Miniaturized microelectronics or small modular reactors (SMRs) simultaneously undergo an increase in heat flux and a decrease in size with the enhancement of device functionalities. Therefore, efficient convection cooling is required to prevent the malfunctioning of electronic devices or small nuclear reactors. Herein, mesh-patterned copper pyramids were fabricated via supersonic spraying for surface cooling applications. Zinc oxide nanowires (ZnO NWs) were patterned on top of the copper pyramids to produce a highly efficient cooling system for a high-heat-flux surface by increasing the overall surface area of the pyramids. In addition, ZnO NW decoration imparted superhydrophilicity to the surface with firm anchoring and spreading of impacting spray liquid drops until complete evaporation. The effects of macroscale pyramid mesh resolution and introduction of ZnO NWs on the spray cooling efficiency were quantified and compared. Herein, the pyramids with coarse resolution and ZnO NWs yielded the highest heat flux during the drop impact heat transfer process and the lowest surface temperature under air and spray cooling conditions owing to the maximized surface area. The use of the pyramids and introduction of ZnO NWs increased the Leidenfrost temperature from 140 to 190 °C, thereby expanding the range of the evaporation cooling regime.

Abstract Our previous work showed that KRAS activation in gastric cancer cells leads to activation of an epithelial-to-mesenchymal transition (EMT) program and generation of cancer stem-like cells (CSCs). Here we analyze how this KRAS activation in gastric CSCs promotes tumor angiogenesis and metastasis. Gastric cancer CSCs were found to secrete pro-angiogenic factors such as vascular endothelial growth factor A (VEGF-A), and inhibition of KRAS markedly reduced secretion of these factors. In a genetically engineered mouse model, gastric tumorigenesis was markedly attenuated when both KRAS and VEGF-A signaling were blocked. In orthotropic implant and experimental metastasis models, silencing of KRAS and VEGF-A using shRNA in gastric CSCs abrogated primary tumor formation, lymph node metastasis, and lung metastasis far greater than individual silencing of KRAS or VEGF-A. Analysis of gastric cancer patient samples using RNA sequencing revealed a clear association between high expression of the gastric CSC marker CD44 and expression of both KRAS and VEGF-A, and high CD44 and VEGF-A expression predicted worse overall survival. In conclusion, KRAS activation in gastric CSCs enhances secretion of pro-angiogenic factors and promotes tumor progression and metastasis.

In this study, a vertical shaft is coated using silver nanowires (AgNWs) together with reduced graphene oxide (rGO) via the supersonic spray-coating technique to enhance the absorption of solar radiative heat for use in solar air heaters. The rGO/AgNW-coated surface induces multiple light reflections, similar to that in a perfect black body inside a Helmholtz jar, thus enhancing the collection of solar radiation. Air fed into the vertical heated shaft ascends owing to buoyancy. The air temperature difference, ΔT, between the outlet and inlet is measured for various mass flow rates to quantify the heat transferred to the air from the solar-heated vertical shaft. The longitudinal air velocity and temperature distributions inside the shaft are numerically simulated using the fire dynamics simulator. Both two- and three-dimensional simulations are performed, and the results are compared with experimental data for various mass flow rates. The results confirm that the trends observed in both the experiments and simulations agree well for all cases. Multiple metal meshes are installed inside the vertical shaft to induce turbulence, which enhances the heat transfer intensity. This turbulence enhancement is confirmed via smoke visualization and infrared images.

Sarcomas are rare tumours with many different subtypes and clinical outcomes; a broader knowledge of their genetic features is required. Here, the authors analyse 2138 soft tissue and bone sarcomas across 45 subtypes using MSK-IMPACT targeted sequencing and find genomic groups that are distinct from histological subgroups.

Gastric cancer is a highly prevalent and lethal disease worldwide. Given the insidious nature of the presenting symptoms, patients are frequently diagnosed with advanced, unresectable disease. However, many patients will present with locally advanced gastric cancer (LAGC), which is often defined as the primary tumor extending beyond the muscularis propria (cT3-T4) or having nodal metastases (cN+) disease and without distant metastases (cM0). LAGC is typically treated with surgical resection and perioperative chemotherapy. The treatment of LAGC remains a challenge, given the heterogeneity of this disease, and the optimal multimodal treatment regimen may be different for different LAGC subtypes. However, many promising treatments are on the horizon based on knowledge of molecular subtypes and key biomarkers of LAGC, such as microsatellite instability, HER2, Claudin 18.2, FGFR2, and PD-L1. This review will expand upon the discussion of current standard neoadjuvant and adjuvant therapies for LAGC and explore the ongoing and future clinical trials for novel therapies, with information obtained from searches in PubMed and ClinicalTrials.gov.

