Water scarcity is an extreme global concern due to changing climate and rapid population growth. Worldwide, freshwater sources are depleting and there is a need to implement water-conserving approaches in water-limited or arid areas for sustainable crop production. Therefore, this study aimed to investigate the impact of deficit irrigation on cucumber plant physiology, growth, and fruit yield traits grown under greenhouse conditions. The experiment was conducted in a complete randomized design with four water regimes, 100%, 80%, 60%, and 40% crop water requirement (ETc). Results showed that the highest plant leaf chlorophyll contents (43.2 µmol m-2), plant biomass (12.43 g), plant height (172.39 cm), number of leaves (19.85), fruit yield (2.49 kg), and total soluble solids (3.73 °Brix) were in plants irrigated at 100% level. In comparison, the lowest plant height (115.73 cm), number of leaves (15.77) and fruit yield (1.81 kg) were recorded at 40% irrigation. Results revealed that reduced irrigation regimes up to 80% or 60% have no significant inhibitory effects on cucumber agro-morphological traits. However, 40% of irrigation levels showed significantly reduced plant morphological and fruit yield traits. Overall, the result demonstrated that regulated deficit irrigation can significantly conserve water without negatively impacting cucumber yield.

Leafy vegetables are very healthy for human beings having high contents of phyto- and physico-chemicals and are low in fat and calories. Mountainous regions worldwide have diverse agroclimatic conditions conducive for the cultivation of different leafy vegetables. Present research study assessed six leafy vegetables for their composition at fifteen different agroecological sites. Fresh leaves of six leafy vegetables, i.e. alfalfa, water clover, spinach, leaf mustard, pennywort, and fenugreek, were collected from the mountainous region. Procured leaf samples were brought to analytical laboratory and were subjected to physicochemical and phytochemical analysis, i.e. moisture content, pH, TSS, ascorbic acid, carotenoids, phenolic content, flavonoids, and antioxidant potential. Among all the sampling sites and vegetables, a significant variation was observed for the physicochemical and phytochemical parameters. In samples procured from different agroecological sites, the better retention of quality parameters was observed among the leaf sample collected from fifteen different agroecological sites. The highest values were recorded for moisture content in spinach at site 1, for pH in leaf mustard at sites 4, 5 and 6, for TSS in spinach at sites 6 and 9, for ascorbic acid in alfalfa at site 6, for total phenolics in spinach at site 5, for carotenoid in leaf mustard at site 5, for flavonoids in fenugreek at site 5, and for antioxidant in spinach at site 6. It can be concluded that mountainous regions are rich in agroecological conditions suitable for producing leafy vegetables carrying healthful constituents for the consumption by human beings.

Background and Objectives: Healthcare workers in hospitals are exposed to infectious diseases that occur in the hospital making them a source of infection for the patients. It is interfaced as cross-contamination agents for MRSA and MR-CoNS, and preventive measures need to be adapted accordingly. The study aimed to assess Methicillin-Resistant Staphylococcus (MRS) on the skin and nasal cavities of healthcare workers (HCWs) and identifying isolates to the species level. Materials and Methods: Swab samples were cultured on mannitol salt agar (MSA) to obtain MRS and determine their ability to produce coagulase. Their susceptibility to antibiotics were determined by agar screening and disk diffusion methods and further identification was done at the species level. Results: The highest percentage of methicillin resistant coagulase positive Staphylococci (MRCoPS) was reported among skins of male HCWs, (71.4%) were identified as MRSA. The highest levels of methicillin resistant coagulase negative Staphylococci (MRCoNS) were mainly detected in both nasal cavities, (75%) were identified as MRSE. MRSA was reported from doctors (p-value 0.033), whereas the highest incidence of MRSE was obtained from the nurses (p-value 0.048). Conclusion: This study highlighted that incidence of MRSA was mainly detected in doctors and MRCoNS in both nasal cavities. The highest percentage of MRCoNS was recovered from the patients’ room followed by the reception table. Moreover, vancomycin is suggested to be highly effective in managing and controlling S. aureus, MRSA- and MRSE related infections.

