Abstract

The XL-Calibur balloon-borne hard X-ray polarimetry mission comprises a Compton-scattering polarimeter placed at the focal point of an X-ray mirror. The polarimeter is housed within a BGO anticoincidence shield, which is needed to mitigate the considerable background radiation present at the observation altitude of ~40 km. This paper details the design, construction and testing of the anticoincidence shield, as well as the performance measured during the week-long maiden flight from Esrange Space Centre to the Canadian Northwest Territories in July 2022. The in-flight performance of the shield followed design expectations, with a veto threshold <100 keV and a measured background rate of ~0.5 Hz (20-40 keV). This is compatible with the scientific goals of the mission, where %-level minimum detectable polarisation is sought for a Hz-level source rate., Comment: Submitted to Nuclear Instruments and Methods A

Abstract

The MuSEUM collaboration is planning measurements of the ground-state hyperfine structure (HFS) of muonium at the Japan Proton Accelerator Research Complex (J-PARC), Materials and Life Science Experimental Facility. The high-intensity beam that will soon be available at H-line allows for more precise measurements by one order of magnitude. We plan to conduct two staged measurements. First, we will measure the Mu-HFS in a near-zero magnetic field, and thereafter we will measure it in a strong magnetic field. We have developed two microwave cavities for this purpose. Furthermore, we evaluated systematic uncertainties from such a fluctuation of microwave fields and confirm the requirement of the microwave system, we use a microwave field distribution calculated from the finite element method.

Patients of ≥ 75 years of age were considered to be older patients (n=40), and patients with PS ≥ 2 were considered to have a poor PS (n=10). The objective response and disease control rates were 42.5% and 52.5%, respectively, in older patients and 0% and 10.0%, respectively, in patients with a poor PS. Overall survival (OS) in the older and younger groups did not differ to a statistically significant extent. However, a poor PS was significantly associated with poor survival. Safety analyses demonstrated no significant difference in the occurrence of any immune-related adverse events (irAEs), including grade ≥ 3, between the older and younger groups. However, a poor PS was significantly associated with the low occurrence of any irAEs. The change of the eosinophil count, the increase of the relative eosinophil count (REC) and the decrease of the neutrophil-to-eosinophil ratio (NER) did not differ to a statistically significant extent between the older and younger groups, but showed significant differences between the poor and good PS (PS 0– 1) groups.Conclusion: Pembrolizumab for advanced UC demonstrated similar changes in the eosinophil count, efficacy and toxicity in both older and younger patients. The association between the occurrence of irAEs and the efficacy of pembrolizumab was investigated. The change in the REC from before the initiation of pembrolizumab therapy, to three weeks after treatment and the incidence of irAEs were determined.Results: Overall irAEs were associated with a significantly higher objective response rate (ORR) (58.8% vs 25.4%, P< 0.001), a longer progression-free survival (PFS) (25.1 months vs 3.1 months, P< 0.001) and overall survival (OS) (31.2 months vs 11.5 months, P< 0.001) compared to patients without irAEs; however, grade ≥ 3 irAEs were not associated with the ORR (36.4% vs 36.2%, P=0.989), PFS (9.5 vs 5.5 months, P=0.249), or OS (not reached vs 13.7 months, P=0.335). Compared to a decreased REC at 3 weeks after pembrolizumab, an increased relative REC at 3 weeks was not associated with the incidence of any-grade irAEs (32.3% vs 32.5%, P=0.984) or of grade ≥ 3 irAEs (10.8% vs 10.0%, P=0.900). Multivariate analyses revealed a female sex (P=0.005), Eastern Cooperative Oncology Group Performance Status ≥ 1 (P=0.024), albumin < 3.7 g/dl (P< 0.001), decreased REC (3 weeks later) (P< 0.001), and the absence of irAEs of any grade (P=0.002) to be independently associated with a worse OS.Conclusion: Patients with irAEs showed a significantly better survival compared to patients without irAEs in advanced UC treated with pembrolizumab. Abstract

