Abstract Oral squamous cell carcinoma (OSCC) is a prevalent oral malignancy, which poses significant health risks with a high mortality rate. Regulatory T cells (Tregs), characterized by their immunosuppressive capabilities, are intricately linked to OSCC progression and patient outcomes. The metabolic reprogramming of Tregs within the OSCC tumor microenvironment (TME) underpins their function, with key pathways such as the tryptophan-kynurenine-aryl hydrocarbon receptor, PI3K-Akt-mTOR and nucleotide metabolism significantly contributing to their suppressive activities. Targeting these metabolic pathways offers a novel therapeutic approach to reduce Treg-mediated immunosuppression and enhance anti-tumor responses. This review explores the metabolic dependencies and pathways that sustain Treg function in OSCC, highlighting key metabolic adaptations such as glycolysis, fatty acid oxidation, amino acid metabolism and PI3K-Akt-mTOR signaling pathway that enable Tregs to thrive in the challenging conditions of the TME. Additionally, the review discusses the influence of the oral microbiome on Treg metabolism and evaluates potential therapeutic strategies targeting these metabolic pathways. Despite the promising potential of these interventions, challenges such as selectivity, toxicity, tumor heterogeneity, and resistance mechanisms remain. The review concludes with perspectives on personalized medicine and integrative approaches, emphasizing the need for continued research to translate these findings into effective clinical applications for OSCC treatment.

Background and purposeIt is unknown the neural mechanisms of developmental stuttering (DS). The aim of this study was to investigate the changes in the structural morphology of the brain in Chinese children who stutter.MethodsA case–control study was conducted to collect magnetic resonance imaging data from stuttering and non-stuttering children, thereby analyzing whole-brain gray matter volume and cortical morphological changes in stuttering children.ResultsA total of 108 subjects were recruited (stuttering group: control group = 1:1). Comparing to healthy controls, the gray matter volume was significantly decreased in right temporal gyrus and bilateral cerebellum. Additionally, there was a significant reduction in cortical folds in the right insula and right superior temporal gyrus. Moreover, the gray matter volume of the right cerebellum and right temporal gyrus is related to the severity score of stuttering.ConclusionThe present study proposes that the neural mechanisms underlying DS are intricately linked to the cortico-basal ganglia-thalamo-cortical loop and the dorsal language pathway. This finding is expected to provide reference value for the clinical treatment of DS.

Abstract Gas Insulated Switchgear (GIS) was a crucial electrical equipment that provides the safety and security of power systems, and finding the eco-friendly alternatives of sulphur hexafluoride (SF6) as insulating medium in GIS has been an urgent demand in past decades. However, few studies reported the specific dielectric strength under various electrodes and partial discharge (PD) decomposition products detection of applying the eco-friendly gas in GIS. In this paper, the insulation properties among C4F7N/CO2 mixtures, dry air and CO2 were explored under the reduced-scale gas-insulated experimental device. The relationship between AC breakdown voltage and gas pressure was gained under the different electrodes and gap distances, and 0.6 MPa 7% C4F7N/93% CO2, 0.9 MPa dry air, 0.9 MPa CO2 all possess the capacity to be the insulating medium in eco-friendly gas insulated switchgear. Furthermore, the partial discharge experimental was also carried out to realize the decomposition products detection of dry air, which designs three common defect types including metal protrusion defects, air gap defects, and metal contamination defects. The decomposition products detection result shows that the contains of CO2, CO, and NO2 linearly increase with the increasing applied voltages and times, and the partial discharge defects are distinguished according to the ratios of c(CO2 + CO)/c(NO2) and c(CO2)/c(CO). The results can provide the basis for the further development of eco-friendly gas insulated switchgear.

As older adults age, changes in their physiological, psychological, and cognitive functions often lead to inherent anxiety and fear regarding the use of digital technologies. Cognitive age, reflecting an older adult’s mental perception of their chronological age, is a crucial moderating factor in shaping their willingness and behavior towards technology adoption. However, the mechanisms through which cognitive age impacts the behavior of older adults within the realm of digital technology utilization remain unclear. Thus, grounded in the Extended Technology Acceptance Model (ETAM) and employing structural equation modeling, this study intends to investigate mechanisms which influence older adults’ behavioral intentions towards the use of museum digital displays. Consequently, it confirms that attitudes mediate the correlation between perceived usefulness, subjective norm, perceived self-efficacy, and the behavioral intention towards museum digital display utilization. Perceived ease of use does not directly affect attitudes, but instead exerts an indirect impact on attitude through perceived usefulness. Cognitive age negatively moderates the relationship between attitudes and the behavioral intention to use digital technologies. Additionally, the mediating role of attitudes in the correlation between older adults’ perception of museum digital displays and behavioral intention is moderated by cognitive age. Specifically, older adults with a higher cognitive age value (who perceive themselves to be younger than their chronological age) exhibit a weaker mediating effect of attitudes on the relationship between perceived usefulness and behavioral intention compared to those with a lower cognitive age value (who perceive themselves to be older than their chronological age). The findings seek to unbox the “black box” of how cognitive age mediates the correlation between attitudes and behavioral intention towards the use of museum digital displays among older adults, providing valuable theoretical insights for the bidirectional enhancement of digital technology utilization, the overall well-being of older adults, and high-quality development in museums.

