18. Caraway Nanoemulsion Gel: A Potential Antibacterial Treatment against Escherichia coli and Staphylococcus aureus.

Abstract

Novel antibiotics are needed due to the rise of antibiotic-resistant pathogens. Traditional antibiotics are ineffective due to antibiotic-resistant microorganisms, and finding alternative therapies is expensive. Hence, plant-derived caraway (Carum carvi) essential oils and antibacterial compounds have been selected as alternatives. In this, caraway essential oil as an antibacterial treatment was investigated using a nanoemulsion gel. Using the emulsification technique, a nanoemulsion gel was developed and characterized in terms of particle size, polydispersity index, pH, and viscosity. The results showed that the nanoemulsion had a mean particle size of 137 nm and an encapsulation efficiency of 92%. Afterward, the nanoemulsion gel was incorporated into the carbopol gel and was found to be transparent and uniform. The gel had in vitro cell viability and antibacterial activity against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). The gel safely delivered a transdermal drug with a cell survival rate of over 90%. With a minimal inhibitor concentration (MIC) of 0.78 mg/mL and 0.78 mg/mL, respectively, the gel demonstrated substantial inhibition for E. coli and S. aureus. Lastly, the study demonstrated that caraway essential oil nanoemulsion gels can be efficient in treating E. coli and S. aureus, laying the groundwork for the use of caraway essential oil as an alternative to synthetic antibiotics in the treatment of bacterial infections. 22. Anti-inflammatory and hepatoprotective potentials of the aerial parts of Silene villosa Caryophyllaceae methanol extract in rats

Abstract

Purpose: To explore the anti-inflammatory and hepatoprotective potentials of Silene villosa Caryophyllaceae methanol extract in rats.Methods: Toxicity of S. villosa extract was evaluated in rats. Inflammation was induced in rats by injection of 0.1 mL carrageenan (1 %) in the left hind paws. Carbon tetrachloride (CCl4) was used to induce liver damage. Five groups of rat were used. The 1st (normal control) and 2nd (hepatotoxic) groups received the vehicle. The 3rd, 4th, and 5th groups received silymarin, 250 and 500 mg/kg of S. villosa extract, respectively, for 7 days. Liver injury was induced on the 7th day by intraperitoneal administration of 1 mL/kg of CCl4 to rats in groups 2 - 5.Results: The results showed that S. villosa is safe. It significantly reduced carrageenan-induced edema compared to normal (p < 0.01) and standard (p < 0.01) groups. The extract protected (p < 0.01) rats against the deleterious effect of CCl4. It decreased (p < 0.01) the elevated serum activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), γ-glutamyl transferase (γ-GT) and alkaline phosphatase (ALP) as well as elevated serum levels of bilirubin (BRN), compared to CCl4 control rats. Reduced activities of the antioxidant enzymes were significantly increased (p < 0.01) in rat liver, compared with CCl4 control group. The results were confirmed by histological findings in rat liver as the extract reduced necrosis and hydropic degeneration of hepatic tissue compared to CCl4 control group.Conclusion: The results suggest that S. villosa possesses anti-inflammatory and hepatoprotective activities in rats, and therefore, has therapeutic potentials in humans.Keywords: S. villosa, anti-inflammatory, carrageenan, CCl4, antioxidant, hepatotoxicity.

Abstract

Artemisia monosperma is a plant with many traditional uses including some affecting smooth muscles. The A. momosperma essential oil (AMEO) prepared by hydrodistillation from fresh and dry aerial parts was compared qualitatively and quantitatively using GC/MS. The drying process affected the yield and composition of AMEO. The bronchodilator potential was explored using isolated guinea-pig trachea in ex-vivo organ bath setup. In the tracheal contractions induced by different spasmogens, AMEO was able to completely relax contractions induced by carbachol (CCh; 1 μM) and high K+ (80 mM) at closely related doses (p > 0.05) indicating a dual inhibition of phosphodiesterase enzyme (PDE) and voltage-mediated L-type Ca++ channels blocker (CCB) as papaverine. Abstract

Introduction: Fenugreek- the seeds of Trigonella foenum-graecum- are known for their medicinal value since the very early-recorded human history. This study is conducted to explore the effect of environmental conditions on the trigonelline content of the plant seeds. Methods: Trigonelline contents in different samples of fenugreek were estimated by HPTLC. The established method was proved to be accurate, precise and robust according to ICH guidelines. The used mobile phase was a mixture of ethyl acetate: methanol: water: formic acid 7.5: 2.5: 3: 1 (%, v/v/v/v) on 20 x 10 cm glass coated silica gel 60 F254 plates. The plates were scanned and quantified densitometrically at λ = 267 nm after development. Results: A valid linear relationship correlating peak area and amount of trigonelline in the range of 100- 700 ng/spot was obtained. The amount of trigonelline varied according to the country of origin. Conclusion: The Indian sample has the highest amount of trigonelline followed by the Moroccan sample. The two locations represent areas with moderate temperature, low altitude and rainy winter.

Abstract

We have recently reported on the effect of the environmental conditions on the quantity of diosgenin. Attempts for the simultaneous quantification of trigonelline and diosgenin using normal-phase silica gel plates were not successful. A high-performance thin-layer chromatography (HPTLC) method was developed using glass-backed plates coated with RP-18 silica gel 60 F254Sand acetonitrile—water (7.5:2.5, V/V) as the mobile phase. Trigonelline and diosgenin peaks were well separated with RFvalues 0.29 ± 0.02 and 0.17 ± 0.01, respectively. The TLC plates were directly scanned at 267 nm for trigonelline and at 430 nm after derivatization with vanillin—sulfuric acid for diosgenin. Linear regression analysis revealed a good linear relationship between the peak area and the amounts of trigonelline and diosgenin in the range of 200–1400 and 50–300 ng per band, respectively. The method was validated in accordance with the International Conference on Harmonization (ICH) guidelines for precision, accuracy, and robustness.

Abstract

Fenugreek is one of the oldest medicinal plants known by mankind. The aim of this study is to evaluate the effect of environmental conditions on the saponin contents of the plant. The amount of diosgenin in different samples of fenugreek was estimated by high-performance thin-layer chromatography (HPTLC). The developed method was validated, in accordance with the International Conference on Harmonization (ICH) guidelines for precision, accuracy, and robustness. HPTLC was carried out using hexane-acetone (8:2%, v/v) on 20 cm × 10 cm glass coated silica gel 60 F254plates. The developed plates were scanned and quantified densitometrically at λ= 430 nm. Linear regression analysis revealed a good linear relationship between the area under the peak and the amount of diosgenin in the range of 50–400 ng per band. The amount of diosgenin which reflects the total saponin contents varied according to the country of origin. The sample obtained from Yemen showed the highest amount of diosgenin followed by the Saudi sample. Both locations represent areas with the highest altitude (Yemen) and the highest temperature (Saudi).