Monoamine oxidase B (MAO-B) plays a pivotal role in the deamination process of monoamines, encompassing crucial neurotransmitters like dopamine and norepinephrine. The heightened interest in MAO-B inhibitors emerged after the revelation that this enzyme could potentially catalyze the formation of neurotoxic compounds from endogenous and exogenous sources. Computational screening methodologies serve as valuable tools in the quest for novel inhibitors, enhancing the efficiency of this pursuit. In this study, 43 acefylline derivatives were docked against the MAO-B enzyme for their chemotherapeutic potential and binding affinities that yielded GOLD fitness scores ranging from 33.21 to 75.22. Among them, five acefylline derivatives, namely, MAO-B14, MAO-B15, MAO-B16, MAO-B20, and MAO-B21, displayed binding affinities comparable to the both standards istradefylline and safinamide. These derivatives exhibited hydrogen-bonding interactions with key amino acids Phe167 and Ile197/198, suggesting their strong potential as MAO-B inhibitors. Finally, molecular dynamics (MD) simulations were conducted to evaluate the stability of the examined acefylline derivatives over time. The simulations demonstrated that among the examined acefylline derivatives and standards, MAO-B21 stands out as the most stable candidate. Density functional theory (DFT) studies were also performed to optimize the geometries of the ligands, and molecular docking was conducted to predict the orientations of the ligands within the binding cavity of the protein and evaluate their molecular interactions. These results were also validated by simulation-based binding free energies via the molecular mechanics energies combined with generalized Born and surface area solvation (MM-GBSA) method. However, it is necessary to conduct in vitro and in vivo experiments to confirm and validate these findings in future studies.

Abstract The Peshawar Basin is a part of the lower Himalayas that contains an enormous amount of groundwater storage. The evaluation of groundwater potential in the southern Peshawar district was done using well logging, lithostratigraphic properties, and combined hydrogeological and geophysical techniques. A total of 13 Vertical Electrical Sounding (VES) profiles were utilised to assess potential groundwater zones for surface resistivity studies. The aquifer system was delineated by comparing the data from five boreholes with the VES findings. An exploration of super-saturated groundwater potential was conducted, utilising parameters such as transmissivity (T), hydraulic conductivity (K), storativity, and the Dar Zarrouk analysis. The Dar Zarrouk analysis yielded average values of transverse resistance (TR), longitudinal conductance (S), and anisotropy (λ), which were determined to be 8069.12, 0.51, and 0.561, respectively. Similarly, average values of transmissivity (T), hydraulic conductivity (K), and storativity were obtained, resulting in 28.67, 0.24, and 0.000177, respectively. The saturated confined layer, characterized by highly saturated zones, was identified to begin at a depth of approximately 119 m and extend down to the lower boundary of the aquifer. The examined aquifer is composed of clay, sand, gravel, boulders, and loose layers of lacustrine mud that are interlayered to form an unconsolidated groundwater aquifer system. The aquifers in the region are highly developed and consisted of unconfined, semi-confined, and confined aquifer systems. As a result, it is possible to use the aquifer for groundwater development in the study area because of its low -to-medium discharge.