In the energy-saving retrofit of existing buildings, investors are particularly concerned about the energy-saving performance of exterior windows and the payback period of additional costs. This study evaluates representative cities in four different climate zones in China to simulate the energy consumption of large office buildings after replacing different glass windows and conducting energy-saving and economic feasibility assessments. The research method includes the following steps: First, a baseline model of large office buildings in four cities was established using AutoBPS and OpenStudio. Then, the baseline and retrofit models of replacing glass windows were simulated using the EnergyPlus V9.3.0 to obtain multiple hourly energy consumption results. The commercial electricity and gas prices in the four cities were adjusted to calculate the total cost within 20 years after replacing different types of windows. Using the discounted payback period (DPP), net present value (NPV), and profitability index (PI) as evaluation indicators, a feasibility analysis was conducted in the four regions to evaluate the economic feasibility of replacing building windows. The simulation results show that considering economic feasibility and meeting energy-saving standards, it is more economical to choose windows with moderate U-value and SHGC value in the four regions than to choose windows with the smallest U-value and SHGC value, and that both energy savings and economic benefits are closely related to building age, with older buildings (especially those in Changsha and Shenzhen) showing greater benefits. Furthermore, the optimal window types in the four cities determined in this study can recover the investment cost within the window life, with Harbin (SC), Beijing (C), Changsha (HC), and Shenzhen (HW) with the payback period of 6.60, 15.66, 10.16, and 11.42 years, respectively. The research model established in this study provides a useful evaluation path for selecting windows for the energy-saving retrofit of large office buildings in cities in different climate zones and provides data support for the decision making of energy-saving retrofit investors.

The Omicron strains of SARS-CoV-2 pose a significant challenge to the development of effective antibody-based treatments as immune evasion has compromised most available immune therapeutics. Therefore, in the ‘arms race’ with the virus, there is a continuing need to identify new biologics for the prevention or treatment of SARS-CoV-2 infections. Here, we report the isolation of nanobodies that bind to the Omicron BA.1 spike protein by screening nanobody phage display libraries previously generated from llamas immunized with either the Wuhan or Beta spike proteins. The structure and binding properties of three of these nanobodies (A8, H6 and B5-5) have been characterized in detail providing insight into their binding epitopes on the Omicron spike protein. Trimeric versions of H6 and B5-5 neutralized the SARS-CoV-2 variant of concern BA.5 both in vitro and in the hamster model of COVID-19 following nasal administration. Thus, either alone or in combination could serve as starting points for the development of new anti-viral immunotherapeutics.

Introduction: Antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) and its subtypes, granulomatosis with polyangiitis (GPA), microscopic polyangiitis (MPA), and eosinophilic GPA (EGPA), frequently present with acute kidney injury and can often lead to kidney failure, even with successful induction therapy. Few contemporary, nationally representative studies have described hospital complications of AAV. Methods: Using data from the 2016–2020 National Inpatient Sample, a nationally representative database, we identified hospitalizations from adults with a new diagnosis of AAV (subtype or unspecified) and an inpatient kidney biopsy during the index hospitalization. We described baseline characteristics, associated inpatient procedures and complications, and compared lengths of stay and costs by geographic region, hospital characteristics, and AAV subtype. Results: We identified an average of 1,329 cases of hospitalized AAV with a concurrent kidney biopsy per year over the 5-year period. More than 50% were not designated as having a specific subtype, likely owing to delays in documentation of histopathology. Kidney involvement was severe as the majority of patients developed acute kidney injury, and the proportion of patients who required inpatient dialysis was approximately 24%. Approximately 20% of patients developed hypoxia. Inpatient plasmapheresis was delivered to 20.4% and 20.6% of patients with GPA and MPA, respectively. There were no clinically meaningful or statistically significant differences in adjusted length of stay or inpatient costs among AAV subtypes. Abstract Thermochromic perovskite smart windows (TPWs) are a cutting-edge energy-efficient window technology. However, like most perovskite-based devices, humidity-related degradation limits their widespread application. Herein, inspired by the structure of medical masks, a unique triple-layer thermochromic perovskite window (MTPW) that enable sufficient water vapor transmission to trigger the thermochromism but effectively repel detrimental water and moisture to extend its lifespan is developed. The MTPW demonstrates superhydrophobicity and maintains a solar modulation ability above 20% during a 45-day aging test, with a decay rate 37 times lower than that of a pristine TPW. It can also immobilize lead ions and significantly reduce lead leakage by 66 times. Furthermore, a significant haze reduction from 90% to 30% is achieved, overcoming the blurriness problem of TPWs. Benefiting from the improved optical performance, extended lifespan, suppressed lead leakage, and facile fabrication, the MTPW pushes forward the wide applications of smart windows in green buildings.

Abstract Alzheimer's disease (AD) is a pressing concern in neurodegenerative research. To address the challenges in AD drug development, especially those targeting Aβ, this study uses deep learning and a pharmacological approach to elucidate the potential of pyrroloquinoline quinone (PQQ) as a neuroprotective agent for AD. Using deep learning for a comprehensive molecular dataset, blood–brain barrier (BBB) permeability is predicted and the anti‐inflammatory and antioxidative properties of compounds are evaluated. PQQ, identified in the Mediterranean‐DASH intervention for a diet that delays neurodegeneration, shows notable BBB permeability and low toxicity. In vivo tests conducted on an Aβ₁₋₄₂‐induced AD mouse model verify the effectiveness of PQQ in reducing cognitive deficits. PQQ modulates genes vital for synapse and anti‐neuronal death, reduces reactive oxygen species production, and influences the SIRT1 and CREB pathways, suggesting key molecular mechanisms underlying its neuroprotective effects. This study can serve as a basis for future studies on integrating deep learning with pharmacological research and drug discovery.