We observed the atomic $1s$ and $2p$ states of $\pi^-$ bound to ${}^{121}{\rm Sn}$ nuclei as distinct peak structures in the missing mass spectra of the ${}^{122}{\rm Sn}(d,{}^3{\rm He})$ nuclear reaction. A very intense deuteron beam and a spectrometer with a large angular acceptance let us achieve potential of discovery, which includes capability of determining the angle-dependent cross sections with high statistics. The $2p$ state in a Sn nucleus was observed for the first time. The binding energies and widths of the pionic states are determined and found to be consistent with previous experimental results of other Sn isotopes. The spectrum is measured at finite reaction angles for the first time. The formation cross sections at the reaction angles between 0 and $2^\circ$ are determined. The observed reaction-angle dependence of each state is reproduced by theoretical calculations. However, the quantitative comparison with our high-precision data reveals a significant discrepancy between the measured and calculated formation cross sections of the pionic $1s$ state.

A receiver operating characteristic curve was used to determine the optimal cut-off values for analyzing the risk.Results: There were no significant differences in the overall survival (OS) between the REC ≥ 4.8% and < 4.8% groups and the NER ≥ 13.7 and < 13.7 groups before pembrolizumab (p=0.997 and 0.669, respectively). However, a significant difference in the OS was confirmed between the increased and decreased REC groups and between the decreased and increased NER groups at 3 weeks after pembrolizumab (p< 0.001 and 0.002, respectively). Multivariate analyses revealed that an Eastern Cooperative Oncology Group Performance Status ≥ 2 (P=0.003), albumin < 3.7 g/dl (p=0.002), LDH > 246 U/L (p=0.011), disease site ≥ 3 organs (p=0.019), decreased posttreatment REC (3 weeks later) (p=0.002) and increased posttreatment NER (3 weeks later) (p=0.022) were independent prognostic factors for a worse OS.Conclusion: An increased REC and decreased NER after pembrolizumab may be significant early predictive markers of improved clinical outcomes in patients with advanced UC receiving pembrolizumab.Keywords: urothelial carcinoma, pembrolizumab, eosinophil, neutrophil-to-eosinophil ratio

13. Formation of an ultracarbonaceous Antarctic micrometeorite through minimal aqueous alteration in a small porous icy body

Abstract

A comprehensive study of the organic chemistry and mineralogy of an ultracarbonaceous micrometeorite (UCAMM D05IB80) collected from near the Dome Fuji Station, Antarctica, was carried out to understand the genetic relationship among organic materials, silicates, and water. The micrometeorite is composed of a dense aggregate of ∼5 µm-sized hollow ellipsoidal organic material containing submicrometer-sized phases such as glass with embedded metal and sulfides (GEMS) and mineral grains. There is a wide area of organic material (∼15 × 15 μm) in its interior. Low-Ca pyroxene is much more abundant than olivine and shows various Mg/(Mg + Fe) ratios ranging from ∼1.0 to 0.78, which is common to previous works on UCAMMs. By contrast, GEMS grains in this UCAMM have unusual chemical compositions. They are depleted in both Mg and S, which suggests that these elements were leached out from the GEMS grains during very weak aqueous alteration, without the formation of phyllosilicates. The organic materials have two textures—smooth and globular with an irregular outline—and these are composed of imine, nitrile and/or aromatic nitrogen heterocycles, and amide. The ratio of nitrogen to carbon (N/C) in the smooth region of the organics is ∼0.15, which is five times higher than that of insoluble organic macromolecules in types 1 and 2 carbonaceous chondritic meteorites. In addition, the UCAMM organic materials are soluble in epoxy and are thus hydrophilic; this polar nature indicates that they are very primitive. The surface of the material is coated with an inorganic layer, a few nanometers thick, that consists of C, O, Si, S, and Fe. Sulfur is also contained in the interior, implying the presence of organosulfur moieties. There are no isotopic anomalies of D, 13C, or 15N in the organic material. Interstellar photochemistry alone would not be sufficient to explain the N/C ratio of the UCAMM organics; therefore, we suggest that a very small amount of fluid on a comet must have been necessary for the formation of the UCAMM. The GEMS grains depleted in Mg and S in the UCAMM prove a very weak degree of aqueous alteration; weaker than that of carbonaceous chondrites. Short-duration weak alteration probably caused by planetesimal shock locally melted cometary ice grains and released water that dissolved the organics; the fluid would likely have not mobilized because of the very low thermal conductivity of the porous icy body. This event allowed the formation of the large organic puddle of the UCAMM, as well as organic matter sulfurization, formation of thin membrane-like layers of minerals, and deformation of organic nanoglobules.