ABSTRACTThe quality of water significantly affects the health and walefare of all orginisms, highlighting the importance to develop low-cost and efficient wastewater treatment methods. Herein, we report the fabrication, characterization, and utilization of a polymer-based ternary nanocomposite (CuO–SiO2/PVA) for the removal of Nile Blue (NB) and Methylene Blue (MB) contaminants from wastewater, along with exploring its potential biological activities. We have successfully employed the cost-effective sol–gel and in-situ polymerization approaches to fabricate the CuO–SiO2/PVA based ternary composite, utilizing Cu(NO3)2·3H2O:Glycerol:TEOS:PVA in a ratio of 8:2:3:4. The desired fabrication of nanocomposite was confirmed through UV-Visible spectroscopy, SEM (scanning electron microscope), TEM (transmission electron microscope), EDX (energy dispersive X-ray diffraction), FTIR (Fourier transform infrared), DSC (differential scanning calorimetry), and TGA (thermogravimetric analysis). In addition to its biological potentialthe performance of the nanocomposite in catalytic / photocatalytic removal of NB and MB dyes is investigated and compared. The higher photodegradation performance of the composite for NB (85%) dye than for MB (76%) dye indicates that variables such as chemical structure, charge, molecular mass, and pH sensitivity of the dyes can influence the catalyst's removal potential. This composite is considered to have a higher capability for removing pollutants and microorganisms from wastewater.

Background and Objectives: Microbial biofilm is characterized by the irreversible attachment of planktonic cells to a surface and is usually associated with high antimicrobial resistance with worsening the wound healing. The objective of the study was to determine the prevalence of Staphylococcus aureus in diabetic foot ulcers (DFUs) of diabetic patients and to investigate antibiotic susceptibility patterns of these isolates. In addition to screen biofilm forming ability of isolated S. aureus. Materials and Methods: A total of 112 non-healing wound swabs of diabetic foot patients were collected and cultured on different culture media to identify and characterize 98 isolates. The S. aureus isolates were examined for their antibiotic susceptibility to different antimicrobial agents. Furthermore, S. aureus isolates were evaluated for their biofilm production capability using the Tissue Culture Plate Method (TPC). The level of icaA gene expression was determined by RT-PCR. Results: The results of this study showed that these non-healing wounds yield positive cultures, with an average of 1.67 organisms per sample. The isolates showed highest resistance against oxacillin (95.2%) and lowest resistance against linezolid (3.7%). All isolates were biofilm producers and a significant association with the icaA gene expression level was recorded. Conclusion: This study showed that S. aureus isolates have a great ability to produce biofilms that are associated with the chronicity of wounds in diabetic patients. Routine screening for biofilm formers in chronic wounds and their antibiotic susceptibility testing will help in early treatment and prevent any other complications.

Introduction and Background: Diabetes is a chronic metabolic disease characterized by elevated blood glucose levels and is one of the main global health concerns. Synthetic sugar substrate has many side effects such as leukemia, bladder cancer, hepatotoxicity, breast cancer, headache, and brain toxicity. The WHO and FDA has recently banned some of the synthetic sugar alternatives due to their carcinogenic effects.Objective and Methodology: Therefore, the main objective of the current study was to investigate the safety and binding affinity of Stevioside with Glucose Transpoter-4 (GLUT-4), Akt, Insulin Receptor (IR) and Insulin Receptor Substrate-1 (IRS-1) to confirmed that Stevioside is one the potent natural sweetener/drug for diabetes. This study delves into the molecular interaction between Stevioside and key diabetic proteins: GLUT-4, Akt, IR and IRS-1. A precise molecular docking approach was used to simulate the binding affinity of Stevioside to these proteins. The pharmacokinetic properties of the molecule should be taken into consideration as important variables throughout the virtual screening process.Results: The result of active site analysis of GLUT-4, Akt, IR and IRS-1 showed a zone of 2158.359 Ǻ2, 579.259 Ǻ2, 762.651 Ǻ2, and 152.167 Ǻ2 and a volume of 2765.094 Ǻ³, 355.567 Ǻ³, 686.806 Ǻ³, and 116.874 Ǻ³, respectively. Docking analysis of the Stevioside compound showed the highest docking energy with scores of −9.9 with GLUT-4, −6.7 with Akt, −8.0 with IR and −8.8 with IRS-1. Studies indicated that it remains undigested by stomach acids and enzymes and is not absorbed in the upper small intestine. Further, tests revealed no hepatotoxicity, AMES toxicity, or skin sensitivity, making it a promising candidate for safe consumption as drug metabolism.Conclusion and Recommendations: Instead of other sugar alternatives, Stevioside will help diabetic patients with a lower chance of infections, lowered blood pressure/blood sugar, and increased glucose uptake in diabetic muscles. Stevioside is a natural sweetener, and the current study recommends its usage in various dietary products for diabetic patients.