Abstract A wearable sensing system that can reconstruct dynamic 3D human body models for virtual cloth fitting is highly important in the era of information and metaverse. However, few research has been conducted regarding conformal sensors for accurately measuring the human body circumferences for dynamic 3D human body reshaping. Here, we develop a stretchable spring‐sheathed yarn sensor (SSYS) as a smart ruler, for precisely measuring the circumference of human bodies and long‐term tracking the movement for the dynamic 3D body reconstruction. The SSYS has a robust property, high resilience, high stability (>18 000), and ultrafast response (12 ms) to external deformation. It is also washable, wearable, tailorable, and durable for long‐time wearing. Moreover, geometric, and mechanical behaviors of the SSYS are systematically investigated both theoretically and experimentally. In addition, a transfer learning algorithm that bridges the discrepancy of real and virtual sensing performance is developed, enabling a small body circumference measurement error of 1.79%, noticeably lower than that of traditional learning algorithm. Furtherly, 3D human bodies that are numerically consistent with the actual bodies are reconstructed. The 3D dynamic human body reconstruction based on the wearing sensing system and transfer learning algorithm enables excellent virtual fitting and shirt customization in a smart and highly efficient manner. This wearable sensing technology shows great potential in human‐computer interaction, intelligent fitting, specialized protection, sports activities, and human physiological health tracking.

Carposina coreana is an important pest of Cornus officinalis, distributed in China, Korea, and Japan. In recent years, its damage to C. officinalis has become increasingly serious, causing enormous economic losses in China. This study and prediction of current and future suitable habitats for C. coreana in China can provide an important reference for the monitoring, early warning, prevention, and control of the pest. In this study, the potential distributions of C. coreana in China under current climate and future climate models were predicted using the maximum entropy (MaxEnt) model with ArcGIS software. The distribution point data of C. coreana were screened using the buffer screening method. Nineteen environmental variables were screened using the knife-cut method and variable correlation analysis. The parameters of the MaxEnt model were optimized using the kuenm package in R software. The MaxEnt model, combined with key environmental variables, was used to predict the distribution range of the suitable area for C. coreana under the current (1971–2000) and four future scenarios. The buffer screening method screened data from 41 distribution points that could be used for modeling. The main factors affecting the distribution of C. coreana were precipitation in the driest month (Bio14), precipitation in the warmest quarter (Bio18), precipitation in the coldest quarter (Bio19), the standard deviation of seasonal variation of temperature (Bio4), minimum temperature in the coldest month (Bio6), and average temperature in the coldest quarter (Bio11). The feature class (FC) after the kuenm package optimization was a Q-quadratic T-threshold combination, and the regularization multiplier (RM) was 0.8. The suitable areas for C. coreana under the current climate model were mainly distributed in central China, and the highly suitable areas were distributed in southern Shaanxi, southwestern Henan, and northwestern Hubei. The lowest temperature in the coldest month (Bio6), the average temperature in the coldest quarter (Bio11), and the precipitation in the warmest quarter (Bio18) all had good predictive ability. In future climate scenarios, the boundary of the suitable area for C. coreana in China is expected to shift northward, and thus, most of the future climate scenarios would shift northward.

Rationale & Objective: Intradialytic hypotension (IDH) is associated with mortality in adults with kidney failure requiring hemodialysis (HD); however, large-scale pediatric studies are lacking. Moreover, there is no evidence-based consensus definition of IDH in pediatric literature. We aimed to examine the association of commonly used definitions of IDH with mortality in adolescents and young adults. Study Design: This was a retrospective observational cohort study. Setting & Participants: In total, 1,199 adolescents and young adults (N = 320, aged 10-18 years and N = 879, aged 19-21 years) who initiated HD in a large dialysis organization were included. Exposures: This study used different definitions of IDH. Outcome: The study outcome was 2-year all-cause mortality. Analytical Approach: Several definitions of IDH were selected a priori based on a literature review. Patients were classified as having IDH if it was present in at least 30% of HD treatments during the first 90 days after dialysis initiation. Cox proportional hazards regression was used to test whether IDH associated with 2-year all-cause mortality. Results: Over a 2-year follow-up period, 54 (4.5%) patients died. Dependent on its definition, IDH was present in 2.9%-61.1% of patients. After the multivariable adjustment for sociodemographic and clinical characteristics, we found no association of IDH with mortality. Results were consistent across subgroups stratified by age (aged 150 mm Hg). We also examined IDH as occurring in 50% of baseline treatments, and did not find a dose-response association with mortality (P > 0.05). Limitations: Owing to low event rates, our current sample size may have been too small to detect a difference in mortality. Conclusions: Our study found that IDH was not associated with mortality in adolescents and young adults. Plain-Language Summary: Intradialytic hypotension (IDH), or hypotension experienced during dialysis, is common among children and young adults with kidney failure requiring hemodialysis. We examined the association of commonly used definitions of IDH with death in adolescents and young adults with kidney failure receiving hemodialysis. Intradialytic hypotension was present in 2.9%-61.1% of patients, depending on the definition, and over a 2-year follow-up period, 4.5% of the patients died. Rationale & Objective: Atrial fibrillation is the most common arrhythmia and is increasing in prevalence. The prevalence of atrial fibrillation is high among patients receiving dialysis, affecting ∼21.3% of the patients receiving hemodialysis and 15.5% of those receiving peritoneal dialysis. The association of previous dialysis modality with incident atrial fibrillation in patients after receiving their first kidney transplant has not been studied. Study Design: We used the United States Renal Data System to retrospectively identify adult, Medicare-insured patients who received their first kidney transplant between January 1, 2005, and September 30, 2012 and who had not previously been diagnosed with atrial fibrillation. Setting & Participants: The study included 43,621 patients who were aged 18 years older when receiving a first kidney transplant between January 1, 2005, and September 30, 2012 and whose primary payer was Medicare (parts A and B) at the time of transplantation and the 6 months preceding it. Exposure: Dialysis modality used before transplant. Outcome: Time to incidence of atrial fibrillation up to 3 years posttransplant. Analytical Approach: Multivariable Cox regression was used to estimate HRs. Results: Of 43,621 patients, 84.9% received hemodialysis and 15.1% received peritoneal dialysis before transplant. The mean ± SD age was 51 ± 13.6 years; 60.8% were male, 55.6% White, and 35.8% Black race. The mean dialysis vintage was 4.3 ± 2.8 years. Newly diagnosed atrial fibrillation after kidney transplant occurred in 286 patients (during 15,363 person-years) who had received peritoneal dialysis and in 2,315 patients (during 83,536 person-years) who had received hemodialysis. After multivariable adjustment, atrial fibrillation was 20% (95% CI, 4%-38%) more likely in those who had been receiving hemodialysis versus peritoneal dialysis, regardless of whether death was considered a competing risk or a censoring event. Each year of pretransplant dialysis vintage increased the risk of posttransplant atrial fibrillation by 6% (95% CI, 3%-9%). Limitations: Residual confounding; data from billing claims does not specify the duration of atrial fibrillation or whether it is valvular. Conclusions: Pretransplant hemodialysis, as compared with peritoneal dialysis, was associated with higher risk of newly diagnosed atrial fibrillation after a first kidney transplant. Plain-Language Summary: New-onset atrial fibrillation (AF) occurs in 7% of kidney transplant recipients in the first 3 years posttransplantation. We conducted this study to determine whether pretransplant dialysis modality was associated with posttransplant AF. We identified 43,621 patients; 84.9% used hemodialysis and 15.1% used peritoneal dialysis pretransplant. Multivariable Cox regression was used to estimate hazard ratios. We found that patients receiving hemodialysis pretransplant were at 20% increased risk of developing posttransplant AF as compared with patients receiving peritoneal dialysis. As our understanding of transplant-specific risk factors for AF increases, we may be able to better risk-stratify transplant patients and develop monitoring and management strategies that can improve outcomes.

