There has been an increased concern on indoor air quality (IAQ) in residences since the majority of individuals’ time is mainly spent indoors. We inspected and measured indoor environmental parameters in 399 homes in northeast China in order to study IAQ. We systematically measured multilevel environmental parameters (physical, chemical, and biological) in children’s bedrooms during all seasons. The results indicated that the median values for indoor temperature, relative humidity, total volatile organic compounds (TVOC), and formaldehyde concentrations throughout the year were within the Chinese national standards. However, the median carbon dioxide concentrations exceeded 1000 ppm during spring, autumn, and winter. In the same seasons, the air change rate (ACR) was below the minimum required level of 0.5 h−1. Di-2-ethylhexyl phthalate (DEHP), di-n-butyl phthalate (DnBP), and di-isobutyl phthalate (DiBP) were predominantly detected in settled dust, displaying median concentrations of 126.9, 41.5, and 16.3 μg/g, respectively. Notably, phthalate concentrations were significantly higher in urban houses as compared to rural houses. Furthermore, median concentrations of Dermatophagoides farinae (Der f) and endotoxin were 689.4 ng/g and 3689.1 EU/g, respectively, trending higher in winter than summer. There was a negative correlation between ACR and chemical pollutants (TVOC, formaldehyde, and DiBP). In conclusion, northeast Chinese homes had poor indoor air quality with ubiquitous exposure to modern chemical compounds and insufficient ventilation.

Autophagy, apoptosis, and the unfolded protein response (UPR) are fundamental biological processes essential for manifold cellular functions in health and disease. Idiopathic pulmonary fibrosis (IPF) is a progressive and lethal pulmonary disorder associated with aging that has limited therapies, reflecting our incomplete understanding. We conducted an observational study linking molecular markers of cell stress response pathways (UPR: BiP, XBP1; apoptosis: cleaved caspase-3; autophagy: LC3β) in lung tissues from IPF patients and correlated the expression of these protein markers to each subject's lung function measures. We hypothesized that changes in lung tissue expression of apoptosis, autophagy, and UPR markers correlate with lung function deficits in IPF. The cell stress markers BiP, XBP1, LC3β puncta, and cleaved caspase-3 were found to be elevated in IPF lungs compared to non-IPF lungs, and, further, BiP and cleaved caspase-3 co-localized in IPF lungs. Considering lung function independently, we observed that increased XBP1, BiP, and cleaved caspase-3 were each associated with reduced lung function (FEV1, FVC, TLC, RV). However, increased lung tissue expression of LC3β puncta was significantly associated with increased diffusion capacity (DLCO), an indicator of alveolar-capillary membrane function. Similarly, the co-localization of UPR (XBP1, BiP) and autophagy (LC3β puncta) markers was positively correlated with increased lung function (FEV1, FVC, TLC, DLCO). However, the presence of LC3β puncta can indicate either autophagy flux inhibition or activation. While the nature of our observational cross-sectional study design does not allow conclusions regarding causal links between increased expression of these cell stress markers, lung fibrosis, and lung function decline, it does provide some insights that are hypothesis-generating and suggests that within the milieu of active UPR, changes in autophagy flux may play an important role in determining lung function. Further research is necessary to investigate the mechanisms linking UPR and autophagy in IPF and how an imbalance in these cell stress pathways can lead to progressive fibrosis and loss of lung function. We conclude by presenting five testable hypotheses that build on the research presented here. Such an understanding could eventually lead to the development of much-needed therapies for IPF.

Due to several advantages that Fiber Reinforced Polymer (FRP) composites offer in comparison with other strengthening techniques for steel structures, they have attracted considerable interest over the past three decades. However, the efficiency of FRP-strengthening for structures was found to be highly dependent on the quality and durability of the adhesive bonding between FRP and steel surface. This paper presents a state-of-the art review on the bond degradation in FRP-steel joints under different environmental conditions. Topics reviewed in this paper comprise the effect of different environmental conditions and their combination on bond durability, synergic effect of combining environmental condition with mechanical load, and the recent developments in the numerical simulation of aged bond. The paper concludes with the research gaps and uncertainties for further investigations.