The miniaturization of electronic devices with ever-increasing functionalities inevitably results in greater thermal load on the devices. Hence, pool boiling, which utilizes the powerful latent heat of vaporization, is introduced as an effective method to manage severe thermal loads on miniaturized electronics. Hydrophobic surfaces tend to augment sufficient nucleation sites via roughening or low surface energy, while hydrophilic surfaces are prone to ample liquid supply, thus facilitating rapid bubble release and preventing surface dry-out. Herein, we compared the heat removal performance of TiO2-coated superhydrophilic and Teflon-coated superhydrophobic surfaces fabricated by aerosol deposition and supersonic spraying, respectively. TiO2 and Teflon particles were deposited on evenly patterned frustums. The resolution of the frustum is categorized as no-frustum, coarse, medium, or fine. We found that the surface wettability had little effect on the critical heat flux (CHF) for the no-frustum and fine cases. However, the CHF and effective heat transfer coefficient (heff) of the superhydrophilic surface increased considerably in the medium case. For the superhydrophobic surface, the presence of frustum deteriorated the pool boiling performance. Frustum texturing enhanced the pool boiling performance by increasing the number of nucleation sites and pathways of fresh liquid supply for the bare and superhydrophilic surfaces, but not for the superhydrophobic surface.

Abstract To identify drivers of sarcoma cancer stem-like cells (CSCs), we compared gene expression using RNA sequencing between HT1080 fibrosarcoma and SK-LMS-1 leiomyosarcoma spheroids (which are enriched for CSCs) compared with the parent populations. The most overexpressed survival signaling-related gene in spheroids was phosphoinositide-3-kinase regulatory subunit 3 (PIK3R3), a regulatory subunit of PI3K, which functions in tumorigenesis and metastasis. In a human sarcoma microarray, PIK3R3 was also overexpressed by 4.1-fold compared with normal tissues. PIK3R3 inhibition using shRNA in the HT1080, SK-LMS-1, and DDLS8817 dedifferentiated liposarcoma in spheroids and in CD133+ cells (a CSC marker) reduced expression of CD133 and the stem cell factor Nanog and blocked spheroid formation by 61–71%. Mechanistic studies showed that in spheroid cells, PIK3R3 activated AKT and ERK signaling. Inhibition of PIK3R3, AKT, or ERK using shRNA or inhibitors decreased expression of Nanog, spheroid formation by 68–73%, and anchorage-independent growth by 76–91%. PIK3R3 or ERK1/2 inhibition similarly blocked sarcoma spheroid cell migration, invasion, secretion of MMP-2, xenograft invasion into adjacent normal tissue, and chemotherapy resistance. Together, these results show that signaling through the PIK3R3/ERK/Nanog axis promotes sarcoma CSC phenotypes such as migration, invasion, and chemotherapy resistance, and identify PIK3R3 as a potential therapeutic target in sarcoma.

Abstract The self-renewal transcription factor Nanog and the phosphoinositide 3-kinase (PI3K)–Akt pathway are known to be essential for maintenance of mesenchymal stem cells. We evaluated their contribution to the maintenance of CD133(+) cancer stem-like cells (CSCs) and spheroid-forming cells in patient-derived cell lines from three human sarcoma subtypes: HT1080 fibrosarcoma, SK-LMS-1 leiomyosarcoma, and DDLS8817 dedifferentiated liposarcoma. Levels of Nanog and activated Akt were significantly higher in sarcoma cells grown as spheroids or sorted for CD133 expression to enrich for CSCs. shRNA knockdown of Nanog decreased spheroid formation 10- to 14-fold, and reversed resistance to both doxorubicin and radiation in vitro and in H1080 flank xenografts. In the HT1080 xenograft model, doxorubicin and Nanog knockdown reduced tumor growth by 34% and 45%, respectively, and the combination reduced tumor growth by 74%. Using a human phospho-kinase antibody array, Akt1/2 signaling, known to regulate Nanog, was found to be highly activated in sarcoma spheroid cells compared with monolayer cells. Pharmacologic inhibition of Akt using LY294002 and Akt1/2 knockdown using shRNA in sarcoma CSCs decreased Nanog expression and spheroid formation and reversed chemotherapy resistance. Akt1/2 inhibition combined with doxorubicin treatment of HT1080 flank xenografts reduced tumor growth by 73%. Finally, in a human sarcoma tumor microarray, expression of CD133, Nanog, and phospho-Akt were 1.8- to 6.8-fold higher in tumor tissue compared with normal tissue. Together, these results indicate that the Akt1/2–Nanog pathway is critical for maintenance of sarcoma CSCs and spheroid-forming cells, supporting further exploration of this pathway as a therapeutic target in sarcoma.

Gastric adenocarcinoma is by far the most common form of gastric cancer (GC) and is a highly lethal form of cancer arising from the gastric epithelium. GC is an important area of focus of the medical community, given its often late-stage of diagnosis and associated high mortality rate. While surgery and chemotherapy remain the primary treatments, attention has been drawn to the use of immunologic therapies, which have shown promise in the treatment of other malignancies. The role for immune-based therapies has become clearer as we obtain a greater understanding of the role of the immune system in gastric cancer formation and growth. A variety treatment to augment the immune system are under evaluation in clinical trials, and these include immune checkpoint inhibitors, antibody-drug conjugates, and immune cell-based therapies. Here, we review the immune landscape and immune-based therapies for GC.