Goji berry (Lycium barbarum L.) is a traditional Chinese herbal medicinal plant that is extensively cultivated in the arid and semiarid regions of northwest China. In this study, a novel cytorhabdovirus, tentatively named “goji cytorhabdovirus A (GCVA),” was identified from the goji berry plant exhibiting leaf crinkle symptoms through high-throughput sequencing (HTS). GCVA contains a linear, negative sense single-stranded RNA genome of 14,812 nucleotides and encodes six open reading frames in the order of 3′ leader-N-P-P4-M-G-L-5′ trailer. The genome of GCVA shares the highest nucleotide (nt) identity of 65.80% (16% query coverage) with yerba mate virus A (YmVA) (NC_076472). Super strength and toughness, excellent deformation resistance, and high-temperature service performance are the key factors to determine the practical application of new thermal barrier coatings (TBCs). The limited mobility of dislocations and the internal inherent defects in ceramics will inevitably lead to the decline of strength–plasticity and the reduction of service performance. Introducing preexisting twin boundaries and stacking faults (SFs) or preparing ceramic materials with high configuration entropy has demonstrated to be an effective strategy for enhancing the mechanical properties of ceramics. However, due to the positive thermal expansion coefficient of most ceramics and the remarkable increase of structural disorder at elevated temperature, the problem of elastic softening has become a bottleneck restricting the high-temperature service life of new TBCs. In this paper, the deformation behavior of high configuration entropy Zr6Ta2O17 ceramics at 25 to 1,200 °C was in situ monitored via digital image correlation technique and three-point bending test platform in high-temperature environment. A remarkable Elinvar-like effect appears in the Zr6Ta2O17 ceramic. More interestingly, mechanical deformation dominates the severe lattice distortion (deformation twins, SFs) and the disorder–order transition of chemical order at the atomic scale, while temperature can further enhance the degree of lattice distortion and ordering of Zr6Ta2O17 ceramics. Furthermore, the atomic fluctuations at high temperature promotes the comprehensive improvement of mechanical properties in the Zr6Ta2O17 ceramics.

Since its discovery, COVID-19 has rapidly spread across the globe and has had a massive toll on human health, with infection mortality rates as high as 10%, and a crippling impact on the world economy. Despite numerous advances, there remains an urgent need for accurate and rapid point-of-care diagnostic tests and better therapeutic treatment options. To contribute chemically distinct, non-protein-based affinity reagents, we report here the identification of modified DNA-based aptamers that selectively bind to the S1, S2, or receptor-binding domain of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein. Several aptamers inhibit the binding of the spike protein to its cell-surface receptor angiotensin-converting enzyme 2 (ACE2) and neutralize authentic SARS-CoV-2 virus in vitro, including all variants of concern. With a high degree of nuclease resistance imparted by the base modifications, these reagents represent a new class of molecules with potential for further development as diagnostics or therapeutics.

Amyotrophic Lateral Sclerosis (ALS) is a debilitating neurodegenerative condition characterized by the progressive degeneration of motor neurons. Despite extensive research in various model animals, the cellular signal mechanisms of ALS remain elusive, impeding the development of efficacious treatments. Among these models, a well-characterized and diminutive organism, Caenorhabditis elegans (C. elegans), has emerged as a potent tool for investigating the molecular and cellular dimensions of ALS pathogenesis. This review summarizes the contributions of C. elegans models to our comprehension of ALS, emphasizing pivotal findings pertaining to genetics, protein aggregation, cellular pathways, and potential therapeutic strategies. We analyze both the merits and constraints of the C. elegans system in the realm of ALS research and point towards future investigations that could bridge the chasm between C. elegans foundational discoveries and clinical applications.