Objective To evaluate the Descemet membrane endothelial keratoplasty (DMEK) preparation performance of trainee surgeons in an ex vivo human donor cornea DMEK wet lab simulation setting.Methods Human donor corneoscleral rims unsuitable for transplantation were obtained from Moorfields Lions Eye Bank. At the wet lab, graft stripping was performed by scoring the peripheral endothelium. The trypan blue positive cells (TBPC) and cell density (cells/mm2—reticule count) were counted manually before and after stripping. The procedural time, peripheral and central tears and complete peel-off were also recorded and analysed.Results Eight trainee surgeons attended the wet lab each attempting three DMEKs. This study aims to explore the adoption of Open-Source Software’s (OSS) in the academic libraries of Pakistan. It also focuses on the challenges encountered during the application and management of these software’s in both public and private sectors academic libraries around the country. Quantitative data was collected through structured questionnaire developed and distributed online to the target population. The findings indicate that the library's adoption of OSS, such as Koha, SLIMS, GREENSTONE, and DSpace, was motivated by the need to distribute information to library users efficiently, modernize library staff workflows, and enhance user accessibility to library information resources. The OSS has been instigated specially in the cataloguing and circulation and digital library management functions of the library. The major hurdles impeding the software's full implementation include lack of planning, lack of technical expertise, and security concerns related to library software’s. Study recommends that, to successfully adopt and implement any integrated library Software, proper planning and prior feasibility studies, along with proper training of the library professionals are recommended.

More recently Kohat's fold-thrust belt, including the Karak area has emerged as an important hydrocarbon fertile region of the western Himalayan orogenic system. The Eocene and Paleocene strata, including mechanical weak Eocene units representing a shallow decollement have a noticeable contribution to the deformation style and petroleum system of the area. In the present study, surface mapping is integrated with seismic information to elucidate the structural style of the Karak area, the data reveal that there are two different structural entities, separated by Karak Fault Zone. The northern domain portrays salt diapiric structures representing doubly plunging and complexity overturned, and fault bounded folds' geometries. However, in the subsurface below the Eocene sequence, the deformation is characterized by south-directed imbricate thrust system. In contrast, because of the absence of Eocene salt horizon, the southern domain of the Karak area illustrates a brittle style of deformation, mimicking fault propagation folding. In addition, an array of south-verging imbricate system from a regional basal decollement translates rock as old as Permian along the Surghar range frontal thrust. It can be observed that fault geometry and fold styles in the north and south of Karak Fault Zone are entirely different. Restored cross-sections across the area indicate 35% of shortening has been accommodated in the area. Moreover, from the present study it has been concluded that in the purlieu of Karak Fault Zone, possibly occupy auspicious structural traps at top Paleocene Lockhart level.