Vascular Ehlers-Danlos syndrome (vEDS) is a genetic disease caused by a pathogenic mutation in the COL3A1 gene. Despite its severe course, the rarity and extreme clinical variability of the disease can pose significant obstacles to a timely diagnosis. Early and accurate diagnosis may lead to improved patient outcomes by providing access to targeted pharmacological treatments like celiprolol and enhancing the management of vEDS-related complications. Herein, we report a patient harboring a novel de novo COL3A1 missense variant, in which the diagnosis was only possible belatedly due to delayed referral for genetic evaluation. The patient developed pulmonary complications, aneurysms, and vascular malformations, and died at the age of 26 years due to massive pulmonary bleeding.

Investigation of aerodynamic characteristics of wind enhanced by bushfires is of great significance due to their destructive impacts on buildings located in bushfire-prone areas. Despite the abundance of studies in the fire-wind interaction domain, there have been limited studies concerning the effects of fire on wind aerodynamics. Fire source shape is one of the main factors affecting enhanced wind by fire. This study reports on the effects of fire source asymmetry on aerodynamic changes of wind by fire using a large eddy simulation analysis based on fireFoam solver of OpenFOAM platform. Wind aerodynamic analysis was performed by implementing a module to the solver to extract the corresponding components of fire-induced pressure gradient and acceleration. The results revealed that deviation from fire source symmetry results in asymmetric behaviour of counter-rotating vortices where the maximum cross-sectional wind enhancement occurs. Moreover, the concept of the first-moment area was used to quantify the level of fire source deviation from symmetry and it was shown that the higher first-moment area (about the equivalent symmetry axis) corresponds to a higher deviation from symmetry which delays the realignment of counter-rotating vortices toward the horizontal vortex line.

A novel hybrid framework of optimized deep learning models combined with multi-sensor fusion is developed for condition diagnosis of concrete arch beam. The vibration responses of structure are first processed by principal component analysis for dimensionality reduction and noise elimination. Then, the deep network based on stacked autoencoders (SAE) is established at each sensor for initial condition diagnosis, where extracted principal components and corresponding condition categories are inputs and output, respectively. To enhance diagnostic accuracy of proposed deep SAE, an enhanced whale optimization algorithm is proposed to optimize network meta-parameters. Eventually, Dempster-Shafer fusion algorithm is employed to combine initial diagnosis results from each sensor to make a final diagnosis. A miniature structural component of Sydney Harbour Bridge with artificial multiple progressive damages is tested in laboratory. The results demonstrate that the proposed method can detect structural damage accurately, even under the condition of limited sensors and high levels of uncertainties.

IRE1α cleaves several mRNAs upon accumulation of misfolded proteins. Here the authors show that active IRE1α cleaves DGAT2 mRNA encoding the rate-limiting enzyme in the synthesis of triacylglycerols, suggesting a role of IRE1α in reprogramming lipid metabolism in cancer cells.