The objective response rate (ORR) and organ-specific response rate (OSRR) were evaluated according to Response Evaluation Criteria in Solid Tumors, version 1.1.Results: We analyzed 69 patients (male, n=51; median age, 71 years) with 226 metastases. The ORR was 23.2%. In total, 32, 31, 16, 14, 13 and 7 patients had measurable lung (OSSR 31.3%), lymph node (OSSR 29.0%), local recurrence (OSSR 12.5%), primary tumor organ (OSSR 7.1%), liver (OSSR 23.1%) and bone (OSSR 28.6%) disease, respectively. The median overall survival (OS) for pembrolizumab was 10.9 months (95% confidence interval, 5.9‑13.7 months). Regarding organ-specific OS, a Log rank test significant differences in OS were confirmed between patients with and without primary tumor organ disease (p=0.046) and liver metastasis (p< 0.001).Conclusion: Metastases and primary tumor organ disease showed different tumor responses to pembrolizumab. The most prominent tumor response was found in lung metastasis and the least response was found in primary organ sites. The mechanisms of these different responses were unclear and there does not appear to be a constant trend between tumor shrinkage and OS in tumor sites. Abstract

A complete set of analyzing powers for the dd->3Hp reaction at the kinetic beam energy of 200 MeV has been measured in the full angular range in the c.m. frame. The observed signs of the tensor analyzing powers Ayy, Axx, and Axz at forward and backward directions have clearly demonstrated the sensitivity to the ratio of the D- and S-wave components of the triton and deuteron, respectively. The new high-precision data are compared with the prediction of the relativistic multiple-scattering model by using standard wave functions of the three-nucleon bound state and of the deuteron.

Abstract

A complete high precision set of deuteron analyzing powers for elastic deuteron-proton ($dp$) scattering at 250 MeV/nucleon (MeV/N) has been measured. The new data are presented together with data from previous measurements at 70, 100, 135 and 200 MeV/N. They are compared with the results of three-nucleon (3N) Faddeev calculations based on modern nucleon-nucleon (NN) potentials alone or combined with two models of three nucleon forces (3NFs): the Tucson-Melbourne 99 (TM99) and Urbana IX. At 250 MeV/N large discrepancies between pure NN models and data, which are not resolved by including 3NFs, were found at c.m.\ backward angles of $\theta_{\rm c.m.}\gtrsim 120^\circ$ for almost all the deuteron analyzing powers. These discrepancies are quite similar to those found for the cross section at the same energy. We found small relativistic effects that cannot resolve the discrepancies with the data indicating that other, short-ranged 3NFs are required to obtain a proper description of the data., Comment: submitted to Phys. Abstract