Abstract Computer-aided drug design has been employed to get the medicinal effects against Corona virus from different pyridine derivatives after synthesizing the new compounds. Additionally, various computational studies are also employed between the newly prepared pyridine derivatives and three controls against three proteins (6Y2E, 6M71 and 6M3M). Different methods were employed to synthesize new pyridine derivatives according to the literature using different reaction mediums. MTT was performed for cytotoxicity study and IC50 for inhibitory concentration. Additionally, in-silico studies including DFT, molecular docking, molecular dynamics, MMPBSA, ADME, pharmacokinetics and Lipinski rules were evaluated. The chemical structures of all new compounds were elucidated based on spectroscopic investigation. A molecular docking study demonstrated that compounds 5, 11, and 12 have the best binders of the SARS-CoV-2 main protease enzyme, with energy scores of − 7.5 kcal/mol, − 7.2 kcal/mol, and − 7.9 kcal/mol, respectively. The net binding energy values of the 11-Mpro, 12-Mpro, and 5-Mpro complexes revealed their highly stable nature in terms of both intermolecular interactions and docked conformation across the simulation time. ADME properties, besides the pharmacokinetics and Lipinski rules, showed that all seven newly synthesized compounds follow Lipinski rules with high GI absorption. The In Vitro antiviral study against SARS-CoV-2 using MTT methods confirms that compound 5 has more potential and is safer than other tested compounds. The study shows that the newly synthesized pyridine derivatives have medicinal properties against SARS-CoV-2 without violating Lipinski rules. Compounds 5, 11, and 12, particularly compound 5, may serve as promising potential candidate for COVID-19.

Abstract Physicochemical and phytochemical assessment of leaf mustard (Brassica juncea L.) grown in different agroclimatic conditions is essential to highlight their compositional variability and evaluate the most suitable bunch of agroclimatic and agronomic practices. B. juncea is one of the important leafy vegetables that serve as source of vitamin A and C and iron, and plenty of antioxidants. This in situ research was executed to assess the quality variability of B. juncea grown in different agroecosystems. Leaves’ samples of B. juncea were procured from 15 farmers’ fields exhibiting different agroclimatic conditions i.e., elevation, nutrient management, temperature, irrigation, and tillage practices. Leaves’ samples were subjected to physicochemical and phytochemical analysis, i.e., moisture, pH, TSS, ascorbic acid, carotenoids, phenolics, flavonoids, and antioxidant potential. In the leaves’ samples of B. juncea, the target properties were found to vary significantly (P ≤ 0.05) in different agroclimatic conditions. The moisture content, ascorbic acid, phenolic content, carotenoids, and antioxidants were found in the range of 62.7–79.3%, 74–91 mg/100 g, 49.2–49.2 mg GAE/100 g, 436.3–480 mg β carotene/100 g, 32.7–46.67%, respectively. This study elaborates the significant variation of physicochemical and phytochemical attributes of B. juncea due to the prevailing agroclimatic conditions. This necessitates the appropriate choice of B. juncea concerning its composition and ecological conditions of its cultivation in the prospective health benefits.

The tri-allelic pattern is a genotyping abnormality that can be observed during routine short tandem repeat (STR) profiling in the field of forensic science. Fourteen tri-allelic patterns have been observed in nine different loci during routine forensic casework consisting of 20,000 STR profiles. All these 20,000 STR unrelated profiles were profiled using AmpFlSTR Identifiler Plus® and Global Filer® kits. Tri-allelic patterns can be divided into two types based on RFUs (relative fluorescent units) in peaks of three component alleles. Unequal RFUs of all three peaks are observed in the Type-I pattern, whereas in the Type-II pattern, RFUs of all three peaks are nearly equal. A total of nine novel tri-allelic genotypes were observed out of 20,000 unrelated individuals in the Pakistani population. All of these forensic cases belonged to the Type-I pattern which means that the sum of the height of two smaller peaks is nearly equal to the height of the third larger peak. The frequency of occurrence for all these patterns was compared with already reported data. In this study, eight novel tri-allelic patterns have been reported which are not listed in the National Institute of Standards and Technology (NIST) database as well as in any published article.