Herein, we report a facile and efficient method for fabricating porous carbon flakes (PCFs)-supported nickel nanoplates (Ni NPs) as electrocatalysts for methanol oxidation in alkaline media. The catalyst was fabricated in one step using molten salt synthesis. Various techniques were used to characterize the morphology and composition of the Ni NPs@PCFs catalyst, and these revealed that the Ni NPs were dispersed finely across the PCFs with a highly crystalline structure. The Ni NPs@PCFs catalyst demonstrated high electrocatalytic activity for methanol oxidation (121 mA/cm2 vs. Ag/AgCl), and it had an onset potential of 0.35 V. It also exhibited high stability in an alkaline electrolyte for the duration of the experiment (up to 2000 s).

Self-healing materials have been developed since the 1990s and are currently used in various applications. Their performance in extreme environments and their mechanical properties have become a topic of research interest. Herein, we discuss cutting-edge self-healing technologies for hard materials and their expected healing processes. The progress that has been made, including advances in and applications of novel self-healing fiber-reinforced plastic composites, concrete, and metal materials is summarized. This perspective focuses on research at the frontier of self-healing structural materials.

Abstract Controlling the thickness of quantum dot (QD) films is difficult using existing film formation techniques, which employ pre-ligand-exchanged PbS QD inks, because of several issues: 1) poor colloidal stability, 2) use of high-boiling-point solvents for QD dispersion, and 3) limitations associated with one-step deposition. Herein, we suggest a new protocol for QD film deposition using electrical double-layered PbS QD inks, prepared by solution-phase ligand exchange using methyl ammonium lead iodide (MAPbI3). The films are deposited by the supersonic spraying technique, which facilitates the rapid evaporation of the solvent and the subsequent deposition of the PbS QD ink without requiring a post-deposition annealing treatment for solvent removal. The film thickness could be readily controlled by varying the number of spraying sweeps made across the substrate. This spray deposition process yields high-quality n-type QD films quickly (within 1 min) while minimizing the amount of the PbS QD ink used to less than 5 mg for one device (300-nm-thick absorbing layer, 2.5 × 2.5 cm2). Further, the formation of an additional p-layer by treatment with mercaptopropionic acid allows for facile hole extraction from the QD films, resulting in a power conversion efficiency of 3.7% under 1.5 AM illumination.

Purpose: To conduct phase 1 and 2 trials with photon intensity modulated radiation therapy and intensity modulated proton therapy (IMPT) arms to selectively escalate the retroperitoneal sarcoma preoperative radiation dose to tumor volume (clinical target volume [CTV] 2) that is judged to be at a high risk for positive margins and aim to reduce local recurrence. We report on the IMPT study arm in phase 1. Methods and materials: Patients aged ≥18 years with primary or locally recurrent retroperitoneal sarcoma were treated with preoperative IMPT, 50.4 GyRBE in 28 fractions, to CTV1 (gross tumor volume and adjacent tissues at risk of subclinical disease) with a simultaneous integrated boost to CTV2 to doses of 60.2, 61.6, and 63.0 GyRBE in 28 fractions of 2.15, 2.20, and 2.25 GyRBE, respectively. The primary objective of the phase 1 study was to determine the maximum tolerated dose to CTV2, which will be further tested in the phase 2 study. Results: Eleven patients showed increasing IMPT dose levels without acute dose limiting toxicities that prevented dose escalation to maximum tolerated dose. Acute toxicity was generally mild with no radiation interruptions. No unexpected perioperative morbidity was noted. Eight months postoperatively, one patient developed hydronephrosis that was treated by stent with ureter dissected off tumor and received 57.5 GyRBE. Retained ureter(s) was (were) subsequently constrained to 50.4 GyRBE without further problem. With an 18-month median follow-up, there were no local recurrences. Conclusions: IMPT dose escalation to CTV2 to 63 GyRBE was achieved without acute dose limiting toxicities. The phase 2 study of IMPT will accrue patients to that dose. Parallel intensity modulated radiation therapy phase 1 arm is currently accruing at the initial dose level. Ureters that undergo a high dose radiation and/or surgery are at risk for late hydro-ureter. Future studies will constrain retained ureters to 50.4 GyRBE to avoid ureteral stricture.

Background and Objectives. Atypical lipomas are uncommon, slow-growing benign tumors. While surgery has been the primary treatment modality, we have managed some patients with radiation (RT) as a component of the treatment and have reported their outcomes in this study. Methods. A retrospective review of all cases of extremity and trunk atypical lipomas in The Sarcoma Database at the study institution was conducted. Results. Thirteen patients were identified. All patients underwent surgical resection at initial presentation and received pre- or postoperative radiation for subtotal resection (n=2), local recurrence (n=8), or progressive disease (n=3). The median total radiation dose was 50 Gy. Median followup was 65.1 months. All patients treated with RT remained free of disease at the last followup. No grade 3 or higher late toxicity from radiation was observed. No cases of tumor dedifferentiation occurred. Conclusion. For recurrent or residual atypical lipomas, a combination of reexcision and RT can provide long-term local control with acceptable morbidity. For recurrent tumors, pre-op RT of 50 Gy appears to be an effective and well-tolerated management approach.