SS433, located at the center of the supernova remnant W50, is a close proximity binary system consisting of a compact star and a normal star. Jets of material are directed outwards from the vicinity of the compact star symmetrically to the east and west. Non-thermal hard X-ray emission is detected from lobes lying on both sides. Shock accelerated electrons are expected to generate sub-TeV gamma rays through the inverse-Compton process in the lobes. Observations of the western X-ray lobe region of SS433/W50 system have been performed to detect sub-TeV gamma-rays using the 10m CANGAROO-II telescope in August and September, 2001, and July and September, 2002. The total observation times are 85.2 hours for ON source, and 80.8 hours for OFF source data. No significant excess of sub-TeV gamma rays has been found at 3 regions of the western X-ray lobe of SS433/W50 system. We have derived 99% confidence level upper limits to the fluxes of gamma rays and have set constraints on the strengths of the magnetic fields assuming the synchrotron/inverse-Compton model for the wide energy range of photon spectrum from radio to TeV. The derived lower limits are 4.3 microgauss for the center of the brightest X-ray emission region and 6.3 microgauss for the far end from SS433 in the western X-ray lobe. In addition, we suggest that the spot-like X-ray emission may provide a major contribution to the hardest X-ray spectrum in the lobe., Comment: 7 pages, 8 figures, to be published in Astroparticle Physics

Abstract

A time projection chamber (TPC) is a strong candidate for the central tracker of the international linear collider (ILC) experiment and we have been conducting a series of cosmic ray experiments under a magnetic field up to 4 T, using a small prototype TPC with a replaceable readout device: multi-wire proportional chamber (MWPC) or gas electron multiplier (GEM). We first confirmed that the MWPC readout could not be a fall-back option of the ILC-TPC under a strong axial magnetic field of 4 T since its spatial resolution suffered severely from the so called E x B effect in the vicinity of the wire planes. The GEM readout, on the other hand, was found to be virtually free from the E x B effect as had been expected and gave the resolution determined by the transverse diffusion of the drift electrons (diffusion limited). Furthermore, GEMs allow a wider choice of gas mixtures than MWPCs. Among the gases we tried so far a mixture of Ar-CF4-isobutane, in which MWPCs could be prone to discharges, seems promising as the operating gas of the ILC-TPC because of its small diffusion constant especially under a strong magnetic field. We report the measured drift properties of this mixture including the diffusion constant as a function of the electric field and compare them with the predictions of Magboltz. 24. 3D MONITORING OF COASTAL EROSION CONTROL STRUCTURES USING UAV.

Abstract

Coastal erosion has increasingly become a problem in recent years due to rising sea levels caused by global warming. To prevent further coastal erosion and damage, control structures like seawalls and breakwaters have been installed along vulnerable coastlines. However, it is crucial that these structures are regularly and thoroughly inspected for any abnormalities or deformations. At present, inspections are done manually by visual surveys which are time-consuming and inefficient. There is great potential to optimize this process using drone technology equipped with 3D laser scanners. In this study, we utilized a drone with a green laser scanner to inspect and diagnose control structures along the coast. We conducted surveys to determine the basic performance of this approach and used ICP algorithms to extract any deformations in vanishing wave blocks over two time periods. Our results showed high variability in basic performance due to the influence of waves during the surveys. However, we were still able to detect strain of around 50 cm in a submerged breakwater located 3 meters below the water's surface. Furthermore, an overall settlement of approximately 34 cm was observed in the vanishing wave blocks along with some localized movements. This demonstrates that drones can be successfully implemented for efficient inspection, diagnosis and detection of abnormalities and deformations in coastal structures that are extremely difficult to identify through visual surveys alone. The use of this advanced technology will allow for quicker identification of at-risk structures, enabling timely maintenance and prevention of further coastal erosion. [ABSTRACT FROM AUTHOR]