The fundamental existence of any living organism necessitates the availability of pure and safe water. The ever-increasing population has led to extensive industrialization and urbanization, which have subsequently escalated micropollutants and water contamination. The environmental impact on various life forms poses a dire need for research in effective environmental management. Versatile technologies involving multiple approaches, including physiochemical and biological bioremediation strategies, draw insights from environmental biology. Metabolic annihilation mediated by microbes shows significant potential in the bioconversion of toxic micropollutants to tolerable limits. Environmentally friendly, cost-effective, and sustainable strategies are envisaged for efficient environmental protection. Phytoremediation technology, especially floating wetland treatments, facilitates micropollutant elimination, landscape management, ecosystem conservation, and aesthetic enhancement in diverse environments. The incorporation of nanomaterials in the bioremediation of toxic micropollutants augments novel and innovative strategies for water pollution abatement. This paper offers a novel strategy that combines nanomaterials to improve micropollutant degradation with bioremediation techniques, particularly the creative application of phytoremediation technologies like floating wetlands. Combining these techniques offers a novel viewpoint on long-term, affordable approaches to reducing water pollution. Additionally, the review proposes a forward-looking strategic framework that addresses the accumulation and refractory nature of micropollutants, which has not been thoroughly explored in previous literature.

Natural product synthesis is an important area of chemical study having various prospects for medicinal, industrial, and agricultural industry. Grignard reaction was developed as a flexible and necessary tool for the production of complex organic compounds in synthetic procedures. Additionally, the synthetic flexibility of the Grignard reaction facilitates its usage in the construction of more complicated ring frameworks and stereoselective transformations. It has been substantially applied in natural product synthesis, demonstrating its effectiveness in the generation of crucial C-C, C-X bonds and the development of intricate molecular structures. Use of Grignard reaction has been significantly studied towards the synthesis of biologically active natural products such as terpenoids, polyketides, alkaloids and amino acids. This review outlines the numerous instances where Grignard reagents have been exploited in the synthesis of notable natural products since past two years.

Thyme oil (TO) is a valuable essential oil believed to possess a variety of bioactivities, including antibacterial, anticancer, and antioxidant properties. These attributes grant TO the excellent capability to treat a wide range of diseases, particularly the effective eradication of Helicobacter pylori infection in the stomach. However, its practical use is limited by its low stability under atmospheric conditions. Our current research aims to encapsulate TO in eudragit (EGT) microsponges to enhance its stability and improve its effectiveness against H. pylori. The TO microsponges were prepared using EGT as a polymer, polysorbate 80 as a stabilizer, and dichloromethane (DCM) as a solvent via the quasi-emulsion solvent evaporation method. The product yield, particle size, surface morphology, entrapment efficiency, drug-polymer interaction, in-vitro floating, and in-vitro drug release of the microsponges were evaluated. The most promising microsponge was tested against H. pylori ATCC 43504 strains. The results showed that the microsponges exhibited a high product yield (ranging from 41 % ± 0.75–81.27 % ± 1.13), excellent entrapment efficiency (ranging from 63.01 % ± 0.79–88.64 % ± 0.98), prolonged in-vitro floating time (more than 12 h) and sustained in-vitro drug release for 18 h (81.53 %). Scanning electron microscopy results indicated that the microsponges were spherical in shape with a spongy surface. The average particle size of the selected microsponges was determined to be 49.79 ± 1.4 μm, and their average pore size was measured to be 0.81 ± 0.14 μm. DSC study results revealed that TO was physically entrapped in the microsponges. In-vitro anti-H. pylori activity studies demonstrated that TO in microsponge was more effective against H. pylori than pure TO. In conclusion, the developed microsponges containing thyme oil provide a promising alternative for the efficient targeting and eradication of H. Pylori infection.

