Abstract

This paper tackles the problem of the worst-class error rate, instead of the standard error rate averaged over all classes. For example, a three-class classification task with class-wise error rates of 10\%, 10\%, and 40\% has a worst-class error rate of 40\%, whereas the average is 20\% under the class-balanced condition. The worst-class error is important in many applications. For example, in a medical image classification task, it would not be acceptable for the malignant tumor class to have a 40\% error rate, while the benign and healthy classes have 10\% error rates.We propose a boosting algorithm that guarantees an upper bound of the worst-class training error and derive its generalization bound. Experimental results show that the algorithm lowers worst-class test error rates while avoiding overfitting to the training set.

Abstract

White matter bundle segmentation using diffusion MRI fiber tractography has become the method of choice to identify white matter fiber pathways in vivo in human brains. However, like other analyses of complex data, there is considerable variability in segmentation protocols and techniques. This can result in different reconstructions of the same intended white matter pathways, which directly affects tractography results, quantification, and interpretation. In this study, we aim to evaluate and quantify the variability that arises from different protocols for bundle segmentation. Through an open call to users of fiber tractography, including anatomists, clinicians, and algorithm developers, 42 independent teams were given processed sets of human whole-brain streamlines and asked to segment 14 white matter fascicles on six subjects. In total, we received 57 different bundle segmentation protocols, which enabled detailed volume-based and streamline-based analyses of agreement and disagreement among protocols for each fiber pathway. Results show that even when given the exact same sets of underlying streamlines, the variability across protocols for bundle segmentation is greater than all other sources of variability in the virtual dissection process, including variability within protocols and variability across subjects. In order to foster the use of tractography bundle dissection in routine clinical settings, and as a fundamental analytical tool, future endeavors must aim to resolve and reduce this heterogeneity. Although external validation is needed to verify the anatomical accuracy of bundle dissections, reducing heterogeneity is a step towards reproducible research and may be achieved through the use of standard nomenclature and definitions of white matter bundles and well-chosen constraints and decisions in the dissection process.

Abstract

Myelination plays an important role in the neurological development of infant brain and MRI can visualize the myelination extension as T1 high and T2 low signal intensity at white matter. We tried to construct a convolutional neural network machine learning model to estimate the myelination. Eight layers CNN architecture was constructed to estimate the subjects age with T1 and T2 weighted image at 5 levels associated with myelin maturation in 119 subjects up to 24 months. CNN model learned with all age dataset revealed a strong correlation between the estimated age and the corrected age and the coefficient of correlation, root mean square error and mean absolute error was 0. 81, 3. 40 and 2. 28. Moreover, the adaptation of ensemble learning models with two datasets 0 to 16 months and 8 to 24 months improved that to 0. 93, 2. 12 and 1. 34. Abstract

In this paper, we introduce a lab trial that is conducted to study the feasibility of ultra-low latency for 5G. Using a novel frame structure and signaling procedure, a stabilized 1.5 ms hybrid automatic repeat request (HARQ) round trip time (RTT) is observed in lab trial as predicted in theory. Abstract