Abstract Background Since the implementation of China’s two-child policy in 2016, the number of re-pregnant women after cesarean section has increased significantly. These women are more prone to fear of childbirth compared with primiparas due to their history of scarred uterus leading to a more complicated delivery process, which poses a great threat to their physical and mental health. However, there is currently limited research on the problem in China. The aim of this study was to assess fear of childbirth and its predictors in re-pregnant women after cesarean section in China. Methods A cross-sectional multicenter study was conducted in three hospitals from June 7 to December 7, 2020, in Changsha, China. Study hospitals were selected using a random sampling technique. Participants were selected using a convenience sampling technique. Three hundred fifty-eight women during the third trimester of pregnancy who were older than 18 years, having a history of CS(s), and not having major physical or mental health problems were included. Fear of childbirth and its predictors were evaluated using the Childbirth Attitude Questionnaire, the short form of the 32-item Chinese Childbirth Self-Efficacy Inventory, the Social Support Rating Scale, and the demographic-obstetric data sheet. After checking for completeness, data were exported to statistical software for analysis. 采用高效液氮加注技术对葵花籽油进行灌装，并与气氮加注、气液氮混合加注技术进行对比，研究其对葵花籽油品质和产品包装的影响。结果表明：3种充氮方式样品的酸值和过氧化值变化趋势基本一致；液氮加注样品常温储藏5个月后酸值（KOH）和过氧化值分别为0.07 mg/g和1.45 mmol/kg，与常温储藏相比，液氮加注样品45 ℃储藏5个月后酸值无差别，过氧化值稍有降低，为130 mmol/kg；液氮加注和气液氮混合加注样品初始及储藏过程中的过氧化值比气氮加注样品的稍高；常温下3种充氮方式样品的初始残氧值相差不大，但储藏过程中液氮加注和气液氮混合加注样品的残氧值明显低于气氮加注样品；常温下液氮加注样品瓶内存在正压，其满瓶抗压能力优于气氮加注和气液氮混合加注样品。High efficiency liquid nitrogen filling technology was used to fill sunflower seed oil, and it was compared with gas nitrogen filling and gas-liquid nitrogen mixed filling technology. The effects of nitrogen filling method on quality and product packaging of sunflower seed oil were studied. The results showed that the change trends of acid value and peroxide value of the samples with three nitrogen filling methods were basically the same. The acid value and peroxide value of liquid nitrogen filling samples were 0.07 mgKOH/g and 1.45 mmol/kg respectively after five months storage at room temperature. Compared with room temperature storage, the acid value of liquid nitrogen filling samples after five months storge at 45 ℃ had little difference, while the peroxide value decreased slightly to 1.30 mmol/kg. The peroxide values of liquid nitrogen filling samples and gas-liquid nitrogen mixed filling samples during initial and storage were slightly higher than that of gas nitrogen filling samples. There was little difference in initial residual oxygen values of three nitrogen filling samples, but the residual oxygen values of liquid nitrogen filling samples and gas-liquid nitrogen mixed filling samples during storage were significantly lower than that of gas nitrogen filling samples. There was positive pressure in the bottle of liquid nitrogen filling sample, and its full bottle compression capacity was better than that of gas nitrogen and gas-liquid nitrogen mixed filling samples.

Alzheimer’s disease (AD) is a progressive neurodegenerative disease and linked to abnormal deposition of amyloid-β (Aβ), neurofibrillary tangles (NFTs), synaptic dysfunction, and neuroinflammation. Despite significant progress in unravelling the pathogenesis of AD, currently main therapeutic interventions is limited to symptomatic alleviation. Methylprednisolone (MP), a synthetic glucocorticoid, is recognized for its extensive anti-inflammatory properties. Our study assessed the neuroprotective effect of MP (25 mg/kg) administration to an Aβ1-42-induced AD mouse model. Our findings demonstrate that MP treatment can ameliorate cognitive impairment in Aβ1-42-induced AD mice and suppress microglial activation in the cortex and hippocampus. RNA-Sequencing analysis reveals that MP ultimately rescues cognitive dysfunction through improving the synapse function and inhibiting the immune and inflammatory processes. Our study suggests that MP could be a promising drug alternative for the treatment of AD, either alone or in combination with other existing drugs.

