jedoch eine beschleunigte Abstoßung unter Entwicklung massiver inflammativer Prozesse beobachtet. Die Aktivierung solcher Effektormechanismen des angeborenen Immunsystems könnte auch für die Wirkung von scFv40-Flag-FasL in vivo verantwortlich sein. Diese Ergebnisse demonstrieren, daß das Prinzip der Antigen-abhängigen Immobilisierung von FasL und der lokalen Aktivierung des Rezeptors Fas/CD95 in vivo für die Tumortherapie genutzt werden kann., Conventional cancer therapy, like e. g. radiation and treatment with chemotherapeutics, is hampered by development of resistant tumor cells and furthermore it damages healthy tissue. In recent years selective approaches for cancer therapy have been developed using existing biological mechanisms to selectively attack tumor cells and to circumvent their resistance mechanisms. For construction of antibody derivatives that show selective cytotoxicity the sequences encoding the antigen-binding domains of antibodies have been genetically fused with the corresponding sequences of death ligands of the TNF-superfamily. In case of CD95L/FasL the working principle is based on the observation, that only the membrane bound form of this ligand possesses a strong proapoptotic activity, while the soluble form does not display a significant cytotoxic activity. This kind of fusion proteins display a latent cytotoxic potential, that is activated exclusively upon specific binding to a membrane bound antigen. Therefore by choice of an appropriate antibody targeted and local activation of CD95L/FasL is achieved. This concept has been realized with the fusion protein scFv40-Flag-FasL. Its antigen-binding domain scFv40 specifically binds the tumorstroma marker "fibroblast activation protein" (FAP). The object of this work was to characterize bioactivity of scFv40-Flag-FasL functionally in vitro and to analyze its postulated antitumoral activity in vivo. The recombinant protein was produced in laboratory scale using HEK293 cells, that have been stably transfected with the corresponding expression plasmid. The bioactivity of scFv40-Flag-FasL was analyzed in vitro using HT1080 and HT1080-FAP cells. By FACS analysis it was shown, that scFv40-Flag-FasL specifically binds to HT1080-FAP cells, but not to parental HT1080 cells. In cell-based assays it was demonstrated, that scFv40-Flag-FasL is about 1000 times more cytotoxic towards HT1080-FAP cells than to parental HT1080 cells, despite both cell lines show a similar sensitivity towards FasL-induced cell death in general. Preincubation with the pancaspase inhibitor z-VAD-fmk protects HT1080-FAP-cells from the scFv40-Flag-FasL-induced cell death. A more detailed characterization demonstrated, that stimulation with scFv40-Flag-FasL exclusively induces processing of procaspase-8 and activation of caspase-3 in HT1080-FAP cells. Furthermore treatment with scFv40-Flag-FasL activates the geninductive capacities of CD95/Fas, especially if caspase activity is blocked with z-VAD-fmk. Under these conditions induction of IL8 was demonstrated exclusively in HT1080-FAP cells by ELISA. IL8 is an known target of the transcription factor NFkB, that is also activated via CD95/Fas signaling. Therefore, by binding to a membrane bound antigen and immobilization scFV40-Flag-FasL is converted into a fully signaling competent entity, that is able to induce proapoptotic as well as gene inductive functions of CD95/Fas, in comparable manner to recombinant aggregated FasL. The use of Fas-specific antibodies or certain recombinant FasL preparations in tumor therapy was impossible so far because these reagents induce severe liver damage after intraveneous application. Intraveneous administration of a single high dose of scFv40-Flag-Fasl in mice does not induce lethal liver damage and therefore lacks systemic toxicity in vivo, demonstrating the construction of a Fas-agonist that might be successfully employed in tumor therapy. The antitumoral activity of scFv40-Flag-FasL was investigated by treatment of HT1080 and HT1080-FAP cells, that have been xenotransplanted subcutaneously in nu/nu NMRI mice. Repeated administration of scFv40-Flag-FasL into an area close to the inoculation site selectively inhibited growth of HT1080-FAP cells, while palpable tumors grew from parental HT1080 cells. In vivo the antitumoral activity might not only be a result of a direct proapoptotic action of scFv40-Flag-FasL. In context of the socalled tumor counter attack hypothesis it has been reported, that FasL-expressing tumor cells are rejected even in syngenic mice by recruitment of neutrophils. Other studies tested the feasibility of ectopic FasL expression to protect pancreatic islet cells from rejection in allogenic recipients, but instead observed an accelerated rejection of transplanted cells under development of massive inflammatory processes. Induction of effector mechanisms of the innate immune system might therefore also be responsible for the observed activity of scFv40-Flag-FasL in vivo. Nevertheless, the results described herein demonstrate, that in principle antibody-mediated immobilization of FasL and local activation of CD95/Fas might be a feasible strategy for cancer therapy in vivo.