Introduction: Olive (Olea europaea L.) is one of the important agricultural and medicinal tree species. This study aimed to assess the antifungal efficacy of olive leaf extract (OLE) obtained from different cultivars and during different seasons against Candida albicans strains. Methods: OLE was prepared using four olive cultivars (‘Koroneiki’, ‘Mission’, ‘Rowghani’, and ‘Zard’) obtained in two seasons (spring and autumn) from Golestan province, North of Iran. The phenolic and oleuropein contents of vegetative leaves were measured by colorimetric and HPLC techniques, respectively. The antifungal capacities of OLEs were tested by agar well diffusion and the minimal inhibitory concentration (MIC) was evaluated by micro-dilution assay. Results: The findings of our study showed that the total phenolic (27.45-88.16 mg g-1) and oleuropein (3.64-18.13 mg g-1) contents varied in leaf extracts, respectively. The highest amount was found in ‘Koroneiki’ and ‘Zard’ (spring leaves) and the lowest in ‘Mission’ cultivars (autumn leaves). The inhibition zones and MIC ranged from 1.92 to 15.41 mm and from 6.07 to 27.20 mg mL-1 based on C. albicans strains, respectively. Relationship between total phenolic content as an independent variable (X) and inhibition zones or MIC as dependent variables (Y) fitted polynomial curves. Conclusion: The present study highlighted the phytochemical and anti-candidal efficacy of OLE derived from olive cultivars or the seasons of harvested leaves against C. albicans strains. It is suggested that ‘Koroneiki’ and ‘Zard’ cultivars, especially during the spring season, could be exploited to isolate potential broad-spectrum antifungal drugs.

Design of orthopedic implants is a challenging task to cope with desired properties, therefore, numerous metal-alloy and coating were utilized in the fabrication of implants as alternative options. Magnesium (Mg)-based alloys are among the unique materials owing to their promising properties to avoid cytotoxicity, genotoxicity, and carcinogenicity; however, the fast degradation of Mg limits its clinical applications. In this study, hydroxyapatite (HA) nano-powder was coated on pure Mg as the compositions of both materials are promising biodegradable bone-compatible materials. The electrophoretic deposition (EPD) technique was used to produce a bioactive coating on pure Mg. The research novelty highlighted for the first time, the EPD parameters were optimized via the full Design of Experiment (DoF) approach during HA coating of Mg, while the weight loss of pure Mg and HA coated Mg were investigated in SBF for 7, 14, 21, and 28 days. The surface morphologies of both materials were investigated before and after coating. The bioactivity of the coated samples was examined in Simulated Body Fluid (SBF) and investigated using SEM. Surface roughness, contact angle measurement, scratch test, wear test, an in-vitro study in SBF, electrochemical corrosion, and cellular studies were conducted according to the standard approaches. The results show that HA coated Mg samples are compatible for orthopedic implants in terms of surface roughness, wettability, adhesion, wear sustainability, HA growth, corrosion rate, and cell viability. Thus, the biological, mechanical, and cellular properties of HA-coated implants are suitable and recommended as alternatives to biodegradable implants.

Background: The IL-12/23/ISG15-IFN-γ pathway is the main immunological pathway for controlling intra-macrophagic microorganisms such as Mycobacteria, Salmonella, and Leishmania spp. Consequently, upon mutations in genes of the IL-12/23/ISG15-IFN-γ pathway cause increased susceptibility to intra-macrophagic pathogens, particularly to Mycobacteria. Therefore, the purpose of this study was to characterize the mutations in genes of the IL-12/23/ISG15-IFN-γ pathway in severe tuberculosis (TB) patients. Methods: Clinically suspected TB was initially confirmed in four patients (P) (P1, P2, P3, and P4) using the GeneXpert MTB/RIF and culturing techniques. The patients' Peripheral blood mononuclear cells (PBMCs) were then subjected to ELISA to measure Interleukin 12 (IL-12) and interferon gamma (IFN-γ). Flow cytometry was used to detect the surface expressions of IFN-γR1 and IFN-γR2 as well as IL-12Rβ1and IL-12Rβ2 on monocytes and T lymphocytes, respectively.The phosphorylation of signal transducer and activator of transcription 1(STAT1) on monocytes and STAT4 on T lymphocytes were also detected by flow cytometry. Sanger sequencing was used to identify mutations in the IL-12Rβ1, STAT1, NEMO, and CYBB genes. Results: P1's PBMCs exhibited reduced IFN-γ production, while P2's and P3's PBMCs exhibited impaired IL-12 induction. Low IL-12Rβ1 surface expression and reduced STAT4 phosphorylation were demonstrated by P1's T lymphocytes, while impaired STAT1 phosphorylation was detected in P2's monocytes. The impaired IκB-α degradation and abolished H2O2 production in monocytes and neutrophils of P3 and P4 were observed, respectively. Sanger sequencing revealed novel nonsense homozygous mutation: c.191 G>A/p.W64 * in exon 3 of the IL-12Rβ1 gene in P1, novel missense homozygous mutation: c.107 A>T/p.Q36L in exon 3 of the STAT1 gene in P2, missense hemizygous mutation:: c.950 A>C/p.Q317P in exon 8 of the NEMO gene in P3, and nonsense hemizygous mutation: c.868 C>T/p.R290X in exon 8 of CYBB gene in P4. Conclusion: Our findings broaden the clinical and genetic spectra associated with IL-12/23/ISG15-IFN-γ axis anomalies. Additionally, our data suggest that TB patients in Pakistan should be investigated for potential genetic defects due to high prevalence of parental consanguinity and increased incidence of TB in the country.