AIM: To compare the efficacy between two different silicone hydrogel bandage contact lenses after transepithelial photorefractive keratectomy (T-PRK). METHODS: In this randomized controlled trial, a total of 89 patients (178 eyes) who underwent T-PRK at the Qingdao Eye Hospital from October to December 2019 were selected. One random eye wore a Senofilcon A bandage contact lens after surgery, and the other eye a Balafilcon A bandage contact lens. Pain scores, uncorrected visual acuity (UCVA), spherical equivalent (SE), corneal epithelial healing status, epithelial thickness, bandage lenses deposits, lenses movement, and ocular surface conditions were measured and compared. RESULTS: There were no differences between the two groups in UCVA, SE, corneal epithelial healing status, corneal epithelial thickness, tear river heights and tear film rupture time at each follow-up visit. Abstract Background Faster and motion robust magnetic resonance imaging (MRI) sequences are desirable in fetal brain MRI. T1-weighted images are essential for evaluating fetal brain development. We optimized the radial volumetric interpolated breath-hold examination (VIBE) sequence for qualitative T1-weighted images of the fetal brain with improved image contrast and reduced motion sensitivity. Materials and methods This was an institutional review board-approved prospective study. Thirty-five pregnant subjects underwent fetal brain scan at 3 Tesla MRI. T1-weighted images were acquired using a 3D radial VIBE sequence with flip angles of 6º, 9º, 12º, and 15º. T1-weighted images of Cartesian VIBE sequence were acquired in three of the subjects. Qualitative assessments including image quality and motion artifact severity were evaluated. The image contrast ratio between gray and white matter were measured. Interobserver reliability and intraobserver repeatability were assessed using intraclass correlation coefficient (ICC). Results Interobserver reliability and intraobserver repeatability universally revealed almost perfect agreement (ICC > 0.800). Significant differences in image quality were detected in basal ganglia (P = 0.023), central sulcus (P = 0.028), myelination (P = 0.007) and gray matter (P = 0.023) among radial VIBE with flip angles 6º, 9º, 12º, 15º. Image quality at the 9º flip angle in radial VIBE was generally better than flip angle of 15º. Radial VIBE sequence with 9º flip angle of gray matter was significantly different by gestational age (GA) before and after 28 weeks (P = 0.036). Quantified image contrast was significantly different among different flip angles, consistent with qualitative analysis of image quality. Conclusions Three-dimensional radial VIBE with 9º flip angle provides optimal, stable T1-weighted images of the fetal brain. Fetal brain structure and development can be evaluated using high-quality images obtained using this angle. However, different scanners will achieve different TRs and so the FA should be re-optimized each time a new protocol is employed.

Abstract Background Gestational diabetes mellitus (GDM) is a common disease during pregnancy. The association of vitamin D receptor (VDR) polymorphisms with GDM is still controversial. This study aimed to assess the associations between VDR polymorphisms and GDM risk. Methods We searched Cochrane Library, PubMed, and Embase electronic database for all eligible studies published from Jan 1, 1980 to December 31, 2020 to conduct a Meta-analysis. We analyzed four VDR polymorphisms: BsmI (rs1544410), ApaI (rs7975232), TaqI (rs731236), and FokI (rs2228570). Inclusion Criteria: (1) The data can be evaluated; (2) case–control study; and (3) meeting the Hardy–Weinberg’s law. Exclusion criteria: (1) Insufficient or extractable data; (2) Severe publication bias in the data; and (3) duplicate publications. We eventually included 15 studies in seven articles, including 2207 cases and 2706 controls. Results We eventually included 15 studies in seven articles, including 2207 cases and 2706 controls. The data showed that ApaI (rs7975232) VDR gene polymorphism was related with the risk of GDM for the comparison of CC vs AA and recessive model in overall population and FokI (rs2228570) VDR gene polymorphism was associated with the risk of GDM for recessive model in overall population. BsmI (rs1544410) polymorphism was not related with the risk of GDM in overall population. However, in the analysis of subgroups grouped by race, BsmI (rs1544410) has certain correlations. And, the data suggested the TaqI (rs731236) polymorphism was not associated with GDM. Conclusion Based on the meta-analysis, VDR ApaI (rs7975232) and FokI (rs2228570) polymorphisms increase susceptibility to GDM. In the future, it can be used to diagnose and screen molecular biomarkers for GDM patients.

Gaze input, i.e., information input via eye of users, represents a promising method for contact-free interaction in human-machine systems. In this paper, we present the GazeVending interface (GaVe), which lets users control actions on a display with their eyes. The interface works on a regular webcam, available on most of today's laptops, and only requires a short one-point calibration before use. GaVe is designed in a hierarchical structure, presenting broad item cluster to users first and subsequently guiding them through another selection round, which allows the presentation of a large number of items. Cluster/item selection in GaVe is based on the dwell time, i.e., the time duration that users look at a given Cluster/item. A user study (N=22) was conducted to test optimal dwell time thresholds and comfortable human-to-display distances. Users' perception of the system, as well as error rates and task completion time were registered. We found that all participants were able to quickly understand and know how to interact with the interface, and showed good performance, selecting a target item within a group of 12 items in 6.76 seconds on average. We provide design guidelines for GaVe and discuss the potentials of the system.

Alzheimer’s disease (AD) is a leading neurodegenerative condition causing cognitive and memory decline. With small-molecule drugs targeting Aβ proving ineffective, alternative targets are urgently needed. Neuroinflammation, which is central to AD’s pathology, results in synaptic and neuronal damage, highlighting the importance of addressing inflammation and conserving neuronal integrity. Cannabidiol (CBD), derived from cannabis, is noted for its neuroprotective and anti-inflammatory properties, having shown efficacy in neuropathic pain management for epilepsy. To investigate the therapeutic efficacy of CBD in AD and to elucidate its underlying mechanisms, we aimed to contribute valuable insights for incorporating AD prevention recommendations into future CBD nutritional guidelines. Aβ1–42 was employed for in vivo or in vitro model establishment, CBD treatment was utilized to assess the therapeutic efficacy of CBD, and RNA-seq analysis was conducted to elucidate the underlying therapeutic mechanism. CBD mitigates Aβ-induced cognitive deficits by modulating microglial activity, promoting neurotrophic factor release, and regulating inflammatory genes. The administration of CBD demonstrated a protective effect against Aβ toxicity both in vitro and in vivo, along with an amelioration of cognitive impairment in mice. These findings support the potential inclusion of CBD in future nutritional guidelines for Alzheimer’s disease prevention.