Recently, the proteins associated with ER status were reported using liquid chromatography and tandem mass spectrometry, and microRNA (miRNA) profiling of breast cancer subtype was demonstrated using real-time-PCR. Methods: We present herein two cases with differential protein expression and miRNA profiling in ER-positive and -negative breast cancer. Results: Proteins associated with fatty acid metabolism were uniquely detected in ER-positive breast cancer. The level of miR-181a expression in ER-positive cancer was higher than that in ER-negative cancer, while the expression of miR-27a, miR-107, and miR-195 was lower in ER-positive compared with ER-negative cancer. Conclusion: These cases suggest that fatty acid synthase (FAS) and FAS-related miRNAs are important in ER-positive breast cancer. Recently, a combination regimen consisting of the HCV nonstructural protein 5A inhibitor elbasvir (EBR) and the HCV NS3/4A protease inhibitor grazoprevir (GZR) was approved for the treatment of patients with chronic HCV and genotypes (Gts) 1 and 4 in various countries. In Phase III trials, the combination of EBR/GZR (fixed-dose combination table or single agent) for 12 or 16 weeks of treatment with or without ribavirin resulted in a high sustained virological response at 12 weeks in treatment-naïve and treatment-experienced patients with HCV Gt 1a, 1b, 4, or 6, including special populations, such as individuals with advanced chronic kidney disease, HCV-HIV coinfection, and compensated cirrhosis. The data on the tensor Ayy, Axx, Axz and vector Ay analyzing powers in the dd->3Hen obtained at Td= 270 MeV in the angular range 0 - 110 degrees in the c.m. are presented. The observed negative sign of the tensor analyzing powers Ayy, Axx and Axz at small angles clearly demonstrate the sensitivity to the ratio of the D and S wave component of the 3He wave function. However, the one-nucleon exchange calculations by using the standard 3He wave functions have failed to reproduce the strong variation of the tensor analyzing powers as a function of the angle in the c.m., Comment: 8 pages, 7 figures, 4 tables. Abstract

High precision vector and tensor analyzing powers of elastic deuteron-proton d+p scattering have been measured at intermediate energies to investigate effects of three-nucleon forces (3NF). Angular distribution in the range of 70-120 degree in the center-of mass frame for incident-deuteron energies of 130 and 180 MeV were obtained using the RIKEN facility. The beam polarization was unambiguously determined by measuring the 12C(d,alpha)10B(2+) reaction at 0 degree. Results of the measurements are compared with state-of-the-art three-nucleon calculations. The present modeling of nucleon-nucleon forces and its extension to the three-nucleon system is not sufficient to describe the high precision data consistently and requires, therefore, further investigation.

Abstract

Inelastic $\alpha$ scattering on 16O is studied at 400 MeV by using an ice target. Near the 4-alpha breakup threshold of 14.4 MeV, a broad peak is observed at an excitation energy of 13.6+/-0.2 MeV with a width of 0.6+/-0.2 MeV. The spin-parity is estimated to be 0+ from the momentum-transfer dependence. The observed width is significantly larger than those of the neighboring 0+ states indicating a state with a well-developed alpha cluster structure. The magnitude of the cross section is sensitive to the density distribution of the constituent alpha clusters. The observed cross section is consistent with the theoretical prediction for the alpha cluster condensed state characterized by its dilute density distribution with a large root-mean-square radius of about 4.3 fm., Comment: 12 pages, 5 fugures, submitted to Phys. Abstract

Three precise measurements for elastic pd scattering at 135 MeV/A have been performed with the three different experimental setups. The cross sections are described well by the theoretical predictions based on modern nucleon-nucleon forces combined with three nucleon forces. Abstract

The cross section of the ${}^{16}{\rm O}(p,p'){}^{16}{\rm O}(0^-,T=1)$ scattering was measured at a bombarding energy of 295 MeV in the momentum transfer range of $1.0 \mathrm{fm^{-1}}$ $\le$ $q_{\rm c.m.}$ $\le$ $2.1 \mathrm{fm^{-1}}$. The isovector $0^-$ state at $E_x$ = 12.8 MeV is clearly separated from its neighboring states owing to the high energy resolution of about 30 keV. The cross section data were compared with distorted wave impulse approximation (DWIA) calculations employing shell-model wave functions. The observed cross sections around $q_{\rm c.m.}$ $\simeq$ $1.7 {\rm fm^{-1}}$ are significantly larger than predicted by these calculations, suggesting pionic enhancement as a precursor of pion condensation in nuclei. The data on the tensor analyzing power T20 in the dd->^3Hen and dd-> ^3Hp reactions at 140, 200 and 270 MeV of the deuteron kinetic energy and at zero degree obtained at RIKEN Accelerator Research Facility are presented. The observed positive sign of T20 clearly demonstrates the sensitivity to the D/S wave ratios in the ^3He and ^3H in the energy domain of the measurements. Abstract