Enterococcus faecalis is a prevalent opportunistic pathogen associated with chicken embryonic and neonatal chick mortality, posing a significant challenge in poultry farming. In the current study, E. faecalis strain EF6, isolated from a recent hatchery outbreak, served as the host bacterium for the isolation of a novel phage EFP6, capable of lysing E. faecalis. Transmission electron microscopy revealed a hexagonal head and a short tail, classifying EFP6 as a member of the Autographiviridae family. EFP6 showed sensitivity to ultraviolet radiation and resistance to chloroform. The lytic cycle duration of EFP6 was determined to be 50 min, highlighting its efficacy in host eradication. With an optimal multiplicity of infection of 0.001, EFP6 exhibited a narrow lysis spectrum and strong specificity towards host strains. Additionally, EFP6 demonstrated optimal growth conditions at 40 °C and pH 8.0. Whole genome sequencing unveiled a genome length of 18,147 bp, characterized by a GC concentration of 33.21% and comprising 25 open reading frames. Comparative genomic assessment underscored its collinearity with related phages, notably devoid of lysogenic genes, thus ensuring genetic stability. This in-depth characterization forms the basis for understanding the biological attributes of EFP6 and its potential utilization in phage therapy, offering promising prospects for mitigating E. faecalis-associated poultry infections.

Geographically distributed cloud data centers (DCs) consume enormous amounts of energy to meet the ever-increasing processing and storage demands of users. The brown energy generated using fossil fuels is expensive and significantly contributes to global warming. Considering the environmental impact caused by the high carbon emissions and relatively high energy cost of brown energy, we propose the integration of renewable energy sources (RES), especially solar and wind energy, with brown energy to power cloud data centers. In our earlier study, we addressed the intermittency of renewable energy sources, where we replaced the random initialization of artificial neural network (ANN) edge weights with the harmony search algorithm (HSA)-optimized assignment of weights. This study incorporated reliably forecast solar and wind energy into the input parameters of our proposed green energy manager (GEM), for cost minimization, carbon emission minimization, and better energy management of cloud DCs, to make our current study more reliable and trustworthy. Four power sources, on-site solar energy and wind energy, off-site solar energy and wind energy, energy stored in energy storage devices, and brown energy, were considered in this study and simulations were carried out for three different cases. The simulation results showed that case 1 (all brown) was 58% more expensive and caused 71% higher carbon emissions than case 2.1 (cost minimization). Case 1 (all brown) was 39% more expensive and had 80% higher carbon emissions than case 2.2 (carbon emission minimization). The simulation results justify the necessity and importance of the GEM, and finally the results proved that our proposed GEM is less expensive and more environmentally friendly.