Alzheimer’s disease (AD), an age-related degenerative disorder, is characterized by β-amyloid deposition, abnormal phosphorylation of tau proteins, synaptic dysfunction, neuroinflammation, and oxidative stress. Despite extensive research, there are no medications or therapeutic interventions to completely treat and reverse AD. Herein, we explore the potential of hydrocortisone (HC), a natural and endogenous glucocorticoid known to have potent anti-inflammatory properties, in an Aβ1–42-induced AD mouse model. Our investigation highlights the beneficial effects of HC administration on cognitive impairment, synaptic function enhancement, and neuronal protection in Aβ1–42-induced AD mice. Notably, HC treatment effectively suppresses the hyperactivation of microglia and astrocytes, leading to a reduction in proinflammatory factors and alleviation of neuroinflammation. Furthermore, HC intervention demonstrates the capacity to mitigate the generation of ROS and oxidative stress. These compelling findings underscore the potential therapeutic application of HC in AD and present promising opportunities for its utilization in AD prevention and treatment. The implications drawn from our findings indicate that hydrocortisone holds promise as a viable candidate for adjunctive use with other anti-AD drugs for the clinical management of patients presenting with moderate to severe AD.

Mink has been identified as an animal with susceptibility to SARS-CoV-2 and also as the only animal with evidence to transmit the virus back to humans. Thus, the surveillance of viruses among high-density farmed minks has a significant meaning for the control of zoonotic emerging diseases in humans. Within anal swabs of minks that died of unknown causes in a mink farm, mink calicivirus (MCV) and mammalian orthoreovirus (MRV) were detected and simultaneously observed within MDCK cell culture from the sample of the same lethal mink. The parallel isolation was successfully performed by utilizing cell lines from different host sources with distinct viral sensitivities, i.e. Mv.1.Lu and Vero-E6 and the two viruses were independently separated. The prevalence of the virus among the minks and its genomic characteristics were investigated through deep sequencing technology. Phylogenetic analysis of the viral genome showed a close relationship of the newly isolated MCV-GCCDC8-2020 with MCV strains belonging to the genus Vesivirus, but with unique mutations derived from the major structural protein (VP1). The reovirus MRV-GCCDC9-2020 isolated from the same mink belongs to serotype 3 mammalian orthoreovirus and genome analysis showed a potential reassortment derived from reoviruses in different species. This study provides a beneficial reference on viral co-infection within disease investigation in farmed minks and raises the concern for the virus surveillance among the high-density fed animal farms.

BackgroundDue to the inability to be cultured in vitro, the biological characteristics and pathogenicity of Pneumocystis jirovecii remain unclear. Intestinal microflora disorder is related to the occurrence and development of various pulmonary diseases. This work explores the pathogenesis of pneumocystis pneumonia (PCP) in acquired immune deficiency syndrome (AIDS) patients from a microbiome perspective, to provide better strategies for the diagnosis, treatment, and prevention of PCP.MethodsSubjects were divided into three groups: human immunodeficiency virus (HIV)-infected patients combined with PCP, HIV-infected patients without PCP, and HIV-negative. Stool and bronchoalveolar lavage fluid (BALF) samples were collected, total DNA was extracted, and 16S rRNA high-throughput sequencing was performed using an Illumina MiSeq platform. PICRUSt and BugBase were used to predict microflora functions, and correlation analysis of intestinal and lung bacterial flora was conducted.ResultsCompared with the HIV- group, prevotella and another 21 genera in the intestinal microbiome were statistically different in the HIV+ group; 25 genera including Escherichia-Shigella from HIV+PCP+ group were statistically different from HIV+PCP- group. The abundance of Genera such as Porphyromonas was positively or negatively correlated with CD16/CD56+ (μL). Compared with the HIV- group, identification efficiency based on area under the curve (AUC) >0.7 for the HIV+ group identified seven genera in the gut microbiota, including Enterococcus (total AUC = 0.9519). Compared with the HIV+PCP- group, there were no bacteria with AUC >0.7 in the lung or intestine of the HIV+PCP+ group. The number of shared bacteria between BALF and fecal samples was eight species in the HIV- group, 109 species in PCP- patients, and 228 species in PCP+ patients, according to Venn diagram analysis. Changes in various clinical indicators and blood parameters were also closely related to the increase or decrease in the abundance of intestinal and pulmonary bacteria, respectively.ConclusionsHIV infection and PCP significantly altered the species composition of lung and intestinal microbiomes, HIV infection also significantly affected intestinal microbiome gene functions, and PCP exacerbated the changes. The classification model can be used to distinguish HIV+ from HIV- patients, but the efficiency of bacterial classification was poor between PCP+ and PCP- groups. The microbiomes in the lung and gut were correlated to some extent, providing evidence for the existence of a lung-gut axis, revealing a potential therapeutic target in patients with HIV and PCP.

Pseudostellaria heterophylla is a traditional Chinese herbal medicine, which has been cultivated for hundreds of years. Viral diseases of P. heterophylla occur widely and limit the yield and quality of this medicinal plant. In this study, five leaf samples of P. heterophylla with typical viral symptoms were collected from four main producing regions that are distributed in Fujian, Guizhou, and Anhui Provinces in China and analyzed by next-generation sequencing. Comprehensive bioinformatics analyses revealed that nine viruses in five genera Carlavirus, Potyvirus, Fabavirus, Cucumovirus, and Amalgavirus infected P. heterophylla. Among these viruses, three novel and two known carlaviruses, tentatively designated Pseudostellaria heterophylla carlavirus 1, 2, and 3 (PhCV1, PhCV2, and PhCV3), Jasmine virus C isolate Ph (Ph-JVC) and Stevia carlavirus 1 isolate Ph (Ph-StCV1), respectively, were first identified in P. heterophylla. PhCV1-3 share a similar genomic organization and clear sequence homology with members in the genus Carlavirus and could potentially be classified as new species of this genus. One novel amalgavirus, tentatively designated P. heterophylla amalgavirus 1 (PhAV1), was first identified in P. heterophylla. It had a typical genomic organization of the genus Amalgavirus. In PhAV1, the + 1 programmed ribosomal frameshifting, which is prevalent in most amalgaviruses, was identified and used in the expression of RNA-dependent RNA polymerase (RdRp). Combined with a phylogenetic analysis, PhAV1 could potentially be classified as new species of the genus Amalgavirus. In addition, multiple Broad bean wilt virus 2 (BBWV2) variants, Turnip mosaic virus (TuMV), and Cucumber mosaic virus (CMV), which have been reported in P. heterophylla, were also detected in this study. The distribution of PhCV1-3, Ph-JVC, Ph-StCV1, TuMV, BBWV2, and CMV in four production regions in Fujian, Guizhou, and Anhui Provinces was determined. This study increased our understanding of P. heterophylla virome and provided valuable information for the development of a molecular diagnostic technique and control of viral diseases in P. heterophylla.