The deuteron to proton polarization transfer coefficients for the $d$--$p$ elastic scattering were precisely measured with an incoming deuteron energy of 135 MeV/u at the RIKEN Accelerator Research Facility. The data are compared to theoretical predictions based on exact solutions of three-nucleon Faddeev equations with high--precision nucleon--nucleon forces combined with different three-nucleon forces (3NFs), representing the current, most popular models: the $2\pi$-exchange Tucson-Melbourne model, a modification thereof closer to chiral symmetry TM'(99), and the Urbana IX 3NF. Theory predicts large 3NF effects, especially in the angular range around the cross section minimum, but the present data only partially concurs, predominantly for $K_{xx}^{y'}-K_{yy}^{y'}$ ($K_{xx}^{y'}$, $K_{yy}^{y'}$). For the induced polarization, $P^{y'}$, the TM$'$(99) and Urbana IX 3NFs reproduce the data, but the Tucson-Melbourne 3NF fails to describe the data. For the polarization transfer coefficients, $K_{y}^{y'}$ and $K_{xz}^{y'}$, the predicted 3NF ffects are in drastic conflict to the data. We have measured the cross sections and analyzing powers Ay and Ayy for the elastic and inelastic scattering of deuterons from the 0+(g.s.), 2+(4.44 MeV), 3-(9.64 MeV), 1+(12.71 MeV), and 2-(18.3 MeV) states in 12C at an incident energy of 270 MeV. Abstract

The deuteron single and double spin-flip probabilities, S1 and S2, have been measured for the 12C(pol{d},pol{d}') reaction at Ed = 270 MeV for an excitation energy range between 4 and 24 MeV and a scattering angular range between Theta_lab = 2.5 and 7.5 deg. The extracted S1 exhibits characteristic values depending on the structure of the excited state. The S2 is close to zero over the measured excitation energy range. The SFP angular distribution data for the 2+ (4.44 MeV) and 1+ (12.71 MeV) states are well described by the microscopic DWIA calculations.

Abstract

For backward elastic scattering of deuterons by ^3He, cross sections \sigma and tensor analyzing power T_{20} are measured at E_d=140-270 MeV. The data are analyzed by the PWIA and by the general formula which includes virtual excitations of other channels, with the assumption of the proton transfer from ^3He to the deuteron. Using ^3He wave functions calculated by the Faddeev equation, the PWIA describes global features of the experimental data, while the virtual excitation effects are important for quantitative fits to the T_{20} data. In the present study, we clarify the significance of the number of aspiration risk factors for mortality and recurrence in pneumonia patients.Methods: This study included 322 patients hospitalized with pneumonia between December 2014 and June 2016. We investigated associations between the number of aspiration risk factors present (orientation disturbance, bedridden, chronic cerebrovascular disease, dementia, sleeping medications and gastroesophageal disease) and 30-day and 6-month mortality, and pneumonia recurrence within 30 days.Results: Patients were categorized by number of risk factors present into groups of 0–1, 2, 3, and 4 or more. Of a total of 322 patients, 93 (28.9%) had 0–1 risk factors, 112 (34.8%) had 2, 88 (27.3%) had 3, and 29 (9.0%) had 4 or more risk factors. The percentages of patients with recurrence of pneumonia were 13.0%, 33.0%, 43.2%, and 54.2% in the 0–1, 2, 3, and 4 or more risk factor groups, respectively. The percentages of patients with 30-day mortality were 2.2%, 5.4%, 11.4%, and 24.1%, and those of patients with 6-month mortality were 6.6%, 24.5%, 30.7%, and 50.0%, in the 0–1, 2, 3, and 4 or more risk factor groups, respectively.Conclusions: The number of aspiration risk factors was associated with increases in both mortality and recurrence in pneumonia patients. Therefore, in clinical practice, physicians should consider not only the presence of aspiration risks but also the number of aspiration risk factors in these patients. Abstract