Purpose: To evaluate a vendor-agnostic multiparametric mapping scheme based on 3D quantification using an interleaved Look-Locker acquisition sequence with a T2 preparation pulse (3D-QALAS) for whole-brain T1, T2, and proton density (PD) mapping.Methods: This prospective, multi-institutional study was conducted between September 2021 and February 2022 using five different 3T systems from four prominent MRI vendors. The accuracy of this technique was evaluated using a standardized MRI system phantom. Intra-scanner repeatability and inter-vendor reproducibility of T1, T2, and PD values were evaluated in 10 healthy volunteers (6 men; mean age +/- SD, 28.0 +/- 5.6 y) who underwent scan-rescan sessions on each scanner (total scans = 100). To evaluate the feasibility of 3D-QALAS, nine patients with multiple sclerosis (nine women; mean age +/- SD, 48.2 +/- 11.5 y) underwent imaging examination on two 3T MRI systems from different manufacturers.Results: Quantitative maps obtained with 3D-QALAS showed high linearity (R2 = 0.998 and 0.998 for T1 and T2, respectively) with respect to reference measurements. The mean intra-scanner coefficients of variation for each scanner and structure ranged from 0.4% to 2.6%. The mean structure-wise test-retest repeatabilities were 1.6%, 1.1%, and 0.7% for T1, T2, and PD, respectively. Overall, high inter-vendor reproducibility was observed for all parameter maps and all structure measurements, including white matter lesions in patients with multiple sclerosis.Conclusion: The vendor-agnostic multiparametric mapping technique 3D-QALAS provided reproducible measurements of T1, T2, and PD for human tissues within a typical physiological range using 3T scanners from four different MRI manufacturers., Funding Agencies|Juntendo Research Branding Project, the Sportology Center of Juntendo University Graduate School of Medicine, Leading-edge Research Project for Sports Medicine and Science

Abstract

The comprehension of the glymphatic system, a postulated mechanism responsible for the removal of interstitial solutes within the central nervous system (CNS), has witnessed substantial progress recently. While direct measurement techniques involving fluorescence and contrast agent tracers have demonstrated success in animal studies, their application in humans is invasive and presents challenges. Hence, exploring alternative noninvasive approaches that enable glymphatic research in humans is imperative. This review primarily focuses on several noninvasive magnetic resonance imaging (MRI) techniques, encompassing perivascular space (PVS) imaging, diffusion tensor image analysis along the PVS, arterial spin labeling, chemical exchange saturation transfer, and intravoxel incoherent motion. These methodologies provide valuable insights into the dynamics of interstitial fluid, water permeability across the blood–brain barrier, and cerebrospinal fluid flow within the cerebral parenchyma. Furthermore, the review elucidates the underlying concept and clinical applications of these noninvasive MRI techniques, highlighting their strengths and limitations. It addresses concerns about the relationship between glymphatic system activity and pathological alterations, emphasizing the necessity for further studies to establish correlations between noninvasive MRI measurements and pathological findings. Additionally, the challenges associated with conducting multisite studies, such as variability in MRI systems and acquisition parameters, are addressed, with a suggestion for the use of harmonization methods, such as the combined association test (COMBAT), to enhance standardization and statistical power. Current research gaps and future directions in noninvasive MRI techniques for assessing the glymphatic system are discussed, emphasizing the need for larger sample sizes, harmonization studies, and combined approaches. In conclusion, this review provides invaluable insights into the application of noninvasive MRI methods for monitoring glymphatic system activity in the CNS. It highlights their potential in advancing our understanding of the glymphatic system, facilitating clinical applications, and paving the way for future research endeavors in this field. Abstract

Dictyostelium discoideum cells contain auto-fluorescent vacuoles. To determine the identity of these vacuoles, the fluorescent dye 4-nitro-7-(1-piperazinyl)-2,1,3-benzoxadiazole (NBD-PZ) was used to stain the lysosomes in D. discoideum cells. Neither the auto-fluorescent vacuoles nor lysosomes were observed in D. discoideum cells immediately after they arose from spores or in stationary phase cells. However, both the auto-fluorescent vacuoles and lysosomes were visible in cells that had entered growth phase. Auto-fluorescent vacuoles and lysosomes were also observed in stationary phase cells incubated with chloroquine. When the cells were allowed to phagocytose BioParticles Fluorescent Bacteria (orange fluorescence) for 1 h, orange phagosomes and blue auto-fluorescent vacuoles were observed as independent moieties. However, after an additional 2 h of incubation, we observed vacuoles with mixed fluorescence (orange and blue) in the cells, suggestive of secondary lysosomes. These results suggest that the auto-fluorescent vacuoles in D. discoideum cells are lysosomes.
