31. Statistical analysis and physicochemical characteristics of groundwater ‎quality parameters: a case study.

Abstract

The assessment of groundwater quality was carried out by taking the shallow wells samples near the bank of River Indus at the downstream Kotri barrage; the study area covers the arid region of Hyderabad, Sindh. The space occupied by the wells was about 1-2 km used for the quantification of geochemical characteristics of the shallow aquifer that lies on the left side of the river. A total of 13 groundwater samples were collected during pre-monsoon and post-monsoon seasons. All the groundwater samples were examined for the characterisation of physicochemical parameters such as Total Dissolved Solids ranged from 320 mg/l to 4011 mg/l, Total Hardness ranged from 120 mg/l to 1320 mg/l, Chloride ranged from 160 mg/l to 2260 mg/l, Sulphate ranged from 61 mg/l to 777 mg/l, pH ranged from 6.9 to 8.1. The findings of the study demonstrate the presence of high quantity physicochemical parameters at a shallow aquifer in contrast to guidelines values by the World Health Organization (WHO) for drinking water. Besides, the T-value was applied to check the variation in pre- and post-monsoon results between parameters, which found significant for chloride 2.701 with p-value 0.031, for fluoride 3.039 with p-value 0.019 and for iron 2.957 with p-value 0.021. Moreover, Correlation coefficients among different parameters were determined which showed that as the correlations between parameters become weaker it also becomes insignificant. An attempt has been made to develop linear regression equations to predict the concentration of water quality constituents. A significantly positive correlation at 1 and 5% found between many parameters. The total hardness with 0.969, chloride with 0.988, and sulphate with 0.994 values were found significant using the conventional level of significance (0.05). Abstract

To improve future agricultural production, major technological advances are required to increase crop production and yield. Targeting the coding region of genes via the Clustered Regularly Interspaced Short Palindromic Repeats/CRISPR-associated Protein (CRISPR/Cas) system has been well established and has enabled the rapid generation of transgene-free plants, which can lead to crop improvement. The emergence of the CRISPR/Cas system has also enabled scientists to achieve cis-regulatory element (CRE) editing and, consequently, engineering endogenous critical CREs to modulate the expression of target genes. Recent genome-wide association studies have identified the domestication of natural CRE variants to regulate complex agronomic quantitative traits and have allowed for their engineering via the CRISPR/Cas system. Although engineering plant CREs can be advantageous to drive gene expression, there are still many limitations to its practical application. Here, we review the current progress in CRE editing and propose future strategies to effectively target CREs for transcriptional regulation for crop improvement.

Abstract

Recently food shortage has become the major flagging scenario around the globe. To resolve this challenge, there is dire need to significantly increase crop productivity per unit area. In the present study, 24 genotypes of rice were grown in pots to assess their tillering number, number of primary and secondary branches per panicle, number of grains per panicle, number of grains per plant, and grain yield, respectively. In addition, the potential function of miR156 was analyzed, regulating seed sequence in rice. Furthermore, OsSPL14 gene for miR156 was sequenced to identify additional mutations within studied region. The results demonstrated Bas-370 and L-77 showed highest and lowest tillers, respectively. Bas-370, Rachna basmati, Bas-2000, and Kashmir Basmati showed high panicle branches whereas, L-77, L-46, Dilrosh, L-48, and L-20 displayed lowest panicle branches. Bas-370 and four other studied accessions contained C allele whereas, L-77 and 18 other investigated accessions had heterozygous (C and T) alleles in their promoter region. C-T allelic mutation was found in 3rd exon of the OsSPL14 gene. The sequence analysis of 12 accessions revealed a novel mutation (C-T) present ~2bp upstream and substitution of C-A allele. However, no significant correlation for novel mutation was found for tillering and panicle branches in studied rice accessions. Taken together present results suggested novel insight into the binding of miR156 to detected mutation found in 3rd exon of the OsSPL14 gene. Nevertheless, L-77, L-46, Dilrosh, L-48, and L-20 could be used as potential breeding resource for improving panicle architecture contributing yield improvement of rice crop. [ABSTRACT FROM AUTHOR]

Abstract

The present study authenticates the fabrication of nickel oxide porous shaped nanostructure by hydrothermal method. The novel and functionalized nickel oxide nanomaterial were visualized by using scanning electron microscopy (SEM) and X-ray diffraction techniques (XRD). NiO nanomaterial advertised sensitive, selective and attracted morphology for the development of peptone biosensor. Phenylalanine displays a soft template and growth directing agent for the developing of nickel oxide low dimension nanostructures. The nickel oxide nanomaterial together with protease possesses tremendous role towards the oxidation potential phenomena and transfer of anodic electro-catalytic current for the peptone. The generation of low potential electrochemical signals exhibited the determination of peptone by utilizing different electrochemical techniques for the given concentration ranging from 0.1 mM to 2.5 mM with the measured limit of detection about 0.002 mM with a sensitivity of 107200 μA/mMCm2. The well-defined and highly developed sensor system provides the standard platform for the fabrication and functioning of new devices that are helpful for the determination of many biological macromolecules.