Importance There is a major need for effective, well-tolerated treatments for idiopathic pulmonary fibrosis (IPF). Objective To assess the efficacy and safety of the autotaxin inhibitor ziritaxestat in patients with IPF. Design, Setting, and Participants The 2 identically designed, phase 3, randomized clinical trials, ISABELA 1 and ISABELA 2, were conducted in Africa, Asia-Pacific region, Europe, Latin America, the Middle East, and North America (26 countries). A total of 1306 patients with IPF were randomized (525 patients at 106 sites in ISABELA 1 and 781 patients at 121 sites in ISABELA 2). Enrollment began in November 2018 in both trials and follow-up was completed early due to study termination on April 12, 2021, for ISABELA 1 and on March 30, 2021, for ISABELA 2. Interventions Patients were randomized 1:1:1 to receive 600 mg of oral ziritaxestat, 200 mg of ziritaxestat, or placebo once daily in addition to local standard of care (pirfenidone, nintedanib, or neither) for at least 52 weeks. Main Outcomes and Measures The primary outcome was the annual rate of decline for forced vital capacity (FVC) at week 52. The key secondary outcomes were disease progression, time to first respiratory-related hospitalization, and change from baseline in St George’s Respiratory Questionnaire total score (range, 0 to 100; higher scores indicate poorer health-related quality of life). Results At the time of study termination, 525 patients were randomized in ISABELA 1 and 781 patients in ISABELA 2 (mean age: 70.0 [SD, 7.2] years in ISABELA 1 and 69.8 [SD, 7.1] years in ISABELA 2; male: 82.4% and 81.2%, respectively). The trials were terminated early after an independent data and safety monitoring committee concluded that the benefit to risk profile of ziritaxestat no longer supported their continuation. Ziritaxestat did not improve the annual rate of FVC decline vs placebo in either study. In ISABELA 1, the least-squares mean annual rate of FVC decline was –124.6 mL (95% CI, −178.0 to −71.2 mL) with 600 mg of ziritaxestat vs –147.3 mL (95% CI, −199.8 to −94.7 mL) with placebo (between-group difference, 22.7 mL [95% CI, −52.3 to 97.6 mL]), and –173.9 mL (95% CI, −225.7 to −122.2 mL) with 200 mg of ziritaxestat (between-group difference vs placebo, −26.7 mL [95% CI, −100.5 to 47.1 mL]). In ISABELA 2, the least-squares mean annual rate of FVC decline was –173.8 mL (95% CI, −209.2 to −138.4 mL) with 600 mg of ziritaxestat vs –176.6 mL (95% CI, −211.4 to −141.8 mL) with placebo (between-group difference, 2.8 mL [95% CI, −46.9 to 52.4 mL]) and –174.9 mL (95% CI, −209.5 to −140.2 mL) with 200 mg of ziritaxestat (between-group difference vs placebo, 1.7 mL [95% CI, −47.4 to 50.8 mL]). There was no benefit with ziritaxestat vs placebo for the key secondary outcomes. In ISABELA 1, all-cause mortality was 8.0% with 600 mg of ziritaxestat, 4.6% with 200 mg of ziritaxestat, and 6.3% with placebo; in ISABELA 2, it was 9.3% with 600 mg of ziritaxestat, 8.5% with 200 mg of ziritaxestat, and 4.7% with placebo. Conclusions and Relevance Ziritaxestat did not improve clinical outcomes compared with placebo in patients with IPF receiving standard of care treatment with pirfenidone or nintedanib or in those not receiving standard of care treatment.