Background: Communication impairments are a leading concern for parent caregivers of individuals with rare neurodevelopmental disorders (RNDDs). Clinical trials of disease modifying therapies require valid and responsive outcome measures that are relevant to individuals with RNDDs. Identifying and evaluating current psychometric properties for communication measures is a critical step towards the selection and use of appropriate instruments. Aims: This systematic review offers (1) a description of parent‐reported communication measures and (2) evidence for their psychometric properties, in RNDDs. Methods: The systematic review protocol was registered in the International Prospective Register of Systematic Reviews (PROSPERO; CRD42022334649). MEDLINE (Ovid), Embase, PsychINFO, Web of Science, CINAHL Plus, Cochrane Library, ClinicalTrials.gov, the Australian New Zealand Clinical Trials Registry were searched from inception to August 2023. Methodological assessment of quality was completed using the COnsensus‐based Standards for the selection of health status Measurement INstruments (COSMIN) checklist. Parent‐reported measures used in observational studies and clinical trials were identified. Data on utility, reliability and validity for RNDDs were extracted. Main Contribution: Sixteen parent‐reported communication measures were used in RNDD research, the Vineland Adaptive Behavior Scales being most commonly used. Validation data in RNDDs were identified for six of these measures. Limitations related to sample size or the scope of psychometric testing. Conclusions: Many communication measures have been used for RNDDs but there are few data validating their use. Valid and reliable methods of measuring communication in persons with RNDDs is a priority for future high‐quality clinical trials. WHAT THIS PAPER ADDS: What is already known on the subject: Communication is a critical domain for families with a child with a rare neurodevelopmental disorder (RNDD). Validated outcome measures are essential for accurate evaluation and interpretation of responses to treatments in clinical trials. What this paper adds to existing knowledge: We identified 16 parent‐reported communication measures that have been used with RNDDs, but only six measures had validation data for at least one RNDD. High quality evidence is accumulating, with all validation studies in this review published between 2020 to 2023. Modifications of existing measures may be required to assess communication for RNDDs. What are the clinical implications of this work?: This systematic review catalogues the available psychometric data for communication measures and indicates an ongoing need for new validation studies to ensure they are fit‐for‐purpose for upcoming clinical trials in RNDDs. Background/Objectives: The purpose of this study was to evaluate numerical changes in immune cells after successful kidney transplantation and associate their recovery with clinical and laboratory factors. Methods: In 112 kidney transplant recipients, we performed flow cytometry to evaluate counts of CD4+, CD8+, and regulatory T cells (Tregs), as well as natural killer (NK) cells, before kidney transplantation (T0) and three (T3), six (T6), and twelve (T12) months later. The results were associated with the recipient's age, cold ischemia time (CIT), the type of donor, dialysis method and vintage, and graft function in one year. Results: Total and CD8+ T cell counts increased gradually one year post transplantation in comparison with pre-transplantation levels, whereas the number of CD4+ T cells and Tregs increased, and the number of NK cells decreased in the first three months and remained stable thereafter. The recipient's age was negatively correlated with total, CD4+, and Treg counts at T12, whereas CIT affected only total and CD4+ T cell count. Moreover, recipients receiving kidneys from living donors presented better recovery of all T cell subsets at T12 in comparison with recipients receiving kidneys from cadaveric donors. Patients on peritoneal dialysis had increased numbers of total and CD8+ T cells, as well as NK cells. Finally, estimated glomerular filtration rate was positively correlated with Treg level and potentially CD4+ T cells one-year post transplantation. Conclusions: Successful kidney transplantation results in the recovery of most T cell subsets. Lower recipient age and better graft function contribute to increased T cell counts, whereas donor type and dialysis modality are the most important modifiable factors for optimal immune recovery. Stainless steels are modern high-performance construction materials exhibiting excellent corrosion resistance, recyclability, ductility, and durability which make them appealing to use in the construction industry. However, when used as structural sections, they are subjected to localised failure in the web. This study aims to examine the structural behaviour of cold-formed low-carbon content standard austenitic 304L and 316L stainless steel channels under localised interior bearing loads. The results of 21 tests on unlipped channels with different cross-section sizes and thicknesses are presented. A nonlinear quasi-static Finite Element (FE) model is then developed. The FE model is validated against experimental test results and demonstrated good agreement in terms of bearing strength and failure modes. In addition, the experimental and FE results are used to compare the results against the results predicted in accordance with the American specification SEI/ASCE 8:2002 and European Standard EN 1993-1-4:2006. It is found that the current design equations are unreliable and too unconservative to use for cold-formed austenitic stainless steel unlipped channels, especially when compared to SEI/ASCE 8:2002, as much as 41%.