Abstract As a key component of an integrated energy system (IES), energy storage can effectively alleviate the problem of the times between energy production and consumption. Exploiting the benefits of energy storage can improve the competitiveness of multi-energy systems. This paper proposes a method for day-ahead operation optimization of a building-level integrated energy system (BIES) considering additional potential benefits of energy storage. Based on the characteristics of peak-shaving and valley-filling of energy storage, and further consideration of the changes in the system’s load and real-time electricity price, a model of additional potential benefits of energy storage is developed. Aiming at the lowest total operating cost, a bi-level optimal operational model for day-ahead operation of BIES is developed. A case analysis of different dispatch strategies verifies that the addition of the proposed battery scheduling strategy improves economic operation. The results demonstrate that the model can exploit energy storage’s potential, further optimize the power output of BIES and reduce the economic cost.

Objective To prepare 3D-printed porous materials of titanium alloy by electron beaming melting (EBM) and to confirm its biosafety to osteoblasts (OB). Methods Three topological structures of titanium alloy porous materials (2.0 mm, 2.25 mm and 2.5 mm Dode-Medium structure) were prepared by 3D printing technology. The extracts of titanium alloy porous materials were collected at multiple time points (24,48,72,96 h) and used for in vitro culture of osteoblasts cell line MC3T3-E1. The relative growth rate (RGR) of osteoblasts was cal-culated by MTT test. The morphological observation of osteoblasts was performed by HE staining. The activity of ALP was determined for analyzing the differentiate capacity of osteoblasts. The expressional level of Col Ⅰ in osteoblasts was assessed by immunofluorescence staining. Results Following incubation with extracts of titanium alloy porous materials, the osteoblasts were found to adhere and proliferate actively. There was no significant difference in the RGR of osteoblasts between extract group and the negative control group of DMEM. However, the RGR of osteoblasts increased significantly in extract groups compared with that in the positive control group of DMSO (P＜0.01). The cytotoxic degree of osteoblasts in extract groups was all less than or equal to 1. There was no significant difference in the level of ALP and Col Ⅰ of osteoblasts between extract groups and the negative control group of DMEM. However, the level of ALP and Col Ⅰ of osteoblasts enhanced significantly in extract groups compared with that in the positive control group of DMSO (P＜0.01). Conclusions The extracts of titanium alloy materials do not affect the proliferation and differentiation of osteoblasts in vitro that confirmed its biosafety.

The rock fragmentation involves the inter-block and the intra-block fracture. A simulation method for rock fragmentation is developed by coupling Voronoi diagram (VD) and discretized virtual internal bond (DVIB). The DVIB is a lattice model that consists of bonds. The VD is used to generate the potential block structure in the DVIB mesh. Each potential block may contain any number of bond cells. To characterize the inter-block fracture, a hyperelastic bond potential is employed for the bond cells that are cut by the VD edges. While to characterize the intra-block fracture, an elastobrittle bond potential is adopted for the bonds in a block. By this method, both the inter-block and intra-block fracture can be well simulated. The simulation results suggest that this method is a simple and efficient approach to rock fragmentation simulation with block smash.

Receptor for activated C kinase 1 (RACK1) has been shown to promote oral squamous cell carcinoma (OSCC) progression, and RACK1 expression levels have been negatively correlated with prognosis in patients with OSCC. Here, we investigated the impact of RACK1 OSCC expression on the recruitment and differentiation of tumor‐associated macrophages. High RACK1 expression in OSCC cells correlated with increased M2 macrophage infiltration in tumor samples from a clinical cohort study. Moreover, the combination of RACK1 expression and the M2/M1 ratio could successfully predict prognosis in OSCC. OSCC cells with high RACK1 expression inhibited the migration of THP‐1 cells, promoted M2‐like macrophage polarization in vitro, and increased the proportion of M2‐like macrophages in a xenograft mouse model. Moreover, both M1‐ and M2‐like macrophage polarization‐associated proteins were induced in macrophages cocultured with RACK1‐silenced cell supernatant. A mechanistic study revealed that the expression and secretion of C‐C motif chemokine 2 (CCL2), C‐C motif chemokine 5 (CCL5), interleukin‐6 (IL‐6), and interleukin‐1 (IL‐1) are closely related to RACK1 expression. In addition, blocking nuclear factor‐kappa B (NF‐κB) could promote M2‐like macrophage polarization. These results indicate that RACK1 and the M2/M1 ratio are predictors of a poor prognosis in OSCC. RACK1 promotes M2‐like polarization by regulating NF‐κB and could be used as a potential therapeutic target for antitumor immunity.
